An in silico cortico-basal ganglia-thalamic testbed for comparing visual and deep brain stimulation in Parkinson's disease.

INTRODUCTION: Deep brain stimulation (DBS) is an established intervention for Parkinson's disease (PD), whereas the network-level effects of prescribed periodic visual input implemented with 10 Hz base pulses remain uncertain. METHODS: We developed a conductance-based cortico-basal ganglia-thalamic (CBT) model comprising 11 Hodgkin-Huxley neuronal populations and a prescribed periodic visual-input unit (RET) to compare 130 Hz subthalamic nucleus (STN) DBS with the archived periodic visual-input condition on the same calibrated PD network. Primary conditions used N = 10 independent runs each.

Pharmacological inhibition of REV-ERBα ameliorates motor deficits in the 6-OHDA-lesioned mouse model of Parkinson's disease.

Parkinson's disease (PD) is characterized by nigrostriatal dopaminergic (DAergic) degeneration and motor dysfunction. Many PD patients also show circadian rhythm disturbances. Increasing evidence links circadian clock dysfunction to PD pathology. REV-ERBα is a circadian nuclear receptor that regulates DAergic genes under physiological conditions. However, its role in nigrostriatal dopamine (DA) homeostasis and motor function in PD remains unclear. In this study, we found elevated Rev-erbα mRNA expression in tyrosine hydroxylase (TH)-positive neurons of the substantia nigra (SN) in a 6-hydroxyd

Gramicidin A attenuates oxidative stress, mitochondrial dysfunction, DNA damage, and inflammation in 6-OHDA-induced SH-SY5Y cells.

BACKGROUND: Oxidative stress and mitochondrial dysfunction are major contributors to neurodegenerative diseases, and this study investigated the protective potential of Gramicidin A in a 6-hydroxydopamine (6-OHDA)-induced SH-SY5Y cellular injury model relevant to Parkinson's disease (PD). METHODS AND RESULTS: Following the 3-(4,5-Dimethyl-2-thiazolyl)- 2,5-diphenyl-2H-tetrazolium bromide (MTT) assay to determine the appropriate Gramicidin A concentration, cells were evaluated for oxidative stress parameters, including reactive oxygen species (ROS), total antioxidant status (TAS), total oxidan

Association between IGFBP-3 and longitudinal neuropsychiatric symptom trajectories in early Parkinson's disease: a longitudinal cohort study.

BACKGROUND: Neuropsychiatric symptoms are common in Parkinson's disease (PD), yet biomarkers associated with their long-term progression remain poorly characterized. Insulin-like growth factor binding protein-3 (IGFBP-3) is involved in neurotrophic and metabolic processes. We examined whether baseline IGFBP-3 was associated with subsequent anxiety and depressive symptom trajectories in patients with early PD. METHODS: This longitudinal cohort included 352 patients with early-stage PD followed annually for up to 7 years. Anxiety and depressive symptoms were assessed using the State-Trait Anxie

Predictive factors associated with interval changes in STN-DBS lead-tip position between immediate and delayed postoperative CT.

BACKGROUND: Brain shift during deep brain stimulation (DBS) surgery may contribute to changes in lead position. Although intracranial air volume is considered an important factor, other intraoperative factors may also influence lead displacement. This study evaluated interval changes in subthalamic nucleus (STN)-DBS lead-tip position between immediate and delayed postoperative computed tomography (CT) and investigated factors associated with these changes, including intracranial air volume and intraoperative head position. METHODS: We retrospectively analyzed 42 DBS leads implanted bilaterall

Sensor-derived turning measures for differentiating earlier-stage Parkinson's disease and characterizing motor severity.

INTRODUCTION: Turning is commonly impaired in Parkinson's disease (PD), but whether distinct sensor-derived turning measures support earlier-stage discrimination and severity assessment remains unclear. We examined their complementary clinical value. METHODS: Participants were 79 healthy controls, 51 with earlier-stage PD (Hoehn and Yahr [H&Y] stages 1-2), and 62 with moderate-stage PD (H&Y stage 3). Thirteen sensor-derived motor features were examined using prespecified group comparisons, associations with MDS-UPDRS Part III, and age- and sex-adjusted regression. Random forest classifiers us

Projected burden of cold spell-attributable Parkinson's disease mortality among older adults in China under climate change and population ageing.

