Low bone mineral density in non-ambulatory adolescents relates to lower correction of neuromuscular scoliosis after posterior spinal fusion.

PURPOSE: Non-ambulatory adolescents with neuromuscular scoliosis (NMS) often require surgical intervention. The purpose of this study was to examine whether low bone mineral density (BMD) affects the results of posterior spinal fusion. METHODS: Volumetric BMD (vBMD) was measured in preoperative computed tomography (CT) scans of scoliotic patients with Duchenne Muscular Dystrophy (DMD), Spinal Muscular Atrophy (SMA), Cerebral Palsy (CP) and Rett syndrome (RTT). CT scans of healthy age-matched adolescents were used to define reference vBMD values and to calculate Z-scores. Scoliosis angles were

MeCP2 MBD-ID module: a unified DNA/RNA binding interface disrupted in Rett syndrome.

Rett syndrome neurodevelopmental disorder is caused by mutations in the epigenetic regulator MeCP2. While the MeCP2 methyl-CpG binding domain (MBD) is well-characterized, the function of the adjacent intervening domain (ID) remains largely understudied. The ID has been described as a distinct RNA-binding region, yet evidence also suggests RNA competitively displaces MeCP2 from DNA. Here, we address these conflicting findings by demonstrating the MBD and ID do not function in isolation but as a synergistic functional unit. The ID significantly enhances affinity of the MBD for methylated DNA by

Pubertal development is unaffected in female Mecp2 -heterozygous mice.

Rett syndrome (RTT) affects predominantly females and is characterized by regression after an initial period of typical development. Previously, we showed that while adolescent female Mecp2 -heterozygous mice (Het, mouse model of RTT) perform pup retrieval comparably to wild-type littermate controls (WT), adult Het regress and become inefficient. Here, we analyzed the pubertal trajectory of naïve nulliparous WT and Het to test whether abnormal pubertal development contributes to this regression phenotype. We found no genotypic differences in pubertal onset nor estrous cyclicity during adolesce

Interventions targeting challenges experienced by individuals with Pitt Hopkins syndrome: a scoping review.

BACKGROUND: Pitt-Hopkins syndrome (PTHS) is a rare neurodevelopmental disorder with pronounced impacts on physical health and everyday functioning, including severe intellectual disability, impaired communication, epilepsy, and breathing dysregulation. Despite significant clinical challenges, there is limited evidence to guide interventions for individuals with PTHS. This scoping review aimed to examine the available literature on interventions targeting challenges experienced by individuals with PTHS and to identify gaps in the current knowledge base. RESULTS: Sixteen peer-reviewed publicati

Cerebellar Contributions to Cerebral Connectivity in Severe Mental Illness: A Developmental Cascade and Predictive-Modeling Framework with Autistic Disorder as the Focal Case.

Severe mental illness (SMI) includes neuropsychiatric disorders of adult onset, but also those that begin to show symptoms in adolescence and early childhood. This review treats the psychotic spectrum disorders and autism as neurodevelopmental in origin, each arising from perturbation of early brain development rather than from a process beginning at the age of clinical presentation. Grouping them is a claim about developmental origin and not about shared pathogenesis: they differ in the nature and timing of the perturbation and in the cortical systems being organized when it occurs. The diver

Ganaxolone, an approved therapy for CDKL5-deficiency disorder, is an inhibitor of PTP1B.

CDKL5-deficiency disorder or CDD, which results in intellectual disability, speech, and motor deficits, and seizures that can start as early as 6 weeks after birth, is caused by de novo mutations in the CDKL5 gene. In early 2022, the FDA approved ganaxolone (Commercial name: Ztalmy) for the treatment of seizures in CDD patients 2 years and older. Ganaxolone has been reported to act as a GABAA receptor agonist that helps reduce neuronal excitability; however, based on its chemical structure, we hypothesized that ganaxolone may also act as a PTP1B inhibitor. We observed that ganaxalone was able

Pubertal development and hypothalamic-pituitary-gonadal axis are altered in male mice lacking Mecp2.

Mutations in the MECP2 gene, encoding the epigenetic reader Methyl-CpG binding protein 2, are the main cause of Rett syndrome, a rare neurodevelopmental disorder. Besides severe symptoms such as profound intellectual disability, loss of speech and motor skills, and epilepsy, loss of function of MECP2 has been associated with pubertal dysregulation, but the biological mechanisms leading to this remain unclear. Using a mouse model of Rett, in which males are hemizygous and females heterozygous for Mecp2 loss of function mutation, we assessed the onset and progression of puberty, together with in

When Neurodevelopment Meets Autoimmunity: Pemphigus Foliaceus in Rett Syndrome Expands the Clinical Spectrum-A Case Report.

Rett syndrome (RTT, OMIM 312750) is a complex multisystem neurodevelopmental disorder. Evidence suggests that RTT may have an autoimmune component and inflammatory activation. However, the autoimmune manifestations remain poorly described. Pemphigus foliaceus is a debilitating autoimmune blistering condition caused by IgG autoantibodies that target desmoglein-1 (Dsg1), resulting in widespread skin blistering and lesions. We report a case of pemphigus foliaceus in a 20-year-old female with RTT and discuss its clinical implications. Clinical data obtained from electronic health records were extr

MECP2 Insufficiency Attenuates RUNX2-Dependent Osteoblast Differentiation via miR-126-3p/DKK1-Mediated Canonical Wnt Signaling Inhibition in Rett Syndrome.

