Patients with temporal lobe epilepsy (TLE) frequently experience worsening epilepsy following COVID-19, referred to as post-COVID-19 active TLE. While neuroinflammatory changes are suspected in these patients, measurements of both central and systemic inflammation in the brain remain unexplored. We investigate whether the translocator protein standardized uptake value ratio (TSPO SUVr), a quantifiable marker of neuroinflammation using positron emission tomography (PET), is elevated in the brains of patients with post-COVID-19 active TLE. In addition, we examine correlations between TSPO SUVr a

Depression is thought to emerge as a result of monoamine neuromodulators' deficiency in a specific central nervous system site. Tauroursodeoxycholic acid (TUDCA) has been found to have a protective role against diseases affecting the central nervous system. The potential effects of TUDCA on brain monoamine neurotransmitters in a stress-induced depression model have not been reported. We investigated the effects of TUCDA treatment on serotonin, norepinephrine and dopamine, catecholamine biosynthesis enzyme tyrosine hydroxylase (TH) and degrading enzyme monoamine oxidase A (MAO-A), NLRP3 and pro

BACKGROUND: Excessive stress leads to injury and dysfunction, but the underlying mechanism remains unclear. As a human longevity gene, forkhead box O3a (FoxO3a) is a transcription factor that regulates various cellular processes, including the response to oxidative stress, apoptosis, and autophagy. This study aims to explore whether FoxO3a in the dentate gyrus (DG) of the hippocampus is involved in the formation of anxiety- and depressive-like behavior and cognitive impairment in stressed rats and to investigate the detailed mechanism.
METHODS: This study was conducted using the 6-week chroni

Molecular therapies for colitis-associated mental disorders show limited efficacy because they usually focus on a single pathway and exhibit substantial off-target toxicity toward healthy tissues. To tackle this limitation, bioinformatic approaches are employed to predict that inflammation and metabolism may be potential targets for Fucoidan. Guided by this prediction, we develop oral polysaccharide engineered nanozymes, Fucoidan-cerium nanocomplexes (FucCeNCs), which are capable of targeting the inflamed colon through electrostatic interactions, exerting anti-inflammatory effects, and concurr

Bone cancer pain (BCP) is a debilitating condition driven by spinal neuroinflammation and glial activation. Esketamine (ESK), an NMDA receptor antagonist, exhibits analgesic effects beyond receptor blockade, but its mechanisms in BCP remain unclear. Here, a rat BCP model was established by intramedullary injection of Walker 256 cells, followed by intrathecal ESK administration (20, 40, 60 µg). Pain behavior was assessed using Von Frey, Hargreaves, CatWalk, and open field tests. While histology confirmed tumor-induced osteolysis, ESK did not prevent bone destruction. Instead, ESK significantly

BACKGROUND: Postoperative cognitive dysfunction (POCD) is a common and serious complication in older adult patients. While the tyrosine kinase ABL1 has been implicated in neurodegenerative diseases, its specific role in POCD remains unexplored. This study aims to investigate whether ABL1 influences POCD in aged mice by regulating microglial autophagy and neuroinflammation via the mTOR/ULK1 pathway.
METHODS: An aged mouse model of POCD was established, and ABL1 silencing and 3-Methyladenine (3-MA) were used to intervene in mice. The Novel Object Recognition Test (NORT) assessment and water maz

OBJECTIVE: This article discusses the pathophysiology, diagnosis, and management of neurologic complications of hepatic and gastrointestinal diseases.
LATEST DEVELOPMENTS: Hepatic and gastrointestinal diseases such as hepatitis C, Wilson disease, inflammatory bowel disease, and celiac disease can initially present with neurologic symptoms, and neurologic syndromes may be the only manifestations of these conditions. Renal replacement therapy to decrease ammonia levels reduces mortality in patients with acute hepatic encephalopathy. Patients with chronic liver disease should be screened for cov

High-calorie diets cause metabolic syndrome, obesity, and emotional disturbances, with neurological consequences. These prevalent conditions impair both peripheral and central nervous system function, elevating depression risk. These complications represent prevalent chronic conditions in modern society. The bioactive compound 6-gingerol demonstrates antioxidant and anti-inflammatory properties. This study investigated 6-gingerol’s protective effects against depression-like behavior and metabolic syndrome induced by a high-fat, high-sucrose diet (HFHS) in rats. Male Sprague-Dawley rats were ra

There is a connection between neuroinflammation and panic attacks (PA), as microglia-driven pro-inflammatory responses help detect homeostatic disturbances like CO₂ inhalation. This model has become widely used in research since CO₂ exposure can trigger PA in humans and panic-related behavior in mice. Minocycline inhibits microglia activation, serving as a promising tool to attenuate CO2-induced PA. The locus coeruleus (LC) is a CO₂/pH-sensitive region, and disruptions in its activity are linked to psychiatric conditions such as panic disorder (PD). We investigated the involvement of microglia

