Transferrin Receptor Marks a Foxp3-Low Treg-like Inflammatory T Cell Subset Associated with Disease Severity in HAM/TSP.

Human T-cell leukemia virus type 1 (HTLV-1)-associated myelopathy/tropical spastic paraparesis (HAM/TSP) is a chronic inflammatory disease driven by HTLV-1-infected CD4+ T cells; however, the phenotypic and functional characteristics of disease-associated T-cell subsets remain incompletely understood. We analyzed samples using flow cytometry (n = 3-5 per group) and RNA-seq (n = 13), focusing on CADM1highCD4+ T cells enriched for HTLV-1-infected cells to evaluate a transferrin receptor (TfR)-expressing subset. TfR+CADM1highCD4+ T cells were detected in both asymptomatic carriers and patients wi

Design and in silico evaluation of an mRNA vaccine against HTLV-1 using AI-driven reverse vaccinology approaches.

Human T-lymphotropic virus type 1 (HTLV-1) is the first discovered human oncogenic retrovirus that can cause adult T-cell leukemia/lymphoma, HTLV-1-associated myelopathy/tropical spastic paraparesis, and several other diseases. Due to the poor prognosis of these diseases and the limited therapeutic modalities, the need for an HTLV-1 vaccine is crucial. The current study has used an artificial intelligence-driven reverse vaccinology approach to design an mRNA vaccine against HTLV-1. The two most antigenic proteins of the virus were selected and analyzed using multiple immunoinformatics tools to

Prevalence of human T-lymphotropic virus type 1 (HTLV-1) infection among patients attending a dermatology clinic in Northeastern Brazil.

Human T-cell lymphotropic virus type 1 (HTLV-1) is a retrovirus with a worldwide distribution, infecting approximately 10 to 20 million people. It is associated with serious diseases such as adult T-cell leukemia/lymphoma (ATLL) and HTLV-1-associated myelopathy/tropical spastic paraparesis (HAM/TSP). In addition to neurological and hematological manifestations, HTLV-1 is associated with dermatological manifestations, notably HTLV-1-associated infectious dermatitis (IDH), frequently described in children but also present in adults. Previous studies demonstrate a higher frequency of dermatologic

[HTLV-1-Associated Myelopathy].

Human T-cell leukemia virus type 1 (HTLV-1)-associated myelopathy (HAM) is a neuroimmunological disorder in which the infiltration of HTLV-1-infected T cells into the spinal cord triggers chronic inflammation and neurological damage. HAM predominantly affects the mid-to-lower thoracic cords. Improving long-term outcomes requires accurate assessment of disease activity and timely initiation of therapy. Activity stratification based on cerebrospinal fluid biomarkers is clinically useful.

Use of the 10-meter timed walk for monitoring long-term gait progression in human T-cell leukemia virus type 1-associated myelopathy/tropical spastic paraparesis: a 10-year analysis from the Japanese HAM-net registry.

BACKGROUND: Human T-cell leukemia virus type 1 (HTLV-1)-associated myelopathy/tropical spastic paraparesis (HAM/TSP) is a chronic neuroinflammatory disease that affects approximately 0.3% of individuals with HTLV-1 infection in Japan. Considering the rarity of HAM/TSP, data on the natural progression of walking disability and its correlation with outcome measures, such as the Osame's Motor Disability Score (OMDS), remain limited. This observational study analyzed longitudinal data from the Japanese HAM patient registry "HAM-net" to identify appropriate clinical outcome measures for evaluating

Therapeutic Approaches for HTLV-1-Associated Myelopathy/Tropical Spastic Paraparesis: Current and Emerging Strategies.

Human T-cell leukemia virus-1 (HTLV-1)-associated myelopathy/tropical spastic paraparesis (HAM/TSP) is a chronic inflammatory disease of the spinal cord induced by immunological activation due to high HTLV-1 proviral load in the peripheral blood. HAM/TSP is representative of HTLV-1-related inflammatory diseases, and its main neurological symptoms-namely, motor dysfunction of the lower extremities through spastic paraparesis with urinary disturbance-are progressive and lead to deterioration in the quality of life of patients once these dysfunctions develop. Therefore, novel and safe therapeutic

Whole Blood Volume-Based Absolute Quantification of HTLV-1 Proviral Load: A Comparative Method Evaluation Study.

