The morphology of α-synuclein fibrils changes during formation, storage, and upon exposure to ligands.

Protein fibrils are pathological hallmarks of many neurodegenerative diseases, including Parkinson's disease. The preparation of α-synuclein (αSyn) fibrils in vitro is widely used in research relating to these conditions. However, αSyn fibrils exhibit substantial structural polymorphism. How fibril morphology evolves during formation, storage, or in the presence of small molecule ligands is poorly understood. Here, the evolution of αSyn fibril morphology was investigated using fluorescence assays, circular dichroism, transmission electron microscopy, and ligand profiling. Fibril morphology was

Interpreting peripheral immune phenotypes in Parkinson's disease: an immune-context framework for biomarker qualification.

Peripheral immune alterations are widely reported in Parkinson's disease (PD), but their biological meaning and clinical utility remain uncertain because circulating immune profiles are shaped by aging, latent infection, medication exposure, comorbidity, disease stage, and preanalytical conditions. This concept-driven narrative review examines how technically valid peripheral immune findings should be biologically interpreted and clinically qualified through an immune-context phenotyping framework. Available human evidence suggests alterations in neutrophil- and monocyte-related inflammatory i

Virulence mechanisms of Desulfovibrio bacteria: viability-dependent processes promote alpha-synuclein aggregation and toxicity in C. elegans.

OBJECTIVES: Desulfovibrio (DSV) species have been linked to Parkinson's disease (PD) and demonstrated to enhance alpha-synuclein (alpha-syn) aggregation, a pathological hallmark of PD, in a Caenorhabditis elegans model. However, the bacterial factors underlying this virulence remain unclear. We investigated whether DSV-induced alpha-syn aggregation and toxicity are more strongly driven by bacterial viability-dependent processes or by bacterial structures retained after paraformaldehyde (PFA) treatment. METHODS: Live or paraformaldehyde (PFA)-treated (non-viable) DSV cells, and curli-producing

An adaptive fMRI paradigm of direction-specific, time-critical grip-force transitions.

Rapid transitions between force states, including fast increases and rapid releases, are fundamental to voluntary movement and everyday object interaction. Converging evidence from movement disorders suggests that rapid force reduction can be disproportionately impaired relative to force production, motivating the hypothesis that these transition directions rely on partly distinct control mechanisms. Brain imaging studies suggest direction-sensitive neural contributions, but existing paradigms make it difficult to determine whether differences between force production and force reduction refle

Imaging nanoscopic co-assemblies of α-synuclein and Tau in human samples.

α-synuclein (αS), amyloid-β (Aβ), and Tau aggregates are hallmarks of many neurodegenerative diseases, yet the prevalence and nature of their nanoscopic co-assemblies remain poorly understood. Here, we adapt a single-molecule pull-down (SiMPull) assay to profile αS, Aβ, and Tau co-assemblies across post-mortem brain tissue homogenates, soluble brain fractions, human serum, cerebrospinal fluid, and saliva. We show that αS-Tau co-assemblies are abundant across all sample matrices, accounting for ~10-70% of total αS aggregates in both Parkinson's disease (PD) and control samples. Notably, αS-Tau

Dopamine amplifies subthalamic long-latency responses to cortical stimulation in patients with Parkinson's disease.

The cortico-subthalamic hyperdirect pathway provides a rapid route for cortical influence over basal ganglia processing and is a key target of deep brain stimulation (DBS) in Parkinson's disease (PD). However, to what degree dopamine modulates signal transmission along this pathway in humans remains unclear. Using bidirectional cortical and subthalamic electrical stimulation while invasively recording evoked responses in 15 PD patients undergoing DBS surgery, we examined cortico-subthalamic communication across dopaminergic states. Cortical stimulation evoked a short-latency subthalamic respon

Machine learning image analysis predicts Parkinson disease genotype and mitochondrial lysosomal abnormalities in iPS neurons.

Mitochondrial and lysosomal dysfunction are central features of Parkinson's disease (PD) across major genetic forms including PRKN, SNCA, and LRRK2. We applied cell morphomics, a machine-learning-based framework combining high-content imaging with quantitative feature extraction, to analyse mitochondrial and lysosomal morphology at single-cell resolution in iPS cell-derived cortical neurons from PD patients and healthy controls (13 lines total). Supervised machine-learning models distinguished PD neurons from controls with high accuracy (AUC = 0.87) and reliably separated individual genotypes.

Computerized fine motor assessment in Parkinson's Disease: Task-specific measures provide complementary clinical information.

BACKGROUND: Fine motor impairment is a common feature of Parkinson's disease (PD) and contributes substantially to functional disability. Although the Movement Disorder Society-Unified Parkinson's Disease Rating Scale (MDS-UPDRS Part III) remains the standard clinical tool for motor evaluation, quantitative computerized assessments may provide complementary task-specific information. OBJECTIVES: This cross-sectional case-control study compared fine motor performance measured using the Motorische Leistungs-Serie (MLS) between 19 patients with idiopathic PD and 20 healthy controls and examined

Cortical Cholinergic Pathway Degeneration Reflects Cognitive Impairment and Dementia in Parkinson's Disease.

