Fusobacterium necrophorum Meningitis 27 Pediatric Cases From a French National Observatory.

BACKGROUND: Fusobacterium necrophorum is primarily involved in ear, nose and throat (ENT) infections, and exceptionally causes meningitis. We aimed at describing specific clinical features associated with cases of F. necrophorum meningitis. METHODS: Cases of F. necrophorum meningitis were prospectively collected by a network of 259 pediatric wards covering 60% of French pediatric wards between 2001 and 2024. Children were categorized in 2 groups: "isolated" meningitis and "complicated" meningitis, defined by ≥1 local or systemic complication (mastoiditis, subperiosteal or epidural abscess, Le

Continuous Infusion of Aztreonam-Avibactam After High Loading Dose for an Infection Caused by an OXA-48- and NDM-1-Co-producing ST147 Klebsiella pneumoniae.

INTRODUCTION: Infections caused by metallo-β-lactamase-producing Enterobacterales offer limited therapeutic options. Aztreonam-avibactam (ATM-AVI) provides a promising alternative, but its approved intermittent regimen is complex and can lead to substantial drug waste. METHODS: We describe a case of mastoiditis with a retrotympanic abscess due to OXA-48- and NDM-1-producing Klebsiella pneumoniae, managed with continuous infusion (CI) of ATM-AVI after a full-vial loading dose, supported by therapeutic drug monitoring (TDM) and whole-genome sequencing (EPISEQ CS V2.0, bioMérieux). RESULTS: A 3

Meningitis due to pandrug resistant Chryseobacterium gleum: microbiological cure and clinical response with linezolid-based combination therapy.

We report a rare case of hospital-acquired meningitis caused by pandrug-resistant Chryseobacterium gleum, which presumed susceptibility to linezolid (inhibition zone, 22 mm). Treatment with ciprofloxacin and piperacillin-tazobactam failed, but adding linezolid led to recovery. This case suggests that linezolid may represent a potential adjunctive option for pandrug-resistant C. gleum meningitis.

Recent advancements in pediatric purulent meningitis: diagnosis and treatment.

Purulent meningitis is a common disease of the central nervous system affecting children. It arises from various pathogens transmitted through diverse routes and can lead to significant sequelae, even death. The pathogenesis involves varying degrees of meningeal damage by pathogens, breaching the blood-brain barrier (BBB) and triggering immune responses and other physiological processes. Early diagnosis of purulent meningitis relies on recognizing clinical symptoms and conducting comprehensive examinations in patients. This includes young children presenting with systemic symptoms of infectiou

Immunogenicity and safety of 20-valent pneumococcal conjugate vaccine in pneumococcal vaccine-naive adults ≥18 years of age in India: A phase 3 single-arm trial.

Streptococcus pneumoniae is a leading cause of bacteremia, bacterial meningitis, and pneumonia. In India, there is an unmet need for an expanded valent pneumococcal conjugate vaccine (PCV) to protect older adults. This phase 3, single-arm study evaluated the safety and immunogenicity of 20-valent PCV (PCV20) in adults ≥18 y of age in India. Participants were enrolled into two age groups (18‒49 and ≥50 y old) and received a single dose of PCV20. Immunogenicity endpoints included geometric mean fold rises (GMFRs) in pneumococcal serotype-specific opsonophagocytic activity (OPA) titers from befor

CRISPR interference in a Streptococcus agalactiae multi-locus sequence type 17 strain.

UNLABELLED: Group B Streptococcus (GBS), a common colonizer of the human genital and gastrointestinal tracts, is a leading cause of neonatal bacterial meningitis, which can lead to severe neurological complications. The hypervirulent serotype III, sequence type 17 (ST-17) strain COH1 is strongly associated with late-onset disease due to its unique set of virulence factors. However, genetic manipulation of ST-17 strains remains challenging, limiting the ability to study key pathogenic genes. In this study, we developed a CRISPR interference (CRISPRi) system utilizing an endogenous catalytically

Epidemiology and Outcomes of Culture-proven Neonatal Meningitis in Australian Tertiary Hospitals: A Multicenter Retrospective Cohort Study.

We conducted a 5-year multicenter, retrospective cohort study to describe bacterial meningitis in Australian infants (0-180 days). Of 21 culture-positive infections, Streptococcus agalactiae (29%, 6/21), Staphylococcus aureus (19%, 4/21) and Escherichia coli (19%, 4/21) predominated. Four infants died (19%), including 3 with neurosurgical meningitis. In survivors, neurologic sequelae were common. Novel interventions to reduce the morbidity and mortality burden caused by neonatal meningitis are needed.

Molecular mechanisms of streptococcal disruption of the blood-brain barrier and their pathogenic role in bacterial meningitis.

Bacterial meningitis represents a devastating inflammatory disease of the central nervous system (CNS), characterized by the invasion of pathogens across the blood-brain barrier (BBB) and subsequent dysregulated immune responses. Key inflammatory mechanisms include pathogen recognition by microglial TLRs and NLRP3, neutrophil infiltration, and cytokine storms such as IL-1β and TNF-α, leading to BBB disruption, cerebral edema, and neuronal injury. Despite antimicrobial therapy, excessive inflammation often results in neurological sequelae. Emerging strategies target immunomodulation through inf

Antibiofilm Activity of Phenyllactic Acid Against Cronobacter sakazakii Biofilms: Insights Into Food-Contact Surface and Membrane Integrity.

Cronobacter sakazakii, an opportunistic foodborne pathogen, primarily with the infections that can cause bacteremia, necrotizing enterocolitis, and neonatal meningitis. These pathogens are Gram-negative rods, motile with peritrichous flagella without spore-forming capability, can grow under aerobic and anaerobic conditions, and tolerance to acidic conditions and desiccations stress. Phenyllactic acid (PLA), a phenolic acid with broad-spectrum antibacterial activity, is a promising natural preservative. However, its effects on C. sakazakii biofilms remain unclear. This study evaluated the antib

Deletion of Glaesserella parasuis nanA promotes apoptosis by inhibiting PINK1/Parkin pathway.

Glaesserella parasuis (G. parasuis) is the primary pathogen responsible for Glasser's disease in large-scale pig farms. It causes annual mortality rates of 10-20% in weaned piglets due to meningitis, serositis, and sudden death syndrome. The rapid spread of multidrug-resistant strains has reduced the cure rate of macrolide antibiotics and other drugs, resulting in significant economic losses in the livestock industry. Based on a previously established Tildipirosin-resistant G. parasuis strain (Wang et al., 2025), this study confirmed that nanA is a virulence gene. To investigate its function,

Group A Streptococcus Meningitis, United States, 1997-2022.

Group A Streptococcus (GAS) causes a variety of diseases in humans but is not widely appreciated as a cause of meningitis. During 1997-2022, ten sites participating in the Active Bacterial Core Surveillance network in the United States identified GAS meningitis cases. We calculated annual incidence and case-fatality rates (CFRs) for 320 of those cases and determined antimicrobial resistance by whole-genome sequencing. Annual incidence of GAS meningitis ranged from 0.02 to 0.07 cases/100,000 persons. Children <1 year of age had the highest average annual incidence, 0.23 cases/100,000 children.
