INTRODUCTION: Obesity is a condition that affects a large part of the global population, and especially in the Western world, leading to a significant systemic inflammatory response in the body, characterized by modification of the secretory inflammatory cytokines and adipokines. The combination of fat accumulation and inflammation can lead to concomitant conditions, such as Insulin Resistance (IR) and increased production and release of fatty acids, ultimately enhancing the occurrence of conditions like metabolic and cardiovascular disorders, with inflammation and oxidative stress being impli

Myocardial Infarction (MI) is a severe cardiovascular event, causing not only substantial damage to the heart but also potentially exerting a profound impact on brain function through a complex cardiac-brain interaction mechanism. The pathological process of MI encompasses myocardial cell necrosis, inflammatory cell infiltration, and the release of a substantial amount of inflammatory mediators. Through the bloodstream, these myocardial mediators may traverse the Blood-Brain Barrier (BBB), eliciting a neuroinflammatory response that can lead to cognitive dysfunction. This article proposes a cr

BACKGROUND: Neuroinflammation critically contributes to secondary brain injury after intracerebral hemorrhage (ICH). The skull-dura interface allows immune trafficking from skull bone marrow (SBM) to the brain, but its role in ICH remains unclear.
METHODS: ICH was induced in male C57BL/6J mice by collagenase injection. To compare the immune landscape of SBM and femur bone marrow (FBM), single-cell RNA sequencing was performed on sham-operated and ICH mice. Flow cytometry at serial time points, combined with cell-tracking experiments, was used to assess time-dependent neutrophil accumulation i

Inflammatory bowel disease (IBD) is an enduring inflammatory complaint with extraintestinal consequences, including autism spectrum disorder (ASD). This experiment was directed to test the influence of maternal separation (MS) stress on the comorbidity of ASD-like behaviors provoked following experimental colitis in male mice, emphasizing the relevance of hippocampal structure, mammalian target of the rapamycin (mTOR), and neuroinflammation. The 32 male Naval Medical Research Institute (NMRI) mice were randomly assigned into four groups, including the MS mice with or without induction of colit

Deep brain stimulation (DBS) is a well-established therapy for advanced Parkinson’s disease (PD), yet peri-electrode edema remains a poorly characterized postoperative complication. We present a 51-year-old male with a 9-year history of PD who underwent bilateral subthalamic nucleus (STN)-DBS implantation. Postoperative imaging confirmed accurate electrode placement, and the patient was discharged without immediate complications. However, one week post-surgery, he developed right-sided neck stiffness and spontaneous seizures, followed by progressive right limb weakness and gait impairment. Mag

High altitude with insufficient oxygen supply poses a great threat to public health. Short- or long-term hypobaric hypoxia (HY) exposure significantly impairs cognitive function. Adenosine receptors with four subtypes participate in the pathogenesis of multiple neurodegenerative diseases. However, the functions of adenosine receptors in HY-induced cognitive impairment are poorly explored. A mouse model of HY-induced cognitive impairment was established using a hypobaric chamber to simulated hypoxia conditions equal to an altitude of 7000 m for 14 days. The levels of adenosine receptors (ADORA1

Neurovascular coupling (NVC) provides a new mechanistic perspective into understanding numerous brain diseases. Previous studies revealed the effects of depressive symptoms on NVC and neurotransmitters in certain neuropsychiatric disorders. However, whether the effects of depressive symptoms on NVC and neurotransmitters would be exhibited in inflammatory bowel diseases (IBD) is hitherto unknown. Forty IBD patients and forty-five healthy controls (HCs) underwent multiparametric magnetic resonance imaging scan. NVC metrics were investigated by exploring Pearson correlation coefficients and ratio

Zika virus (ZIKV) infection can cause severe neurological complications, yet the role of CD8+ T cells in controlling viral pathogenesis in the brain remains unclear. Using Ifnar1-/- mice, which lack type I interferon signaling, we demonstrate that ZIKV infection triggers significant infiltration of CD8+ T cells into the brain, accompanied by neurological defects. ZIKV-experienced CD8+ T cells exhibit enhanced cytotoxic potential, and adoptive transfer of these cells improves survival. In contrast, blocking their infiltration exacerbates brain inflammatory and injury-associated signatures, high

RATIONALE: Glial fibrillary acidic protein antibody-associated astrocytopathy (GFAP-A) is an autoimmune central nervous system inflammatory disorder diagnosed via cerebrospinal fluid (CSF) glial fibrillary acidic protein-immunoglobulin G detection. Clinically, it typically presents with meningoencephalitis or myelitis, accompanied by periventricular perivascular enhancement on brain magnetic resonance imaging (MRI) (findings that guide most routine diagnoses). Yet isolated spinal cord abnormalities (with negative brain MRI) in GFAP-A remain rarely reported, especially when occurring in patient

Ulcerative colitis (UC) is a chronic inflammatory bowel disease with systemic manifestations, including cognitive impairment linked to gut‒brain axis dysregulation. While probiotic therapies show promise, their mechanisms in mitigating neuropsychiatric comorbidities remain unclear. Here, we investigated the therapeutic potential of Akkermansia muciniphila-derived extracellular vesicles (AmEVs) in a murine model of dextran sulfate sodium (DSS)-induced colitis and associated cognitive deficits. AmEVs administration significantly alleviated colitis severity, as evidenced by improved weight retent