Speech dysfluency occurs across multiple motor disorders to varying degrees, yet the underlying mechanisms remain incompletely understood. Like locomotion, speech production is relies on basal ganglia circuits, and disruptions in these pathways can lead to varied symptomatic presentations. In this perspective, we discuss evidence from cellular, circuit and network-level studies to highlight shared and disorder-specific mechanisms of speech dysfluency in Parkinson's disease and developmental stuttering. Basal ganglia function and dopaminergic modulation are central to both disorders, while cort

Burn injuries are severe traumas characterized by tissue damage and inflammation, and pain is the common symptom of burn patients. Transforming growth factor beta 1 (TGF-β1) is a multifunctional cytokine involved in organ development, immune response, tumor biology and injury repair. This study investigates the effects of TGF-β1 on the burn injury induced pain and explore the underlying mechanisms. A mouse model of second degree burn injury was established, and spinal intrathecal injection with lentivirus was used to knockdown or overexpress TGF-β1. The assessment of mechanical allodynia and t

BACKGROUND: Neuroinflammation and programmed cell death are two key pathogenic processes that contribute to poor patient outcomes in early brain injury (EBI) following subarachnoid hemorrhage (SAH). Neuronal pyroptosis in SAH has been explored in connection to berberine (BBR), a naturally occurring isoquinoline alkaloid having neuroprotective properties. However, its precise role and molecular mechanisms in this case remain unknown.
OBJECTIVE: To investigate the protective effects of BBR on EBI following SAH and elucidate its potential molecular mechanisms.

Langerhans cell histiocytosis (LCH) is a rare inflammatory myeloid neoplasm with heterogeneous organ and system involvement. Neurodegenerative LCH (ND-LDH) is an uncommon late manifestation with progressive clinical and radiological features, substantial morbidity and an unpredictable course. Historically it was considered a paraneoplastic sequela for which treatment was largely ineffective. Multisystemic disease, central diabetes and orbital and/or skull base bone lesions are risk factors associated with the development of ND-LCH. Early diagnosis, guided by clinical, radiological, electrophys

Sleep deprivation (SD) disrupts systemic homeostasis, but how it drives ocular surface disease remains unclear. Using a male mouse SD model, we show that chronic SD activates the hypothalamic-pituitary-adrenal (HPA) axis, elevates corticosterone, alters gut microbiota, and depletes short-chain fatty acids (SCFAs). These alterations coincide with lacrimal gland atrophy, reduced tear secretion, and increased CD4⁺/CD8⁺ T cell infiltration, accompanied by activation of IL-17-associated inflammatory pathways. Pharmacological inhibition of glucocorticoid synthesis with metyrapone preserves lacrimal

BACKGROUND: Post-traumatic epilepsy (PTE) is a common complication of traumatic brain injury (TBI). Studies have indicated that functional abnormalities of the sodium-activated potassium (KNa) channels are closely related to infantile "refractory epilepsy". However, whether and how the KNa channels are involved in the occurrence of PTE remains unknown.
METHODS: We used adult male C57BL/6 J mice to establish a controlled cortical impact (CCI) mice model with different severity levels. We implanted intracranial electrodes 7 days after injury to observe the spontaneous seizures in the moderately

BACKGROUND: Chronic kidney disease (CKD) is increasingly recognized as a systemic disorder characterized by profound metabolic, inflammatory, and endocrine derangements. Among the endocrine abnormalities observed in CKD, hyperprolactinemia and hypothalamic-pituitary-gonadal axis dysfunction are highly prevalent yet frequently underrecognized. Beyond their reproductive implications, both prolactin excess and sex hormone disturbances appear to contribute to the exceptionally high cardiovascular burden observed in patients with CKD and dialysis-dependent kidney failure.

BACKGROUND: Disorders of gut-brain interactions (DGBI) are increasingly prevalent in children and adults and can significantly impact quality of life starting in childhood and extending into adulthood. DGBI encompass a range of gastrointestinal symptoms in the absence of identifiable structural etiologies with biopsychosocial implications. Ongoing research efforts aim to understand the etiology and pathophysiology of DGBI, with disruptions of the gut-brain axis leading to visceral hypersensitivity and hypervigilance believed to be involved in symptom manifestation. Currently, there are 17 pedi

Chronic sleep deprivation (CSD) is increasingly recognized as a contributor to gut dysbiosis, systemic inflammation, and neurobehavioral impairments via the gut-brain axis. γ-Aminobutyric acid (GABA)-producing postbiotics, derived from microbial fermentation, offer potential in mitigating such dysfunctions. This study investigates the effects of GABA-producing postbiotics produced by Levilactobacillus brevis on CSD-induced gut and brain disturbances in mice. Male C57BL/6 mice were subjected to 30 days of sleep fragmentation and treated with low (250 mg/kg) or high (500 mg/kg) doses of postbiot
