Relationship between locus coeruleus and slow-wave sleep in aging and Alzheimer's disease.

INTRODUCTION: Sleep disruption, particularly loss of slow-wave sleep (SWS), is common in Alzheimer's disease (AD), but its neurobiological underpinnings remain unclear. We investigated whether locus coeruleus (LC) integrity relates to SWS across the AD continuum and whether sex and perivascular spaces (PVSs) modify these associations. METHODS: In a cohort (11 controls, 30 mild cognitive impairment, 17 AD dementia) we combined overnight polysomnography with LC-sensitive magnetic resonance imaging of the LC, basal ganglia and centrum semiovale PVS ratings, and cerebrospinal fluid noradrenaline.

Recommendations from the European interdisciplinary council on ageing on physical activity and diet for mental health conditions in older adults.

The global rise in life expectancy is accompanied by an increase in the prevalence of mental health conditions among older adults, including mild cognitive impairment (MCI), dementia, delirium, depression, anxiety, and other severe mental illness. These conditions significantly impact independence, increase healthcare costs, and increase mortality risk. Mounting evidence underscores the central role of modifiable lifestyle factors-particularly physical activity and diet-in the prevention and management of these conditions. This consensus, developed under the auspices of the European Interdisci

Aggregate learning ratio from the Neuropsychological Assessment Battery distinguishes between amnestic and non-amnestic mild cognitive impairment in a real-world clinical sample.

Mild cognitive impairment (MCI) is a heterogeneous syndrome with varying cognitive profiles and trajectories. Amnestic MCI (aMCI) is associated with memory deficits and increased risk of Alzheimer's disease, while non-amnestic MCI (naMCI) often involves executive dysfunction and may signal risk for other dementias. Encoding deficits, though more prominent in aMCI, are also observed in naMCI. This study examined the utility of the Neuropsychological Assessment Battery (NAB) learning ratio (LR) scores-specifically List LR, Story LR, and an Aggregate LR-to distinguish among cognitively normal ind

Conformation-specific monoclonal antibodies reveal early Tau structural intermediates in Alzheimer's disease.

Tauopathy, including Alzheimer's disease, is the most prevalent neurodegenerative disorder, yet current therapies fail to halt disease progression, partly because of limited understanding of early tau pathological structures. Cryo-EM has clarified distinct Tau structures tied to disease phenotypes but only in their mature forms and at low throughput. Hence, molecular probes that can recognize early Tau conformations in high throughput and in situ hold the potential to transform our understanding of Tau aggregation. Tau undergoes sequential changes, including hyperphosphorylation and misfolding

Identifying Subclinical Transthyretin Cardiac Amyloidosis in V142I TTR Carriers: Design, Rationale, and Methods of the VISTA (Variant Imaging of Subclinical Transthyretin Amyloidosis) Study.

BACKGROUND: Approximately 1.5 million of the 44 million individuals of African descent in the United States are carriers of the p.V142I variant in transthyretin (TTR). This is the most common cause of variant amyloid cardiomyopathy (ATTR-CM) worldwide which leads to heart failure (HF) and premature death. Despite promising new treatments, ATTR-CM is often diagnosed at an advanced stage and conventional diagnostic tools lack specificity to detect early disease. Thus, the overall objectives of the Variant Imaging of Subclinical Transthyretin Amyloidosis (VISTA) study are to determine imaging and

Parkinson's and Alzheimer's disease impairs temporal precision.

Several studies have reported temporal processing deficits in neurodegenerative diseases such as Parkinson's and Alzheimer's disease. These deficits can be quantified by interval timing paradigms that require participants to estimate or produce an interval of several seconds and require working memory for temporal rules as well as attention to time. Timing performance can be quantified by a variety of measures; however, two relatively universal metrics include: 1) temporal accuracy, defined as the mean temporal estimate and 2) temporal precision, reflected by the variability of temporal estima

Associations of dementia polyexposure scores to Alzheimer's disease endophenotypes in diverse populations.

INTRODUCTION: Dementia clinical risk scores (CRS) provide accessible tools for identifying individuals at risk for Alzheimer's disease (AD), yet their performance across diverse populations and relationships to AD endophenotypes remains unclear. METHODS: We evaluated four CRS, mCAIDE, WHICAP, LIBRA, and CogDRisk, in relation to cognitive impairment diagnoses and endophenotypes for AD, including plasma biomarkers, neuroimaging measures, and cognitive composite scores. Logistic and linear regression models stratified by self-reported race/ethnicity were used to assess the associations of CRS wi

Microglial GM3 accumulation impairs Aβ phagocytic activity and promotes neuroinflammation in Alzheimer's disease.

Growing evidence underscores the critical role of lipid metabolism in the pathogenesis of Alzheimer's disease (AD). We previously demonstrated that 5xFAD mice exhibit a marked accumulation of ganglioside GM3 in the cerebral cortex and hippocampus as the disease progresses, with this increase being more pronounced in females than in males. However, the specific brain cell types exhibiting elevated GM3 accumulation, along with GM3's underlying molecular mechanisms and functional significance in AD pathogenesis, remain to be fully elucidated. Here, we report that elevated GM3 levels in 5xFAD are

Analyses of plasma multi-omic data across ancestries identify novel pathways implicated in Alzheimer's disease.

