Nanomaterial Design for Macrophage Reprogramming: Emerging Strategies and Mechanistic Insights for Inflammation and Tumor Therapy.

Macrophages play a paradoxical role in inflammation and tumor. They can protect the host during acute inflammation but act as the main driver for the chronic inflammation which is a common characteristic of several diseases including rheumatoid arthritis, atherosclerosis, type 2 diabetes, inflammatory bowel disease, and neurodegenerative diseases such as Alzheimer's disease and Parkinson's disease. In the tumor microenvironment, macrophages switch their phenotype to the immunosuppressive M2 state that promotes tumor growth, angiogenesis, and metastasis. Emerging nanomaterial platforms are tryi

The Pallidothalamic Tract in Functional Neurosurgery: A Systematic Review.

BACKGROUND: The pallidothalamic tract (PTT), formed by convergence of the ansa lenticularis and fasciculus lenticularis at Forel's field H1, is a major basal ganglia efferent pathway and an emerging functional neurosurgical target. No systematic review has previously synthesised outcomes across PTT-targeted interventions. METHODS: This PRISMA 2020-compliant systematic review was registered on PROSPERO (CRD420251248260). PubMed, OVID, and Web of Science were searched for studies reporting functional neurosurgical outcomes targeting the PTT or Forel's fields. Prospective and retrospective studi

Parkinson's disease mortality in the United States, 1999-2025: a national multiple-cause-of-death analysis with projections to 2030.

BACKGROUND: Parkinson's disease (PD) is the second most common neurodegenerative disorder in the United States and a growing driver of late-life disability and death. We characterized U.S. PD-related mortality from 1999 to 2025 and projected it to 2030. METHODS: In this serial cross-sectional analysis of CDC WONDER Multiple Cause of Death data, PD-related deaths in adults aged ≥45 years were identified using ICD-10 code G20 recorded anywhere on the death certificate. Age-adjusted mortality rates (AAMRs) per 100,000 were decomposed by joinpoint regression and projected to 2030 using a log-scal

A multimodal MRI-based study of early-stage Parkinson's disease across age-at-onset groups.

PURPOSE: This study aimed to investigate distinct iron deposition and neurite microstructural alterations in deep gray matter nuclei between early-onset Parkinson's disease (EOPD) and late-onset Parkinson's disease (LOPD) patients using quantitative susceptibility mapping (QSM) and neurite orientation dispersion and density imaging (NODDI), and to evaluate the ability of these quantitative metrics to distinguish EOPD and LOPD from their corresponding healthy controls. METHODS: Fifty-three early-stage PD patients (15 EOPD and 38 LOPD) and 58 healthy controls (17 young healthy controls and 41 o

An Interpretable Framework for Identifying EEG Spectral Biomarkers of Pathological Brain Aging.

Background: Normal aging involves gradual declines in brain function, whereas neurodegenerative disorders show accelerated deterioration. Electroencephalography (EEG) captures alterations in brain oscillatory activity, making it promising for detecting neurodegenerative disorders. However, existing approaches often emphasize classification performance over spectral biomarker identification, limiting clinical translation. Methods: We propose an EEG-based framework integrating explainable machine learning, analyses of inter-subject heterogeneity and temporal stability, and probabilistic modeling

Cuproptosis: New Mechanisms Linking Endothelial Dysfunction to Neurodegenerative Diseases and Therapeutic Prospects.

Cuproptosis is a copper-dependent form of regulated cell death driven by disrupted copper homeostasis and mitochondrial metabolic reprogramming. It is distinct from other cell death pathways and has been increasingly linked to vascular endothelial dysfunction and neurodegenerative diseases such as Alzheimer's and Parkinson's disease. This article comprehensively reviews the core molecular mechanisms of cuproptosis and its emerging roles in endothelial dysfunction and neurodegeneration, synthesizing recent findings from cellular, animal, and human studies. Cuproptosis exacerbates vascular patho

Neurodegenerative diseases in Sub-Saharan Africa: diagnostic invisibility, biological heterogeneity and a precision-neuroscience agenda.

Neurodegenerative diseases (NDs) are increasingly recognised in Sub-Saharan Africa (SSA), but the principal scientific challenge is no longer simply to document a presumed hidden burden. It is to determine how much of the observed heterogeneity reflects underlying neurobiology and how much arises from age structure, case ascertainment, culturally sensitive phenotyping and unequal access to neurological diagnosis. This critical narrative review synthesises evidence from 2000 to July 2026 across epidemiology, disease mechanisms, genomics, biomarkers, environmental exposures, therapeutics and hea

Impact of Ayurvedic purification (Shodhana) on Mucuna pruriens for primary health care in Parkinson's disease: phytochemical profile, bioavailability of L-DOPA, and comparative UPLC-MS/MS QTOF-Caco-2 analysis.

Parkinson's disease (PD) is an increasing global health challenge, and although L-DOPA remains the cornerstone of treatment, its long-term use in synthetic form is associated with complications such as dyskinesia and wearing-off. In Ayurveda, Mucuna pruriens (Kapikacchu/MP) is used in conditions similar to PD. However, the effects of classical purification methods and different dosage forms on their phytochemical composition and functional outcomes have not been well investigated. The present in vitro study evaluated the qualitative phytochemical profiling (untargeted) of traditionally Purifie

Screening human brain targets for Japanese encephalitis virus NS5 via yeast two-hybrid assay.