Population ageing may amplify cold-related health risks under climate change, particularly for older adults with Parkinson's disease (PD). We developed a climate-ageing projection framework to estimate the burden of cold spell-attributable PD mortality in China by integrating national mortality data, climate projections, and demographic forecasts. We analyzed 59,384 PD deaths among adults aged 60 years and older in China mainland during 2013-2020 using a time-stratified case-crossover design. Cold spells were associated with an increase in mortality, with effects concentrated within lags 0-4;

Cannabidiol attenuates behavioral impairments and deficits in parvalbumin interneurons and their perineuronal nets induced by adolescent stress in adult mice.

Adolescence is a critical period for the development of neural circuits, and exposure to stressors during this stage constitutes a risk factor for psychiatric disorders. Previous studies have shown that adolescent stress in rodents leads to behavioral alterations and functional loss of parvalbumin-containing GABAergic (PV+) interneurons and their perineuronal nets (PNNs) in the medial prefrontal cortex (mPFC) and ventral hippocampus (vHip). Although less explored, somatostatin-expressing (SST+) interneurons may also be impaired. Given the reported anti-stress effects of cannabidiol (CBD) in ad

Modeling disease progression among moderate-stage Parkinson's disease patients: A targeted literature review.

BACKGROUND: Predicting disease progression in Parkinson's disease (PD) is challenging, especially for moderate-stage patients already on dopaminergic therapy. Most models focus on early, untreated PD, but progression rates may differ in moderate-stage PD. Better understanding of these trajectories can improve clinical trial design, access to care, and value demonstration for new therapies that target this population. METHODS: We conducted a targeted literature review of longitudinal human PD studies in PubMed (inception-December 9, 2024) that quantitatively modeled disease progression, extrac

Evaluation of Hydrogen Peroxide as an Environmental Model of Parkinson's Disease in Drosophila Larvae.

Parkinson's disease (PD) is characterized by motor dysfunction and dopaminergic neuron loss and is influenced by genetic and environmental factors. Here, we evaluated hydrogen peroxide (H₂O₂) as an accessible oxidative stress model of PD in Drosophila larvae. High H₂O₂ concentrations caused lethality or severe developmental impairment, whereas animals exposed to 0.005-0.025% H₂O₂ progressed through larval development and to pupation. Pupation timing was similar to untreated controls at 0.005% and 0.01% H₂O₂ but was delayed by 0.025% H₂O₂. Low-dose H₂O₂ reduced crawling speed and impaired navig

Chorea-Acanthocytosis Without Chorea: VPS13A Disease Manifesting as Parkinsonism.

Pathogenic variants in VPS13A cause chorea-acanthocytosis, classically presenting as chorea, dystonia, and orolingual dyskinesias. Parkinsonism is an underreported presenting symptom, particularly in the absence of these classical features, and may lead to misdiagnosis. We report the case of a 52-year-old man evaluated for heart transplantation, during which the neurology service was consulted for recently diagnosed Parkinson's disease to inform transplant candidacy. His history revealed progressive gait difficulty, falls, slurred speech, nonresponse to levodopa, and absence of resting tremor

Integrating graph attention networks and deep learning with multi-stage computational screening: Bisdemethoxycurcumin as a potent A₂A receptor inhibitor for Parkinson's disease.

Parkinson's disease (PD) remains a progressive neurodegenerative disorder with limited disease modifying therapies, as current treatments such as L-DOPA offer only symptomatic relief and are associated with severe adverse effects. This study presents an integrated multi-stage computational pipeline that combines quantitative structure activity relationship (QSAR) modelling enhanced by graph attention networks (GATs), molecular docking, density functional theory (DFT), molecular dynamics (MD) simulations, and ADMET profiling to identify natural inhibitors targeting four key PD related proteins:

Mincle exacerbates neuroinflammation and dopaminergic neurodegeneration via synergistic crosstalk with TLR4.

