Metabolic-based therapeutic strategies to treat Huntington's disease.

Huntington's disease is an autosomal dominant neurodegenerative disorder marked by progressive motor, cognitive, and psychiatric decline. Huntington's disease arises from the expansion of particular DNA sequences (cytosine/ adenosine/guanine repeats that encode glutamine) within the huntingtin (HTT) gene. This expansion leads to the synthesis of a mutant huntingtin protein (mhtt) featuring an excessively long polyglutamine segment, which is harmful and prone to form aggregates or clusters within cells. A range of abnormalities has been observed in both the central nervous system and peripheral

Visual cognition as a marker of longitudinal disease progression in Huntington's disease.

BACKGROUND: Visual cognitive impairment is common in Huntington's disease (HD), but its longitudinal trajectory and prognostic value remain unclear. We aimed to characterize changes in visual cognition in premanifest (preHD) and early manifest HD compared with healthy controls, and to determine whether baseline visual cognition is associated with clinical progression. METHODS: A total of 148 participants (78 controls, 39 preHD, 31 early HD) underwent annual assessments at baseline, 12, and 24 months. Visual cognition was assessed using an assessment protocol focused on attention, perception/p

c-Jun in neurodegeneration: A key transcriptional regulator with therapeutic implications.

c-Jun, a core component of the activating protein-1 (AP-1) transcription factor complex, regulates cellular processes including proliferation, differentiation, survival, apoptosis, and oncogenesis. c-Jun functions by dimerizing to bind DNA and modulates the expression of genes such as Bcl-2, cyclin D1, and pro-inflammatory cytokines, enabling context-dependent transcriptional control. Its role in neurodegenerative diseases has gained attention due to its regulation of oxidative stress, inflammation, and apoptosis. In Parkinson's disease, Alzheimer's disease, and Huntington's disease, dysregula

Circadian disruption as a driver and target in neurodegenerative diseases: from molecular mechanisms to chronotherapeutic strategies.

The Circadian System is a complex network of coordinated clocks that regulates the organism’s internal clock in synchronisation with the outside world. These rhythms are controlled by genetically controlled positive and negative transcriptional-translational feedback loops (TTFL) that generate 24-hour oscillations in the protein level and mRNA of core circadian components. Circadian disruption is recognised as a significant contributor to the molecular pathogenesis of neurodegenerative illnesses, as disease-specific alterations in clock gene expression and melatoninergic signalling have been i

Molecular insights into SEN177 binding to human glutaminyl cyclase: a combined MD and DFT study.

Aberrant upregulation of human glutaminyl cyclase (hQC) is associated with the onset and progression of neurodegenerative disorders, notably Alzheimer's disease and Huntington's disease. Consequently, inhibition of hQC activity represents a potential avenue for treatment. SEN177, a potent small-molecule hQC inhibitor, has demonstrated notable preclinical efficacy but its binding mechanism remains incompletely understood. Herein, an integrated computational investigation was performed to elucidate the molecular basis of SEN177-hQC recognition. Notably, we implemented an integrated MD-energy dec

Axonopathy: mechanisms and potential therapeutic targets for neurodegenerative diseases.

Axons are unique structural and functional features of nerve cells, which play a critical role in regulating neuronal homeostasis. Dysfunction and degeneration of axons (axonopathy) has been established as an early and prominent contributing mechanism to the pathogenesis of neurodegenerative diseases including Alzheimer's disease, Parkinson's disease, Huntington's disease, and amyotrophic lateral sclerosis. In this review, we briefly summarize the structure and function of axons, and highlight recent advances in the understanding of the role of axons in health and disease. We argue that axons

The selective dynamics of interruptions at short tandem repeats.

Short tandem repeats (STRs) are hotspots of genomic instability that mutate at rates orders of magnitude greater than nonrepetitive loci due to frequent replication slippage. Expansions at some STR loci cause Mendelian diseases, while variation at other noncoding loci may affect complex traits, possibly by altering transcription factor occupancy of nearby binding sites. Accordingly, some STRs are inferred to be under purifying selection, regardless of their instability. One or more "interruptions", or bases that disrupt the locus's canonical repeat, significantly decrease an STR's mutability.

Aberrant expression of the MID1 protein in neurons of Huntington's disease brain.

Huntington's disease (HD) is caused by a CAG repeat expansion mutation in the Huntingtin (HTT) gene that transcribes into mRNA and translates into a polyglutamine tract. The mutant HTT gene products drive pathological changes that result in neurodegeneration. The mutant CAG repeat RNA contributes to cellular dysfunction by aberrantly recruiting RNA-binding proteins. For example, the mutant HTT transcript associates with a protein complex containing the MID1 protein. This aberrant recruitment of the MID1 protein complex results in an increased translation of mutant HTT. MID1 expression is abnor

Neuroprotective Effects of Transplanted Induced Pluripotent Stem Cell-Derived Neural Precursors in Huntington's Disease Models.

