A NeuroD1 AAV-Based Gene Therapy for Functional Brain Repair in Alzheimer's Disease-Like Non-Human Primate Model.

There is a pressing demand for neuroregenerative treatment for Alzheimer's disease (AD). Recently, a NeuroD1-mediated neuroregeneration strategy has been proposed, yet its efficacy remains untested in non-human primate (NHP) AD models closely reflecting human pathology. This study evaluates the therapeutic potential of NeuroD1 AAV-based gene therapy in an NHP AD model with hippocampal hTau overexpression, utilizing immunostaining, fluorescence/confocal imaging, MRI and FDG PET scans, Simoa CSF biomarker analysis, behavioral tests, and bulk RNA sequencing. NeuroD1 AAV-based gene therapy prevent

Intracerebroventricular human mesenchymal stem cells induce MMP9-driven transient inflammation in Alzheimer's disease.

BACKGROUND: Mesenchymal stem cells (MSCs) are often considered hypoimmunogenic. However, a transient fever observed after intracerebroventricular (ICV) administration in a clinical trial suggests an acute host response. This study examines the mechanisms underlying this reaction, with a focus on MSC migration and the role of matrix metalloproteinase-9 (MMP9). METHODS: We analyzed cerebrospinal fluid (CSF) from Alzheimer's disease (AD) patients treated with saline (n = 3) or human MSCs (hMSCs) (n = 6) using an exploratory protease array, followed by enzyme-linked immunosorbent assay (ELISA). Reversibility and β-sheet formation are decoupled in tau condensate aging.

Neurofibrillary tangles (NFTs) formed from the protein tau disrupt neuronal function in Alzheimer's disease and are strongly associated with cognitive decline. Early events in tau aggregation are increasingly linked to the formation of biomolecular condensates, which lower the energetic barriers to pathological aggregation by acting as intermediates that transition into insoluble assemblies, a mechanism also implicated in other neurodegenerative diseases, such as amyotrophic lateral sclerosis (ALS) and frontotemporal dementia (FTD). Despite growing evidence for this pathway, the molecular basi

Exploring the Mechanism of Selenium-Biofortified Polygonatum Kingianum in Alzheimer's Disease: An Integrated Metabolomics and Network Pharmacology In Silico Study.

BACKGROUND: Alzheimer's disease [AD] involves multifactorial pathogenesis such as Aβ deposition and Tau hyperphosphorylation, yet effective multi-target therapies remain scarce. The mechanisms by which selenium-biofortified Polygonatum kingianum [Se-PK] modulates AD pathways are poorly understood, limiting its clinical translation. METHODS: An integrated in silico approach was employed: 1] UPLC-MS/MS metabolomics to identify differential metabolites in Se-PK [VIP > 1, fold change ≥2 or ≤0.5]; 2] network pharmacology to construct compound-target-pathway networks; and 3] molecular docking and d

Multivariate patterns of plasma biomarkers predict region-specific Alzheimer's pathology and cognitive decline across independent cohorts.

INTRODUCTION: Alzheimer's disease (AD) is characterized by the progressive accumulation of amyloid-beta (Aβ) plaques, tau tangles, and neurodegeneration (ATN framework). While neuroimaging detects these changes, it is costly and not widely accessible. Blood-based biomarkers offer scalable alternatives for early detection. METHODS: We integrated plasma biomarkers, neuroimaging, and cognitive data from two large cohorts. We examined multivariate patterns of plasma Aβ42/Aβ40, phosphorylated tau (p-tau) 217, p-tau181, neurofilament light (NfL), and glial fibrillary acidic protein (GFAP) that pred

Filamin A phosphorylation at S2152: a molecular switch fueling cancer and neurodegeneration.

