Parkinson's disease is the second leading neurodegenerative disease, pathologically characterized by the accumulation of alpha-synuclein in the brain and the loss of dopaminergic neurons in the substantia nigra pars compacta. Despite the intensive efforts to identify diagnostic biomarkers for Parkinson's disease nominating prospective candidates such as such as CSF α-synuclein seed amplification assay or α-/β-synuclein ratio, establishment of minimally invasive biomarkers remains the focus of Parkinson's disease research. We leveraged transcriptomic data from 4,343 participants from four indep

The membrane-embedded γ-secretase complex is involved in the intramembrane cleavage of ∼150 substrates. Cleavage of amyloid precursor protein (APP)-derived substrate C99 generates 38-43-residue secreted amyloid β-peptides (Aβ), with the aggregation-prone 42-residue form (Aβ42) particularly implicated in the pathogenesis of Alzheimer's Disease (AD). However, whether Aβ42 is the primary driver of neurodegeneration in AD remains unclear. Dominant mutations in APP or presenilin-the catalytic component of γ-secretase-cause early-onset familial AD (FAD) and reduce one or more steps in the multi-step

BACKGROUND: Complement dysregulation is increasingly recognized in Alzheimer's disease (AD). However, the temporal profile of complement alterations preceding AD onset and their distinction from age-related immune changes remain poorly defined. Clarifying these dynamics could provide insights into AD pathogenesis and identify systemic factors that predict disease onset and progression.
METHODS: We conducted a study involving two cohorts: a longitudinal cohort (n = 235; all cognitively normal at baseline) and a cross-sectional cohort (n = 323; including 53 with AD, 54 with vascular dementia, 5

Amyloid precursor protein (APP) plays an essential role in brain function and development. Variants in the APP gene are associated with both familial Alzheimer's disease and cerebral amyloid angiopathy. We report a case of early onset, slowly progressive mixed dementia with a newly identified APP variant. The patient developed mild cognitive impairment at age 51, followed by neuropsychiatric symptoms, seizures, and progressive white matter changes. Despite a fluctuating clinical course, significant deterioration occurred later, culminating in death at age 77. Genetic testing revealed an APP c.

The timing of dementia onset in patients with Parkinson's disease varies significantly. Predictors of the early development of dementia include postural instability, gait disturbance, REM sleep behavior disorder, and visuo-spatial impairment. The main neuropathological basis of dementia in Parkinson's disease involves the co-occurrence of α-synuclein pathology in the cerebral cortex and Alzheimer's disease co-pathology. Cholinergic system degeneration significantly affects symptom expression, making cholinesterase inhibitors effective for alleviating visual hallucinations and cognitive deficit

Alzheimer's disease (AD) -the most common form of dementia- begins with mild memory loss and gradually progresses, eventually resulting in a generalized loss of brain function. The pathological changes of AD in the brain cortex begin decades before the onset of symptoms. Improvements in lifestyle habits and disease-modifying treatments before the onset of the disease have been shown to help prevent or delay the onset of AD. However, diagnosis of AD is difficult at the early stage -or even at the prodromal stage [i.e., mild cognitive impairment due to AD (MCIAD)]- since normal aging and other t

INTRODUCTION: The incidence of Alzheimer's disease (AD) in Down syndrome (DS) exceeds 90%. Approximately 50% of people with DS have congenital heart disease (CHD). Having CHD increases risk for early-onset AD in populations without DS, but it is unclear if CHD influences AD in DS.
METHODS: Data from the Alzheimer Biomarker Consortium-Down Syndrome (ABC-DS) were used. Participants with CHD (n = 82, mean age = 39.9 ± 8.5 years, 97.6% White race) were age- and sex-matched to participants without CHD (n = 82, mean age = 40.5 ± 8.1 years, 98.8% White race). INTRODUCTION: The PSEN1E280A mutation causes autosomal dominant Alzheimer's disease (ADAD) with predictable onset, enabling presymptomatic studies. Extracellular vesicles (EVs) are emerging biomarkers of cognitive decline, but their role in early ADAD is unclear. The rare apolipoprotein E (APOE3) Christchurch (APOE3Ch) variant delays disease onset, yet its effect on EVs is unknown.
METHODS: We analyzed plasma EVs from mild cognitive impairment (MCI) and non-MCI PSEN1E280A-APOE3 carriers and non-MCI PSEN1E280A-APOE3Ch carriers using flow cytometry, proteomics, and co-culture assays.

BACKGROUND: Alzheimer's disease (AD) is characterized by early accumulation of amyloid-β (Aβ) plaques and tau pathology which precede overt neurodegeneration and cognitive decline. Detecting microstructural brain changes associated with Aβ deposition before the onset of atrophy is critical for early diagnosis and intervention.
OBJECTIVE: This study investigates whether diffusion kurtosis imaging (DKI) can detect early microstructural alterations in cortical and subcortical gray matter (GM) associated with Aβ pathology in individuals with mild cognitive impairment (MCI).
METHODS: Using DKI-de

BACKGROUND: Given inconsistent evidence regarding the migraine-Alzheimer's disease association, this Mendelian randomization (MR) study examines their potential causal relationship.
METHODS: GWAS summary statistics for migraine and Alzheimer's disease were acquired from the IEU Open GWAS repository. We implemented a multi-stage MR framework comprising (1) univariate analysis, (2) independent replication, (3) multivariable MR, (4) meta-analysis to evaluate migraine-AD causality.
RESULTS: Univariate results showed OR = 13.43, 95% CI: 2.86-63.16, P < 0.01; replication Mendelian randomization re

