Acute viral infection accelerates neurodegeneration in a mouse model of ALS.

While several viral infections have been associated with amyotrophic lateral sclerosis (ALS), the mechanism(s) through which they promote disease remains elusive. Here we investigate the impact of common, acute viral infections on ALS disease onset and progression in the SOD1G93A mouse model. A single sublethal infection prior to onset of ALS clinical signs is associated with markedly accelerated ALS disease progression characterized by rapid loss of hindlimb function. Prior infection results in gliosis in the lumbar spine and upregulation of transcriptional pathways involved in inflammatory r

Treponema pallidum suppressive microenvironments and central nervous system damage: from mechanisms to tools and prevention.

The global resurgence of syphilis, marked by a dramatic 150% increase in cases over the last decade and the rise of the dominant SS14-Ω strain cluster, represents a critical threat to public health. This review explores the complex molecular mechanisms by which Treponema pallidum functions as a "stealth pathogen," establishing suppressive chemotactic microenvironments that facilitate neuroinvasion and persistent central nervous system (CNS) damage. We delineate a biomechanical and molecular cascade wherein the spirochete utilizes its unique "naked" outer membrane and hypervariable Treponema pa

Escitalopram and N-acetylcysteine alleviate major depressive disorder with cognitive impairment through YTHDC1/ATF4/CHOP-mediated pyroptosis.

BACKGROUND: Cognitive impairment (CI) is an important factor contributing to poor prognosis in major depressive disorder (MDD). Endoplasmic reticulum stress and microglial inflammatory response are closely associated with the occurrence and development of MDD with CI. Escitalopram (ESC) and N-acetylcysteine (NAC) are two drugs used to improve depression. However, the therapeutic efficacy and underlying molecular mechanisms of ESC and NAC in treating MDD with CI are still not fully understood. METHODS: The depression rat was established using the chronic unpredictable mild stress (CUMS) protoc

Stigmasterol ameliorates neurological deficits and neuroinflammation in rats with traumatic brain injury by suppressing STAT3/NF-κB/LCN-2.

PURPOSE: TBI triggers intricate inflammatory processes that lead to secondary damage. Stig, a compound known for its cholesterol-lowering effects, demonstrates significant therapeutic potential, encompassing anti-cancer, antioxidant, and anti-atherosclerotic properties. It possesses an ability to penetrate the blood-brain barrier and accumulate in the brain, where it exerts potent anti-inflammatory, anti-apoptotic, and anti-oxidative stress properties, specifically in conditions impacting the central nervous system. METHODS: This study is a basic research combining in vitro and in vivo experi

A hippocampal-lateral posterior thalamic nucleus neural circuit mediates cognitive impairment and metabolic dysfunction in male mice with type 2 diabetes.

OBJECTIVES: Type 2 diabetes mellitus (T2DM) is a multisystem disorder characterized by metabolic dysregulation and cognitive decline. While hippocampal dysfunction is a recognized feature of cognitive impairment, upstream neural circuits that couple peripheral metabolic signals to central cognition remain poorly defined. This study aimed to determine whether the lateral posterior thalamic nucleus-hippocampal glutamatergic (LP-HPCGlu) circuit mediates the intersection of metabolic and cognitive deficits in T2DM. METHODS: A T2DM mouse model was established using a high-fat diet (HFD) and low-do

Lycium barbarum polysaccharides ameliorate diabetes-associated cognitive impairment through modulation of peripheral glycemic homeostasis and inflammation in mice.

BACKGROUND AND PURPOSE: Diabetes-associated cognitive impairment (DCI) is a devastating complication of type 2 diabetes mellitus (T2DM), driven by abnormal glucose metabolism and chronic systemic inflammation. Lycium barbarum polysaccharide (LBP), a macromolecular bioactive component, has shown neuroprotective potential despite limited blood-brain barrier (BBB) permeability. We hypothesized that LBP ameliorates DCI through peripheral glycemic regulatory and anti-inflammatory mechanisms. METHODS: KKAy mice (a spontaneous polygenic T2DM model) were used. Cognitive function was assessed by the M

Microglial FKBP51 drives immunometabolic dysfunction linking perioperative neurocognitive disorders and Alzheimer's disease.

BACKGROUND: Perioperative neurocognitive disorders (PND) and Alzheimer's disease (AD) are both important aging related cognitive disorders. Accumulating evidence suggests that they share common pathological mechanisms involving Aβ deposition, neuroinflammation, and immunometabolic imbalance. However, the key regulatory molecules linking these two conditions remain unclear. FKBP prolyl isomerase 51 (FKBP51) is an immune regulatory molecule widely involved in inflammation control and metabolic homeostasis maintenance, but its role in PND and AD is still unknown. This study aimed to investigate w

Naringenin ameliorates neuropsychiatric deficits in mice fed a methionine-choline-deficient diet.

Neuropsychiatric dysfunction is increasingly being acknowledged as a disabling complication of non-alcoholic steatohepatitis (NASH), but there are no therapeutic approaches. We investigated in the present study the neuroprotective effectiveness of naringenin, a citrus flavonoid with known anti-inflammatory and neurotrophic effects, in a murine NASH model induced by an 8-week methionine-choline-deficient (MCD) diet. Male C57BL/6 mice (n = 8/group) were treated with naringenin (50 mg/kg/day, i.p.) during the final 4 weeks. In behavioral tests, naringenin counteracted cognitive impairment in nove

Autophagy-related pathway modulation is associated with hippocampal neuroinflammation and depressive-like behaviors in bone cancer pain mice.