Best known for crosslinking actin to form the cytoskeleton, the scaffolding protein filamin A (FLNA) is an unusually complex intracellular regulator, interacting with a vast array of receptors, cytoskeletal proteins and signaling molecules to expand its regulatory function from structural dynamics to focal adhesion, cell migration and diverse cell signaling. This review focuses on the interplay between FLNA’s conformation, its phosphorylation at S2152 and changes in its protein interactions that alter its function and contribute to cancer, metastasis, Alzheimer’s and potentially other age-rela

Diagnostic performance of plasma GFAP in preclinical stages of Alzheimer's disease using the Lumipulse platform.

BACKGROUND: Plasma glial fibrillary acidic protein (GFAP) has emerged as a promising biomarker of astrocytic reactivity in preclinical Alzheimer´s Disease (AD). However, most evidence to date comes from ultrasensitive research assays, with limited data from automated clinical platforms. We aimed to evaluate the diagnostic performance of plasma GFAP in cognitively unimpaired (CU) individuals using the fully automated Lumipulse platform, and to assess its concordance with a highly sensitive research-based immunoassay, Single Molecule Array (SIMOA). METHODS: Plasma GFAP, and cerebrospinal fluid (

Non-physiological potassium concentrations in commercial culture media trigger acute seizure-like activity in human iPSC-derived neurons.

Neuronal in vitro cultures are pivotal for studying brain electrophysiological function and dysfunction. Neuronal activity and communication are regulated by extracellular ion concentrations. Therefore, cell culture medium ion concentrations should ideally mimic those of cerebrospinal fluid (CSF)—considered as the milieu for brain cells in vivo. In this study, we demonstrate that commonly used cell culture media, including NeurobasalTM (+ /− A), Neurobasal PlusTM, and BrainPhysTM media, do not accurately replicate human CSF ion concentrations. Using human iPSC-derived neuronal networks on micr

Ciliary p75 neurotrophin receptor (p75NTR) facilitates the enrichment of exogenous amyloid beta (Aβ 1-42) peptide and promotes oxidative stress in human hippocampal astrocytes.

Accumulation of amyloid beta 1–42 (Aβ42) peptide in the extracellular space in the brain is a major observation in Alzheimer’s Disease (AD)-related pathology. Astrocytes are known to play pivotal role in clearing the extracellular aβ peptide from the brain, and the underlying mechanism of Aβ42 peptide clearance remains underappreciated. Like other cell types in the brain, astrocytes have primary cilia, a nonmotile microtubule-based organelle. Aβ42 peptide is reported to affect cilia length or structure in multiple cell types including neurons and inhibit ciliary p75 neurotrophin receptor (p75N

Astrocytes at the crossroads of obstructive sleep apnea and Alzheimer's disease: from oxygen sensing to neurodegeneration.

PURPOSE: Obstructive sleep apnea (OSA) is a highly prevalent sleep-related breathing disorder characterized by recurrent episodes of intermittent hypoxia and sleep fragmentation, and it has been increasingly linked to cognitive decline and Alzheimer’s disease (AD). Growing evidence suggests that vascular risk factors and sleep-related breathing disorders such as OSA may contribute to AD disease onset and progression. This review aims to explore astrocytes as a potential mechanistic link between OSA and AD. METHODS: A literature search was conducted in the PubMed database using combinations of

Modulation of apolipoprotein E receptor-2 by ApoE4, amyloid β-peptide, reelin, and secreted amyloid precursor protein: a common point of impact in Alzheimer's disease pathogenesis.

INTRODUCTION: Apolipoprotein E (ApoE), reelin, and several other proteins bind ApoE-receptor 2 (apoER2), distinguished from other members of its receptor family by signal transduction which enhances the activity of N-methyl D-aspartate (NMDA) receptors. Evidence indicates that this signal transduction depends upon apoER2 forming dimers or other high-order clusters. It seems noteworthy therefore that protein products of major APOE gene variants differ in their numbers of cysteines capable of forming disulfide dimers, with the allele (ε4) associated with highest rates of Alzheimer's disease (AD)

APP as an innate injury-response molecule.

