CAG-targeting artificial miRNA with reduced off-target risk for efficient lowering of pathogenic polyglutamine proteins.

Huntington's disease (HD) is the best-known example of a neurodegenerative disorder caused by the expansion of a glutamine-encoding CAG repeat in the causative gene. Growing evidence indicates that somatic CAG expansions play a key role in disease progression, providing a strong rationale for therapeutic strategies directly targeting the repeat tract. However, achieving sufficient efficacy while maintaining allele selectivity and minimizing off-target effects remains a major challenge. Here, we developed allele-selective, CAG-targeting artificial microRNA (amiRNA) molecules that exhibit signif

Lipid composition controls the huntingtin exon 1 membrane-association and differentially modulates its flanking regions' dynamics.

The pathological expansion of the polyglutamine (polyQ) repeat within the first exon of huntingtin (Httex1) protein is a defining hallmark of Huntington's disease (HD). Multiple evidence supports that the membrane recruitment of Httex1 is critical for its self-assembly and related toxicity in HD. In this work, we quantitatively examined the early steps of monomeric Httex1(23Q) association with lipid membranes and its impact on the conformational dynamics of the adjacent polyQ regions-the N-terminal N17 segment and C-terminal proline-rich region (PRR). A broad range of membrane physical propert

Remote smartphone-based spoken language screening predicts clinical markers in Huntington's disease.

Assessment and monitoring of Huntington's disease (HD) symptoms remain limited to infrequent, clinic-based evaluations. We evaluated whether fully automated linguistic analysis of speech tasks recorded via smartphone can remotely capture core clinical markers of HD severity. In this cross-sectional multicenter study across Czech and German sites, 53 participants, including 30 HD (9 pre-symptomatic, 3 prodromal, and 18 manifest) and 23 healthy controls, completed a smartphone-based speech assessment, including spontaneous monologue and fairy tale retelling for 7 consecutive days. Recordings wer

Decoding shared pathogenic networks of oxidative stress in neuropsychiatric disorders to prioritize multi-target therapeutics from natural products.

BACKGROUND: Neurodegenerative and psychiatric disorders, including Alzheimer's disease (AD), Parkinson's disease (PD), Huntington's disease (HD), and schizophrenia (SZ), are characterized by progressive neuronal loss and synaptic dysfunction. Despite their severity, effective disease-modifying treatments remain unavailable, largely due to the elusive nature of their underlying molecular mechanisms. METHODS: To elucidate these mechanisms, we conducted an integrative systems biology analysis incorporating transcriptomic datasets, in silico proteomic networks, and inferred metabolomic profiles.

Huntingtin polyglutamine expansions misdirect axonal transport by perturbing motor and adaptor recruitment.

Huntington's disease is caused by polyglutamine (polyQ) expansions in huntingtin (HTT). PolyQ lengths >35Q lead to neurodegeneration, and longer repeats correspond to earlier onset of symptoms. HTT scaffolds kinesin-1 and dynein to organelles directly and through adaptors. We tracked BDNF vesicles, mitochondria, and lysosomes in stem-cell-derived neurons engineered to express HTT with polyQ lengths of 30, 45, 65, and 81. BDNF endosomes were more motile in HTT-45Q and HTT-65Q neurons and misdirected toward the distal tip in HTT-81Q neurons. Under neuroinflammatory stress, polyQ expansions resul

An integrative neuropharmacological review of Huntington's disease challenges and the role of novel formulations in addressing pharmacological‒pharmaceutical limitations.

BACKGROUND: Huntington's disease (HD) is an autosomal dominant neurodegenerative disorder caused by CAG repeat expansion in the huntingtin gene, leading to progressive neuronal dysfunction and neurodegeneration. Although classically defined as a brain-restricted disorder marked by striatal and cortical degeneration, increasing evidence suggests HD as a multisystem disease involving both central and peripheral pathological alterations. OBJECTIVE: This review aims to provide an integrated overview of neuronal and non-neuronal mechanisms underlying HD, focusing on systemic alterations that influ

C57BL/6 BAC-CAG Huntington's disease mice show somatic CAG expansion and responses to small interfering RNAs comparable to the FVB strain.

Huntington's disease (HD) is a neurodegenerative disorder caused by CAG repeat expansion in the huntingtin (HTT) gene, with longer repeats linked to earlier onset. Somatic CAG expansion, particularly in the striatum, contributes to disease progression and is influenced by HTT biology and genetic modifiers. Modulating somatic expansion is emerging as a promising approach to slow or prevent HD, and mouse models have been crucial for preclinical testing of different therapeutic strategies. The BAC-CAG model, developed on the FVB strain, has been used to study somatic expansion of human expanded H

Pertrochanteric Femur Fracture in a Post-Polio Patient treated with a Pediatric Hip Plate: A Case Report.

INTRODUCTION: Fracture fixation in post-polio residual paralysis is technically demanding due to dysplastic proximal femoral anatomy, osteopenia, and distorted trochanteric morphology. Standard cephalomedullary nailing may be impossible when the medullary canal is narrow, or the entry point is malpositioned. CASE REPORT: A 45-year-old man with childhood poliomyelitis and complete paralysis of the right leg sustained an AO 31-A1.2 pertrochanteric fracture. Radiographs revealed coxa valga and a markedly narrow canal, precluding safe cephalomedullary nailing. Closed reduction on a traction table

Mesenchymal stromal cells in age-related neurodegenerative disorders: mechanisms, translational barriers, and precision strategies.

Ageing remodels the neuroimmune, metabolic, and vascular environment in ways that accelerate neuronal vulnerability and constrain tissue repair. Mesenchymal stromal cells (MSCs) and MSC-derived extracellular vesicles (MSC-EVs) are therefore attractive candidates for neurodegenerative disorders because their effects extend beyond cell replacement to coordinated paracrine, immunomodulatory, trophic, and vascular regulation. This critical review evaluates the biological rationale, disease-specific evidence, translational limitations, and precision-development strategies for MSC-based intervention

Beyond the brain barrier: Hybrid nanoparticles orchestrating intelligent neuro-theranostics.

Hybrid nanoparticles (HNPs) that integrate organic and inorganic components have been recognized as one of the most sophisticated solutions in the field of nanomedicine to surpass the physiological limitations of the blood-brain barrier (BBB). By the combination of biocompatibility, flexibility, and the capacity of functionalization of the organic shells with the magnetic, optical, or structural precision of the inorganic cores, HNPs provide receptor-mediated transport, controlled drug release, and multimodal imaging with high efficiency. This review details the molecular mechanisms of HNPs cr

A Dataset of Microelectrode Recordings from Deep Brain Stimulation Procedures.

Precise intraoperative localisation of subcortical brain structures remains a critical challenge in deep brain stimulation, yet openly available microelectrode recording datasets are scarce. We present a dataset of 6,646 processed MER recordings from 132 patients with neurological disorders, including Parkinson's disease, dystonia, Huntington's disease, epilepsy and others, acquired during DBS procedures. Signals were band-pass filtered and cleaned using an automated machine learning-based artifact rejection pipeline; annotation quality was confirmed by independent review. In addition an exper