Bone cancer pain (BCP) is a common and debilitating complication of primary bone tumors and bone metastases that is often accompanied by emotional disturbances and reduced quality of life. However, the hippocampal mechanisms underlying BCP-associated depressive-like behaviors remain unclear. In this study, a mouse model of BCP was established by intratibial inoculation of Lewis lung carcinoma cells. Behavioral assessments, immunofluorescence staining, ELISA, and Western blotting were performed to investigate changes in hippocampal inflammatory markers and autophagy-related protein expression.

Human brain matters: Navigating the neuropathology of COVID-19.

Infection with severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) has caused millions of deaths worldwide. Although the incidence of severe acute cases has declined, the prevalence of long COVID, also known as post-acute sequelae of COVID-19 (PASC), is rising. The pathological mechanisms underlying severe COVID-19, along with the relationship to neurological disorders and potential risk for neurodegeneration, remain poorly understood. The aim of this narrative review is to summarize neuropathological features described in postmortem human COVID-19 brains (n = 352). Furthermore, analy

Maraviroc attenuates inflammation-exacerbated cognitive and amyloid pathology in an early-stage Alzheimer's disease mouse model.

Alzheimer's disease (AD) is an age-related neurodegenerative disorder characterized by progressive cognitive decline, and increasing evidence indicates that systemic inflammation can accelerate disease progression. Maraviroc, a CCR5 antagonist approved for the treatment of human immunodeficiency virus (HIV) infection, has shown neuroprotective effects in several neurological contexts, but its role in AD-related pathology remains unclear. In this study, cognitive performance was assessed in 5 × FAD mice using the Y-maze, novel object recognition, novel location recognition, and social discrimin

Dexmedetomidine attenuates sepsis-associated inflammation and encephalopathy by modulating the MeCP2/NLGN1 pathway.

PURPOSE: Sepsis-associated encephalopathy (SAE) is a common and serious complication in septic patients that significantly reduces quality of life. Besides its sedative and analgesic effects, dexmedetomidine has been suggested to have potential neuroprotective benefits. This study aims to determine whether dexmedetomidine guards against cognitive dysfunction in a murine model of sepsis and to investigate the possible role of the MeCP2/NLGN1 signaling pathway in this neuroprotection. METHODS: An animal model of SAE was established by cecal ligation and puncture (CLP). Sham-operated mice underw

Oleic acid ameliorates the disrupted blood-brain barrier and neuroinflammation following intracerebral hemorrhage in mice by regulating PPARγ.

BACKGROUND: Oleic acid (OA) is demonstrated to have neuroprotective effects and may improve the therapy of neurological disorders. However, the mechanism of OA impact on blood-brain barrier (BBB) and neuroinflammation following intracerebral hemorrhage (ICH) remains indistinct. Here, we aimed to affirm the neuroprotective influence of OA in ICH and to explore the latent molecular mechanism. METHODS: The male C57BL/6 mice (n = 124) was used for this study. ICH model was induced by intracerebral injection of collagenase. OA (60 mg/kg) was administrated 2 h after ICH. Neurobehavioral tests were

A dermo-hypothalamic axis driven by TNF-α mediates sleep disturbances in psoriasis.

Sleep disturbances are a debilitating feature of psoriasis, but whether they arise from itching or from direct effects of inflammation on the brain remains unclear. Here we show, using clinical data and a mouse model of psoriasis-like inflammation, that affected animals exhibit marked wakefulness and fragmented non-rapid eye movement sleep, even when itching is eliminated. This sleep disruption is linked to overactivity of wake-promoting neurons in a brain region called the anterior hypothalamic area. We find evidence of a local inflammatory response in this region, including activation of mic

The role of inflammation in cerebral small vessel disease and vascular cognitive impairment, and therapeutic implications.

BACKGROUND: Increasing evidence suggests that inflammation occurs in cerebral small vessel disease (SVD). Inflammation has been hypothesized to play a role in disease pathogenesis and also in progression to vascular cognitive impairment and dementia (VCID), suggesting that targeting inflammation may offer a novel treatment option. AIMS: In this review, we aimed to summarize the evidence that both systemic and central nervous system (CNS) inflammation occur in SVD and VCID, critically examine whether associations are causal, and review the evidence that anti-inflammatory interventions might re

Downregulating Cdk5 reverses hippocampal neuron ferroptosis by regulating the AMP-activated protein kinase pathway and "M1" polarized microglia.

Aberrant activation of microglia plays a crucial role in neuronal injury after ischemic stroke. Cyclin-dependent kinase 5 (Cdk5) is a serine/threonine-directed kinase that plays a significant role in neuronal damage. We investigated the role of Cdk5 in microglial activation and neuronal ferroptosis in cellular and animal models of hypoxic-ischemic neuronal injury. Treatment with the Cdk5 inhibitor (S)-roscovitine (Ros) and/or an AMPK pathway activator metformin (Met) were investigated in a middle cerebral artery occlusion/reperfusion (MCAO/R) model in C57BL/6J mice. The results showed that Ros
