β-sitosterol and next-generation neuroprotection for multi-target strategies and the gut-brain axis in neurodegenerative diseases.

Neurodegenerative disorders such as Alzheimer's and Parkinson's diseases arise from complex interactions among oxidative stress, neuroinflammation, metabolic dysfunction, and dysregulated signaling networks. This review aim of the synthesize mechanistic evidence on β-sitosterol as a multi-target phytochemical and clarify how its actions connect to gut-brain axis modulation in neurodegeneration. The integrated mechanistic framework linking β-sitosterol's effects on cholesterol homeostasis, neuroinflammation, mitochondrial function, cholinergic signaling, and microbiota-barrier integrity to cogn

Gasdermin E: Bridging Pyroptosis, Immunity, and Disease Pathogenesis Toward Precision Intervention.

Gasdermin E (GSDME), a pivotal executor of pyroptosis, has emerged as a central regulator of immune responses across a spectrum of diseases. Initially identified as a tumor suppressor and a deafness-associated gene (DFNA5), GSDME is now recognized for its complex, context-dependent roles in health and pathology. Its activation, primarily via caspase-3 cleavage, converts apoptotic signals into a lytic, pro-inflammatory form of cell death, releasing damage-associated molecular patterns and cytokines that profoundly shape the immune microenvironment. This review synthesizes the current research l

AMPK Therapy-A Little Goes A Long Way.

AMP-activated protein kinase (AMPK) is a highly conserved serine/threonine kinase that integrates energetic, nutrient, hormonal, redox, and stress signals to coordinate cellular and whole-body energy homeostasis. Although AMPK was initially characterized primarily as a sensor of changes in cellular AMP/ATP ratios, recent studies have revealed additional layers of regulation involving upstream kinases, post-translational modifications, heterotrimeric isoform composition, subcellular compartmentalization, and tissue-specific signaling. In this review, we provide an updated overview of the molecu

Phase Separation Drives Pathological Aggregation in Neurodegenerative Diseases: A 15-Year Bibliometric Landscape (2009-2024).

Liquid-liquid phase separation (LLPS), a biophysical driver of membraneless organelle assembly, is central to pathological aggregation in neurodegenerative diseases. Initially linked to amyotrophic lateral sclerosis (ALS), LLPS dysregulation has now been implicated in Alzheimer's, Parkinson's, and frontotemporal dementia, where aberrant transitions convert dynamic condensates into insoluble fibrils. To systematically map this landscape, we employed CiteSpace-based bibliometrics to analyze 784 Web of Science articles from 2009 to 2024. Our analyses reveal dominant contributions from the United

Molecular programs in human locus coeruleus link APOE and neuromelanin to Alzheimer's vulnerability.

The locus coeruleus (LC) is a small noradrenergic nucleus in the dorsal pons that sends widespread projections across the brain to regulate sleep, arousal, and cognition. LC neurons show early accumulation of phosphorylated tau (pTau) and are particularly vulnerable to degeneration in Alzheimer's disease (AD). AD is also associated with loss of neuromelanin (NM) pigmentation in LC neurons, and NM-sensitive neuroimaging predicts clinical severity and future disease progression. To better understand molecular vulnerability of the LC, we generated spatially resolved transcriptomics data on LC tis

Abortive transcripts in exosomes - A potential biomarker for Alzheimer's disease.

Diagnosis of Alzheimer's disease (AD) relies on invasive cerebrospinal fluid analysis or costly neuroimaging, underscoring the need for minimally invasive blood-based biomarkers for early detection. Exosomes, promising biomarker carriers, are unexplored for ultra-short RNA species like abortive transcripts (ATs). We investigated whether 8-nucleotide ATs are selectively packaged into exosomes to reflect brain transcriptional dysregulation in AD. Using a transgenic AD mouse model and Aβ-stimulated BV2 microglia coupled with Base-Stacking Hybridization Assisted Ligation (BSHAL), we detected signi

Irisin as an Exerkine of Neuroprotection in Aging and Alzheimer's Disease.

Alzheimer's disease (AD) is a neurodegenerative disease impacting over 6 million Americans, with cases projected to increase to over 14 million by 2060. The AD pathology leads to difficulty completing everyday tasks or conversations, and ultimately, progresses to disrupt the most basic bodily functions and require full-time caretaking. While disease-modifying therapy remains elusive, reducing the incidence of AD is crucial to mitigate the projected increase in cases. Exercise has emerged as an effective strategy to promote brain health in late adulthood and to protect against the onset of AD.

Postmortem brain MRI reveals differential associations of subcortical and limbic volumes with cortical thinning and neurodegenerative pathologies.

INTRODUCTION: The impact of different neuropathologies on deep brain structures remains to be understood. We examine subcortical and limbic volumetry in neurodegenerative diseases involving phosphorylated tau (p-tau), α-synuclein, and transactive response DNA binding protein 43 (TDP-43). METHODS: We acquired neuropathological measures and brain segmentations from postmortem analysis of 132 donors with Alzheimer's disease (AD), Lewy body disease (LBD), frontotemporal lobar degeneration with TDP-43 (FTLD-TDP), and FTLD-tau. RESULTS: LBD had the least subcortical, limbic, and cortical atrophy c

Administration of Pgk1 missense mutation leads to more effective mitigation of neurodegenerative effects observed in ALS mice and transgenic zebrafish.

