Unknotting the crosstalk between COPD and neuroinflammation.

COPD is a progressive respiratory illness that is mostly caused by air pollution, biomass fuel exposure, or long-term cigarette smoking. It is increasingly being referred to as a multisystem inflammatory disease rather than a pulmonary-only one. As a result of chronic airway inflammation in COPD, pro-inflammatory cytokines such as IL-1β, IL-6 and TNF-α are released throughout the body, causing systemic oxidative stress. Cardiovascular disease, skeletal muscular atrophy, osteoporosis, metabolic syndrome, and neurological impairment manifested as depression, anxiety, and cognitive impairment are

Anxiety in male psoriasis mouse model is mediated by the interaction between γδ T cell-derived IL-17A and microglia.

OBJECTIVE: Both clinical and animal studies indicate a high incidence of anxiety in psoriasis, though the underlying mechanisms remain poorly understood. Our previous review suggested that T cells, related cytokines, and blood-brain barrier (BBB) disruption might serve as potential links between the two conditions. This study aimed to investigate the mechanistic connection. METHODS: Psoriasis was induced in male C57BL/6J mice using imiquimod (IMQ). Anxiety-like behaviors were assessed through the open field test, elevated plus maze test, and light-dark box test. Microglial activation was eval

Movement disorders due to primary central nervous system lymphoma: An illustrative case and systematic literature review.

INTRODUCTION: Movement disorders (MD) can result from various mechanisms: central nervous system infection (20%), trauma (15%), metabolic dysregulation (7%), vascular causes (22%), or inflammatory, degenerative, and tumoral diseases. MD leading to the diagnosis of primary central nervous system lymphoma (PCNSL) are rare but may be treatable, depending on how quickly the diagnosis is made. The objective of this study was to explore movement disorders in PCNSL and discuss the potential pathophysiological relationships between PCNSL and symptoms. METHODS: We conducted a systematic review from da

Microglial efferocytosis dysfunction contributes to depression-like behaviors and cognitive impairment in aged mice following chronic sleep deprivation.

BACKGROUND: Chronic sleep deprivation (CSD), commonly observed in the elderly, is associated with an increased risk of neuroinflammation, depression, and cognitive impairment. Microglial efferocytosis plays a vital role in maintaining neural homeostasis and tissue repair. However, the mechanistic role of microglial efferocytosis in linking CSD to neurobehavioral impairments in aging remains largely undefined. METHODS: A 21-day CSD model was established in aged mice, and depression-like behaviors and cognitive function were evaluated. Hippocampal neuronal injury and neuroinflammation were eval

β-hydroxybutyrate alleviates motor impairment and neurological damage in hypoxic-ischemic encephalopathy mice.

Hypoxic-ischemic encephalopathy (HIE) is a major reason for neonatal mortality and long-term neurological deficits, with limited efficacy from current standard treatments like therapeutic hypothermia. β-hydroxybutyrate (BHB) is an abundant ketone body synthesized in the liver, possesses energy supply, anti-inflammatory, antioxidant, and anti-apoptotic activities. This study aims to investigate the effect of exogenous BHB on motor impairments and brain damage in HIE mice. Ten-day-old mouse pups underwent left common carotid artery ligation followed by hypoxia to induce HIE. Control underwent Sh

Long COVID neuropathy: The role of mast cells.

Postacute sequelae of SARS-CoV-2 infection (PASC), or Long COVID, is estimated to affect over 60 million individuals globally, with almost half of COVID-19 survivors experiencing persistent symptoms such as neuropathic pain, fatigue, and autonomic dysfunction. Despite its prevalence, the pathophysiology of PASC remains poorly understood. This narrative review highlights activation of mast cells (MCs), the unique tissue immune cells as a central contributor to neuropathic manifestations in PASC. Mast cell locations near nerves and vessels allows them to regulate neuroimmune and neurovascular pr

The 129S1/SvlmJ mouse strain recapitulates severe hypertensive target organ damage under moderate angiotensin II-induced hypertension.

