Metabolism fluctuation coupling can track the progression of dementia and describe MST1 gene-related pathology.

BACKGROUND: Cognitive resilience refers to an individual's capacity to cope with brain aging and pathology and to delay cognitive decline, whereas existing techniques such as functional magnetic resonance imaging capture only macroscopic features without linking them to neurophysiological mechanisms. Recent studies have shown that overexpression of the MST1 gene exacerbates Alzheimer's disease phenotypes by affecting neuronal activity and metabolism; however, its association with cognitive trajectories and imaging biomarkers remains to be further investigated. METHODS: Multimodal imaging data

Energetic implications of fMRI-based nodal complex network metrics: a complex picture unfolds across diverse brain states.

Functional MRI-based graph theory has provided profound insights into the brain's functional organization, yet the neuroenergetic meaning of widely used graph-theoretical metrics remains poorly understood. Although resting-state research suggests a positive coupling between network topology and glucose metabolism, it remains unclear whether this relationship reflects a general principle of brain organization or a state-specific phenomenon. Here, we test the neuroenergetic interpretability of nodal graph-theoretical metrics by linking complex network topology to cerebral glucose consumption acr

Blood biomarkers of frailty and cognition: A scoping review.

Frailty increasingly is recognized as a factor that modifies the relationship between disease biomarkers, including neuropathology, and dementia expression. The mechanisms underlying the relationship between frailty and dementia remain unclear, but blood biomarkers can offer insight into these mechanisms. We completed a scoping review of research examining the associations between blood biomarkers, frailty, and cognition. Three online databases were searched to identify original research examining blood biomarkers in the context of frailty and/or cognitive decline that accounted for the other

Cognability across adulthood: A qualitative investigation of neighborhoods and cognitive health behaviors.

While geographic variation in Alzheimer's Disease and Related Dementias (ADRD) rates suggests that environmental factors are important in the development of dementia, understanding of specific neighborhood sites that impact dementia risk is limited, especially in early and mid adulthood. This paper extends Cognability to a life course perspective to conceptualize how neighborhoods may support cognitive health behaviors including physical activity, diet, cognitive stimulation, and socialization across adulthood. The Neighborhoods and Health at All Ages Study employed stationary and mobile inter

Cognitive data harmonization in the ADRC Network and beyond-Past, present, and future.

The National Alzheimer's Coordinating Center (NACC) Uniform Data Set (UDS) includes extensive cognitive data contributed by Alzheimer's Disease Research Centers (ADRCs) since 2005. The UDS has evolved over time and is on its fourth iteration. In addition to this core dataset, individual ADRCs have administered augmented neuropsychological batteries to research participants that go beyond the UDS. Here we describe ongoing efforts and future plans to optimize use of these data with modern psychometric methods. Modern psychometric methods address challenges from an evolving battery of cognitive t

Targeting of PCSK9 in the Management of Alzheimer's Disease: Expanding the New Potential Key Player.

Alzheimer's disease (AD) represents the most common cause of dementia in the elderly population worldwide. However, most of the anti-AD medications did not resolve the underlying neuropathology. Consequently, targeting other signaling pathways may be helpful in the management of AD. Particularly, preprotein convertase subtilisin/kexin type 9 (PCSK9), which is a regulator protein of low-density lipoprotein (LDL), is intricate in the pathogenesis of AD. Normally expressed PCSK9 in the brain plays a critical role in the regulation of neuronal differentiation and apoptosis, and degradation of LDL

The Interplay Between Nurr1 and Mitochondrial Biogenesis: Implications for Neurodegenerative Therapy.

The transcription factor Nurr1 (NR4A2) serves as an essential element in dopaminergic neuron development since it functions predominantly in the substantia nigra, which becomes severely affected during Parkinson's disease (PD) and Alzheimer's disease (AD). Nurr1 regulates dopamine synthesis, survival-promoting, and oxidative stress genes that affect mitochondrial formation. Nurr1 binds to PGC-1α, allowing for mitochondrial activity regulation. This relationship supports mitochondrial biogenesis. Post-translational changes, including phosphorylation and acetylation, modify Nurr1 transcriptional

Motor deficits in the McGill-R-Thy1-APP transgenic rat model of Alzheimer's disease.

The McGill-R-Thy1-APP rat is a transgenic model of Alzheimer's disease (AD) which expresses APP with two mutations found in cases of familial AD, resulting in the development of amyloid pathology and cognitive deficits. Motor deficits are common symptoms of AD, emerging early in the disease, and are correlated with AD neuropathology and cognitive symptoms. This study evaluated hemizygous and homozygous McGill-R-Thy1-APP rats and their wildtype littermates for spontaneous alternation and locomotion in the T and Y mazes, and motor behaviour on an accelerating rotarod at 12-13 months of age. We f

Excitation-inhibition homeostasis in Alzheimer's disease: a selective multiscale review of mechanisms, sex differences, and therapeutic opportunities.

Alzheimer's disease is increasingly viewed as a breakdown of balanced excitation-inhibition (E/I) homeostasis layered atop classical proteinopathy. Restoring circuit-level neural excitation and inhibition is rapidly becoming a tractable therapeutic strategy, guiding trials of circuit-modulating drugs such as levetiracetam. To date, however, findings across species and modalities remain fragmented, and it is unclear how to contextualize AD-focused E/I findings across scales and methodologies. Synthesizing over 150 studies of E/I homeostasis in AD, we organize the results into several prevailing

Engineering In Situ Activatable Integrated Polymer Dots with a Defined Chiral Microenvironment for In Vivo Enantioselective Monitoring of D-Noradrenaline in Brains with Alzheimer's Disease.

Enantioselective discrimination of enantiomers is very important to reveal the function and concentration fluctuation of chiral isomers in biological processes and disease pathogenesis. It remains challenging to distinguish D/L-enantiomers because they possess identical chemical composition and very similar physicochemical properties. Norepinephrine (NE) is a chiral monoamine neurotransmitter that shows extensive physiological effects on both the peripheral and central nervous systems of mammals and plays significant roles in the pathogenesis of Alzheimer's disease (AD). However, specific fluo

Pyrimidine RNA Homopolymers Promote Amyloid Formation of a Tau Fragment from the Microtubule-Binding Domain.

Abnormal aggregation of microtubule-associated protein tau into β-sheet-rich fibrils is a hallmark feature of Alzheimer's disease and other tauopathies. The pathogenic P301L mutation within the microtubule-binding domain of tau promotes tau filament formation; however, the molecular mechanisms by which intracellular RNAs regulate this aggregation process remain not fully understood. Here, we investigated the mechanistic effects of RNA homopolymers on the aggregation of a tau fragment peptide (residues 298-317) derived from the microtubule-binding region and its P301L mutant. The results showed

Carboxymethyl chitosan for neurological drug delivery: Current trends and future prospects.

The complexity of drug delivery across the blood-brain barrier (BBB) makes disorders of the central nervous system (CNS), such as Alzheimer's disease, Parkinson's disease(PD), brain tumors, and other neurodegenerative conditions, a growing global health burden. Poor brain bioavailability and systemic side effects are common problems with traditional therapies. Delivery systems based on nanotechnology have shown promise as solutions to these problems. These include carboxymethyl chitosan (CMC), a water-soluble, biocompatible, and biodegradable chitosan derivative that has drawn a lot of interes
