PURPOSE: Microglia-mediated neuroinflammation is a shared feature across Alzheimer's disease (AD), Parkinson's disease (PD), and diabetic cognitive impairment (DCI), but the convergent molecular mechanisms remain unclear. This study investigated whether protein S-palmitoylation serves as a common regulator of microglial neuroinflammation across distinct neurodegenerative diseases etiologies and aimed to identify the key palmitoyltransferase involved.
METHODS: In vivo ND models (APP/PS1 for AD, MPTP for PD, STZ/high-fat diet for DCI) and in vitro BV2 microglial models stimulated with amyloid-β

AIM: Abdominal tics are a less commonly described axial or tonic tic phenotype in children and adolescents with Tourette syndrome and chronic tic disorders. Their abdominal location and symptom language may create diagnostic uncertainty with gastrointestinal, epileptic, myoclonic, respiratory or functional presentations.
METHODS: This clinically focused narrative synthesis identified relevant literature through targeted searches of PubMed/MEDLINE, Scopus and Web of Science from database inception through March 2026, supplemented by a targeted PubMed/MEDLINE and citation-chasing update during

INTRODUCTION: Lewy body dementia (LBD), encompassing dementia with Lewy bodies (DLB) and Parkinson's disease dementia (PDD), is progressive and heterogeneous in clinical presentation and underlying pathobiology. LBD substantially impacts patient function, independence, and quality of life. Current therapies are symptomatic, and disease-modifying therapies are not established. Advances in in vivo biomarkers provide an important window for early disease diagnosis and intervention.
AREAS COVERED: This review examines the complex spectrum of LBD, including DLB, PDD, and prodromal states of MCI-LB

The development and evaluation of novel treatment options for freezing of gait (FOG) in Parkinson's disease have been limited by an ambiguous definition and lack of standardized clinical outcome assessments (COAs) that 1) reliably assess FOG severity, 2) are responsive to change, and 3) capture patient-relevant impact. To address these gaps, working groups from the International Consortium on FOG (ICFOG) developed a clinician-rated outcome-the Giladi Protocol for assessing FOG severity (GP-FOG), and a patient-reported outcome-the FOG Quality of Life questionnaire (FOG-QOL). The FOG-COA project

OBJECTIVE: Previous studies using finger-tapping tasks have shown that patients with Parkinson's disease (PD) exhibit a deficit of temporal processing linked to gait disturbance. In this study, we aimed to investigate abnormalities in lower-limb rhythmic control and their association with freezing of gait in PD using a bilateral foot-tapping task.
METHODS: Fifteen patients with PD and 25 healthy controls performed foot-tapping tasks synchronized with auditory tones at fixed intervals (500-2,400 ms). The tasks included unilateral and bilateral foot-tapping, with bilateral movements performed e

BACKGROUND AND OBJECTIVE: Parkinson's disease is a progressive neurodegenerative disorder associated with balance impairment, gait dysfunction, falls, disability, and progressive loss of functional independence. Healthy longevity emphasises the preservation of functional ability and well-being rather than survival alone. Virtual reality-based rehabilitation provides an interactive, technology-assisted approach to balance and gait training, but comparative evidence addressing multiple functional domains simultaneously remains limited. This study aims to compare the effectiveness of virtual real

Neurological disorders arise from interconnected pathological processes, including oxidative stress, neuroinflammation, mitochondrial dysfunction, impaired proteostasis, synaptic injury and blood brain barrier disruption. Increasing evidence indicates that glial cells are not passive bystanders in these diseases, but central regulators of neuroimmune balance, metabolic support, myelin integrity and tissue repair. Polygonatum spp., a traditional medicinal and edible plant, contains diverse bioactive constituents, including polysaccharides, steroidal saponins and flavonoids, which exhibit multi-

Gaucher disease and Parkinson's disease are characterized by β-glucocerebrosidase (GCase) dysfunction and increased oxidative stress. Here, we report two multifunctional hybrids that combine an iminosugar-based pharmacological chaperone scaffold with curcumin- or ferulic acid-derived antioxidant motifs. The curcumin-iminosugar hybrid 2 exhibits pH-dependent GCase modulation, enhanced GCase activity under lysosomal conditions, enhanced enzyme activity in L444P Gaucher and wild-type fibroblasts, and reduced intracellular reactive oxygen species (ROS) levels. By integrating GCase enhancement and

Three novel fluorinated benzoylthiourea derivatives, namely FBTU1-FBTU3, were successfully synthesized in 79-86% yields and fully characterized using spectroscopic techniques including FT-IR, 1H, 13C and 19F NMR, UV-Vis spectroscopy, elemental analysis, and single-crystal X-ray diffraction (SCXRD). All three compounds crystallize in the monoclinic system (P21/c for FBTU1 and FBTU3; P21/n for FBTU2) with Z = 4. The crystallographic analysis revealed well-defined molecular geometries stabilized by intramolecular and intermolecular interactions. Hirshfeld surface and energy framework analyses ind