Alzheimer's disease (AD) is a complex neurodegenerative condition and the leading cause of dementia worldwide. Treatments that safely and effectively counteract disease progression are currently lacking. While the formation of amyloid plaques has long been considered the leading hypothesis of disease onset, growing evidence suggests that the emergence of AD could be driven by a combination of underlying factors that promote chronic neuroinflammation, including pathogenic infections, environmental toxicants, and disruptions along the gut-brain axis. Traditional nonclinical models of AD, such as

Alzheimer's disease (AD) is characterized by an insidious onset and complex pathophysiology, necessitating the development of effective strategies for early detection and intervention. This exploratory study aimed to identify differentially expressed genes (DEGs) and disrupted molecular pathways in AD by analyzing blood samples from participants recruited in Valle del Cauca, Colombia, a region with high genetic admixture and persistent underrepresentation in genomic research. A total of 41 individuals (AD, n = 14; cognitively healthy controls (CHC), n = 27) were included. Groups did not differ

We aimed to understand longitudinal associations between Alzheimer's disease (AD) biomarkers in Autosomal Dominant AD (ADAD) across estimated years to symptom onset (EYO). Forty-five individuals (19 mutation carriers [EYO = -7.9 ± 11.7 years, APP N = 11; PSEN1 N = 8]) from Swedish ADAD families participated. All received baseline 18F-Flurodeoxyglucose (FDG) PET and cognitive testing, and a subset (N = 26) plasma glial fibrillary acidic protein (GFAP) measurement. Follow-up data collection (including 106 FDG scans) was performed over 7.4 ± 6.4 years (visits ranged from 1-5, EYO = -25.8 to +10.3

Emerging evidence suggests that Parkinson's disease (PD) may have its origin in the enteric nervous system (ENS), from where α-synuclein (αS) pathology spreads to the brain1-4. Decades before the onset of motor symptoms, patients with PD suffer from constipation and present with circulating T cells responsive to αS, suggesting that peripheral immune responses initiated in the ENS may be involved in the early stages of PD1,5-7. However, cellular mechanisms that trigger αS pathology in the ENS and its spread along the gut-brain axis remain elusive. Here we demonstrate that muscularis macrophages

Toxic signaling of oligomeric protein species via binding to the cellular prion protein (PrPC) is implicated in various neurodegenerative diseases, including Alzheimer’s (AD) and Parkinson’s disease (PD). Metabotropic glutamate receptor 5 (mGluR5) has been identified as a PrPC signaling partner, and pharmacological inhibition of mGluR5 was shown to improve cognitive performance and rescue long-term-potentiation (LTP) impairment in both in vitro and in vivo models of AD and PD. Prion diseases are another group of fatal neurodegenerative disorders that are characterized by templated misfolding o

INTRODUCTION: Olfactory dysfunction is common in the Alzheimer's Disease continuum, and olfaction may be altered before clinical syndrome onset. The present study aimed at investigating the functional connectivity of the olfactory cortex and its correlation with olfaction performance in a group of patients with Mild Cognitive Impairment (MCI) who subsequently converted or not converted to Alzheimer's Disease (AD) dementia.
METHODS: At baseline, 30 MCI patients were evaluated with the Sniffin' Sticks (threshold, discrimination, and identification) to assess olfactory capacities, and they were

BACKGROUND: Mitochondrial dysfunction and protein aggregation are central features of brain aging and Alzheimer's disease (AD). To define how AD seed proteins modulate these processes, we applied quantitative proteomics to sarkosyl-insoluble aggregates from C. elegans models of normal aging and from worms expressing human Aβ or Tau transgenes.
RESULTS: Normal aging produced a late-onset accrual of mitochondrial proteins within aggregates, implicating impaired energy metabolism and proteostasis collapse. Aβ expression caused a striking expansion and included glycolytic enzymes, tricarboxylic a

INTRODUCTION: Sleep-wake disturbances frequently occur at early stages of Alzheimer's disease (AD) and accelerate disease progression, but the underlying neural mechanisms are not fully understood.
METHODS: We examined sleep-wake behavior and locus coeruleus (LC) activity in young 5xFAD mice using electrophysiology and pharmacological approaches targeting adrenergic signaling and potassium channels.
RESULTS: 5xFAD mice displayed dark phase-specific hyperarousal and impaired brain state transitions by 2 months of age. LC neurons exhibited increased tonic firing due to impaired Kv4 and Kv7 pot

Aging is the strongest risk factor for Alzheimer's disease (AD). Identifying reliable biomarkers of brain aging helps to predict functional decline and dementia onset. Evaluations of aging-related biomarkers in plasma and neuronal-derived extracellular vesicles (nEVs) from cognitively healthy and AD subjects, alongside post-mortem IPL brain samples from control (Ctr), pre-clinical AD (PCAD), mild cognitive impairment (MCI) and AD cases were performed. Cognitive tests, functional assessments, and MRI data were also included. Biomarkers in nEVs more accurately reflected brain pathology than thos

The rising burden of neurodegenerative diseases has exposed critical gaps in the development of diagnostic and therapeutic tools, particularly regarding their limited sensitivity, poor spatiotemporal resolution, and lack of treatment specificity. These shortcomings highlight the need for technologies that enable early disease detection, real-time monitoring of regenerative processes, and precise therapeutic interventions. This review focuses on recent advances in photonic biosensing and optogenetics, examining how these technologies are reshaping neural regeneration research and assessing thei
