Isoliensinine ameliorates postoperative neurocognitive disorder in aged mice by restoring hippocampal IGF-1 receptor signaling and suppressing oxidative stress and neuroinflammation.

Postoperative neurocognitive disorder (PND) is a significant complication in elderly surgical patients, primarily driven by hippocampal neuroinflammation and oxidative stress. This study investigated the role of Insulin-like Growth Factor-1 Receptor (IGF1R) signaling in the pathogenesis of PND and evaluated the therapeutic potential of isoliensinine (ISL). Using a tibial surgery model in aged mice, we assessed cognitive function, hippocampal IGF-1/IGF1R pathway activity, and the effects of daily intraperitoneal ISL administration (5 or 10 mg/kg). Surgery induced significant cognitive deficits

Third ventricle volume and psychometric alterations in patients with alcohol usage.

BACKGROUND AND OBJECTIVES: Alcohol use disorder (AUD) is one of the deadliest psychiatric conditions. Previous studies indicate that AUD is linked to several neurological biomarkers in the human brain; however, how alcohol use manifests in brain volumes and the relationships between the subcortical brain regions affected by AUD and clinical outcomes are not fully understood. This study attempts to compare subcortical brain volume differences between patients with AUD and healthy controls (HC) to understand how alcohol use relates to brain volume changes. METHODS: Patients with AUD, as determi

Chronic social defeat stress accelerates lung cancer progression and induces hippocampal metabolic dysregulation in a duration-optimized comorbid model.

Chronic social defeat stress (CSDS) is a commonly used mouse model of depression, yet lung cancer - CSDS combined model remains underexplored. Research on the duration of CSDS to induce persistent depressive phenotypes is also relatively limited and calls for further study. This study aims to elucidate the relationship between stress duration and depressive severity, and to clarify the neurobiological links between tumor burden and depression-like behaviors. A combined lung cancer - CSDS mouse model was established by integrating stress exposure and Lewis lung carcinoma implantation. Depressiv

Vogt-Koyanagi-Harada Disease in Paediatric Population: Case Series and Literature Review.

Vogt-Koyanagi-Harada (VKH) is a multisystem autoimmune disorder affecting the eyes, central nervous system, ears, and skin. VKH accounts for approximately 0.4% to 1.7% of paediatric uveitis cases. This case series aims to describe seven paediatric VKH cases with markedly different courses, with the oldest presenting at 17 years old and the youngest at 3 years old, all followed at a tertiary eye care centre, and to review current paediatric VKH literature. Consistent with previous reports, this series shows a female predominance. Two cases involved systemic symptoms; one patient experienced a h

The novel IDO-1 inhibitor 3-047 combined with icaritin ameliorates neuroinflammation and diabetes-associated cognitive dysfunction with suppression of TLR4/MyD88/NF-κB signaling.

BACKGROUND: Diabetes-associated cognitive dysfunction (DACD) is one of the common chronic complications of diabetes mellitus in the central nervous system. Given that current therapeutic agents are limited, exploring novel therapeutic agents is particularly important. The novel IDO-1 inhibitor 3-047 and icaritin (ICT) demonstrate efficacy in ameliorating neuroinflammation and cognitive dysfunction, and their combination exhibits certain hypoglycemic effects. However, the impact of this combination therapy on DACD remains unclear. METHODS: The db/m mice were used as the control group, and the

Therapeutic Potential of Saikosaponins in MASLD and Cross-Organ Protection: A Research Update.

Metabolic dysfunction-associated steatotic liver disease (MASLD), formerly known as non-alcoholic fatty liver disease (NAFLD), has emerged as the most prevalent chronic liver disorder in China, posing a significant threat to public health. While current Western pharmacological therapies demonstrate efficacy, they don't comprehensively ameliorate pathological components of MASLD and are often associated with substantial adverse effects. Saikosaponins (SSs), as the main active components of Bupleurum chinense, a traditional Chinese medicine (TCM), have been demonstrated to exert potential therap

A Protective Role of Hydrogen Sulfide Donor GYY4137 Through Activated Microglia-induced Anti-inflammatory Cytokines in the Spinal Cord of Diabetic Rats.

The increasing significance of inflammation in the pathophysiology of diabetes and its complications, such as diabetic peripheral neuropathy, has generated significant interest in targeting inflammation for disease prevention and control. Hydrogen sulfide (H₂S) has emerged as a promising therapeutic candidate due to its regulatory role in neuroinflammation. The present study investigated the anti-inflammatory effects of prolonged treatment with H₂S donor GYY4137 on microglial and astrocyte activation, along with the subsequent release of anti-inflammatory cytokines, including interleukin (IL)-

Oral Pain and Alzheimer's Disease: Prospective Cohort and Cross-Sectional Analyses.

Given the limited effectiveness of Alzheimer's disease (AD) treatments, modifiable risk factors warrant attention. Oral pain (OP), a common inflammatory symptom, may be linked to neurodegeneration. This study aimed to investigate associations between OP and AD risk, cognitive performance, and brain structure. Data from 490,035 UK Biobank participants free of AD at baseline and with complete OP records were analyzed, while other dementias were excluded. Participants were categorized into an OP group (n = 33,697; defined as self-reported toothache or painful gums via touchscreen questionnaire) a

A sex-biased response of the adult brain to Zika virus infection: from down-regulation of neuronal IRF3 to enhanced pathology in adult female mice.