The proviral load of human T-cell lymphotropic virus type 1 (HTLV-1) is an important biomarker associated with the monitoring and risk stratification of adult T-cell leukemia/lymphoma (ATL) and HTLV-1-associated myelopathy/tropical spastic paraparesis (HAM/TSP). However, the lack of standardized quantification methods limits its broader application. This study evaluated a novel absolute quantification approach based on whole blood volume and compared its performance with established protocols. A total of 66 HTLV-1-infected individuals were analyzed using six qPCR-based methodologies, including

PD-1 and TIM-3 Expression on Peripheral Blood T Cells in HTLV-1-Associated Myelopathy/Tropical Spastic Paraparesis (HAM/TSP) and Their Correlations With Disease Stage and Proviral Load.

HTLV-1-associated myelopathy/tropical spastic paraparesis (HAM/TSP) is a long-term neuroinflammatory condition without effective therapy, partly due to limited understanding of its immunopathogenesis. Although higher HTLV-1 proviral load (PVL) is linked to disease susceptibility, the contribution of T-cell immune checkpoints remains unclear. PD-1 and TIM-3 regulate T-cell function, and evaluating them alongside cytokine balance may provide insight into disease mechanisms. This study included 30 HAM/TSP patients, 30 HTLV-1 asymptomatic carriers, and 30 healthy controls. Distribution of PD-1 and

Asymptomatic is not silent: proposal of HTLV-1-associated multiple inflammatory disorder as an early neuroinflammatory state.

BACKGROUND: Human T-lymphotropic virus type 1 (HTLV-1) infection is classically categorized as either asymptomatic or associated with HTLV-1-associated myelopathy (HAM). However, accumulating clinical evidence suggests that a proportion of individuals labeled as asymptomatic present early inflammatory and neurological manifestations that do not fulfill HAM diagnostic criteria. RESEARCH DESIGN AND METHODS: We conducted an observational study within a large, long-standing Brazilian HTLV-1 cohort. Between January 2015 and December 2025, adults previously classified as asymptomatic were systemati

[Steroid responsive rapidly progressive HTLV-1-associated myelopathy with HTLV-1-associated bronchioloalveolar disease: a case report].

An 84-year-old woman presented with a two months history of progressive bilateral lower limb weakness and bladder bowel disfunction. Neurological examination revealed spastic paraplegia and anti HTLV-1 antibodies were positive in the serum and cerebrospinal fluid. Due to the rapid progression along with the elevation of neopterin and CXCL-10 in the cerebrospinal fluid, the diagnosis of rapidly progressive HTLV-1 associated myelopathy (HAM) was made. Chest X-ray and CT coincidentally revealed diffuse micronodular opacities, nodular in various sizes, and interlobular septal thickening. HTLV-1 as

Vitamins as modulators of Deltaretrovirus infection: implications for HTLV-1 and bovine leukemia virus-associated diseases.

Deltaretroviruses, particularly human T-cell leukemia virus type 1 (HTLV-1) and bovine leukemia virus (BLV), are oncogenic retroviruses responsible for severe lymphoproliferative and inflammatory diseases, including adult T-cell leukemia/lymphoma (ATLL) and HTLV-1-associated myelopathy/tropical spastic paraparesis (HAM/TSP). Despite decades of research, effective curative therapies remain limited, and disease progression is strongly influenced by host immune and metabolic factors. Accumulating evidence indicates that vitamins play critical roles in modulating viral pathogenesis, immune dysregu

Investigating the Mechanisms Underlying Cell Death Induction by a Tributyltin Molecule in HTLV-1-Infected Cells Dependent or Not on IL-2 as a Growth Factor.

Human T-Lymphotropic virus type 1 (HTLV-1) lifelong infects at least 5-10 million people worldwide, a minority of whom develop severe lethal diseases including adult T-cell Leukemia/lymphoma and HTLV-1-associated myelopathy or tropical spastic paraparesis. Currently, no vaccines or curative therapies to fight HTLV-1 infection or diseases are available. Recently we found that a tributyltin molecule, Bu3SnOCOCF3 (TBT), which is more potent than cisplatin in inducing cytotoxic effects towards a panel of cell lines including high-tumorigenic cells, also exerted potent cytotoxic effects even toward

HTLV-1-Derived Exosomes Drive Transcriptional Reprogramming of Monocytes Toward a Mixed M1/M2 Phenotype in HAM/TSP.

Human T-lymphotropic virus type 1 (HTLV-1)-associated myelopathy/tropical spastic paraparesis (HAM/TSP) is a chronic neuroinflammatory disorder often leading to demyelination of the spinal cord. Progression to HAM/TSP is closely associated with the high proviral load and the presence of virally infected CD4+ T cells that release extracellular vesicles (EVs). Exosomes, an EV subtype released by many cell types, transport proteins and nucleic acids that regulate intercellular communication and have been implicated in the progression of cancer and neuroinflammatory diseases.
