Muscone Exerts Rapid Antidepressant Effects Through Modulating Glutamatergic Neural Transmission in the Anterior Cingulate Cortex Against Peripheral Inflammation-induced Depression.

Depression associated with peripheral inflammation is characterized by severe symptoms and high relapse rates, highlighting the urgent need for rapid-acting antidepressants. Here, we demonstrate that muscone, the principal bioactive component of musk, exerts rapid antidepressant effects in murine models of peripheral inflammation-induced depression [lipopolysaccharide (LPS) challenge; dextran sulfate sodium-induced colitis] rather than in chronic restraint stress-induced depression. Acute muscone administration did not possess innate anti-inflammatory activity but selectively suppressed the hy

Rutin ameliorates perioperative neurocognitive disorders by inhibiting microglial HIF-1α/PKM2‑driven glycolysis.

BACKGROUND: Perioperative neurocognitive disorders (PND) are common complications in elderly patients, largely driven by microglia-mediated neuroinflammation. Rutin, a natural flavonoid, exhibits anti-inflammatory and neuroprotective effects, but its role in PND remains unclear. METHODS: An abdominal surgery-induced PND model was established in aged mice. Behavioral performance was assessed using the Morris Water Maze and Fear Conditioning Test. Hippocampal tissues were collected to evaluate microglial activation, synaptic integrity, and neuroinflammatory markers. Glycolytic metabolism was as

Temporal dynamics of the cerebrospinal fluid proteome in tick-borne encephalitis and enteroviral meningitis.

BACKGROUND: Tick-borne encephalitis (TBE) is a severe viral infection of the central nervous system (CNS), causing acute meningoencephalitis and often long-term neurocognitive sequelae. The lack of specific treatment for TBE highlights the need to improve understanding of disease mechanisms and identify novel biomarkers. Enteroviral meningitis is typically a milder infection of the CNS and rarely causes encephalitis. OBJECTIVES: In this hypothesis-generating study, we aimed to characterise temporal changes in the cerebrospinal fluid (CSF) proteome in TBE and identify candidate biomarkers asso

Ganoderma lucidum polysaccharides alleviate cognitive impairment in sleep-deprived mice: Association with gut microbiota remodeling.

Sleep deprivation (SD) can impair cognitive function through multiple processes, including systemic inflammation, oxidative stress, and synaptic dysfunction, yet effective interventions remain limited. Therefore, this study investigated whether Ganoderma lucidum polysaccharides (GLP) could alleviate SD-induced cognitive impairment and whether such effects were accompanied by changes in gut microbiota, fecal metabolism, and indicators related to the microbiota-gut-brain (MGB) axis. In a chronic SD mouse model, GLP administration not only improved memory performance and anxiety-like behaviors bu

Chronic kidney disease as a potential risk factor for pituitary hormone dysregulation: a review of the underlying mechanisms.

Chronic Kidney Disease (CKD) is a condition of abnormal kidney structure or function, pathologically characterized by chronic inflammation, tubular atrophy, vascular rarefaction, glomerulosclerosis, and interstitial fibrosis. CKD potentially represents a risk factor for several disease conditions, such as cardiovascular disease, mineral bone disorder, and electrolyte imbalance. Hormone dysregulation within the pituitary axis is yet another condition that CKD influences. Notably, CKD dysregulates several anterior pituitary hormones, either by directly influencing hypothalamic-pituitary secretio

Neuroimmune crosstalk in HIV-associated neurocognitive disorders and the impact of antiretroviral therapy.

HIV-associated neurocognitive disorders (HAND) remain prevalent among people living with HIV (PLWH) despite effective combination antiretroviral therapy (cART) and durable systemic viral suppression. The central nervous system (CNS) represents a unique immunological and pharmacological compartment in which HIV persists within long-lived reservoirs, particularly microglia and perivascular macrophages, thereby sustaining chronic neuroimmune activation. Current evidence indicates that HAND is driven primarily by indirect mechanisms rather than direct neuronal infection. Persistent viral transcrip

Histone neutralization protects the ischemic brain against the consequences of stroke-associated pneumonia.

Bacterial pneumonia aggravates ischemic stroke via mechanisms that still remain to be determined. In ischemic stroke patients, we show that stroke-associated pneumonia is associated with poor clinical outcome and long-term neutrophil deregulation. In mice exposed to transient middle cerebral artery occlusion (MCAO), experimental S. pneumoniae pneumonia induced at 3 days after stroke impaired neurological recovery and increased blood-brain barrier breakdown, brain neutrophil infiltrates, cerebral microvascular thrombosis, and progressive brain atrophy. The antibiotic amoxicillin only partially

Effects of High-Phenolic Extra Virgin Olive Oil on Sensory Processing in Children With Autism Spectrum Disorder: A Pilot Study.

No approved treatments currently exist for the core symptoms of autism spectrum disorder (ASD), prompting clinicians and families to seek alternative approaches, including dietary and nutritional interventions. The Mediterranean diet (MeDi), characterized by a high intake of antioxidant and anti-inflammatory components, is associated with improved health outcomes and reduced risk of chronic diseases. A key element of the MeDi is extra virgin olive oil (EVOO), which is rich in bioactive phenolics, including oleocanthal, oleuropein and hydroxytyrosol. Through its antioxidant and anti-inflammator

BMAL1 Overexpression in the SCN Confers Protection Against Anxiety-Like Behaviors of Chronic Sleep Restricted-Mice.

