Jean-Noël Seignot (1925-2022): Pioneer in the treatment of Gilles de la Tourette syndrome.

In 1961, Jean-Noël Seignot (1925-2022) published a clinical observation in the Annales Médico-Psychologiques of a patient with a severe form of Gilles de la Tourette syndrome. He included the results of his clinical test with R.1625, a new compound then in an experimental phase, developed by Janssen Laboratories in Belgium-that is, haloperidol. Seignot's article is constantly cited in the bibliographies on the treatment of this syndrome. Here, I present the first-ever complete translation of Seignot's publication, followed by his biography, based on family archives kindly made available to me.

Tourette syndrome.

Tourette syndrome is characterized by the presence of motor and phonic tics. Individuals with Tourette syndrome can have a spectrum of related behaviours including attention deficit hyperactivity features, obsessions, compulsions, impulsions and complex tics. The pathophysiology of Tourette syndrome is thought to reflect genetic influences on the development and function of cortical-striatal-thalamic-cortical and broader cortical-subcortical circuitry implicated in motor and inhibitory control and interoceptive processing, which may contribute to the characteristic 'urge-tic-relief' cycles. Brain-heart coupling in tourette syndrome: a precision functional mapping study of discordant siblings.

Tourette syndrome (TS) is a neurodevelopmental disorder characterized by motor and vocal tics that are modulated by internal physiological state and interoceptive state, including premonitory urges and associated changes in short-timescale parasympathetic variability. Since tic expression is thought to be linked to fluctuations in autonomic state, understanding how brain networks integrate physiological signals may provide insight into the neural mechanisms contributing to tic generation and suppression. In recent years, there has been increasing interest in understanding interactions between

"Just Right" Perceptions and Obsessive-Compulsive Symptoms in Tourette Syndrome and Chronic Tic Disorder: Implications for Tourettic Obsessive-Compulsive Disorder.

OBJECTIVES: Perceptions that something is not "just right" and feelings of incompleteness are forms of premonitory urges that extend beyond simple bodily sensations; they involve subjective interpretations of those sensations and may therefore be regarded as mental phenomena. These phenomena may precede tics but can also trigger compulsive behaviors. Investigating these phenomena is essential for understanding intermediate clinical phenotypes between tic disorders and obsessive-compulsive disorder (OCD). METHODS: We retrospectively reviewed the medical records of 71 patients older than 10 yea

The Association Between Depression and Tourette Syndrome: A Bibliometric Analysis of Research Hotspots and Emerging Trends from 2001 to 2025.

BACKGROUND: Depression is a clinically relevant comorbidity of Tourette syndrome (TS), yet the organization and development of research on this topic remain poorly characterized. This study aimed to map the knowledge structure, research hotspots, and emerging trends in the literature on TS and depression. It also sought to identify questions that warrant further investigation. METHODS: English-language articles and reviews published from 2001 to 2025 were retrieved from the Web of Science Core Collection and Scopus. Bibliometrix, VOSviewer, and CiteSpace were used to analyze publication and c

Obsessive-Compulsive Disorder in Adulthood: Etiology, Diagnosis, and Treatment.

BACKGROUND: Obsessive-compulsive disorder (OCD) is common, with a lifetime prevalence of 1.3-4.1%, and it usually markedly impairs the quality of life of those affected. The rationale behind this article is to present an up-to-date review of the etiology, diagnosis, and treatment recommendations of OCD in adults. METHODS: This narrative review is based on relevant publications, including the revised German clinical practice guideline on OCD. RESULTS: Genetic factors are estimated to account for 50% of the variance in OCD; up to 30 independent associated loci have been identified across the g

Somatosensory Gating of Pneumatic Stimuli in Adults with Chronic Tic Disorder.

Individuals with chronic tic disorder (CTD) frequently report sensory over-responsivity, defined as heightened awareness of and/or behavioral reactivity to stimuli that are non-aversive to most neurotypical individuals. Within CTD populations, sensory over-responsivity is associated with severity of tics and co-occurring psychiatric symptoms. Despite its prevalence and association with core facets of the phenotype, the neurobiological basis of sensory over-responsivity in CTD remains unclear. Sensory gating, the physiological process of filtering out repetitive stimuli in early perceptual stag

The Use of Cannabis-Based Medicine in Selected Neurological Disorders.

Cannabis-based medicine (CBM) is used in a wide variety of different neurological disorders. While the use of CBM in the treatment of pain, AIDS wasting, loss of appetite, and spasticity is well established, CBM application in movement disorders and neurodegenerative disorders is still an emerging topic. The purpose of this chapter is to summarize current evidence behind the use of CBM in selected neurological diseases, mainly movement and neurodegenerative disorders. The best evidence for efficacy of CBM is for Tourette syndrome resulting in an improvement of tics and psychiatric comorbiditie

Distinct cerebellar networks underpin clinical improvement in adolescent Tourette disorder.