Microglial activation and subsequent neuroinflammation are critically implicated in the pathogenesis and progression of Parkinson's disease (PD). Macrophage-inducible C-type lectin (Mincle) is a pivotal pattern recognition receptor that participates in the regulation of innate immune responses. Nevertheless, the functional role of Mincle in mediating dopaminergic neuronal injury in PD remains poorly defined. In the present study, we observed a marked upregulation of Mincle expression in microglia in PD mouse models. Pharmacological inhibition of Mincle via Anti-Mincle antibody treatment effect

A critical mechanistic synthesis of PINK1-mediated mitophagy signaling and its therapeutic targeting in Parkinson's disease neurodegeneration.

Parkinson's disease (PD) is a progressive neurodegenerative disorder that is associated with loss of dopaminergic neurons, mitochondrial dysfunction and disruption of cellular homeostasis. PTEN-induced kinase 1 (PINK1) plays a crucial role in mitochondrial quality control by regulating mitophagy, mitochondrial dynamics, and calcium signaling to maintain neuronal bioenergetic homeostasis. This review critically evaluates the molecular pathways underlying PINK1-mediated mitochondrial integrity and the mechanisms by which loss-of-function mutations contribute to the pathogenesis of PD. Drug-Specific Associations of Angiotensin Receptor Blockers With Parkinson's Disease Risk in a Multicenter Electronic Health Record Cohort.

BACKGROUND: Angiotensin receptor blockers (ARBs) have demonstrated neuroprotective effects in preclinical models of Parkinson's disease (PD), but individual ARBs differ in pharmacological properties that may influence their neuroprotective potential. OBJECTIVES: Using large-scale hospital electronic health records of the University of California health system, we tested whether specific ARBs are associated with lower PD risk. METHODS: We conducted a new-user, active-comparator cohort study in the University of California Health Data Warehouse, encompassing 890 712 hypertensive patients acros

Identification of platelet-derived biomarker in a cohort of Parkinson's disease.

Evidence has suggested that platelets (PLTs) share biochemical properties with neurons. However, whether PLTs might serve as biomarkers of Parkinson's disease (PD) remains uncertain. In this study, we first analyzed RNA data from a publicly available microarray of the substantia nigra (SN) and RNA-seq data from the PLTs of newly recruited PD patients using weighted gene coexpression network analysis (WGCNA). We found that in the SN, the majority of network modules were associated with chromatin organization, energy metabolism, and synaptic signaling, whereas those in the PLTs were related to c

Feedback Control of Arterial Pressure Using Deep Brain Stimulation: Transfer Function Analysis and Simulation Study.

BACKGROUND: Deep brain stimulation (DBS) is an established treatment for Parkinson disease. Emerging evidence suggests that DBS of the subthalamic nuclei may be a target for neuromodulation therapy for orthostatic hypotension due to efferent baroreflex failure. To develop a central algorithm for an artificial baroreflex system, we first estimated the dynamic responses of arterial pressure to DBS, and then determined the feedback correction factors for DBS. METHODS: A total of 14 Parkinson disease patients treated with DBS participated in submaximal stimulation screening tests for 20 s to iden

Mechanistic Evaluation of Psilocybin in Neurodegenerative Disorders: Relevance of 5-HT2A Receptors and Downstream Pathways.

More than fifty million individuals worldwide are experiencing neurodegenerative disorders, which include Parkinson's disease, Alzheimer's disease, amyotrophic lateral sclerosis, Huntington's disease, multiple sclerosis, and frontotemporal dementia. These disorders have a significant role in the gradual degradation of cognition and motor skills. Most current treatment approaches have little effect on changing the course of the disease and are largely symptomatic. Psilocybin, a naturally occurring tryptamine alkaloid from psilocybin mushrooms, has become a promising medicinal agent because of i
