Modeling striatal development and disease with human pluripotent stem cells.

The striatum plays a central role in motor control, cognition, reward processing, and habit formation, and its dysfunction is implicated in a broad spectrum of neurological and psychiatric disorders. Although animal models have provided important insights into striatal development and disease mechanisms, species-specific differences in cellular composition, developmental timing, and circuit organization limit their translational relevance to the human brain. In this context, human pluripotent stem cells (PSCs), including embryonic stem cells and induced pluripotent stem cells, have emerged as

Focused Ultrasound Modulation of PGC-1α Pathways in Neurological Disease: Mechanistic Rationale and Translational Opportunities.

The brain's disproportionate energy demand creates an enduring bioenergetic imperative where mitochondrial performance directly determines synaptic resilience and neuronal survival. Peroxisome proliferator-activated receptor gamma coactivator 1 alpha (PGC-1α) has emerged as a master transcriptional regulator orchestrating mitochondrial biogenesis, antioxidant defenses, proteostasis, and neuroplasticity, with dysregulation of this axis representing a convergent pathogenic mechanism across Parkinson's disease, Alzheimer's disease, Huntington's disease, stroke, and neuropsychiatric disorders. Assessment of Dose Proportionality and the Effect of Food on a Once-Daily Extended-Release Tablet Formulation of Deutetrabenazine in Healthy Participants.

BACKGROUND AND OBJECTIVE: Deutetrabenazine is approved as a treatment in adults for tardive dyskinesia and chorea associated with Huntington's disease. A once-daily tablet (Austedo XR® Teva) was recently approved in addition to the twice-daily tablet (Austedo®), and studies were conducted to evaluate dosage strength proportionality and food effect of the once-daily formulation. METHODS: In a randomized cross-over study of dosage strength proportionality, healthy participants (n = 116) received single doses of the once-daily deutetrabenazine formulation and a power model was fitted to describe

Somatic CRISPR editing of Msh3 mitigates Huntington's disease pathology in mice.

Huntington's disease (HD) is a fatal, dominantly inherited neurodegenerative disorder caused by a CAG repeat expansion in Huntingtin (HTT) exon 1. Further progressive CAG repeat expansion occurs in somatic cells, particularly in neurons, and drives the timing of clinical onset. Therefore, therapeutic strategies to slow somatic expansion are predicted to be disease-modifying. Somatic CAG expansion is driven by mismatch repair protein MSH3, a leading therapeutic target supported by human genetic data. To gain insight into the impact of targeting MSH3 at different stages of the disease process we

Mismatch repair dissection by in vivo RNAi reveals dose-dependent modulators of somatic instability and proteome remodeling in Huntington's disease.

Human and mouse genetics have established mismatch repair (MMR) as a central mediator of somatic repeat expansion, a key pathogenic process in Huntington's disease (HD) and related disorders. How individual MMR components function within the intact mammalian brain and interact with broader cellular networks remains poorly understood. We screened more than 500 chemically stabilized siRNAs targeting 10 MMR genes and used interventional RNAi in the Q111 HD mouse model to systematically dissect MMR function in vivo. MSH3 and PMS1 emerged as the most dose-sensitive regulators of somatic expansion b

Data mining revealed no other trinucleotide repeat expansion associated with the Huntington's disease biomarker (CAG).

The CAG trinucleotide repeat expansion is believed to be the only expansion associated with Huntington's Disease and other phenotypically similar genetic diseases. Here, we mine a large set of data that contains samples from patients of Huntington's disease as well as control to find associated trinucleotide repeat expansions of CAG. The total number of pathological and control datasets is 83 and 74, respectively. The total number of DNA base pairs in these samples is 210 and 162 Gigabases (1 × 109 bases). Predicated on association rules, most of the trinucleotide repeats found in these datase

Development of an in vivo, screenable, split-luciferase based model of huntingtin multimerization.

Huntington's disease is a neurodegenerative disorder caused by a polyglutamine (polyQ) expansion in exon one of the gene that encodes for the protein huntingtin (HTT). PolyQ expansion drives HTT aggregation into multimeric species that range from soluble oligomers to fibrillar, insoluble inclusion bodies. Cellular mechanisms facilitating HTT aggregation are incompletely understood, hindering efforts to develop strategies that prevent inclusion body formation or promote clearance of misfolded protein. To enable future unbiased in vivo screening approaches to identify genetic modifiers and pharm

Leucine rich repeat containing protein 8B (LRRC8B) - A novel modulator of endoplasmic reticulum (ER) stress response.

The endoplasmic reticulum (ER), a major calcium ion (Ca2+) reservoir, plays a pivotal role in lipid synthesis, protein synthesis, and secretion. Disruption of ER function leads to the accumulation of misfolded proteins, resulting in ER stress. To restore cellular homeostasis, the unfolded protein response (UPR) is activated. However, prolonged or irreversible ER stress can trigger apoptosis and cell death. Cellular dysfunction and apoptosis arising from disrupted ER Ca2+ homeostasis are often implicated in neurodegenerative and metabolic disorders. We investigated the role of LRRC8B, an ER-res

Machine learning applications in Huntington's disease prognosis: A review.

Understanding the trajectory of Huntington's disease (HD) is critical for patient stratification and the development of targeted interventions. Traditionally, studies relied on age-CAG models to estimate disease onset and progression, based on the well-established relationship between CAG repeat length and age at onset. However, additional genetic, environmental, and clinical factors can cause substantial variability. Recent machine learning approaches integrate clinical, imaging, and molecular data for more precise prediction of disease progression. Following PRISMA guidelines, we systematica

Enhancing the detection of HTT1a with neoepitope antibodies in mouse models of Huntington's disease.

Huntington's disease is an inherited neurodegenerative disorder caused by a CAG repeat expansion in exon 1 of the huntingtin (HTT) gene, encoding an expanded polyglutamine tract in the huntingtin (HTT) protein. The pathogenic CAG repeat of HTT is unstable and undergoes progressive somatic expansion in specific brain cells and peripheral tissues throughout life. Genes involved in DNA mismatch repair pathways, which promote repeat expansion, have been identified as genetic modifiers of the disease. Consequently, the rate of CAG repeat expansion is a key determinant driving the age of onset and d

A Novel Plate Reader-Based Protocol for Measurement of DNAJB6 Dimerization Activity.

Progressive neurodegeneration linked to the accumulation of misfolded proteins is a hallmark of several neurodegenerative disorders, including Parkinson's disease, Huntington's disease, and Alzheimer's disease. Dysfunction in the protein homeostasis machinery correlates with pathology. The chaperone protein DNAJB6 is expressed in neurons and oligodendrocytes and has been shown to play a key role in preventing amyloid aggregation by binding to amyloidogenic proteins and facilitating their refolding or degradation, in cooperation with other chaperones. Here, we describe a simple and feasible ass

Mitochondrial dynamics in Huntington's disease.

Emerging evidence suggests a central and early role of mitochondrial dysfunction, including altered mitochondrial dynamics, in Huntington's disease (HD) pathogenesis. Processes such as mitochondrial fission, fusion, transport and mitophagy are vital for proper mitochondrial function and seem to be key mediators of neuronal vulnerability in HD. In this review, we summarize mechanistic insights into mitochondrial dynamics in HD, highlighting how mutant huntingtin (mHTT) impairs mitochondrial biogenesis and morphology, disrupts Drp1-dependent fission, compromises fusion, transport and organelle c
