AA and Parkinson's Disease.

Parkinson's disease (PD) is due to degeneration of dopaminergic neurons in substantia nigra pars compacta. PD neuropathology is characterized by the presence of Lewy bodies that contain aggregated α-synuclein, which has been implicated in a range of interactions with phospholipid membranes and free fatty acids. I propose that α-synuclein interacts with arachidonic acid (AA) to regulate the function of dopaminergic neurons. α-Synuclein sequesters AA, whereas AA induces the formation of oligomers that are alpha-helical; resistant to fibril formation; more prone to disaggregation, enzymatic diges

AA in Brain-Memory, Dementia, Depression, Anxiety, Alzheimer's Disease, Affective and Psychiatric Disorders, and Coronary Heart Disease (CHD).

The human brain is rich in AA and DHA, which are crucial for normal brain growth and development and memory. They form a major structural component of neurons of the cerebral cortex and photoreceptor cells in the retina. AA and DHA support synaptic plasticity and contribute to efficient neurotransmission. Adequate amounts of AA and DHA support neuronal growth and the formation of new synapses and thus are crucial for memory and cognitive function.Depression, anxiety, schizophrenia, and other neurological diseases occur more frequently in those with coronary heart disease (CHD) and vice versa.

Systematic Dissection of Key Driver Perturbation Signatures in Single Cells via ECCITE-seq.

CRISPR screens, such as expanded CRISPR-compatible cellular indexing of transcriptomes and epitopes by sequencing (ECCITE-seq), enable the simultaneous measurement of transcriptomes, gRNA identity, and cell-surface protein expression at single-cell resolution to systematically interrogate gene function. This platform provides a powerful and scalable experimental approach for validating disease-associated regulators identified by large-scale association studies and other computational methods, including network-based analyses of multi-omics data. Here, as an example application, we describe an

A Second Pathogenic Protein, PolyGN2C-iso2, Reveals a Dual-Protein Pathology in Neuronal Intranuclear Inclusion Disease.

BACKGROUND: Neuronal intranuclear inclusion disease (NIID) pathogenesis has been strongly linked to uN2CpolyG translated from NOTCH2NLC transcript variant 1. However, emerging evidence suggests that NOTCH2NLC transcript variant 2 may also generate a disease-relevant protein, PolyGN2C-iso2, but isoform-discriminating and antibody-independent evidence remains incomplete. METHODS: We characterize the aggregation propensity of PolyGN2C-iso2 in vitro, develop isoform-discriminating monoclonal antibodies for its detection in patient tissues, and perform targeted proteomic analysis of laser-microdis

Tau-induced mitochondrial reverse electron transport drives neurodegeneration.

Hyperphosphorylation and aggregation of tau are pathological hallmarks of tauopathies. Mitochondrial dysfunction is also a common feature of tauopathies. The mechanistic link between tau abnormalities and mitochondrial dysfunction and its relationship to the physiological function of tau, however, is unclear. Here, we demonstrate that tau regulates mitochondrial reverse electron transport (RET), which produces excess reactive oxygen species (ROS), reduces the NAD+/NADH ratio, and is activated by aging or stress. In flies, mice, and human induced pluripotent stem cell (hiPSC)-derived neurons, t

Astrocyte lipid metabolism: From physiological homeostasis to reactive phenotype plasticity in neurodegeneration.

Astrocytes are one of the most abundant types of glial cells in the central nervous system (CNS) and play pivotal roles in metabolic support, synaptic regulation, and neurotransmitter homeostasis. Astrocytic dysfunction is closely associated with the pathogenesis of neurodegenerative disorders such as Alzheimer's disease (AD), and lipid metabolism constitutes a central axis for maintaining astrocyte function and CNS homeostasis. Here, we provide a systematic review of recent advances in astrocyte lipid metabolism, summarizing the fundamental roles of astrocytes in the CNS and focusing on the b

Unraveling the role of lipid metabolism in ALS risk: a Mendelian randomization analysis using GWAS data.

BACKGROUND: Amyotrophic lateral sclerosis (ALS) is a progressive neurodegenerative disease. Lipid metabolism is closely related to neuronal function and energy homeostasis, but the genetic association between specific lipid species and ALS risk remains unclear. OBJECTIVE: This study aimed to investigate the potential causal associations between genetically predicted lipid species and ALS risk using a two-sample Mendelian randomization (MR) approach. METHODS: Summary-level GWAS data for 179 lipid species were obtained from 7,174 Finnish participants in the GeneRISK cohort. ALS GWAS data inclu

A Blood-Derived Factor Rescues ALS: Platelet Factor 4 Activates OPTN-Dependent Autophagy to Clear SOD1 Aggregates Independently of PINK1.

Peripheral factors that systemically regulate amyotrophic lateral sclerosis (ALS) have remained elusive-until now. Here, by integrating population-scale epidemiology with mechanistic dissection, we identify platelet factor 4 (PF4) as the central driver of a circulating neuroprotective axis that restores proteostasis and rescues ALS. In a prospective cohort of >500 000 UK Biobank participants, platelet indices were strongly associated with ALS risk, and serum PF4 levels were significantly reduced in ALS patients. Systemic administration of recombinant PF4 in hSOD1G93A mice produced dramatic the

Toward a Behavioral Reserve Model in Amyotrophic Lateral Sclerosis.

OBJECTIVE: Behavioral impairment is common in amyotrophic lateral sclerosis (ALS) and strongly affects autonomy, caregiver burden, and outcomes, yet predictors of vulnerability remain unclear. We investigated whether premorbid regulatory traits and socio-educational exposures are associated with behavioral phenotypes in ALS within a behavioral reserve framework. METHODS: We analyzed 965 consecutively assessed patients with ALS from a prospective tertiary-center cohort. Behavioral impairment was measured using the Frontal Systems Behavior Scale (FrSBe; family-rated before and after onset) and,

Genotype-phenotype relationship in a cohort of 131 Chinese patients with myotonic dystrophy type 1.

