INTRODUCTION: Trofinetide is an approved treatment in the USA and Canada for Rett syndrome in patients aged ≥ 2 years. Trofinetide is currently supplied as a ready-to-use oral solution (TOS). A new trofinetide powder for oral solution (TPOS) offers potential advantages: smaller configuration of unit-dose packs; excludes preservatives, synthetic red dye, and maltitol; can be dissolved in a range of volumes in cold/room temperature water or water-based beverages; does not require refrigeration. To ensure that the formulation change does not impact trofinetide pharmacokinetics, a bioequivalence s

Nonsense mutations, responsible for ~11% of gene lesions causing human monogenic diseases, introduce premature termination codons (PTCs) that lead to truncated proteins and nonsense-mediated mRNA decay (NMD). In the central nervous system (CNS), these mutations drive severe, progressive neurological conditions such as spinal muscular atrophy, Rett syndrome, and Duchenne muscular dystrophy. Readthrough therapies-strategies to override PTCs and restore full-length protein expression-have evolved from early aminoglycosides to modern precision tools including suppressor tRNAs, RNA editing, and CRI

The serotonin receptor 7 (5-HT7R) has been indicated as a key modulator of neuronal structure and function, playing critical roles in synaptic plasticity, dendritic spine formation, and cytoskeletal remodeling. 5-HT7R activation promotes neurite outgrowth, enhances long-term potentiation (LTP), stimulates local protein synthesis at synapses, and regulates mitochondrial functions, and the mTOR pathway. These properties make the 5-HT7R a compelling candidate for therapeutic intervention in neurodevelopmental disorders characterized by synaptic dysfunctions. Angelman syndrome (AS) is a severe neu

Neonatal encephalopathy is a common morbidity witnessed by neonatologists globally. Though hypoxic ischemic encephalopathy (HIE) is the most common cause of neonatal encephalopathy, the absence of a specific history of hypoxia-ischemia necessitates knowledge about other diseases causing encephalopathy among neonates. These include a plethora of central nervous system, neuromuscular, vascular, metabolic, and genetic disorders. We describe a rare case of neonatal encephalopathy caused by a Methyl CpG binding protein 2 (MECP2) gene mutation in a female neonate. MECP2 mutations mostly lead to clas

INTRODUCTION: Nodding Syndrome (NS) is a childhood-onset epileptic encephalopathy of unknown etiology, occurring in clustered outbreaks across East Africa. Despite extensive investigation, its molecular underpinnings remain unresolved.
METHODS: We performed an 18-plex tandem mass tag (TMT)-based quantitative proteomic analysis of immunodepleted cerebrospinal fluid (CSF) from Ugandan NS patients (n = 9) and age-comparable Ugandan Controls (n = 9). Differential protein abundance and pathway-level enrichment analyses were conducted to identify dysregulated molecular networks.
RESULTS: A total o

Rett syndrome (RTT) is a neurodevelopmental disorder caused by mutations in methyl-CpG binding protein 2 (MeCP2). MeCP2 is a non-cell type-specific DNA binding protein, and its mutation influences not only neural cells but also non-neural cells in the brain, including vasculature-associated endothelial cells. Vascular integrity is crucial for maintaining brain homeostasis, and its alteration may be linked to the pathology of neurodegenerative diseases, but a non-neurogenic effect, such as the relationship between vascular alteration and RTT pathogenesis, has not been shown. Here, we developed

This dataset contains mass spectrometry-based proteomic profiles of human brain organoids cultured on Earth for 30 days, then maintained aboard the International Space Station (ISS) for an additional 30 days, with matched ground controls that remained on Earth for the equivalent duration. Brain organoids were derived from induced pluripotent stem cell (iPSC) lines: Q83X, carrying a nonsense mutation in MECP2 from a male patient with Rett syndrome, and WT83, derived from the patient's unaffected familial control. Rett syndrome is a severe X-linked neurodevelopmental disorder caused by loss-of-f

INTRODUCTION: Rett syndrome (RTT) is a severe neurodevelopmental disorder mainly affecting females. The patients could experience many comorbidities, including gastrointestinal, breathing, cardiovascular disorders, and seizures, which are often drug-resistant. Many antiseizure medications (ASMs) can be utilized as monotherapy or in combination. Fenfluramine (FFA), has a unique mechanism of action that targets the serotonergic system and sigma-1 receptors, has shown benefit in other epileptic encephalopathies, but data on RTT are lacking.
METHODS: We retrospectively reviewed the charts of four