Children and adolescents globally experience inadequate sleep, which harms their cognitive functions and emotional regulation. The role of circadian clock genes in influencing the antioxidant and anti-inflammatory capabilities has been well-established. Previous studies reported that the impairment of neural activity within the suprachiasmatic nucleus (SCN) can lead to anxiety-like behaviors in mice. Our study was designed to explore the role of BMAL1 expression in SCN, which could be responsible for anxiety-like behaviors induced by chronic sleep restriction (SR) in young mice. Here, 5-week-o

Chemotherapy-Induced Skeletal Muscle Dysfunction and Cognitive Impairment: A Muscle-Brain Mechanistic Framework With Implications for Exercise Oncology.

Chemotherapy commonly induces skeletal muscle dysfunction and cognitive impairment, complications that reduce treatment tolerance and quality of life. Skeletal muscle mass frequently declines during chemotherapy, with cohort studies reporting losses of 3%-10% across treatment courses, while chemotherapy-induced cognitive impairment affects up to one-third of patients. Emerging evidence indicates that chemotherapeutic agents impair mitochondrial function in peripheral tissues, including skeletal muscle, increasing systemic oxidative stress and inflammatory cytokines. These factors may promote o

Glymphatic dysfunction as a potential mechanistic link to cognitive ımpairment in Behçet's disease: a hypothesis.

Behçet's disease (BD) is a chronic multisystem inflammatory disease in which cognitive impairment is increasingly recognized but remains incompletely understood. Although cognitive dysfunction is more common in patients with Neuro-Behçet's disease (NBD), it has also been reported in patients without clinically apparent neurological involvement, suggesting that mechanisms beyond focal inflammatory lesions may contribute to cognitive impairment. The glymphatic system is a brain-wide perivascular clearance pathway that maintains cerebral homeostasis by facilitating the exchange of cerebrospinal a

Type I Interferon Mitigates ZIKV-Induced Neuroinflammation and Cell Death in the Central Nervous System.

Zika virus (ZIKV) infection can cause severe neurological complications, particularly when antiviral immune responses are impaired. Here, we investigated how type I interferon (IFN-I) signaling influences the cellular inflammatory response and immunopathology in the central nervous system (CNS) during ZIKV infection. ZIKV infection induced early IFN-α and IFN-β expression in the CNS of wild-type (WT) mice. In contrast, mice lacking the IFN-I receptor (IFNAR-/-) exhibited higher CNS viral loads, severe clinical symptoms, weight loss, increased mortality, and histopathological alterations charac

ANCA-associated vasculitis presenting with tongue necrosis and severe gastrointestinal involvement.

OBJECTIVE: Neuroinflammation in the hippocampus has been determined to contribute to postoperative cognitive dysfunction (POCD) occurrence in elderly individuals. Histone deacetylases (HDACs) have been identified as important regulators of inflammation. However, the roles of different types of HDACs in POCD have never been fully explored. MATERIALS AND METHODS: POCD mouse models were established using isoflurane and validated by the Morris water maze test. The mice were pretreated with UF010 [a Class I HDAC inhibitor (HDACi)], MC1568 (a Class II HDACi) and SAHA (a Class I and II HDACi) before

Single-cell and single-nucleus transcriptomics in ischemic stroke: cellular mechanisms and therapeutic implications.

Ischemic stroke remains a major cause of death and long-term disability and is defined by complex, dynamic pathophysiological processes involving diverse cell types and interconnected molecular networks. In recent years, single-cell and single-nucleus RNA sequencing (sc/snRNA-seq) have provided unmatched resolution for dissecting these processes. A growing body of studies has applied these approaches to ischemic stroke, generating extensive insight into cellular heterogeneity, molecular regulation, and disease-associated cell states. This review systematically integrates current evidence from

Emerging preclinical evidence supports a potential role for cannabidiol in the management of sickle cell disease.

Sickle cell disease (SCD) imposes a substantial global health burden, with acute and chronic pain representing a major component of morbidity. Standard pain management, largely opioid-based, carries significant risks and often provides inadequate long-term relief, highlighting an unmet need for alternative analgesics as well as disease modifiers. Medicinal cannabinoids have analgesic and anti-inflammatory properties; most clinical studies so far have used Δ9-tetrahydrocannabinol (THC)-containing products with conflicting outcomes. In contrast, purified cannabidiol (CBD) has a broader spectrum

Mesenchymal stem cell-derived extracellular vesicles ameliorate heat stroke-induced hippocampal injury and modulate VIM-AS1-associated miR-34a-5p/Per2 signaling.

BACKGROUND: Central nervous system (CNS) injury is a major determinant of mortality and persistent disability after heat stroke. Mesenchymal stromal/stem cell-derived extracellular vesicles (MSC-EVs) are candidate cell-free immunomodulatory therapeutics, but their effects and molecular correlates in heat stroke-associated brain injury remain incompletely defined. We investigated whether MSC-EVs attenuate hippocampal inflammation and alter microglial phenotype-associated markers after heat stroke. METHODS: Human umbilical cord MSC-EVs were isolated by differential ultracentrifugation and chara

Correlation between perinatal asphyxia-induced brain injury, number of enlarged perivascular spaces, and sleep quality in later life.

BACKGROUND: Perinatal asphyxia (PA) is increasingly recognized as a significant contributor to neonatal brain injury. Identifying reliable biomarkers for assessing the severity of injury is crucial. Enlarged perivascular spaces (EPVSs), which are considered imaging markers of glymphatic dysfunction are reliable indicators of disease severity and prognosis. However, the application of EPVSs among neonates remains unclear. This study aims to investigate whether numbers of EPVSs can serve as a novel biomarker for the severity of PA-induced brain injury (PABI) and the sleep quality in later life.