INTRODUCTION: Myotonic dystrophy type 1 (DM1) is an autosomal dominant dynamic mutation disorder characterized by myotonia and muscle weakness. The genotype-phenotype relationship has not been clearly defined in Chinese patients. METHODS: The clinical and genetic characteristics of 131 patients from 105 pedigrees were retrospectively analyzed. Triplet-primed Polymerase Chain Reaction (PCR) and Flanking PCR were performed to analyze the CTG repeats in DMPK gene. RESULTS: Among 131 DM1 patients, 64.9% were male, indicating a higher male admission rate, possibly due to limited diagnostic awaren

Modification of locomotion-respiration coupling in patients with obstructive sleep apnea syndrome.

INTRODUCTION: Locomotion is a complex motor task integrating sensory-motor and cognitive inputs through central pattern generator control. Locomotion-respiration coupling (LRC), defined as the synchronization between gait and breathing events, has been described during running and walking. Patients with obstructive sleep apnea syndrome (OSAS) exhibit altered gait patterns during wakefulness, which appear to be partially independent of nocturnal symptomatic treatment. While intermittent hypoxia may induce long-term ("fixed") central nervous system alterations, OSAS patients also display "revers

Learning global and local representations for generalized category discovery in neurodegenerative diseases.

BACKGROUND: Artificial intelligence (AI) has achieved notable success in closed-set classification of neurodegenerative diseases, but its performance degrades significantly in more realistic open-set settings, such as discovering rare or novel disease subtypes from unlabeled data that are not seen during training. PURPOSE: This study introduces, to our knowledge, the first framework for generalized category discovery (GCD) in neurodegenerative diseases using MRI. It aims to leverage labeled data from known categories to automatically identify novel categories within unlabeled datasets. METHO

From Structurally Diverse Sigma-1 (σ1) Receptor Reference Ligands to Live-Cell Probes: Fluorescent Phenoxyalkylpiperidines for the Detection of σ1 Receptor.

Sigma-1 (σ1) receptor is increasingly recognized as a 'pluripotent' chaperone mainly localized at the mitochondrial-associated endoplasmic reticulum membranes, where it contributes to the regulation of several pathophysiological pathways through a not fully undisclosed interactome. σ1 modulation with small molecules is progressively appearing promising against diverse pathologies that include neurodegenerative, oncology diseases and pain. However, only a deeper understanding of its mechanism of action may lead to σ1 full therapeutic exploitation and specific σ1 fluorescent tracers may help tow

Intelligent exogenous predictive networks for nonlinear Parkinson's disease model involving delay with effects of immunotherapy.

Artificial intelligence is used to simulate, solve, or approximate the dynamics of complex neurodegenerative diseases through accurate prediction. Parkinson's disease (PD) is a progressive disorder caused by the loss of dopaminergic neurons, leading to motor impairments such as muscle stiffness, tremors, and Bradykinesia. In this study, artificial intelligence-driven nonlinear autoregressive exogenous neural networks (AI-NARX-NN) are applied to the existing mathematical model reflecting the dynamics of innate as well as adaptive immune responses to PD under immunotherapy (IAIR-PDI). The IAIR-P

Liquid-liquid phase separation in neurodegenerative diseases: Mechanisms, pathological transitions, and therapeutic opportunities.

Liquid-liquid phase separation (LLPS) provides a biophysical framework for understanding biomolecular organization, yet its pathological relevance in neurodegeneration remains incompletely resolved. In this narrative review, we critically synthesize current evidence on LLPS in Huntington's disease, Parkinson's disease, Alzheimer's disease, and amyotrophic lateral sclerosis, focusing on mutant huntingtin, α-synuclein, amyloid-β, tau, TDP-43, and FUS. We first outline the molecular principles-including intrinsic disorder, multivalent interactions, and environmental sensitivity-that govern their

Repurposing Ionophores to Tackle Antimicrobial Resistance with a Focus on PBT2.

Antimicrobial resistance (AMR) remains a growing global health challenge. While resistant bacterial strains continue to emerge and spread, the development of new antibiotics has remained slow. This gap has increased interest in drug repurposing as a promising strategy to combat multidrug-resistant (MDR) pathogens. Ionophores are a structurally diverse group of natural and synthetic compounds that transport ions across biological membranes. Many ionophores were originally developed and widely used for the prevention and treatment of coccidiosis, a common protozoan parasitic intestinal disease i

Receptor-Specific ER-Phagy in Neurodegenerative Diseases: Molecular Mechanisms and Emerging Therapeutic Opportunities.

Neurodegenerative diseases (NDs) constitute a major global health burden driven by rising life expectancy and the lack of disease-modifying therapies. Central to these progressive pathologies is the accumulation and aggregation of conformationally altered proteins, categorizing them as 'proteinopathies'. These aberrant aggregates disrupt cellular homeostasis and trigger chronic stress responses that drive progressive neuronal dysfunction and loss. The endoplasmic reticulum (ER) is a dynamic organelle defined by remarkable structural diversity and functional plasticity, that preserves cellular

Microglia constitute a major innate defense mechanism against prion infection; however, the molecular pathways regulating microglial responses during disease progression remain incompletely understood. Galectin-3 (Gal3), a β-galactoside-binding lectin associated with reactive microglia in multiple neurodegenerative disorders, has been implicated in phagocytosis, inflammatory signaling, and lysosomal homeostasis. Here, we investigated the role of Gal3 in prion disease pathogenesis using prion-infected mice. Gal3 expression was undetectable in healthy brain but became upregulated beginning at la