Post-acute sequelae of COVID-19 (PASC) encompasses persistent neurological disease, including olfactory and cognitive dysfunction. The basis for this dysfunction is poorly understood. Here, we report neurological dysfunction for at least 120 d postinfection in mice infected with a virulent nonneurotropic mouse-adapted SARS-CoV-2. Long after recovery from nasal infection, we observed diminished tyrosine hydroxylase expression in olfactory bulb glomeruli and in substantia nigra. Similar changes were observed in brains of COVID-19 deceased patients. Vulnerability of dopaminergic neurons in these

BACKGROUND: Mitochondrial dysfunction was reported as a potential factor that contributes to the development of migraine and peripheral hypersensitivity. The recent accumulation of knowledge on mitochondrial involvement in migraine paved the way for a new non-pharmacological-therapeutic strategy like hyperbaric oxygen therapy (HBOT), which targets mitochondrial dysfunction. Yet, the efficacy of HBOT in modaulting migraine associated pain remains unclear. METHODS: Rats were injected with nitroglycerin (NTG, 10 mg/kg), a human migraine trigger, every other day for 9 days. Mechanical and thermal

Hemolytic uremic syndrome (HUS), a fatal complication of Shiga toxin-producing Escherichia coli (STEC) infection, is classically characterized by acute renal failure, but frequently accompanied by central nervous system (CNS) dysfunction. Because the CNS is normally protected by the blood-brain barrier (BBB), the toxin-mediated BBB injury is considered to be a major cause of neurologic sequelae in STEC infection. Here, we delineate how Shiga toxin type 1a (Stx1a) and Shiga toxin type 2a (Stx2a) compromise BBB-like endothelial barrier integrity with the human brain microvascular endothelial cel

Ischemic stroke is among the important causes of death globally, with many survivors experiencing considerable cognitive deficits. This study explored metformin (Met)'s therapeutic effects in a global cerebral ischemia-reperfusion (I/R) injury model in male rats. One hundred and twelve rats randomly divided into four groups were subjected to a global cerebral I/R or sham surgery followed by 2 weeks of metformin treatment. We induced global cerebral ischemia in rats by employing a four-vessel occlusion (4VO) model, which lasted for 20 min. Before occlusion, just removing the small vessel clips

Parthenolide (PTL) and salicin (SA), the main active components in Feverfew (Tanacetum parthenium) and White Willow (Salix alba), respectively, have been traditionally used as a remedy for various types of pain, including headaches. Because PTL and SA have different mechanisms of action, we hypothesize that a combination of these drugs would result in an additive effect. We investigated the effects of local and/or systemic administration of PTL, SA, or their combination on cephalic mechanical hypersensitivity (MH) in acute and chronic model of migraine induced by dural application of inflammat

Numerous studies report cognitive impairment in COVID-19 patients from the acute to post-acute phases, linked to blood inflammation affecting blood-brain barrier (BBB) permeability and causing leakage of glial and neuronal proteins. However, a clear classification of these cognitive deficits and molecular blood events over time is still lacking. This narrative review summarizes the neuropsychological consequences of COVID-19 and evidence of altered cytokines and BBB disruption as potential mediators of cognitive impairment across post-infection phases. Post-COVID-19 cognitive dysfunction appea

Traumatic brain injury (TBI) induces profound neuroinflammation, leading to secondary brain damage and neurological dysfunction. Emerging evidence highlights the critical role of neutrophil extracellular traps (NETs) in amplifying inflammatory responses after injury. This study investigates the involvement of the NLRP3 inflammasome and gasdermin D (GSDMD) in regulating NET formation and subsequent microglia-mediated neuroinflammation after TBI. Using a male mouse model of TBI, we demonstrate that activation of the NLRP3/GSDMD axis significantly enhances NET release from neutrophils. These NETs

Zika virus (ZIKV) is an arbovirus associated with neurological complications such as neonatal microcephaly and fetal anomalies, collectively referred to as congenital Zika syndrome. In the context of co-circulation with other arboviruses, it remains unclear whether pre-infection with a virus could be a risk factor for increased severity of ZIKV-induced disease. Here, we aimed to investigate whether there is differential gene expression in offspring of ZIKV-infected mothers pretreated with panflavivirus (4G2) at subneutralizing concentrations. Twelve weeks after birth, the offspring of these mo

Chorioamnionitis or infection of the amniotic fluid and membranes surrounding the fetus can cause fetal systemic inflammation and adverse neurological outcomes. To investigate the pathogenesis of chorioamnionitis-induced neurological injury, we developed a non-human primate model by infusion of live Escherichia coli. A micro-osmotic pump was placed in pregnant Rhesus macaques at ~ 85% term gestation via hysterotomy. Live E. coli (105 CFU/h, 1µL/h, n = 7) or sterile broth (Controls, 1µL/h, n = 7) infusions were given over 72 h followed by necropsy. A third group of pregnant macaques received mi

BACKGROUND: Parkinson's disease (PD) is a neurodegenerative disorder that affects millions of people worldwide. PD is widely known for its motor symptoms and non-motor symptoms. The long-term clinical course of PD is complicated by the onset of dementia in 80 % of patients. The diagnosis of PD is based on the patient's medical history and clinical examination, but molecular imaging can play an important role in the study of disease progression and mechanisms. [18F] -fluorodeoxyglucose (FDG) -PET, as one of the important molecular imaging methods, was adopted by our research to compare the diff
