A masking effect of vitamin D on ASD symptom severity: mediation by developmental level and altered predictive features in vitamin D insufficiency.

BACKGROUND: Symptom severity in children with autism spectrum disorder (ASD) correlates with developmental level. Vitamin D is crucial to developmental level. Studies show inconsistent findings on serum 25(OH)D and ASD symptom severity. This study explores their association and the mediating role of developmental level. METHODS: A total of 699 children aged 24-71 months were enrolled. Serum 25(OH)D levels were measured and children were categorized into normal vitamin D (NVD) and vitamin D insufficiency (VDI) groups. Symptom severity and developmental level were assessed using the Childhood A

An Association study of the SOD2 rs4880 (Ala16Val) Variant with Autism Spectrum Disorder.

Autism Spectrum Disorder (ASD) is a complex and highly heterogeneous neurodevelopmental disorder. Among various contributing factors, oxidative stress plays a major role in ASD pathophysiology. The study of antioxidant genes, therefore, may contribute to revealing important aspects of ASD. The Superoxide Dismutase (SOD) gene family (well-known antioxidant) plays a crucial role in protecting cells from oxidative damage. This study aimed to investigate the functional impact of the SOD2 rs4880 (C>T) variant in children with ASD. A total of 595 participants were recruited, including 366 children w

Dual-target deep brain stimulation for severe pediatric autism: Sustained core symptom reduction in a 12-month case study.

OBJECTIVE: Core autism spectrum disorder (ASD) symptoms lack effective interventions. While single-target DBS helps comorbid conditions, it fails to improve social deficits. Here we aimed to explore the efficacy of bilateral dual-target DBS targeting the anterior limb of the internal capsule (ALIC) and nucleus accumbens (NAcc) in a pediatric severe ASD patient. METHODS: A 9-year-old girl diagnosed with ASD, presenting with severe symptoms and intellectual disability, underwent bilateral NAcc/ALIC DBS implantation. Standardized assessments, including Autism Treatment Evaluation Checklist (ATEC

Evaluating the therapeutic efficacy of a modified ketogenic diet in children with autism spectrum disorder: a randomized controlled trial.

BACKGROUND: Autism spectrum disorder (ASD) is a heterogeneous neurodevelopmental disorder marked by persistent impairments in social communication and interaction, together with restricted and repetitive behavior or interests. Recent epidemiological data indicate that the prevalence of ASD has continued to increase worldwide. Current behavioral and pharmacological interventions remain limited in efficacy, particularly for associated comorbidities. In this context, metabolic interventions, especially the ketogenic diet (KD), have attracted growing interest as a potential therapeutic strategy.

Objective and subjective sleep characteristics across severity subgroups in children with autism spectrum disorder.

BACKGROUND: Sleep disturbances are common in children with autism spectrum disorder (ASD), yet the relationship between objective sleep characteristics and ASD severity remains unclear. We examined polysomnography-derived and caregiver-reported sleep characteristics in children with ASD and typically developing children (TDC), with additional analyses across ASD severity subgroups. METHODS: We included 111 children aged 3-13 years: 81 with ASD and 30 TDC. Participants underwent home PSG, and second-night PSG data were used for the primary analyses. Caregiver-reported sleep behaviors were asse

Glyphosate exposure and epigenetic-environmental interactions in ASD: The role of circTiam1.

Glyphosate (Gly), the most widely used agrochemical, is epidemiologically linked to escalating autism spectrum disorder (ASD) prevalence, yet mechanisms remain elusive. A mouse model of gestational and lactational glyphosate-based herbicide (GBH) exposure (0.10-1.00%) was established to assess ASD-like neurodevelopmental effects. Neural and microglial cell lines (mouse BV2, human HMC3) were used to elucidate mechanisms of GBH-induced neurodevelopmental deficits. GBH induced dose-dependent ASD-like behaviours, including stereotyped repetitive actions and social impairments, without anxiety-like

Clinical Improvement and Taxonomic-Functional Gut Microbiome Remodeling After Six Months of Multi-Strain Synbiotic Supplementation in Mexican Children with Autism Spectrum Disorder.