BACKGROUND: Cognitive impairment is a prevalent and debilitating feature of Parkinson's disease (PD). Increasing evidence from postmortem and in vivo positron emission tomography imaging studies supports cholinergic dysfunction in PD. There is, however, little evidence regarding the influence of the structural integrity of cortical cholinergic pathways that emanate from the nucleus basalis of Meynert (NBM). OBJECTIVES: To investigate the association between cognition and the structural integrity of medial and lateral cortical cholinergic projection pathways in PD. METHODS: Neuropsychological

Targeting BCL11B-mediated JAK2/STAT3 activation: A novel therapeutic strategy for Parkinson's disease.

Parkinson's disease (PD) is driven in part by neuroinflammation. We investigated whether the transcription factor BCL11B contributes to dopaminergic neurodegeneration in this context. Transcriptome sequencing of MPTP-induced PD mice revealed BCL11B upregulation, which was confirmed in MPP+-treated MN9D cells, MPTP-lesioned mice, and dopaminergic neurons exposed to inflammatory conditioned medium. BCL11B knockdown attenuated dopaminergic neuron loss, reduced apoptosis, improved motor deficits, and partially blocked JAK2/STAT3 pathway activation in both inflammatory and MPTP-induced PD models. Neuromelanin-sensitive magnetic resonance imaging in the diagnosis and assessment of Parkinson's disease.

Parkinson's disease (PD) is a progressive neurodegenerative disorder characterized primarily by the progressive loss of dopaminergic neurons in the substantia nigra pars compacta (SNpc); noradrenergic neurons in the locus coeruleus (LC) are also frequently affected. Because clinical motor symptoms usually emerge only after substantial neurodegeneration has occurred, there is a compelling need for noninvasive imaging biomarkers capable of detecting early pathological changes. Neuromelanin-sensitive magnetic resonance imaging (NM-MRI) generates neuromelanin-related image contrast in the SNpc and

The Role of Astrocytes in the Inflammatory Response in Neurodiseases.

Astrocytes have emerged as central regulators of neuroinflammation, extending far beyond their traditional roles as structural and metabolic support cells. In neurological disorders, astrocytes undergo reactive remodeling that encompasses morphological, transcriptional, metabolic, and functional changes. Rather than fitting into a fixed A1/A2 dichotomy, reactive astrocytes exist along a disease-, region-, and stage-dependent continuum shaped by local pathology and intercellular crosstalk. This review summarizes how astrocytes contribute to neuroinflammatory processes in Alzheimer's disease, Pa

A Data-Driven Operational Definition of Square-Wave Jerks and Its Diagnostic Utility in Progressive Supranuclear Palsy.

BACKGROUND: The Movement Disorder Society diagnostic criteria for progressive supranuclear palsy (PSP) list frequent square-wave jerks (SWJ) as a suggestive oculomotor feature. A standardised operational definition of SWJ is however presently lacking. OBJECTIVE: To create a data-driven optimal SWJ definition and to assess its diagnostic utility in PSP. METHODS: We prospectively recruited PSP n = 25, Parkinson's disease (PD) n = 32, Alzheimer's disease (AD) n = 35, and controls n = 88. SWJ were recorded with a visual fixation task using video-oculography. Multiple candidate definitions of SWJ

Neurodegeneration epicenters: Distinct network mechanisms of dopaminergic treatment in Parkinson's disease.

Parkinson's disease exhibits heterogeneous spatiotemporal patterns of neurodegeneration. Dopaminergic therapy improves motor symptoms by modulating brain function in affected regions, yet whether the neural mechanisms through which dopaminergic therapy exerts its effects are linked to this heterogeneity of neurodegeneration remains unclear. A connectivity-based epicenter framework combined with spectral dynamic causal modeling was applied to resting-state functional magnetic resonance imaging data from 160 drug-naïve Parkinson's disease patients and 174 healthy controls to identify subtype-spe

Cerebrospinal fluid and dural accumulation of α-synuclein impairs CNS clearance in Parkinson's disease.

Parkinson's disease (PD) is characterized by the build-up of α-synuclein (α-syn) in the brain, where it drives neurodegeneration. Importantly, aggregated α-syn also accumulates in the cerebrospinal fluid (CSF) of PD patients, and while increasingly used for diagnosis, its pathological role remains poorly understood. Here, we demonstrate that α-syn accumulation in central nervous system (CNS) border tissues promotes CNS clearance deficits, as observed in PD patients. We find that aggregated α-syn accumulates not only in the CSF but also in the dura mater of individuals with PD, and, using acute