Japanese encephalitis (JE) is the most important vaccine-preventable acute meningoencephalitis in tropical and subtropical Asia with no known antiviral therapeutics. Available vaccines offer limited protection against emerging Japanese encephalitis virus (JEV) genotypes, posing a persistent public health threat. Disruption of the interactions between JEV non-structural protein 5 (NS5) protein and human brain cellular proteins could be explored to develop flavivirus druggable host targets. This study aimed to identify the human brain cellular proteins that interact with the JEV NS5 protein. A h

Frontiers in Neurotransmitter Probes: Innovations and Perspectives in Norepinephrine Detection Technology.

Norepinephrine (NE), the primary neurotransmitter of locus coeruleus projections, plays a crucial role in regulating arousal, emotion, memory, and stress responses. Its dysregulation is closely associated with various neurological disorders such as depression, Parkinson's disease, and epilepsy. However, conventional detection techniques such as microdialysis coupled with liquid chromatography are constrained by limited spatiotemporal resolution and inherent invasiveness, largely due to NE's low concentration, rapid metabolism, and complex distribution in vivo. These limitations make real-time,

What happens in vagus.

The parasympathetic nervous system is phylogenetically ancient. In humans, the vagus nerve is the brain's only pathway for bidirectional parasympathetic signaling with the thorax, abdomen, and upper pelvis. Unsurprisingly, the vagus nerve has played a significant role in scientific discovery and therapeutic applications, attracting the attention of Galen, Darwin, and Pavlov, among others. For example, Otto Loewi used the vagus nerve to gather data that confirmed Santiago Ramón y Cajal's Neuron Doctrine. This intensive focus has led to a number of clinical advances related to vagus nerve functi

Striatal network dysfunction underlying speech impairment in early Parkinson's disease.

The pathophysiology underlying Parkinson's disease (PD) speech impairments and its potential sex differences remain unclear. We investigated sex-specific alterations in speech and striato-cortical networks in 104 patients with early PD (68M/36F), and 44 healthy controls (23M/21F), using quantitative speech markers and multimodal neuroimaging. Speech features related to prosody, phonation, articulation, fluency, and rhythm, were extracted from voice recordings; striatal dopaminergic loss was quantified using DaT-SPECT, and functional connectivity between striatal and cortical language networks

Evaluating multivariable clinical prediction models for Parkinson's disease prognosis: A scoping review.

Multivariable clinical prediction models combine two or more patient predictor variables to calculate the probability of a clinical outcome. In persons with Parkinson's disease (PD), clinical prognostic models can be used to predict disease progression or the occurrence of a clinical event. These models can aid in PD prognostication and complement clinical judgment by informing medical decision-making and resource allocation. In this scoping review of the literature from inception through May 2025, we present data from 74 multivariable prognostic models in PD, organized by clinical outcome. We

Eye-movement-based machine learning for identifying Parkinson's disease dementia in a retrospective cohort: an interpretable model-development study.

BACKGROUND: Parkinson's disease dementia (PDD) is a major non-motor complication of Parkinson's disease (PD), but conventional cognitive screening is influenced by education, language, motor impairment, fatigue, and testing conditions. We developed and interpreted machine-learning models using eye-movement and demographic features to identify PDD among patients with PD. METHODS: In this retrospective single-center study, 486 patients were screened between December 2024 and February 2025. After exclusions, 297 patients with idiopathic PD were analyzed, including 84 without dementia and 213 wit

Digital mobility assessment using wearable devices in people with Parkinson's disease: a feasibility and implementation study in Ghana (TraPCAf) compared with a UK cohort.

INTRODUCTION: Life expectancy is rising in Africa, increasing Parkinson's disease (PD) prevalence. PD in Africa is often underdiagnosed/undertreated due to limited specialists, awareness and medication access. Gait impairments are among the earliest and most disabling PD symptoms, yet their presentation in African populations is poorly understood. This study aimed to (1) compare real-world mobility between PD and older controls in a population from Ghana (GHA-PD and GHA-CL), (2) examine associations between digital mobility outcomes (DMOs) and clinical/demographic factors between GHA-PD and a

Harnessing cGAS-STING in neurodegeneration: From mitochondrial DNA leakage and microglial reactivity to species-specific translational barriers.

The cyclic GMP-AMP synthase (cGAS)-stimulator of interferon genes (STING) pathway, a cornerstone of innate immunity, has emerged as a critical driver of sterile neuroinflammation in the aging brain. Age-related decline in mitochondrial quality control, coupled with nuclear and lysosomal erosion, fosters the aberrant accumulation of cytosolic self-DNA, establishing a permissive environment for chronic cGAS-STING activation. This review synthesizes recent breakthroughs elucidating how major neurodegenerative diseases-Alzheimer's disease, Parkinson's disease, amyotrophic lateral sclerosis, and Hu

Pathological significance of converting Zn2 +dysregulation to H2O2 in AMPA-induced neurotoxicity.

The causative mechanism of nigrostriatal dopaminergic degeneration, which is a characteristic in Parkinson's disease (PD), remains to be solved, while it has been widely accepted that excessive reactive oxygen species (ROS) are the key mediator of the PD pathogenesis. In contrast, pathological significance of Zn2+-mediated ROS generation is poorly understood in the PD pathogenesis. Because one pathway of ROS generation is mitochondrial dysfunction, we focused on mitochondrial Zn2+-mediated ROS generation and examined pathological role of H2O2 signaling, which derived from mitochondrial ROS, af