Rett syndrome (RTT) is a rare neurodevelopmental disorder caused by loss-of-function mutations in the gene encoding methyl-CpG-binding protein 2 (MECP2) that is located on the X chromosome. Affected individuals also exhibit a variety of non-neurological symptoms such as kyphoscoliosis and osteoporosis. Thus, MECP2 may play a functional role in bone remodeling and osteoblast differentiation. This study aimed to clarify the molecular mechanisms underlying the deregulation of bone remodeling in RTT. Human deciduous tooth-derived mesenchymal stem cells that exhibit osteoblast plasticity were used

Co-Occurring Genetic Mutations in Rett Syndrome and MECP2-Related Disorders-Clinical and Diagnostic Implications from a Case Series.

Background/Objectives: Factors modulating phenotypic variability in Rett syndrome (RTT, OMIM 312750) include X chromosome inactivation (XCI), type of MECP2 variant, and/or disease modifiers. Emerging evidence also points to multi-locus genetic variants. Understanding the phenotypic variability associated with multi-locus genetic diagnoses in individuals with RTT and MECP2-related disorders would be important not only for accurate diagnosis, risk stratification and clinical management but also to explain symptoms that might not be typically associated with RTT. Methods: We present a case series

Reduction in Seizure Generalization Associated With Long-Term Low-Dose Immunomodulatory Therapy Using Prednisolone and Methotrexate in Rett Syndrome: A Case Report.

OBJECTIVE: Rett syndrome is a severe neurodevelopmental disorder most commonly associated with pathogenic variants of the MECP2 gene and frequently accompanied by epilepsy. Seizures occur in a substantial proportion of patients and may be resistant to conventional antiepileptic therapy. Emerging evidence suggests that neuroinflammatory processes may contribute to seizure propagation and represent a potential therapeutic target. CASE REPORT: A girl born at term with initially normal early development is described. At approximately 2 years of age, she developed regression of speech and stereoty

Active learning for the accelerated discovery of complex concentrated NiCoCr alloys in additive manufacturing.

BACKGROUND: According to the World Health Organization, air pollution is closely associated with climate change and, in particular, with global warming. In addition to melting of ice and snow, rising sea level, and flooding of coastal areas, global warming is leading to a tropicalization of temperate marine ecosystems. Moreover, the effects of air pollution on airway and lung diseases are well documented as reported by the World Allergy Organization. METHODS: Scientific literature was searched for studies investigating the effect of the interaction between air pollution and climate change on

Progress and challenges in intrathecal gene therapy for neurological disorders.

Gene therapy for neurological disorders is a rapidly evolving field with many preclinical studies and some successful clinical trials. Selectivity of cell specific targeting, efficiency and safety of administration route, and vector dosing are some of the most challenging aspects that need to be addressed for a successful treatment approach. In clinical practice, intravenous delivery is the most commonly used route of administration of viral vectors but adverse events led to the development of intrathecal delivery as a safer delivery method. Preclinical studies confirm the efficacy of intrathe

Burn injury is often a devastating event with long-term physical and psychosocial effects. Burn scars after deep dermal injury are cosmetically disfiguring and force the scarred person to deal with an alteration in body appearance. In addition, the traumatic nature of the burn accident and the painful treatment may induce psychopathological responses. Depression and post-traumatic stress disorder (PTSD), which are prevalent in 13-23% and 13-45% of cases, respectively, have been the most common areas of research in burn patients. Risk factors related to depression are pre-burn depression and fe

A call for minimum standards in methodological reporting for studies using animal-borne accelerometers.

Fault-tolerant connectivity labelings are schemes that, given an n-vertex graph G = ( V , E ) and a parameter f, produce succinct yet informative labels for the elements of the graph. Given only the labels of two vertices u, v and of the elements in a faulty-set F with | F | ≤ f , one can determine if u, v are connected in G - F , the surviving graph after removing F. For the edge or vertex faults models, i.e., F ⊆ E or F ⊆ V , a sequence of recent work established schemes with poly ( f , log n ) -bit labels for general graphs. This paper considers the color faults model, recently introd

Human Brain Organoid-Enabling Discoveries Reach the Clinic: A Translational Inflection Point.

Human brain organoids have evolved from early neurodevelopmental models into platforms for therapeutic discovery. Here, we highlight two cases in which organoid-derived findings enabled FDA-approved clinical trials. Patient-derived organoids modeling Pitt-Hopkins syndrome revealed human-specific, TCF4-dependent abnormalities and supported the development of a regulated AAV gene therapy. In parallel, Rett syndrome organoids cultured aboard the International Space Station uncovered space-induced neural senescence, characterized by retroelement-associated neuroinflammation, prompting evaluation o

Lysosomal metabolism in pancreatic cancer and its crosstalk with host physiology.

Transcription factors from the family of Signal Transducers and Activators of Transcription (STAT) are activated by numerous cytokines. Two members of this family, STAT5A and STAT5B (collectively called STAT5), have gained prominence in that they are activated by a wide variety of cytokines such as interleukins, erythropoietin, growth hormone, and prolactin. Furthermore, constitutive STAT5 activation is observed in the majority of leukemias and many solid tumors. Inactivation studies in mice as well as human mutations have provided insight into many of STAT5's functions. Disruption of cytokine