BACKGROUND: Zika virus (ZIKV) is an emerging, neurotropic flavivirus susceptible to induce brain damage and neurological disorders in adults. In human and mice adult brain tissue, ZIKV predominantly infects neurons. The host type I interferon (IFN-I) response that efficiently restricts ZIKV replication in peripheral cells is impaired in neurons through mechanisms that remain unknown. Since epidemiological studies reveal a greater susceptibility of adult females to ZIKV, investigating the influence of biological sex on the brain’s response to infection may provide new insights into this impairm

Persistent Abdominal Pain in Patients with Inactive Inflammatory Bowel Disease: A Clinical Conundrum.

BACKGROUND: Gastrointestinal tract inflammation may cause deep alterations of the gut homeostasis, even after the acute phase is resolved. Indeed, there is at present substantial evidence that a considerable percentage of patients with inactive inflammatory bowel disease may experience persistent abdominal symptoms (often resembling those of irritable bowel syndrome) long after the intestinal inflammatory flares had successfully treated and resolved. METHODS: The pertinent literature on this topic was reviewed and critically examined. RESULTS: Persistent symptoms can be disabling and signifi

Activation of M1mAChR's improves spatial learning and memory deficits in rats exposed to chronic intermittent hypoxia.

Obstructive sleep apnea (OSA) is characterized by chronic intermittent hypoxia (CIH), which drives neurodegeneration through oxidative and inflammatory stress and heightened synaptic vulnerability. However, the molecular circuitry linking CIH to hippocampal dysfunction remains incompletely defined. In this study, Sprague–Dawley rats were exposed to CIH for 28 consecutive days, achieved by repeated hypoxia–reoxygenation cycles in a normobaric chamber. Hematoxylin–eosin (HE) staining was used to assess hippocampal histopathology. Hippocampal protein abundance of total STAT3 (t-STAT3), phosphoryl

Chronic Stress Leads to Time-Dependent Bone Loss Through HPA Axis Dysregulation and GR Nuclear Translocation Disorder.

Chronic stress and sustained hypothalamic-pituitary-adrenal (HPA) axis activation are major contributors to metabolic bone diseases, including osteoporosis. However, the precise molecular mechanisms by which chronic stress-induced HPA axis dysregulation drives bone deterioration remain unclear. A Chronic Unpredictable Mild Stress (CUMS) model was established in male rats to simulate prolonged stress exposure. Animals were randomly allocated into three groups: control, 10-week CUMS, and 20-week CUMS (n = 10/group). Model validity was confirmed via behavioral assessments. Bone mineral density (B

Neutrophil extracellular traps (NETs) in cerebrovascular disease mechanisms, detection, and therapeutic targeting.

Neutrophil extracellular traps (NETs) play a vital role in the pathophysiology of cerebrovascular diseases. During cerebral ischemia-reperfusion injury, NETs enhance the stability of immunothrombi by forming DNA-histone scaffolds, thereby conferring resistance to thrombolysis. They also exacerbate damage to the blood-brain barrier (BBB) by degrading tight junction proteins via activation of inflammatory cascades. Additionally, the persistent formation of NETs impedes vascular remodeling and neural repair, thereby compromising long-term functional recovery. This review comprehensively examines

Status epilepticus-induced 12/15-lipoxygenase drives neuroinflammation and contributes to neuronal injuries and behavioral comorbidities.

Status epilepticus (SE) is a serious neurological condition defined as a continuous seizure lasting longer than 5 min or multiple seizures without full recovery of consciousness between them. Uncontrolled SE causes severe brain inflammation and damage, leading to life-long epilepsy and behavioral comorbidities. 12/15-Lipoxygenase (12/15-LOX), an enzyme that generates bioactive lipid metabolites from polyunsaturated fatty acids, plays pathogenic role in oxidative and inflammatory processes that can aggravate tissue injuries. However, its involvement in SE-triggered neuroinflammation and long-te

Pain mechanisms in PsA: differentiating inflammation-related pain in Achilles enthesitis using US and functional MRI.

OBJECTIVES: Pain in PsA is common and multifactorial, involving nociceptive, neuropathic and nociplastic mechanisms. We hypothesized that PsA patients with entheseal pain exhibit distinct brain activation patterns on functional MRI (fMRI), depending on the presence of US-detected inflammation. METHODS: PsA patients underwent clinical evaluation, US assessment of the Achilles tendon and fMRI brain imaging. Participants were grouped as follows: Group 1-clinical Achilles enthesitis with active US findings (hypoechogenicity and Doppler ≥grade-2); Group 2-clinical enthesitis but negative US; Group

Chronic airway inflammation is associated with sex-dependent changes in behaviour, neuroinflammation and the gut microbiome.

Asthma is a chronic inflammatory lung disease linked to a higher prevalence of comorbid mood disorders, such as anxiety and depression, though the underlying pathophysiology and the influence of biological sex remain poorly understood. To investigate this complex relationship, we employed a chronic house dust mite (HDM) exposure model in male and female C57BL/6 mice, examining the sex-dependent effects of allergic airway inflammation across systemic, neuroimmune, behavioural, and gut microbiome axes. Chronic HDM exposure successfully induced hallmark features of allergic airway disease in both

Proposal for Diagnostic Criteria for Manifestations of Chronic Graft-versus-Host Disease in Peripheral Nervous System and Muscles.

Atypical neurological graft-versus-host disease (cGVHD) is a rare complication after allogeneic hematopoietic stem cell transplantation (allo-HSCT). In addition to central nervous system (CNS) involvement, patients with cGVHD may also exhibit pathology of the peripheral nerves, muscles or neuromuscular junction. In this report, we propose more detailed diagnostic criteria for these manifestations and apply them in a case series of peripheral nervous system and muscles (PNSM) involvement of cGVHD. Based on an interdisciplinary evaluation applying the recent provisional criteria for atypical man