INTRODUCTION: Few genetic studies on Alzheimer's disease (AD) have incorporated multiple ancestries and omic datasets to pinpoint actionable AD risk effectors for each ancestry. METHODS: Here, we first performed genetic colocalization between molecular phenotypes (proteomics and metabolomics) from two ancestral groups (European [EUR] and African [AFR]) and the two largest EUR AD genome-wide association studies. We next performed pathway enrichment analyses to identify biological mechanisms. RESULTS: We found 21 proteins and one metabolite colocalized with AD risk that were shared between the

Modeling and application of alzheimer's disease complex trait prediction based on multi-task learning.

Alzheimer’s disease (AD) is a complex disorder influenced by genetic factors, and related phenotypes often share common genetic mechanisms. However, traditional genetic risk prediction models typically focus on a single phenotype, neglecting valuable cross-phenotype information. This study aims to explore the effectiveness of multi-task learning (MTL) in modeling genetic data for AD and to assess whether aggregating genetic signals at the gene level can further improve prediction accuracy. We applied two representative MTL models, including the Sluice Network and the Learning to Branch (LTB) m

Beyond the Leaderboard: Evaluating the Robustness of Deep Learning Models for Detecting Freezing of Gait.

Freezing of gait is a common, debilitating symptom that affects many patients with Parkinson's disease. The lack of a standard, objective method to quantify freezing obstructs research and treatment. Wearable sensors combined with automatic detection algorithms have demonstrated increasingly promising results; nonetheless, video annotation remains the gold standard. After organizing a global machine learning contest to expedite the development of acceleration-based algorithms designed to automatically detect freezing, we tested the transferability of the winning models to a new dataset. Ferroptosis and Alzheimer's disease: unraveling the molecular mechanisms and therapeutic opportunities.

Ferroptosis is a novel form of regulated cell death. Compared with other types of cell death, it shows great differences in structure and biochemistry. This type of cell death is receiving increasing attention. For example, studies have found that it plays a key role in the development of neurodegenerative diseases underlying brain atrophy, such as Alzheimer's disease (AD). AD is a chronic and worsening neurodegenerative disease. It poses a serious threat to the health and quality of life of the elderly. The pathology of AD is mainly the presence of extracellular beta-amyloid (Aβ) plaques and

Pharmacological inhibition of O-GlcNAcase reduces pS129-α-synuclein positive aggregates in the substantia nigra of mThy1-hSNCA mice.

BackgroundThe aggregation and spread of α-synuclein within brain are associated with the loss of dopaminergic neurons and the formation of Lewy bodies as seen in Parkinson's disease. Blocking the initiation of α-synuclein aggregation, or the spread of such aggregates, may offer disease-modifying approaches to slow disease progression. Previous studies have demonstrated that modification of aggregation prone proteins, including α-synuclein, with O-linked β-N-acetylglucosamine (O-GlcNAc) reduces their aggregation. Small molecule inhibitors of the enzyme O-GlcNAcase (OGA), which removes O-GlcNAc

Ultrasound-mediated blood-brain barrier opening for targeted neurological drug delivery.

Neurological disorders represent a devastating global health crisis, and the blood-brain barrier (BBB) remains a major obstacle for their treatment. Conventional strategies for BBB opening, including direct intracranial injection, osmotic disruption, receptor-mediated transcytosis, and nanoparticle carriers, often suffers from surgical invasiveness, systemic toxicity, poor biodistribution, and off-target effects. Ultrasound-mediated drug delivery has emerged as a revolutionary non-invasive technology for transient and targeted BBB opening, enabling enhanced penetration of therapeutic agents in

Alzheimer's disease prediction algorithm based on hippocampal longitudinal hybrid morphological features.

BACKGROUND: Alzheimer's disease (AD) is a progressive neurodegenerative disorder characterized by cognitive decline; this decline is closely linked to hippocampal morphological changes observed in structural magnetic resonance imaging (MRI). However, the existing AD prediction models have not fully explored the spatiotemporal correlation of hippocampal morphological features. To address this limitation, this study aims to develop a longitudinal prediction framework that captures both the temporal evolution and spatial distribution of hippocampal morphological alterations. METHODS: In this pap

Targeted Protein Degrader from Ginkgo to Mitigate Amyloid β-Induced Neurotoxicity.

Protein degradation through the autophagy-lysosome process by eukaryotic cells is a major pathway to remove unwanted proteins, organelles, and invading pathogens. It is also an emerging intervention strategy to selectively eliminate inaccessible toxic amyloid proteins to prevent amyloid β (Aβ)-induced neurotoxicity. Currently, there is no natural product-derived peptide that targets amyloid proteins for degradation through the autophagy-lysosome pathway. We recently discovered a new peptide family from Ginkgo biloba nuts, which we termed β-ginkgotides. The prototype β-gB1 is 20-residue in leng
