Multimodal brain network topology and enhanced computer-aided diagnosis in Parkinson's Disease: a systematic review and meta-analysis.

Parkinson's disease (PD) is increasingly recognized as a brain network-disconnection syndrome. However, there is little consistent evidence on multimodal global topological alterations and their diagnostic value. We systematically searched PubMed, Embase and Web of Science up to March 2025 for articles reporting brain network topology in PD, to which we applied a multilevel random-effects meta-analyses with robust variance estimation to account for statistical dependencies. Our case-control meta-analysis included 80 studies (42 fMRI, 25 dMRI, 10 EEG, 4 sMRI, 3 others) involving 3736 PD patient

Impact of Mobility Loss on Independence and Psychosocial Wellbeing in People With Parkinson's Disease and Carers: A Meta-Ethnography.

BackgroundMobility impairments in Parkinson's disease (PD) significantly impact individuals' physical health, independence, and psychosocial wellbeing. In this review, mobility loss is used to refer to altered or impaired mobility (eg, reduced gait, balance difficulties, freezing of gait), rather than a complete loss of mobility. These changes affect daily functioning and quality of life. Carers also experience significant burden as they manage mobility issues. Understanding the implications of mobility loss in PD is essential for improving support for both people with PD and carers. This revi

Agonism of FFA4/GPR120 activates tyrosine hydroxylase and confers neuroprotection from 6-OHDA-induced cytotoxicity in PC12 cells and in a rat 6-OHDA model of Parkinson's disease.

Free-fatty acid receptor 4 (FFA4/GPR120), a G-protein-coupled receptor activated by medium to long-chain free-fatty acids (FFA) including ω-3 polyunsaturated fatty acids, regulates many metabolic and anti-inflammatory processes, and has also been linked to diverse neurophysiological functions. Here, we investigated the expression and functional role of FFA4 in dopaminergic systems using PC12 cells, rat striatal tissue, and an in vivo 6-hydroxydopamine (6-OHDA) model of Parkinson's disease (PD). FFA4 mRNA and protein were expressed in rat caudate-putamen and colocalized with tyrosine hydroxylas

Plants vs. GPCRs: A workflow for ligand discovery from an extract library illustrated by dopamine receptors.

Plants have served as source of medicines for millennia, yielding structurally diverse and biologically active small molecules with high therapeutic potential. Many plant natural products modulate the most druggable target class, G protein-coupled receptors (GPCRs). Traditionally, the discovery of these plant natural products relies on ethnopharmacological guidance and trial-and-error based medicinal use. However, the vast majority of plants remain unexplored for their pharmacological and therapeutic potential. This suggests that an unbiased approach based on screening plant extracts at GPCRs

Propensity-matched multicenter comparison of Parkinson's disease outcomes with and without deep brain stimulation.

Subthalamic deep brain stimulation is established for Parkinson's disease, reducing funding for randomized trials and necessitating complementary approaches to assess outcomes in broader clinical contexts. Using publicly available multicenter data, we compared propensity-matched patients with and without stimulation. The intervention was associated with improved mid-term patient-reported motor and non-motor experiences of daily living, while ongoing follow-up will clarify long-term effects. This demonstrates feasibility of matched comparisons in open observational cohorts.

Excessive daytime sleepiness in parkinsonism is associated with cortical and basal ganglia beta oscillatory activity.

Excessive daytime sleepiness, i.e., the inability to sustain wakefulness during the day, is a common Parkinson's disease (PD) non-motor symptom. Elevated cortico-basal ganglia beta oscillatory power (8-35 Hz) is a pathophysiological signature of PD motor signs. The relationship between beta oscillatory power and daytime sleepiness, however, remains unclear. Here, we examined the relationship between beta power and daytime vigilance in parkinsonian nonhuman primates. We administered dopaminergic medication to alter cortico-basal ganglia beta power and measured its effect on daytime vigilance. T

Research Prospects for mTOR in Neuroinflammation: A Bibliometric Analysis.

INTRODUCTION: Neuroinflammation is a key pathological process in neurological disorders, from acute injuries to chronic diseases such as Alzheimer's Disease (AD), Parkinson's Disease (PD), and stroke. The mTOR pathway regulates neuroinflammation by influencing microglial activation, autophagy, and synaptic plasticity. To address the scarcity of systematic analyses in this growing field, our study employs bibliometric analysis to map the landscape, identify key themes and collaborations, and highlight therapeutic targets for future neurological research. METHODS: A Web of Science search retrie

Video-based machine learning models for predicting deep brain stimulation outcomes in Parkinson's disease patients.

