OBJECTIVE: Meningeal lymphatic vessels (mLVs) play a significant role in neurological homeostasis and disease. However, their contribution to brain injury following cerebral venous sinus thrombosis (CVST) remains unknown. This study investigated whether mLV dysfunction influences the pathological progression of CVST by regulating the endoplasmic reticulum (ER) and oxidative stress(OS)pathways.
METHOD: A total of 65 male C57BL/6J mice were randomly assigned to four groups: sham-operated, CVST; CVST combined with cervical lymph node ligation (CVST + Ligation); and 4-phenylbutyric acid (4-PBA) i

Abnormal activation of the immune system and oxidative stress are crucial factors in neurodegenerative disorders, such as Parkinson's disease and Alzheimer's disease. Microglia, neutrophils, oxidative stress mediators such as reactive oxygen species (ROS), lipid peroxidation products (e.g., malondialdehyde), and nitrosative stress markers (e.g., nitrite and nitrate) play important roles in neuroinflammatory mechanisms. Microglial cells acquire a proinflammatory phenotype through interactions with endogenous or exogenous compounds, including cell debris, abnormally modified proteins (including

Mesial temporal lobe epilepsy with hippocampal sclerosis (MTLE-HS) is an intractable form of epilepsy involving the hippocampus, and temporal lobectomy remains an effective treatment. Human herpesvirus 6B (HHV-6B) establishes latency in the hippocampus and may contribute to MTLE-HS pathogenesis by altering host gene expression; however, transcriptomic data from healthy controls remain limited. This study investigated the role of HHV-6B to MTLE-HS pathogenesis by analyzing gene expression in resected hippocampal tissues. Samples were collected from 12 to 43 HHV-6 DNA-positive and -negative pati

OBJECTIVE: Recent studies have identified persistent immune inflammation as the third core pathological change in Alzheimer’s disease (AD). Progesterone receptor membrane component 2 (PGRMC2) is a member of the membrane-associated progesterone receptor family. This study aimed to investigate the effects of the expression of PGRMC2 in astrocytes in an AD mouse model. METHODS: PGRMC2 overexpression in astrocytes was induced in male APPswe/PSEN1dE9 transgenic (AD) mice and C57BL/6J wild-type mice via an adeno-associated virus vector. The Morris water maze and Y-maze tests were used to evaluate sp

AIMS: Mitochondrial homeostasis is essential for maintaining central nervous system function. Both inflammation and oxidative stress induced by chronic intermittent hypoxia (CIH) can result in neuronal mitochondrial injury in obstructive sleep apnea syndrome (OSAS)-related cognitive dysfunction. Recent evidence implicates neuronal impairment caused by abnormal activation of microglia as a key contributor to CIH-induced cognitive dysfunction. However, the mechanism between microglia and neuronal injury remains elusive. This study seeks to elucidate the underlying mechanism of microglia-mediated

INTRODUCTION: Neurological complications are increasingly recognized as a significant consequence of COVID-19; however, time-resolved, brain-specific characterization of transcriptional alterations underlying SARS-CoV-2-associated neuroinflammation and neuronal injury remain limited. We hypothesized that brain transcriptional responses evolve dynamically during acute SARS-CoV-2 infection, resulting in temporal transcriptional programs.
METHODS: KRT18-hACE2 transgenic mice were intranasally inoculated with SARS-CoV-2. Brain was harvested at 4 and 6 days post-infection (dpi) for analyses.

Fatigue in patients with Graves' disease (GD) is characterized by a profound lack of mental energy that affects daily functioning, including work. This symptom is particularly prominent in the early stages of the disease, affecting more than 50% of patients, but in some cases, it persists even after successful endocrinological treatment and restoration of euthyroidism. Individuals with persistent fatigue often seek support. Because this tiredness originates in the brain, we refer to it as brain fatigue. It is accompanied by a cluster of interconnected symptoms as cognitive, sensory, and emotio

BACKGROUND: Burn injury (BI) is one of the most devastating traumas, causing acute and long-term immune dysregulation and organ dysfunction. BI, by polygenic pathways, affects the central nervous system, which is manifested as chronic neurological dysfunction that extends well beyond complete wound healing. In this study, we examined BI-induced systemic inflammation and brain neuroinflammation and their impact on neurobehavior in mice.
METHODS: Male C57BL/6 J mice were subjected to BI. Cytokine array and enzyme-linked immunosorbent assay were utilized to evaluate cytokine levels in the serum

BACKGROUND: Visceral hypersensitivity is a prevalent and debilitating symptom in inflammatory bowel disease (IBD), severely impairing quality of life. Although mechanisms remain incompletely understood, recent studies have reported abnormal copper metabolism and elevated serum copper in IBD patients. However, whether copper homeostasis contributes to visceral hypersensitivity remains unknown.
METHODS: We established a dextran sulfate sodium (DSS)-induced colitis model in C57BL/6J mice to induce visceral hypersensitivity. Copper content in the terminal colon and dorsal root ganglia (DRG) was m

