Neuroinflammation, Autophagy, and Neurodegeneration: Mechanisms and Therapeutic Insights.

Neuroinflammation and autophagy dysregulation are critical in the pathogenesis of neurodegenerative diseases like Alzheimer's, Parkinson's, and Huntington's disease. Neuroinflammation occurs after a sustained immune response, which transitions into a chronic pathological state, leading to the sustained generation of pro-inflammatory cytokines and oxidative stress, causing neuronal damage. Meanwhile, defective autophagy exacerbates disease by promoting protein accumulation, e.g., amyloid-β, tau, and α-synuclein, thereby enhancing neuroinflammation. In this review, we focus on critical pathways,

Long non-coding RNAs in neurodegenerative diseases - Molecular mechanisms, liquid biopsy biomarkers, and therapeutic targets: A review.

Neurodegenerative diseases (NDDs), such as Alzheimer's disease (AD), Parkinson's disease (PD), amyotrophic lateral sclerosis (ALS), and Huntington's disease (HD), are age-related disorders characterized by progressive neuronal loss, cognitive decline, and limited options for disease-modifying treatments. Increasing evidence suggests that long non-coding RNAs (lncRNAs) play significant roles in neurodevelopment, neuronal homeostasis, and disease progression; however, their involvement in shared pathogenic pathways and clinical applications remains inadequately defined. This review consolidates

ATF3-dependent formation of inclusion bodies in polyQ-expressing human iPSC-derived neurons confers cellular protection.

Huntington's disease (HD) is an incurable, neurodegenerative disorder. While the causative mutation - CAG expansions within the coding region of the Huntingtin (HTT) gene - has been identified over 30 years ago, the pathological mechanisms underlying HD are still not clear. The abnormal CAG track encodes a polyglutamine (polyQ) expanded protein, which leads to HTT protein misfolding. These polyQ aggregates can form insoluble inclusion bodies (IBs); however, whether IBs are protective or detrimental remains debatable. Here we developed fluorescent iPSC-based human neuronal models for polyQ-rela

Clinical implications of malnutrition in Huntington's disease progression: evidence from a Chinese cohort and Mendelian randomization.

BACKGROUND: Huntington's disease (HD) is a neurodegenerative disorder associated with progressive motor, cognitive, and psychiatric dysfunction. Peripheral metabolic disturbances, including malnutrition, are commonly observed in HD. However, the prevalence, clinical correlations, and prognostic value of malnutrition in HD, especially among Chinese patients, remain inadequately explored. METHODS: This cohort study recruited 113 genetically confirmed HD patients and 113 age/sex-matched healthy controls (HCs). Nutritional status was assessed using the Controlling Nutritional Status (CONUT) score

Scn4b Modulates Huntington's Disease Phenotype Severity in vivo.

Although it has been known for over 30 years that CAG trinucleotide repeat expansions in the HTT gene are the cause of Huntington's disease (HD), it is still not understood how these mutations lead to the loss of striatal spiny projection neurons (SPNs) and other vulnerable neuronal cell types in HD. Here we show that SCN4B , a gene that is enriched in neurons that influence motor function, including striatal SPNs, modulates HD-associated phenotypes in vivo . Loss of Scn4b in wild-type mice mimics and Scn4b overexpression in an HD mouse model rescues several HD-associated phenotypes, including

Organelles storing Ca 2+ in the brain cells: New druggable targets in neurodegenerative diseases.

Several lines of evidence suggest that targeting dysfunctional calcium (Ca 2+ )-storing organelles and their defective connections may represent a promising therapeutic strategy counteracting neurodegeneration. Dysfunction in these compartments converges to promote oxidative and endoplasmic reticulum stress, energy failure, autophagy blockade or hyperactivation, and progressive neurodegeneration. Within the intracellular scenario, several dysfunctional organelles have been characterized in terms of their capability to hijack Ca 2+ signaling during neurodegeneration to deadly impact on neuronal

Targeting Non-Coding RNAs as a Potential Therapeutic and Delivery Strategy Against Neurodegenerative Diseases.