BACKGROUND: Autism spectrum disorder (ASD) is a heterogeneous neurodevelopmental condition. Increasing studies examine whether gut microbiota alterations and the gut-brain axis are linked to ASD-relevant phenotypes. As the literature expands rapidly, a quantitative mapping is needed to clarify influential work and evolving themes.
OBJECTIVE: To map global research on ASD and the gut microbiota, identify major contributors and knowledge bases, and characterize thematic evolution and emerging fronts.
METHODS: We analyzed 1,391 English-language articles and reviews indexed in the Web of Science

Rett syndrome neurodevelopmental disorder is caused by mutations in the gene encoding the epigenetic regulator MECP2. While the MECP2 methyl-CpG binding domain (MBD) is well-characterized, the function of the adjacent intervening domain (ID) remains largely understudied. The ID has been described as a distinct RNA-binding region, yet evidence also suggests RNA competitively displaces MECP2 from DNA. Here, we address these conflicting findings by demonstrating the MBD and ID do not function in isolation but as a synergistic functional unit, establishing a new model for MECP2 function. We show t

This year, the selection criteria for highly cited articles in the 'Molecular and Cellular Neuroscience' section of Brain Sciences were focused on publications that achieved a citation count of 10 or more during 2024. Applying this metric, the Editorial Office, in collaboration with myself as Associate Editor of the 'Molecular and Cellular Neuroscience' section of the journal, identified eight articles that not only exemplified the mission of this section but also made significant scientific contributions by advancing our current understanding of the molecular and cellular mechanisms underlyin

Chromatin organization during postnatal development is very important for establishing neuronal function and may be disrupted in neurodevelopmental disorders that are associated with impaired brain function. Both the Methyl CpG-binding protein 2 (MeCP2) and the linker histone H1 are important chromatin regulators. Still, their developmental expression patterns and functional interactions across diverse genetic backgrounds are not well understood. This study examined changes in histone H1, histone H3, and MeCP2 levels in CD1 and C57BL/6 mice in two different strains, in the liver, cerebellum, a

BACKGROUND: Comprehensive profiling of epigenetic states is essential for understanding gene regulation and disease mechanisms. Sequencing-based methods such as ChIP-seq, Hi-C, and RNA-seq provide genome-wide views of histone modifications and 3D genome organization, but lack spatial resolution within single nuclei. RESULTS: Here we present an image-based epigenetic profiling framework that combines high-speed super-resolution microscopy with deep learning. Using models of (i) histone deacetylase inhibition in HEK293T cells and (ii) Rett syndrome iPS cells carrying MECP2 mutations, our approac

Rett syndrome (RTT) is a neurological disorder caused by loss-of-function mutations in methyl-CpG-binding protein 2 (MECP2), which encodes a transcriptional regulator essential for maintenance of normal neuronal function. The current US Food and Drug Administration-approved treatment for RTT, trofinetide, mildly alleviates some symptoms. In contrast, reintroducing MeCP2 or increasing its amount through transgenesis in mouse RTT models improves most neurological phenotypes and enhances survival. Here, we devised a therapeutic strategy to moderately increase MeCP2 protein by modulating the alter

Rett syndrome is a severe, progressive developmental condition of the nervous system. It is linked to the X chromosome and affects mainly girls aged 6 to 18 months. The syndrome is usually caused by pathogenic mutations in the MECP2 gene, which encodes a methyl-CpG-binding protein. The prevalence of the disease is 1:10.000 in the female population. This syndrome is characterized by a progressive course, loss of acquired motor, mental, and speech functions, and multisystem comorbidities. A case of Rett syndrome in a 3-year-old child, confirmed by molecular genetic analysis, is presented. The fe

INTRODUCTION: Rett syndrome is a neurodevelopmental disorder associated with epilepsy that influences motor/communication skills, behavior, and other systems. Clinical experiences for these patients are not well described.
METHODS: This post hoc analysis consists of a patient subgroup with Rett syndrome enrolled in an open-label, single-arm, safety study. Age- and weight-based doses of diazepam nasal spray (5-20 mg) were administered to patients (aged 6-65 years) for seizure clusters. Treatment-emergent adverse events (TEAEs) were recorded, and days between seizure clusters (SEIzure interVAL

BACKGROUND: Rett syndrome (RTT) is an X-linked neurodevelopmental disease with clear diagnostic criteria and mainly affects females. Mutations of the methyl-CpG-binding protein 2 (MECP2) gene cause most RTT cases. While numerous MECP2 mutations have been reported, the pathogenicity of novel identified variants often requires functional validation. To obtain a definite genetic diagnosis result and determine the specific pathogenesis, it is essential to conduct functional validation of the novel mutation.
CASE DESCRIPTION: We reported one clinical case of a female child diagnosed with typical R