Hypertension remains the leading cause of cerebral, cardiac, renal, and retinal vascular damage. However, genetic determinants underlying organ-specific vulnerability are poorly understood, and commonly used mouse models, notably C57BL/6J, often fail to recapitulate severe hypertensive complications seen in humans. This study compares the widely used C57BL/6J mouse strain with the genetically distinct 129S1/SvlmJ strain under hypertensive stress, aiming to identify a model that better reproduces hypertensive target organ damage. Moderate hypertension was induced in 129S1/SvlmJ and C57BL/6J mic

From Gums to Brain: Unveiling the Hidden Inflammatory Connection Between Periodontal and Cerebrovascular Diseases.

Periodontal disease (PD) encompasses inflammatory conditions affecting the supporting structures of the teeth, with gingivitis and periodontitis being the most prevalent forms. Gingivitis, characterized by gum inflammation and bleeding, can progress to periodontitis, which involves the destruction of connective tissue and alveolar bone. PD is a global health burden, affecting over a billion individuals, with 42.2% of adults aged ≥30 years in the United States experiencing some form of periodontitis. Without proper management, PD can lead to severe systemic consequences. The cause of PD is mult

Pharmacological advances in multi-targeted strategies for type 2 diabetes mellitus: a systematic perspective based on traditional Chinese medicine.

Type 2 diabetes mellitus (T2DM) is a complex systemic metabolic disease driven by insulin resistance, β-cell dysfunction, chronic low-grade inflammation, oxidative stress, and neuro-immune dysregulation. It frequently progresses to multi-organ complications affecting the kidneys, retina, heart, and central nervous system. This review synthesizes mechanistic and translational evidence on Traditional Chinese Medicine (TCM)-related botanical drugs and botanical preparations (formula-based interventions), along with representative plant metabolites that are frequently investigated in the TCM resea

Peripheral cannabinoid receptor activation attenuates frostbite-induced chronic pain via modulation of TRP channels, neuroinflammation, and autophagy.

Frostbite injury is a debilitating cold injury encountered in extreme high-altitude and subzero environments, frequently resulting in persistent chronic pain even after tissue healing. Using our previously validated frostbite-induced chronic pain model, we further investigated the contribution of neuroimmune, excitatory mechanisms and evaluated the therapeutic efficacy of peripheral cannabinoid receptor activation. Frostbite produced significant mechanical allodynia, mechanical hyperalgesia, and cold hypersensitivity, along with increased spontaneous ongoing pain behaviors. Local peripheral ad

Recurrent cerebral venous thrombosis associated with IgG4-related hypertrophic pachymeningitis: Case report and literature review.

IgG4-related disease (IgG4-RD) is a systemic inflammatory condition marked by tissue infiltration of IgG4-positive plasma cells, often resulting in fibrosis. While IgG4-RD commonly affects organs such as the pancreas and salivary glands, central nervous system involvement, particularly hypertrophic pachymeningitis (HP), is rare and can present unique clinical challenges. This case report describes a 67-year-old male with IgG4-related hypertrophic pachymeningitis complicated by recurrent cerebral venous thrombosis (CVT), a rarely reported association. The patient initially responded to high-dos

Gastrointestinal dysfunction after brain injury: Mechanisms and the role of the brain-gut axis.

Brain injury (BI) encompasses traumatic BI and stroke, and is a leading cause of mortality and morbidity worldwide. In addition to primary brain damage, BI can cause a series of sequelae, of which gastrointestinal (GI) dysfunction is one of the most important affecting patient outcomes. GI dysfunction, including delayed gastric emptying, increased intestinal permeability, and gut dysbiosis, is common among patients with BI. GI dysfunction not only impairs nutrient absorption and increases the risk for infection, but also enhances secondary BI by aggravating the systemic inflammatory response.

Neuroprotective Effects of Esketamine in Central Nervous System Disorders: Mechanisms and Cellular Targets.