Background/Objectives: Gut dysbiosis in children with autism spectrum disorder (ASD) has been associated with alterations in microbial ecology and metabolic function that may contribute to gastrointestinal dysfunction and the severity of clinical manifestations. Synbiotic and probiotic supplementation has emerged as a promising microbiome-targeted strategy for ASD; however, its effects on gut microbiome composition, functional potential, and clinical outcomes remain incompletely understood. We conducted a longitudinal study of Mexican children diagnosed with ASD to analyze changes in the compo

Investigation of the Relationship Between Prodynorphin Gene Polymorphisms and Sensory Differences in Children With Autism Spectrum Disorder.

BACKGROUND: Autism spectrum disorder (ASD) is a complex neurodevelopmental disorder characterized by deficits in social communication and repetitive behaviours. Sensory processing abnormalities are now recognized as a core feature of ASD and have been extensively studied. While differences in sensory profiles between individuals with and without ASD are well established, the genetic underpinnings of sensory variability within the ASD population remain unclear. The opioid system is believed to play a role in processes such as social behaviours, pain, addiction, reward, mood, cognition and perce

Deficits in emotion knowledge can impact children's social-emotional interactions, which are particularly evident in children with autism spectrum disorders (ASD). However, consistent evidence regarding emotion knowledge and its individual components across ASD subtypes, as defined by ICD-10, remained limited. This study investigates emotion knowledge development - both as a whole and in seven specific components - in children with ASD, categorized by subtype, and compares them with a matched group of children without ASD using nearest neighbor matching based on gender, age, and language backg

Association of prenatal co-exposure to OPEs and PAEs with ASD symptom trajectories in preschool children: the modifying role of vitamin D.

INTRODUCTION: Few studies have examined prenatal co-exposure to organophosphate esters (OPEs) and phthalic acid esters (PAEs) in relation to childhood autism spectrum disorder (ASD) symptoms, and whether vitamin D modifies these associations. METHODS: Included 2400 mother-child dyads from the Ma'anshan Birth Cohort and examined levels of maternal OPEs, PAEs, and vitamin D across the three trimesters. Used the Clancy Autism Behavior Scale to assess preschoolers' ASD symptoms at ages 3, 5, and 6, then used group-based trajectory modeling to fit ASD symptom trajectories. RESULTS: ASD symptom sc

Fetal exposure to persistent organic pollutants and childhood autism risk.

Exposure to persistent organic pollutants (POPs) during fetal development may increase the risk of autism spectrum disorder, but previous studies have relied on maternal pregnancy samples as proxies instead of directly measuring fetal exposures. Here, we used naturally shed deciduous teeth from autism cases and typically developing controls born between 1998 and 2014 from the California, USA, population-based case-control Childhood Autism Risk from Genetics and the Environment study. Using gas chromatography-tandem mass spectrometry, we quantified fetal second and third trimester and postnatal

Association Between First MMR Vaccination Before Age 2 Years and Childhood Autism in a U.S. EHR Cohort of 2.5 Million Children.

BACKGROUND: Concerns about potential associations between measles-mumps-rubella (MMR) vaccination and autism persist despite extensive prior research. Observational studies evaluating vaccine timing are susceptible to bias from healthcare utilization patterns, neurodevelopmental concerns and time-dependent exposure classification. METHODS: We conducted a retrospective cohort study using the Cosmos electronic health record database, including 2,560,035 US children with linked birth-parent data and follow-up through age 8 years. To minimize bias related to exposure timing and healthcare utiliza

Towards credible digital twins for basic and preclinical research.

Atomoxetine (Strattera(R)) is a selective norepinephrine (noradrenaline) reuptake inhibitor that is not classified as a stimulant, and is indicated for use in patients with attention-deficit hyperactivity disorder (ADHD). Atomoxetine is effective and generally well tolerated. It is significantly more effective than placebo and standard current therapy and does not differ significantly from or is noninferior to immediate-release methylphenidate; however, it is significantly less effective than the extended-release methylphenidate formulation OROS(R) methylphenidate (hereafter referred to as osm