Huntington's disease (HD) is characterized by progressive striatal degeneration associated with mutant huntingtin (mHTT)-related proteostatic disruption and chronic neuroinflammation. Although mHTT-lowering approaches hold therapeutic promise, their capacity to restore the degenerating neural microenvironment remains limited. Here, we evaluated the therapeutic potential of human induced pluripotent stem cell (iPSC)-derived neural precursor cells (s513-NPCs) in two complementary HD models, the acute R6/2 transgenic fragment model and the protracted, full-length YAC128 genomic model. Fidelity of DNA ligase I is sensitive to physiological Mg2+ level.

The pool of free intracellular Mg2+ varies among tissues with the highest concentration measured in muscle tissue and the lowest measured in immune cells and in the brain. Here we investigate the impact of free Mg2+ on the fidelity of human DNA ligase I (LIG1). LIG1 is the major DNA ligase and is required to complete DNA replication, recombination and repair pathways. Biallelic hypomorphic variants of LIG1 cause immunodeficiency-96. We employed steady-state kinetics to compare fidelity of LIG1 towards a damaged nucleobase at the 3'- hydroxyl side of a nicked DNA substrate. The fidelity for dis

The experiences of being a young carer to a parent with Huntington's disease: a thematic synthesis.

PURPOSE: This thematic synthesis explored the experiences of being a young carer to a parent with Huntington's disease. METHODS: A systematic search strategy was utilised across eight databases, resulting in 13 papers that met the inclusion criteria. Thomas and Harden's thematic synthesis approach was used to guide the methodological decisions and analyse the data. RESULTS: The analysis resulted in three themes (1) Lack of awareness, undervalued and unsupported: barriers to young carers accessing support (2) Adult responsibilities: impact on developmental stages and worries about the future,

m6A RNA methylation in neural plasticity, brain aging, and neurodegenerative vulnerability.

m6A is a pervasive post-transcriptional RNA modification that regulates RNA splicing, stability, localization, and translation in the brain. In this review, we outline the core m6A regulatory machinery and summarize its spatial organization across neurons and glial cells, highlighting established roles in brain development, synapse formation, and axon growth. We then focus on experience-dependent plasticity, synthesizing evidence that neuronal activity and environmental inputs dynamically reshape m6A to regulate immediate-early transcription and local translation at synapses across sensory, co

Neuroinflammation, Autophagy, and Neurodegeneration: Mechanisms and Therapeutic Insights.

Neuroinflammation and autophagy dysregulation are critical in the pathogenesis of neurodegenerative diseases like Alzheimer's, Parkinson's, and Huntington's disease. Neuroinflammation occurs after a sustained immune response, which transitions into a chronic pathological state, leading to the sustained generation of pro-inflammatory cytokines and oxidative stress, causing neuronal damage. Meanwhile, defective autophagy exacerbates disease by promoting protein accumulation, e.g., amyloid-β, tau, and α-synuclein, thereby enhancing neuroinflammation. In this review, we focus on critical pathways,

Long non-coding RNAs in neurodegenerative diseases - Molecular mechanisms, liquid biopsy biomarkers, and therapeutic targets: A review.

Neurodegenerative diseases (NDDs), such as Alzheimer's disease (AD), Parkinson's disease (PD), amyotrophic lateral sclerosis (ALS), and Huntington's disease (HD), are age-related disorders characterized by progressive neuronal loss, cognitive decline, and limited options for disease-modifying treatments. Increasing evidence suggests that long non-coding RNAs (lncRNAs) play significant roles in neurodevelopment, neuronal homeostasis, and disease progression; however, their involvement in shared pathogenic pathways and clinical applications remains inadequately defined. This review consolidates

ATF3-dependent formation of inclusion bodies in polyQ-expressing human iPSC-derived neurons confers cellular protection.

Huntington's disease (HD) is an incurable, neurodegenerative disorder. While the causative mutation - CAG expansions within the coding region of the Huntingtin (HTT) gene - has been identified over 30 years ago, the pathological mechanisms underlying HD are still not clear. The abnormal CAG track encodes a polyglutamine (polyQ) expanded protein, which leads to HTT protein misfolding. These polyQ aggregates can form insoluble inclusion bodies (IBs); however, whether IBs are protective or detrimental remains debatable. Here we developed fluorescent iPSC-based human neuronal models for polyQ-rela
