BMAL1 Overexpression in the SCN Confers Protection Against Anxiety-Like Behaviors of Chronic Sleep Restricted-Mice.

Children and adolescents globally experience inadequate sleep, which harms their cognitive functions and emotional regulation. The role of circadian clock genes in influencing the antioxidant and anti-inflammatory capabilities has been well-established. Previous studies reported that the impairment of neural activity within the suprachiasmatic nucleus (SCN) can lead to anxiety-like behaviors in mice. Our study was designed to explore the role of BMAL1 expression in SCN, which could be responsible for anxiety-like behaviors induced by chronic sleep restriction (SR) in young mice. Here, 5-week-o

Chemotherapy-Induced Skeletal Muscle Dysfunction and Cognitive Impairment: A Muscle-Brain Mechanistic Framework With Implications for Exercise Oncology.

Chemotherapy commonly induces skeletal muscle dysfunction and cognitive impairment, complications that reduce treatment tolerance and quality of life. Skeletal muscle mass frequently declines during chemotherapy, with cohort studies reporting losses of 3%-10% across treatment courses, while chemotherapy-induced cognitive impairment affects up to one-third of patients. Emerging evidence indicates that chemotherapeutic agents impair mitochondrial function in peripheral tissues, including skeletal muscle, increasing systemic oxidative stress and inflammatory cytokines. These factors may promote o

Glymphatic dysfunction as a potential mechanistic link to cognitive ımpairment in Behçet's disease: a hypothesis.

Behçet's disease (BD) is a chronic multisystem inflammatory disease in which cognitive impairment is increasingly recognized but remains incompletely understood. Although cognitive dysfunction is more common in patients with Neuro-Behçet's disease (NBD), it has also been reported in patients without clinically apparent neurological involvement, suggesting that mechanisms beyond focal inflammatory lesions may contribute to cognitive impairment. The glymphatic system is a brain-wide perivascular clearance pathway that maintains cerebral homeostasis by facilitating the exchange of cerebrospinal a

Type I Interferon Mitigates ZIKV-Induced Neuroinflammation and Cell Death in the Central Nervous System.

Zika virus (ZIKV) infection can cause severe neurological complications, particularly when antiviral immune responses are impaired. Here, we investigated how type I interferon (IFN-I) signaling influences the cellular inflammatory response and immunopathology in the central nervous system (CNS) during ZIKV infection. ZIKV infection induced early IFN-α and IFN-β expression in the CNS of wild-type (WT) mice. In contrast, mice lacking the IFN-I receptor (IFNAR-/-) exhibited higher CNS viral loads, severe clinical symptoms, weight loss, increased mortality, and histopathological alterations charac

ANCA-associated vasculitis presenting with tongue necrosis and severe gastrointestinal involvement.

OBJECTIVE: Neuroinflammation in the hippocampus has been determined to contribute to postoperative cognitive dysfunction (POCD) occurrence in elderly individuals. Histone deacetylases (HDACs) have been identified as important regulators of inflammation. However, the roles of different types of HDACs in POCD have never been fully explored. MATERIALS AND METHODS: POCD mouse models were established using isoflurane and validated by the Morris water maze test. The mice were pretreated with UF010 [a Class I HDAC inhibitor (HDACi)], MC1568 (a Class II HDACi) and SAHA (a Class I and II HDACi) before

Depression in neurodegenerative disease: neurobiological mechanisms and emerging treatments.

Depression is one of the most common and disabling neuropsychiatric complications of neurodegenerative diseases. In Alzheimer's disease (AD) and Parkinson's disease (PD), depressive syndromes affect more than one-third of patients and are associated with accelerated cognitive decline, reduced quality of life, and increased healthcare utilisation. Despite this burden, current antidepressant treatments show little or no efficacy in these populations, suggesting that depression arising in the context of neurodegeneration may reflect distinct underlying neurobiological mechanisms. Emerging evidenc

Single-cell and single-nucleus transcriptomics in ischemic stroke: cellular mechanisms and therapeutic implications.

Ischemic stroke remains a major cause of death and long-term disability and is defined by complex, dynamic pathophysiological processes involving diverse cell types and interconnected molecular networks. In recent years, single-cell and single-nucleus RNA sequencing (sc/snRNA-seq) have provided unmatched resolution for dissecting these processes. A growing body of studies has applied these approaches to ischemic stroke, generating extensive insight into cellular heterogeneity, molecular regulation, and disease-associated cell states. This review systematically integrates current evidence from

Emerging preclinical evidence supports a potential role for cannabidiol in the management of sickle cell disease.

Sickle cell disease (SCD) imposes a substantial global health burden, with acute and chronic pain representing a major component of morbidity. Standard pain management, largely opioid-based, carries significant risks and often provides inadequate long-term relief, highlighting an unmet need for alternative analgesics as well as disease modifiers. Medicinal cannabinoids have analgesic and anti-inflammatory properties; most clinical studies so far have used Δ9-tetrahydrocannabinol (THC)-containing products with conflicting outcomes. In contrast, purified cannabidiol (CBD) has a broader spectrum

The Potential Functions and Beneficial Effects of Melatonin on Cognitive Impairment, Neuroinflammation, Blood-Brain Barrier Leakage, and Synaptic Dysfunction in the Offspring of Mice Exposed to Gestational Intermittent Hypoxia.