Neurodegenerative diseases (NDs), including Alzheimer's disease, Parkinson's disease, Huntington's disease, and amyotrophic lateral sclerosis (ALS), represent a growing global health challenge characterized by progressive neuronal loss and a lack of definitive disease-modifying treatments. This review explores the emerging potential of targeting non-coding RNAs (ncRNAs), such as microRNAs (miRNAs), long non-coding RNAs (lncRNAs), and exosomal RNAs, to modulate pathogenic molecular pathways and address the underlying molecular origins of neurodegeneration. We evaluate the integration of advance

Personalized health predictions challenge existing insurance frameworks: Time to revisit life and disability insurance regulation?

Personalized health predictions are developing rapidly across a wide range of diseases, challenging the existing legal frameworks for life and disability insurance. These frameworks were originally developed with a focus on presymptomatic genetic testing for severe monogenic diseases such as Huntington's disease. The rapid expansion of personalized medicine raises questions about whether existing legal frameworks remain adequate. This paper examines the underlying ethical values of current legal frameworks that rely on financial thresholds to balance stakeholders' interests and explores how th

Effect of tetrabenazine on Stroop interference in Huntington's disease: Protocol.

BACKGROUND: Tetrabenazine has been shown to improve gating of abnormal visual stimuli and improve postural stability in Huntington's disease (HD) patients as measured by computerized dynamic posturography testing. This open label pilot study aims to elucidate whether partial dopaminergic depletion via low dose tetrabenazine has a similar effect on masking out of abnormal visual stimuli on the Stroop interference test in HD patients. The study endpoint is the Golden formula calculated from off and on tetrabenazine Stroop test raw scores. METHODS: The study will be performed as a single arm med

Iloperidone treatment mitigates the Juvenile Huntington's Disease phenotype possibly via Sigma-1 Receptor Modulation.

Juvenile Huntington's disease (jHD) is a severe genetic disorder with combined neurodegenerative and neurodevelopmental features, characterized by early onset and markedly reduced life expectancy. Despite extensive efforts, drug development has been largely unsuccessful and no effective therapy capable of mitigating the pathological phenotype is currently available. Here, we investigated the potential of the FDA-approved drug iloperidone to ameliorate disease-related phenotypes in cortical neurons differentiated from patient-derived induced pluripotent stem cells. Given previous reports indica

Ethnic Bridging of Votoplam, a HTT Gene Splicing Modifier, Using Pharmacokinetic Comparison Between Caucasian and Japanese Subjects.

Votoplam is a novel, orally bioavailable, small molecule HTT gene splicing modifier that is being developed for the treatment of Huntington's disease. A single-dose, open-label, three-dose level (5, 10, 20 mg), parallel group ethnicity study was conducted to evaluate the pharmacokinetics and safety of votoplam in healthy Caucasian versus Japanese participants. Six participants per ethnic group were enrolled at each dose (12 participants per dose level overall). Thirty-five participants (N = 18 Japanese; N = 17 Caucasian) completed the study. Study results showed slightly higher exposure for Ja

Pathogenic human huntingtin expression causes prolific intramuscular aggregation, leading to nuclear, metabolic, and physiological dysregulation in striated muscle.

Huntington's disease is caused by expansion of a CAG repeat in the human HTT gene, producing a mutant huntingtin protein that misfolds and forms intracellular aggregates. Although Huntington's disease is primarily characterized as a neurodegenerative disorder, mutant huntingtin is ubiquitously expressed, and peripheral tissues such as skeletal muscle exhibit pathological abnormalities. To define the muscle-intrinsic consequences of pathogenic huntingtin expression, we expressed caspase-6 truncated pathogenic human huntingtin in body wall muscle of Drosophila melanogaster larvae and performed q

scVIP: personalized modeling of single-cell transcriptomes for developmental and disease phenotypes.

Single-cell transcriptomics resolves cellular heterogeneity within individuals, but connecting molecular states to individual-level phenotypes requires frameworks that explicitly bridge these scales. We present scVIP, a generative model that links gene expression, cell-type composition, and phenotypic measurements within a single probabilistic model, which enables accurate phenotype prediction and interpretable trajectory inference. A cell-type-aware multi-instance learning architecture learns donor embeddings that capture progression while localizing phenotype-associated signals to specific c