Esketamine (ESK), the dextrorotatory enantiomer of ketamine, is an antagonist of the N-methyl-D-aspartic acid (NMDA) receptor. ESK is considered an effective anesthetic because of its ability to mitigate postoperative pain and opioid use. Recently, it has attracted significant research attention for its fast-acting antidepressant effects. Given the advantages of ESK in both perioperative and postoperative contexts, foundational research into this molecule is progressively moving forward. Accumulating evidence suggests that ESK possesses anti-inflammatory, antiapoptotic and antioxidant characte

[Mechanism of Wuyou Decoction in improving hippocampal injury in chronic sleep deprivation model rats by inhibiting neuroinflammation and protecting blood-brain barrier].

This study explored the mechanism by which Wuyou Decoction(WYD) ameliorated hippocampal injury in a chronic sleep deprivation model rat by suppressing neuroinflammation and protecting the blood-brain barrier(BBB). Fifty 2-month-old male SD rats were randomly divided into five groups: normal, model, low-dose WYD, medium-dose WYD, and high-dose WYD. Except for the normal group, all rats underwent a 7-day adaptation period for the water platform sleep deprivation model, which was followed by 21 days of formal sleep deprivation modeling. Concurrently with the start of formal modeling, rats in the

Lipocalin-2 perpetuates postoperative and post-infectious neuroinflammation and anxiety-like behavior.

Lipocalin-2 (LCN2) is a pleiotropic cytokine with well-established roles in immune defense, iron metabolism, and inflammation that has been used as a biomarker for kidney injury, cancer, and infection. Recent work has also suggested a role for LCN2 in brain function and behavior, suggesting that LCN2 excess could be a driver of neuropsychiatric complications of infectious and inflammatory conditions. Here, we show that LCN2 levels remain elevated in both the brain and circulation for weeks after an acute infection or simple laparotomy in male and female mice. We used an inducible knockout mous

Empagliflozin and dapagliflozin, sodium glucose cotransporter 2 inhibitors, may improve cognitive dysfunctions: in silico and in vivo findings.

Type 2 Diabetes Mellitus (T2D) accelerates cognitive decline through a complex interaction of metabolic, oxidative, inflammatory, and vascular pathways and is widely recognized as a significant risk factor for Alzheimer's disease (AD). Sodium-glucose cotransporter (SGLT)2 inhibitors are widely used in the treatment of T2D, and accumulating evidence suggests that they may influence neurodegenerative processes beyond their glycemic effects. In this context, the present study investigated the potential contributions of empagliflozin (EMPA) and dapagliflozin (DAPA) to cognitive function by evaluat

Case Report: Delayed gastrointestinal structural complications requiring surgery following massive paint thinner ingestion in an adult.

BACKGROUND: Paint thinners, which contain volatile aromatic hydrocarbons like toluene and xylene, are primarily associated with acute systemic toxicity (e.g., central nervous system depression, metabolic acidosis) following inhalation. Case reports detailing severe, delayed gastrointestinal (GI) structural complications in adults after massive oral ingestion, which necessitate extensive surgical intervention, remain exceedingly rare in the medical literature. CASE DESCRIPTION: A 68-year-old male with a history of hypertension and chronic alcoholism presented 8 h after mistakenly ingesting app

Evidence of White Matter Neuroinflammation in Myalgic Encephalomyelitis/Chronic Fatigue Syndrome: A Diffusion-Based Neuroinflammation Imaging Study.

Myalgic encephalomyelitis/chronic fatigue syndrome (ME/CFS) is a debilitating disorder with suspected neuroinflammatory pathophysiology. However, previous diffusion tensor imaging (DTI) studies have reported inconsistent white matter abnormalities in ME/CFS, and specific white matter inflammatory changes remain poorly characterised. This study employed an advanced diffusion-based neuroinflammation imaging (NII) model to investigate white matter neuroinflammation in ME/CFS. Diffusion MRI data from 67 ME/CFS patients (median age, 38; and 54 women) and 67 rigorously matched healthy controls (HCs)