INTRODUCTION: Gestational intermittent hypoxia (GIH), which serves as a model for obstructive sleep apnea (OSA), is associated with adverse maternal and neonatal outcomes, especially cognitive impairments in offspring. Growing evidence supports that the anti-inflammatory actions of melatonin significantly influence the peripartum environment and contribute to the mitigation of neurodegeneration. However, the full impact of GIH on offspring cognition and the molecular mechanisms by which melatonin modulates these effects remain uncertain. Thus, in this study, we explored the neurobiological cha

Understanding neuroinflammation in post-COVID-19 syndrome: biological mechanisms, diagnostic biomarkers, and therapeutic prospects.

Post-COVID-19 syndrome (PCS) is an escalating global health concern, marked by persistent cognitive, neurological, and psychiatric symptoms following acute SARS-CoV-2 infection. Although its underlying mechanisms remain incompletely understood, mounting evidence implicates chronic neuroinflammation as a key driver. Sustained microglial and astrocyte activation, blood-brain barrier disruption, and aberrant cytokine signaling contribute to prolonged immune dysregulation within the central nervous system, promoting long-term brain dysfunction. In this expert review, we synthesize emerging insight

iPLA2β-dependent phospholipid remodeling drives ZIKV-induced neuroinflammation and neurological injury.

Zika virus (ZIKV) infection of the developing brain induces neuroinflammatory responses that can restrict viral replication but may also contribute to neurological injury when dysregulated. Although neuronal phospholipid homeostasis is critical for membrane integrity and synaptic function, how ZIKV infection reprograms lipid metabolism to drive neuropathogenesis remains unclear. Here, we identify calcium-independent phospholipase A2β (iPLA2β) as a key mediator of ZIKV-induced neuronal lipid remodeling. iPLA2β is rapidly upregulated in neurons and in the neonatal brain following infection, and

Mesenchymal stem cell-derived extracellular vesicles ameliorate heat stroke-induced hippocampal injury and modulate VIM-AS1-associated miR-34a-5p/Per2 signaling.

BACKGROUND: Central nervous system (CNS) injury is a major determinant of mortality and persistent disability after heat stroke. Mesenchymal stromal/stem cell-derived extracellular vesicles (MSC-EVs) are candidate cell-free immunomodulatory therapeutics, but their effects and molecular correlates in heat stroke-associated brain injury remain incompletely defined. We investigated whether MSC-EVs attenuate hippocampal inflammation and alter microglial phenotype-associated markers after heat stroke. METHODS: Human umbilical cord MSC-EVs were isolated by differential ultracentrifugation and chara

Correlation between perinatal asphyxia-induced brain injury, number of enlarged perivascular spaces, and sleep quality in later life.

BACKGROUND: Perinatal asphyxia (PA) is increasingly recognized as a significant contributor to neonatal brain injury. Identifying reliable biomarkers for assessing the severity of injury is crucial. Enlarged perivascular spaces (EPVSs), which are considered imaging markers of glymphatic dysfunction are reliable indicators of disease severity and prognosis. However, the application of EPVSs among neonates remains unclear. This study aims to investigate whether numbers of EPVSs can serve as a novel biomarker for the severity of PA-induced brain injury (PABI) and the sleep quality in later life.

Hydrogen alleviates sepsis-associated encephalopathy via ATG9B-dependent mitophagy.

BACKGROUND: Sepsis-associated encephalopathy (SAE) is a severe complication with high mortality and limited therapeutic options. Molecular hydrogen (H₂) has shown neuroprotective potential, but its mechanism remains elusive. METHODS: SAE was induced in C57BL/6J mice by cecal ligation and puncture, with or without 2% H2 inhalation. HT22 hippocampal neurons were challenged with lipopolysaccharide in hydrogen-rich medium. H₂ intervention was administered via inhalation or hydrogen-rich medium. Cognitive function was evaluated by novel object recognition and Y-maze tests. Transcriptome sequencing

MgSO4 alleviates anxiety-like behaviours in mice after traumatic brain injury by inhibiting astrocytic TRPM7 in the amygdala.

BACKGROUND: Reports indicate anxiety as one of the most prevalent mental modifications that occur following traumatic brain injury (TBI). Transient Receptor Potential Melastatin 7 (TRPM7) is extensively distributed throughout the nervous system. It regulates intracellular homeostasis of calcium and magnesium ions, regulates the functions of glial cells, including astrocytes in the central nervous system, influences the inflammatory cascade, and may play a significant role in anxiety pathogenesis. However, the potential of astrocytic TRPM7 in the treatment of TBI-induced anxiety remains largely

Peripheral and central neuroinflammatory mechanisms regulating vagal sensory circuit processing and cough in lung disease.

BACKGROUND: The pulmonary system can be exposed to a diverse range of injurious insults which drive pathological events including inflammation, tissue damage and structural remodelling within the airways and lungs. SUMMARY: Patients with lung inflammation and injury often experience sensory neurological complications, suggesting that the pathological processes in the lung either impact or extend into the nervous system. KEY MESSAGES: Cough is a common presenting neurological symptom in patients with lung disease and there is growing evidence that neuroinflammation, either within the vagal ne

Chronic Spontaneous Urticaria and Psychiatric Symptoms: The Blood-Brain Barrier Hypothesis.

Chronic spontaneous urticaria (CSU) is a mast cell-driven inflammatory disease frequently accompanied by anxiety, depression, sleep disturbance, fatigue, and impaired quality of life. Pruritus, disturbed sleep, psychosocial burden, and treatment-related effects are established contributors to this neuropsychiatric and functional burden. Whether immune-to-brain signalling provides an additional mechanism in a susceptible subgroup remains unknown. This hypothesis-driven narrative review examines the blood-brain barrier (BBB) as one potential interface linking CSU-associated inflammation to centr