INTRODUCTION: Although there have been significant advances in the genetic etiology of Parkinson's disease (PD), most of this research has been conducted in European populations. This study aimed to characterize the genetic landscape of PD in South Africa, whose populations are renowned for their extensive genetic diversity and admixture.
METHODS: We recruited 689 PD probands across South Africa. These participants were genotyped using the NeuroBooster array in collaboration with the Global Parkinson's Genetics Program (GP2). An in-house computational workflow was used to identify pathogenic

INTRODUCTION: Deep brain stimulation (DBS) is an established intervention for Parkinson's disease (PD), whereas the network-level effects of prescribed periodic visual input implemented with 10 Hz base pulses remain uncertain.
METHODS: We developed a conductance-based cortico-basal ganglia-thalamic (CBT) model comprising 11 Hodgkin-Huxley neuronal populations and a prescribed periodic visual-input unit (RET) to compare 130 Hz subthalamic nucleus (STN) DBS with the archived periodic visual-input condition on the same calibrated PD network. Primary conditions used N = 10 independent runs each.

Parkinson's disease (PD) is characterized by nigrostriatal dopaminergic (DAergic) degeneration and motor dysfunction. Many PD patients also show circadian rhythm disturbances. Increasing evidence links circadian clock dysfunction to PD pathology. REV-ERBα is a circadian nuclear receptor that regulates DAergic genes under physiological conditions. However, its role in nigrostriatal dopamine (DA) homeostasis and motor function in PD remains unclear. In this study, we found elevated Rev-erbα mRNA expression in tyrosine hydroxylase (TH)-positive neurons of the substantia nigra (SN) in a 6-hydroxyd

BACKGROUND: Neuropsychiatric symptoms are common in Parkinson's disease (PD), yet biomarkers associated with their long-term progression remain poorly characterized. Insulin-like growth factor binding protein-3 (IGFBP-3) is involved in neurotrophic and metabolic processes. We examined whether baseline IGFBP-3 was associated with subsequent anxiety and depressive symptom trajectories in patients with early PD.
METHODS: This longitudinal cohort included 352 patients with early-stage PD followed annually for up to 7 years. Anxiety and depressive symptoms were assessed using the State-Trait Anxie

BACKGROUND: Brain shift during deep brain stimulation (DBS) surgery may contribute to changes in lead position. Although intracranial air volume is considered an important factor, other intraoperative factors may also influence lead displacement. This study evaluated interval changes in subthalamic nucleus (STN)-DBS lead-tip position between immediate and delayed postoperative computed tomography (CT) and investigated factors associated with these changes, including intracranial air volume and intraoperative head position.
METHODS: We retrospectively analyzed 42 DBS leads implanted bilaterall

INTRODUCTION: Turning is commonly impaired in Parkinson's disease (PD), but whether distinct sensor-derived turning measures support earlier-stage discrimination and severity assessment remains unclear. We examined their complementary clinical value.
METHODS: Participants were 79 healthy controls, 51 with earlier-stage PD (Hoehn and Yahr [H&Y] stages 1-2), and 62 with moderate-stage PD (H&Y stage 3). Thirteen sensor-derived motor features were examined using prespecified group comparisons, associations with MDS-UPDRS Part III, and age- and sex-adjusted regression. Random forest classifiers us

Population ageing may amplify cold-related health risks under climate change, particularly for older adults with Parkinson's disease (PD). We developed a climate-ageing projection framework to estimate the burden of cold spell-attributable PD mortality in China by integrating national mortality data, climate projections, and demographic forecasts. We analyzed 59,384 PD deaths among adults aged 60 years and older in China mainland during 2013-2020 using a time-stratified case-crossover design. Cold spells were associated with an increase in mortality, with effects concentrated within lags 0-4;

Adolescence is a critical period for the development of neural circuits, and exposure to stressors during this stage constitutes a risk factor for psychiatric disorders. Previous studies have shown that adolescent stress in rodents leads to behavioral alterations and functional loss of parvalbumin-containing GABAergic (PV+) interneurons and their perineuronal nets (PNNs) in the medial prefrontal cortex (mPFC) and ventral hippocampus (vHip). Although less explored, somatostatin-expressing (SST+) interneurons may also be impaired. Given the reported anti-stress effects of cannabidiol (CBD) in ad

BACKGROUND: Predicting disease progression in Parkinson's disease (PD) is challenging, especially for moderate-stage patients already on dopaminergic therapy. Most models focus on early, untreated PD, but progression rates may differ in moderate-stage PD. Better understanding of these trajectories can improve clinical trial design, access to care, and value demonstration for new therapies that target this population.
METHODS: We conducted a targeted literature review of longitudinal human PD studies in PubMed (inception-December 9, 2024) that quantitatively modeled disease progression, extrac