Diabetes-associated cognitive impairment (DACI) represents a debilitating complication lacking effective therapeutics. Extracellular vesicles (EVs) demonstrate a paradoxical dual role in DACI pathophysiology. Under diabetic conditions, EVs secreted by metabolically dysregulated adipose tissue and activated circulating inflammatory cells deliver neurotoxic cargo (miR-9-3p, miR-125a-5p) inducing neuronal ferroptosis, synaptic loss, and blood-brain barrier disruption via MEK/ROCK signaling. Brain-derived EVs enriched in complement proteins and mitochondrial markers serve as minimally invasive dia

Nanoencapsulation, especially in the form of PEGylated niosomes, is considered a novel strategy in drug delivery that improves the solubility and bioavailability of compounds. Silibinin, as a flavonoid, has low bioavailability despite its antioxidant, anti-inflammatory, and neuroprotective properties. Therefore, the present study was designed to investigate the effect of PEGylated niosomal nanoparticles containing Silibinin (Sili-PEG-Nios-NPs) on memory and learning, inflammatory pathways, and morphological changes in the hippocampus in an animal model of Alzheimer's disease. In this study, Si

Post-stroke depression (PSD) is a complex neuropsychiatric complication driven by neuroinflammation and ferroptosis, yet effective therapies remain limited. Sinensetin (SIN), a polymethoxylated flavone derived from citrus fruits, possesses potent anti-inflammatory and antioxidant properties. However, its therapeutic efficacy and underlying mechanisms in PSD have not been explored. To investigate this, a mouse model of PSD was established by combining photothrombotic stroke with low-dose lipopolysaccharide (LPS) administration. Mice were treated with SIN (25 and 50 mg/kg) for 14 days. Depressiv

BACKGROUND: Sepsis associated encephalopathy is a poorly understood yet common complication of sepsis. Immune challenge and animal models implicate blood-brain barrier (BBB) dysfunction and microglial activation in the hippocampus as key drivers of pathology. In this exploratory study, we used bloodstream infections (BSI) as a model of severe infection to investigate the relationship between changes in cognition with plasma biomarkers of brain injury, BBB permeability and microglial activation quantified using advanced neuroimaging.
METHODS: In this longitudinal exploratory study we recruited

In forensic pathology, accurately estimating the time since injury is essential. Current histological and imaging approaches commonly miss subtle temporal changes, especially in deaths occurring within hours of injury. This review discusses the timing of neuroinflammation after traumatic brain injury and emphasizes possible markers for estimating the time of injury in forensic cases. Promising markers include microglial activation (allograft inflammatory factor 1 and transmembrane protein 119, detectable within 10 min to 2 h), β-amyloid precursor protein accumulation (20-35 min), high-mobility

Neuroinflammation in Alzheimer’s disease (AD) extends beyond a secondary response to amyloid-β plaques and neurofibrillary tangles, representing a fundamental driver of disease pathogenesis. The NLRP3 inflammasome has emerged as a central mediator of neuroinflammation in AD, highlighting the importance of understanding its downstream effectors. Interleukin-18 (IL-18), a key proinflammatory cytokine activated by NLRP3 inflammasome, plays a crucial role in innate immunity and neuroinflammatory responses in neurodegenerative diseases and aging. However, its specific contribution to AD pathogenesi

Hyperammonemia (HA) is a metabolic disorder characterized by elevated ammonia levels in the blood. Ammonia produced from the metabolism of amino acids is mainly detoxified in the liver through the urea cycle. However, defects in this cycle result in the buildup of ammonia in the blood, which is highly neurotoxic and disrupts multiple signaling pathways in the brain, including nitric oxide (NO). NO is produced from the enzyme nitric oxide synthase (NOS), which exists in three different isoforms: neuronal NOS (nNOS), endothelial NOS (eNOS), and inducible NOS (iNOS). NO is an important signaling

Central diabetes insipidus (CDI) is a rare condition characterized by arginine vasopressin deficiency, leading to persistent polyuria and polydipsia. Although it can present at any age, idiopathic CDI in young adults is uncommon and diagnostically challenging. Inflammatory conditions such as infundibuloneurohypophysitis must be considered, particularly when magnetic resonance imaging (MRI) reveals a thickened pituitary stalk and absence of the posterior pituitary bright spot. Early recognition and differentiation from diabetes mellitus, nephrogenic diabetes insipidus, and primary polydipsia ar

The protective impact and unique mechanisms of human umbilical cord mesenchymal stem cells (HUC-MSCs) transplantation following hypoxic-ischemic (HI)-induced brain white-matter injury (WMI) were explored. To establish a WMI model, Sprague-Dawley rats with three days after birth underwent unilateral carotid artery ligation, followed by hypoxic exposure (8% oxygen and 92% nitrogen). Subsequently, HUC-MSC transplantation was performed into the lateral ventricle. Molecular and behavioral experiments were conducted to assess how it would influence NLRP3 inflammasome activation, M1/M2 microglial pol