Adolescence is frequently called the second period of brain maturation. In Tourette disorder (TD), the clinical trajectory of tics and associated psychiatric co-morbidities varies significantly across individuals during the transition from adolescence to adulthood. In this study, we aimed to identify patterns of resting-state functional connectivity that differentiate adolescents with Tourette disorder from their neurotypical peers, and to monitor symptom-specific functional changes over time. We employed a multivariate analysis based on a support vector machine (SVM) to predict patterns of re

Systematic Literature Review on Public Health Impacts of Persistent Tic Disorders: Education and Employment.

Tourette syndrome and persistent tic disorders (TS/PTD) begin in childhood and can contribute to negative outcomes across the lifespan. A systematic review was conducted to summarize current evidence on education and employment outcomes among individuals with TS/PTD. The review summarized education and/or employment outcomes from 69 articles published between 2003 and March 5, 2025 that reported these outcomes for individuals with TS/PTD and a comparison group without TS/PTD. Of these studies, most included small samples of individuals with TS/PTD (less than 100), and those that reported on ra

Providing tailored support to neurodivergent nursing students during their placements.

The term neurodivergence encompasses conditions such as attention deficit hyperactivity disorder (ADHD), autism, developmental language disorder, dyscalculia, dyspraxia, dyslexia and Tourette syndrome. In recent years, there has been an increase in the reported numbers of neurodivergent nursing students. The Equality Act 2010 promotes inclusivity by placing a duty on organisations, including education providers, to make reasonable adjustments aimed at removing or reducing the disadvantages faced by people with disabilities. However, there are several barriers to making reasonable adjustments f

Autism in children and young people with functional tic-like behaviours: a retrospective cross-sectional study.

BACKGROUND: Functional tic-like behaviours (FTLB) are characterised by multiple, complex, vocal and/or motor tics, typically with adolescent onset. Emerging research highlights neurodevelopmental conditions as vulnerability markers for FTLB development, yet no studies have specifically studied this subgroup. This study (1) investigates the prevalence of autism in young people with FTLB, (2) compares demographic and clinical features between patients with FTLB with and without autism and (3) examines the diagnostic accuracy of the Development and Well-Being Assessment (DAWBA) in identifying aut

Online Remote Behavioural Intervention for Tics (ORBIT-UK): protocol of a single cohort usability study.

INTRODUCTION: Tourette syndrome is a common, disabling childhood-onset condition. Exposure and response prevention (ERP) is an effective treatment for tics, yet access remains limited due to a shortage of trained therapists and uneven geographical distribution of services. The ORBIT trial demonstrated that internet-delivered ERP is both clinically and cost-effective, but was developed on a university research platform, not suitable for widescale roll-out. To enable adoption by the National Health Service (NHS) in England, ORBIT has been redeveloped on an NHS compliant platform. This study will

Prefrontal 5-HT 2A receptors directly contribute to tic ontogeny: translational evidence.

Tourette syndrome (TS) is a neuropsychiatric disorder characterized by motor and vocal tics and frequently accompanied by comorbidities such as obsessive-compulsive disorder (OCD) and pathological aggression. Although approved pharmacotherapies often reduce tic severity, they frequently cause adverse effects and are insufficient for managing comorbid symptoms, underscoring the need for improved treatments. Recent evidence indicates that the serotonin 5-HT 2A receptor (5-HT 2A R) antagonist pimavanserin reduces tic severity and improves quality of life, yet its mechanism of action in TS remains

Psychiatric Comorbidity, Tic Severity and Quality of Life in Adults with Tourette's Syndrome.

BACKGROUND: Psychiatric comorbidities are common in adults with Tourette's Syndrome (TS), and appear to influence both tic severity and quality of life (QOL). OBJECTIVE: To assess the association between psychiatric comorbidities, tic severity and QOL in TS. METHODS: We used data from 227 participants in the Calgary and Paris Adult Tic Disorders Clinical Registry. Our aim was to investigate how comorbid psychiatric symptoms impact tic severity, impairment, and QOL. We measured tic severity and impairment with the Yale Global Tic Severity Scale (YGTSS), and QOL with the Gilles de la Tourette

Remote delivery of a mindfulness-based intervention for adults with tics: Protocol of a randomized controlled trial.

Tourette Syndrome and persistent tic disorders (collectively, TS) are characterized by involuntary motor and/or vocal tics that onset in childhood. Existing evidence-based treatments-including behavior therapy and pharmacotherapy-are often only partially effective, associated with burdensome side effects, and/or inaccessible. The current paper describes a randomized controlled trial designed to compare a novel, remotely delivered mindfulness-based group intervention for tics (MBIT) to psychoeducation with relaxation and supportive therapy (PRST) in 150 adults with TS. All interventions and ass
