Silencing of the Metabolic Gene HKDC1 Is Associated With Aging and Neurodegeneration in Mice and Humans.

Increased life expectancy brought about by improved healthcare and lifestyle has heightened the challenge of neurodegenerative disorders like Alzheimer's disease (AD) and other age-related disorders. Neurodegeneration is known to be accompanied by loss of memory, changes in brain morphology, and neuroinflammation, and multiple factors contribute to the progression and pathogenesis of the condition. Of these factors, metabolic dysregulation is known to influence the process, but the precise mechanisms remain unexplored. In this study, we investigated the brain-specific role of the metabolic enz

Magnetic resonance spectroscopy as a non-invasive tool for assessing brain and muscle adaptation to exercise training in older age: a scoping review into existing research.

BACKGROUND: Exercise training has attracted increasing attention as a non-pharmacological intervention approach to counteract age-related deterioration of brain and muscle function, yet objective biomarkers are needed to understand mechanisms and optimize interventions. Magnetic resonance spectroscopy (MRS) provides non-invasive, in vivo assessment of metabolic profiles altered by aging and exercise. However, MRS-based exercise research in older populations remains limited. This scoping review aims to identify brain and muscle metabolites detectable by MRS that can serve as markers of exercise

LKB1-AMPK Signaling Pathway in Cardiovascular and Other Diseases.

The LKB1-AMPK signaling pathway is a master regulator of cellular energy homeostasis and a central hub in stress adaptation. As a conserved metabolic sensor, this pathway coordinates glucose, lipid, and protein metabolism, thereby sustaining physiological function across diverse tissues. Beyond its canonical role in energy balance, growing evidence highlights its dysregulation in multiple pathological conditions. Despite extensive mechanistic studies, the disease-specific regulation and translational potential of the LKB1-AMPK pathway remain incompletely understood. This review systematically

HEXIM1/P-TEFb complex controls RNA polymerase II pause release and immediate early gene induction following neuronal depolarization.

Cognitive processes require de novo gene transcription in neurons. Memory requires the rapid and robust transcription of a class of genes called immediate early genes (IEGs). IEG transcription is facilitated by the formation of a poised basal state, in which RNA polymerase II (RNAP2) initiates transcription, but remains paused downstream of the promoter. Upon neuronal depolarization, the paused RNAP2 is released to complete the synthesis of mRNA transcripts, a process stimulated by positive transcription elongation factor b (P-TEFb). In many cell types, P-TEFb is sequestered into a large inact

N-Salicyloyl Tryptamine Derivatives Improve Mitochondrial Function To Treat Alzheimer's Disease.

Alzheimer's disease (AD) is a neurodegenerative disease (NDD) characterized by complex pathogenesis and remains incurable to date. Emerging evidence has demonstrated that mitochondrial dysfunction not only serves as a central component in AD initiation but also precedes other pathological processes, thereby playing a pivotal role in both the pathogenesis and progression of AD. Previous studies have confirmed that N-salicyloyl tryptamine derivatives can regulate mitochondrial function and effectively treat NDDs. Among them, compound N2 showed the best efficacy. This study first applied compound

Sex-specific effects of acetylation on tauopathy in aging htau mice.

Alzheimer’s disease involves extracellular β-amyloid accumulation and intracellular phosphorylated tau aggregates, with higher disease prevalence and neuropathological burden in aging females. While tau phosphorylation contributes to tau pathology, other modifications, such as acetylation, also promote aggregation. Aging disrupts proteostasis, in part through acetylation, a post-translational modification affecting protein function and stability; however, its role in sex-specific tauopathy remains unclear. This study investigated acetylation in an age-, sex-specific manner across presymptomati

Cognitive screening biases in a secondary prevention Alzheimer's disease clinical trial.

INTRODUCTION: Alzheimer's disease (AD) prevention trials have multiple steps to identify cognitively unimpaired individuals with AD biomarker evidence. Cognitive/functional screening tests may be biased in ethnoracial minorities, impacting trial eligibility. METHODS: A total of 6669 participants screened for the Anti-Amyloid Treatment in Asymptomatic Alzheimer's (A4) study were grouped by ethnoracial background and testing language. Ethnoracial/language differences in ineligibility reason, cognitive/functional test performance, and amyloid positivity rates were examined. RESULTS: Ethnoracial

Association of Aβ monomers with cerebral amyloid angiopathy in brains without parenchymal Aβ deposition.

β-Amyloid (Aβ) deposition is a hallmark of both Alzheimer's disease and cerebral amyloid angiopathy. Whilst insoluble Aβ aggregates have been extensively studied, the role of soluble Aβ monomers in vascular amyloid pathology-and their association with cognitive decline-remains unclear in plaque-free brains. This study examined whether soluble cortical Aβ species are associated with cognitive outcomes and amyloid-related pathologies, including cerebral amyloid angiopathy, in the absence of parenchymal Aβ deposition. We examined post-mortem cortical tissue from nearly 200 individuals without par

Transcriptomic signature of frontotemporal lobar degeneration with TDP-43 type C pathology.

Semantic variant of primary progressive aphasia is a clinical subtype of frontotemporal lobar degeneration and is marked by TDP-43 subtype C pathology (FTLD-TDP C). It is a sporadic disease, yet has a strikingly homogeneous clinicopathological presentation, suggesting a common pathophysiology. The aim of this study was to discover dysregulated pathways in FTLD-TDP C through transcriptomics of the temporal cortex, its most affected region. Bulk RNA sequencing was conducted on temporal cortices of a post-mortem cohort of 18 FTLD-TDP C patients and 23 sex- and age-matched controls. Differential e

Neuro-Dynamic Quantitative Systems Pharmacology (QSP) model describing Alzheimer's disease pathophysiology and treatment effects.

Lecanemab, an anti-amyloid antibody, has demonstrated a significant clinical benefit in slowing cognitive decline in early Alzheimer's disease (AD). A mechanistic Neuro-Dynamic Quantitative Systems Pharmacology (QSP) model was developed to capture the temporal and biological complexity of AD progression. This QSP model incorporates three interlinked modules reflecting core aspects of AD pathology: Aβ accumulation, tau pathology, and cognitive decline, where Aβ accumulation promotes tau pathology, which leads to neuronal damage and cognitive impairment. A large multivariate dataset was assemble

Psychiatric and Cognitive Effects of Testosterone Therapy in Adult Men: A Systematic Review of Clinical Evidence and Mechanistic Insights.

Testosterone plays a critical role in male physical and psychological health, influencing not only reproductive and metabolic functions but also mood, cognition, and overall mental well-being. Age-related testosterone decline has been associated with depressive symptoms, cognitive impairment, and reduced quality of life. Although testosterone therapy (TT) is well established for somatic outcomes, its psychiatric and cognitive effects remain inconsistently characterized across clinical trials. The objective is to systematically review the evidence on the effects of TT on psychiatric and cogniti

Effects of caregiving experience on expectations of becoming an Alzheimer's disease caregiver in the future.

OBJECTIVES: Research often focuses on persons who are or have been caregivers to a person with dementia. Little is known about stigma reactions in persons who expect to care for a person with Alzheimer's disease (AD). Understanding these beliefs may help guide clinical discussions with family members of persons with AD, particularly those diagnosed in the early stages. METHODS: A sample of 3,527 adults was randomized to read a vignette describing a fictional person with either a positive or a negative AD biomarker test result. Bivariate and multivariable analyses examined associations between