Amyloid precursor protein (APP) is best known as the percussor for amyloid beta (Aβ), a hallmark of pathology in Alzheimer's disease and related disorders. Nevertheless, APP did not likely evolve to serve this purpose. This article reviews available evidence for functions of APP and its enzymatically generated proteolytic derivatives, sAPPα, CTFα, sAPPβ, CTFβ, p3 peptide, AICD, and Aβ. These functions include not only effects on the development and function of neurons in the central nervous system (CNS), but also innate roles in the response to diverse types of cell and tissue injury in multip

Performance of a fully automated plasma tau phosphorylated at threonine 217 immunoassay to reflect amyloid-beta burden in an unselected cohort representative of clinical practice.

BACKGROUND: With the emergence of disease-modifying anti-amyloid-beta (Aβ) therapies for Alzheimer's disease (AD), early and accurate quantitative measures of Aβ burden are critical. Blood-based biomarkers are a scalable and minimally invasive diagnostic solution; plasma tau phosphorylated at threonine 217 (pTau217) is a promising marker for Aβ pathology. The clinical performance of the prototype ElecsysⓇ Phospho-Tau (217P) Plasma immunoassay (Roche Diagnostics) to detect Aβ burden was investigated in an unselected cohort reflective of clinical practice. Neuronal overexpression of Nrf2 reduces dystrophic neurites in 5XFAD Alzheimer's disease model mice.

BACKGROUND: The hallmark lesions of the Alzheimer's disease (AD) brain are amyloid plaques consisting of the β-amyloid protein and neurofibrillary tangles comprised of hyperphosphorylated, aggregated tau protein, which both cause neuronal dysfunction and loss. One goal of neuroprotective therapies is to maintain normal neuronal function and survival in the presence of toxic pathologies such as plaques and tangles. A potential neuroprotective target is nuclear factor erythroid 2-related factor 2 (Nrf2) transcription factor, which regulates the expression of many antioxidant and detoxification g

Parvalbumin Neuron-Targeted Loss of Alzheimer's Disease Risk Gene BIN1 Is Insufficient to Drive Cognitive or Network Excitability Changes.

Bridging integrator 1 (BIN1) is one of the strongest genetic risk factors for Alzheimer's disease (AD), yet its function in the brain and role in AD remain unclear. Neuronal BIN1 isoform levels are decreased in AD, and recent data show an important role of BIN1 in inhibitory neurons. Inhibitory neurons are key regulators of cognition and network excitability, with parvalbumin-expressing (PV) neurons as the most abundant subtype. We tested the hypothesis that loss of BIN1 from PV neurons contributes to AD-related cognitive dysfunction and network hyperexcitability. We generated a cell type-spec

Tau Protein Aggregation Inhibitors-Therapeutic Strategy for Concurrent Tau and Amyloid Aggregation Inhibition.

Tau protein, a microtubule-associated protein widely distributed in the central nervous system, aggregates abnormally and forms neurofibrillary tangles in neurodegenerative diseases. Particularly in Alzheimer's disease, pathological tau protein aggregates disrupt the structure and function of neurons, triggering other neurodegenerative-related processes such as neuroinflammation and amyloid plaque formation, and finally leading to neuronal death. Several classes of drugs targeting neurofibrillary tangles have recently been studied, with tau protein aggregation inhibitors as a key research dire

Immunohistochemical Markers of Mitochondrial Electron Transport Chain Instability in Human Brain Regions: A Study of Aging and Alzheimer's Disease.

Expanding research indicates that oxidative stress, particularly mitochondrial oxidative stress, is one of the key components in the pathogenesis of Alzheimer's disease (AD). Mitochondrial oxidative stress is largely driven by impaired function of electron transport chain (ETC) complexes and their regulators. This study conducted an immunohistochemical analysis of ETC proteins (α-subunit of complex V, subunits MTCO1 and MTCO2 of complex IV) and mitochondrial complex V inhibitor IF-1 in the neurons of the caudate nucleus head, hippocampus, anterior cingulate gyrus, middle frontal gyrus, and inf