Amyotrophic lateral sclerosis (ALS) is characterized by progressive degeneration of motor neurons (MNs) with few available therapeutic options. Previous ALS studies demonstrated a decrease in phosphoglycerate kinase 1 (Pgk1) secreted from NogoA-overexpressing muscle cells, thus reducing interaction between extracellular Pgk1 (ePgk1) and neural membranous Enolase 2 (Eno2) with consequent inhibition of neurite outgrowth of MNs (NOMN). The negatively charged 419th aspartic acid of receptor Eno2 (Eno2-D419) is a critical residue interacting with the positively charged 353rd lysine of ligand ePgk1-

Association of VTN Genotype with Plasminogen Activator Inhibitor-1 Activity in Late-Onset Alzheimer's Disease.

BACKGROUND/OBJECTIVES: Late-onset Alzheimer's disease (LOAD) is a multifactorial neurodegenerative disorder involving the interaction of genetic and environmental factors. Dysregulation of the fibrinolytic system, particularly an increase in plasminogen activator inhibitor-1 (PAI-1) levels, may contribute to Alzheimer's pathology. Vitronectin (VTN) regulates fibrinolysis by stabilizing PAI-1. This study investigated the relationships between plasma PAI-1 activity and VTN, SERPINE1 (PAI-1), and APOE gene variants in nineteen LOAD patients (>65 years) and ten cognitively normal age-matched contr

APOE4 Alters Early Transcriptional Programs and Inflammatory Signaling in Human Induced Pluripotent Stem Cells.

The APOE4 allele represents the strongest genetic risk factor for late-onset Alzheimer's disease (AD), yet its influence on early cellular programs remains poorly understood. In this study, we investigated transcriptional differences between human induced pluripotent stem cells (iPSCs) carrying the APOE3 or APOE4 genotype. RNA sequencing revealed pronounced genotype-dependent transcriptional changes, with enrichment of genes associated with neural development and metallothioneins in APOE4 cells, while genes related to extracellular matrix organization and cell adhesion were downregulated. Non-clinical safety evaluation of coenzyme I (NAD⁺): A GLP-compliant systematic study.

Nicotinamide adenine dinucleotide (NAD⁺) is a core coenzyme in cellular energy metabolism and redox homeostasis, with pharmacological effects in cardiovascular and neurodegenerative diseases and in aging. Its non-clinical safety was evaluated through GLP-compliant studies that included safety pharmacology (single intramuscular injection, 3-30 mg/kg, Beagle dogs, telemetry), acute toxicity (single intramuscular injection, 45-180 mg/kg, SD rats), repeated-dose toxicity (10-100 mg/kg intramuscular, once daily for 4 weeks plus a 4-week recovery, SD rats), muscular irritation (intramuscular, 14 con

Cognitive and Behavioural Profile of SOD1-ALS Across the ALS-FTD Spectrum: A Systematic Review.

Amyotrophic Lateral Sclerosis (ALS) is increasingly recognized as a multisystem disorder. However, SOD1-associated ALS has traditionally been regarded as an exclusively motor phenotype. This systematic review challenges that paradigm by characterizing the cognitive and behavioural profiles of SOD1 mutation-related ALS. Following PRISMA guidelines, a systematic PubMed search identified 17 eligible studies. Across the included studies, encompassing 222 patients with SOD1 variants, three of the analyzed subjects fulfilled the diagnostic criteria for frontotemporal dementia (FTD), and a larger pro

Phase separation and protein aggregation in neurodegenerative diseases.

Neurodegenerative diseases such as Alzheimer's, Parkinson's, frontotemporal dementia, and ALS are characterized by amyloid protein aggregation involving intrinsically disordered proteins that are also capable of liquid-liquid phase separation (LLPS). LLPS, known to drive the formation of dynamic membraneless organelles essential for cellular functions, can play a role in limiting fibrillation process or aberrantly transition into solid aggregates under pathological conditions. Here we review how mutations, post-translational modifications, and environmental factors can modulate LLPS of protein

Peripheral-central inflammatory crosstalk in Alzheimer's disease: immunometabolic functions of circulating soluble metabolites under dynamic blood-brain barrier states.

Immunometabolic dysregulation orchestrating bidirectional peripheral-central inflammatory crosstalk has been increasingly recognized as a contributor to Alzheimer's disease (AD) pathogenesis, challenging the conventional view of AD as an isolated central disorder dominated by amyloid-beta (Aβ) and tau pathology. However, although central immune-metabolic disturbances are well established, this perspective largely overlooks peripherally derived regulatory signals from systemic circulation. Classical peripheral-to-central pathways, such as immune cell infiltration and cytokine leakage, depend on

Exploring Endotoxemia in Age-Related Macular Degeneration, Glaucoma, and Diabetic Retinopathy.

Lipopolysaccharide (LPS), a component of Gram-negative bacteria, is a potent activator of the innate immune system and has been implicated in the pathogenesis of eye diseases, including age-related macular degeneration (AMD), primary open-angle glaucoma (POAG), and diabetic retinopathy (DR). LPS enters systemic circulation through bacterial lysis or active secretion, leading to endotoxemia-either acute or chronic. Chronic endotoxemia, often resulting from gut dysbiosis, may link microbial imbalances to eye diseases. Indeed, chronic LPS exposure may contribute to both systemic and local inflamm