Current levodopa challenge test (LCT) for deep brain stimulation (DBS) candidate screening in Parkinson's disease (PD) relies on subjective clinical scales, limiting its predictive capacity for postoperative motor outcomes. We developed video-based machine learning models using quantified kinematic metrics during preoperative LCT in seventy PD patients who underwent DBS surgery. Objective multi-domain motor features were extracted via validated motor assessment software. Binary classification defined patients' outcomes as DBS+ (≥30% improvement in MDS-UPDRS Part III) or DBS- (<30%). Ternary cl

The Impact of Dance-Based Physical Activity on Sensorimotor and Psychological Function in Parkinson's Disease: A Narrative Review.

Background and Objectives: Parkinson's disease (PD) is a progressive neurodegenerative disorder characterized by a wide range of motor and non-motor symptoms that significantly compromise daily functionality, psychophysical wellbeing, and quality of life. Currently, a number of pharmacological and surgical treatments can reduce the clinical severity of motor impairments, but they are limited or poorly tolerated for non-motor symptoms, thus highlighting the need for non-medical complementary approaches. In this context, dance-based interventions have emerged as promising and enjoyable integrati

Genetic Engineering and Encapsulation Strategies for Lacticaseibacillus rhamnosus Enhanced Functionalities and Delivery: Recent Advances and Future Approaches.

This review addresses the recent advances made through various genetic engineering techniques to improve the properties of Lacticaseibacillus rhamnosus, not only for industrial applications, but also for the health-related benefits. However, due to the strict regulations on microorganisms intended for human consumption, concerning the insufficient characterization degree of the newly isolated strains and the lack of data regarding the safety of the genetically modified (GM) variants, the feasibility of bringing such L. rhamnosus strains to the market and their safety prospects were evaluated.

Elevated FKBP5 expression associates with epilepsy-related molecular changes and promotes neuronal hyperexcitability.

OBJECTIVE: Epilepsy is one of the neurological disorders, characterized by recurrent, spontaneous seizures arising from neuronal hyperexcitability and hypersynchrony in the brain. The mechanisms of epilepsy are intricate and remain elusive. FKBP5 has emerged as a significant protein implicated in neurological disorders such as Alzheimer's disease (AD) and Parkinson's disease (PD). This study aims to investigate the role of FKBP5 in a kainic acid (KA)-induced intrahippocampal epilepsy model and assessed how FKBP5 gain-of-function and FKBP51 inhibition influence neurotransmitter dynamics and neu

Metabolomic breath landscape analysis unravels lipid biomarker candidates in patients with genetic and idiopathic Parkinson's disease.

Parkinson's disease (PD) is the fastest growing neurodegenerative disorder. The current lack of efficient early diagnostic tools necessitates novel approaches to biomarker discovery. We propose an untargeted metabolomics approach using non-invasive exhaled breath analysis. Breath samples, collected from 73 PD patients, encompassing both genetic (LRRK2: n = 12, GBA1: n = 35, PRKN: n = 6) and idiopathic PD (n = 20), 4 unaffected LRRK2 pathogenic variant carriers, and 90 controls underwent extreme-resolution FT-ICR-MS analysis. Findings were compared with metabolomics data from blood plasma. Bios

Subthalamic nucleus in patients with Parkinson's disease encodes changes and magnitude of applied force.

The subthalamic nucleus (STN), a primary target for Parkinson's disease (PD) neuromodulation therapies, contributes to movement dynamics but it is unknown how STN relates to specific kinematic parameters. We investigated the neuronal activity in the dorsolateral STN during a visually cued isometric grip-force task in the contralateral hand in patients with PD who were undergoing deep brain stimulation surgery. Patients applied various levels of grip force during a task that separated a sustained applied force vs periods of changing force (yank) while we recorded single unit activity with a mic

Microengineering of the capillary interface of midbrain dopaminergic neurons to study Parkinson's disease vascular alterations.

Parkinson's Disease (PD) involves not only α-synuclein pathology in dopaminergic neurons but also vascular impairments that remain underexplored due to limitations of traditional in vitro models. Here we present a microengineered 3D neurovascular midbrain model that reconstructs the capillary interface of substantia nigra dopaminergic neurons. In our proof-of-concept demonstration, we successfully recapitulated neuronal pathology in PD, including α-synuclein aggregation, inflammatory responses, and progressive neuronal degeneration, by exposing our model to specially generated PD-associated α-

Evaluating the interactions of L-DOPA and droxidopa toward D3 dopamine receptor through computational study: A comparison with dopamine.

The D3 dopamine receptor (D3R), a member of the G protein-coupled receptor family, plays a critical role in neurophysiological processes and has emerged as a potential target for treating neurological disorders such as Parkinson's disease. In this study, we employed computational approaches to investigate the molecular interactions of D3R with dopamine and two key dopamine precursors: L-DOPA and droxidopa. Molecular docking was conducted to evaluate binding affinities and identify key receptor-ligand interactions, followed by molecular dynamics simulations to assess the stability and dynamic b
