Bioinformatic analysis of differentially expressed mitochondrion-related genes, immune cell infiltration, and diagnostic value in Alzheimer's disease.

BACKGROUND: Mitochondrial dysfunction and neuroinflammation are critically implicated in the pathogenesis of Alzheimer's disease (AD). However, a systematic exploration of key mitochondrion-related genes (MRGs) in AD, and their specific roles in reshaping the immune microenvironment and serving as diagnostic biomarkers, remains insufficient. METHODS: To address this, we conducted an integrative bioinformatics analysis. Differentially expressed MRGs were identified from public AD transcriptomic datasets. Their biological functions were elucidated through enrichment analyses. The correlations b

The association between social determinants of health and Parkinson's disease prevalence among U.S. adults: A cross-sectional study.

BACKGROUND: Social determinants of health (SDOH) encompass the economic, social, and environmental conditions that shape health outcomes. While SDOH are increasingly recognized as fundamental drivers of chronic disease, their relationship with Parkinson's disease (PD) remains poorly characterized. OBJECTIVE: To examine the association between individual and cumulative adverse SDOH and PD prevalence in a nationally representative sample of U.S. adults. METHODS: We conducted a cross-sectional analysis of 24,018 adults from the 2003-2016 National Health and Nutrition Examination Survey. A cumul

An Icelandic pangenome reference.

Reference bias is an issue that affects most genomic studies analysing short reads mapped to a reference genome1,2. It can be mitigated by mapping to multiple haplotypes represented in a pangenome3-5. Here we introduce two new methods to address reference bias: Emblask for pangenome construction and Weaver for mapping to pangenomes at scale. Emblask is a hybrid long- and short-read haplotype-resolved dual assembly pipeline for parent-offspring trio data. Using Emblask, we assembled 698 Icelandic haplotypes and added them to the Human Pangenome Reference Consortium (HPRC) pangenome4 to construc

Astragalin confers coordinated neuroprotection in Parkinson's disease models through modulation of neurotrophic, anti-inflammatory, and synuclein-related pathways.

Parkinson's disease (PD) remains a neurodegenerative disorder without effective disease-modifying therapies, largely due to its multifactorial pathogenesis. We report that the natural flavonoid Astragalin (AST) concurrently addresses three core pathological processes in PD, namely dopaminergic neuron degeneration, α-synucleinopathy, and neuroinflammation, through coordinated modulation of interconnected molecular pathways. In both subacute MPTP- and chronic rotenone-induced murine PD models, AST preserved 85% of nigral tyrosine hydroxylase-positive neurons, fully prevented motor deficits, and

Helicobacter pylori eradication status is associated with lower motor severity despite similar levodopa pharmacokinetics in Parkinson's disease.

BACKGROUND: Helicobacter pylori (H. pylori) infection has been associated with impaired levodopa absorption and worse motor symptoms in Parkinson's disease (PD). However, the relationships among H. pylori infection, eradication history, levodopa pharmacokinetics, and motor severity remain unclear. METHODS: We retrospectively analyzed 40 patients with PD who underwent standardized levodopa/carbidopa pharmacokinetic testing. Participants were classified as HP-positive, never infected, or prior eradication. Plasma levodopa and carbidopa concentrations were measured serially after administration

Predicting biological age using an accumulated neurotoxicity biomarker for amyloid beta oligomers.

This study proposes using accumulated neurotoxicity, defined as the time integral of Aβ oligomer concentration, as a biomarker for neuronal aging. A relationship between biological age and accumulated neurotoxicity is proposed. Numerical analysis guided the development of a new analytical solution linking the biological and calendar ages of neurons. The effects of Aβ monomer and oligomer half-lives, key indicators of proteolytic efficiency, on biological age are examined. Both constant and age-dependent (exponentially increasing) half-life scenarios are considered. The findings indicate that i

A single-cell meta-analysis evidences transposable element dysregulation in sex-based differences in Parkinson's disease.

Transposable elements (TEs) (mobile genetic elements comprising ∼45% of the human genome) have recently emerged as potential contributors to Parkinson's disease (PD); however their role and sex-specific impact remain poorly understood. Here, we present the first integrative meta-analysis of TE expression across 4 substantia nigra single-nucleus RNA-seq datasets, comprising a total of 66 donors, generating a cell-type-resolved atlas of TE dysregulation in PD. We identified widespread TE activation across major brain cell types (i.e. neurons, astrocytes, oligodendrocytes and microglia), with mar

Dissecting the Lewy body.

How Lewy pathology forms and diversifies remains a central question in Parkinson's disease research. A recent study by Mahul-Mellier and Colleagues proposes a dynamic model in which inclusion maturation and organelle remodeling drive pathological heterogeneity, reshaping concepts of disease mechanisms and providing new perspectives on therapeutic approaches.

Fatigue and fatigability in Parkinson's disease: a neurophysiological perspective.

Fatigue is one of the most frequent and disabling non-motor symptoms in Parkinson's disease (PD). Its multidimensional nature and overlap with related symptoms contribute to diagnostic and therapeutic challenges. A distinction has been proposed between fatigue, defined as the subjective perception of lack of energy and fatigability, defined as an objective decline in performance during sustained activity. However, their underlying pathophysiological mechanisms are still unclear. We conducted a narrative review of studies investigating PD-related fatigue and fatigability with neurophysiological

Targeting astrocyte-mediated neurotoxicity induced by ALS/FTD-associated RNA binding proteins.

BACKGROUND: Amyotrophic lateral sclerosis (ALS) and frontotemporal dementia (FTD) are neurodegenerative disorders characterized by reactive astrocytes that contribute to neuronal injury through TAR DNA-binding protein 43 (TDP-43)-or fused in sarcoma (FUS)-driven neuroinflammatory signaling. Dehydrocostus lactone (DHE), a blood-brain barrier-permeable sesquiterpene lactone with established anti-inflammatory activity, represents a promising but unexplored therapeutic candidate for ALS/FTD. METHODS: The therapeutic effects of DHE were evaluated in primary mouse and human astrocytes expressing AL

A dual-function guanidinium scaffold for copper sequestration and redox protection in Wilson disease models.

Wilson disease (WD) is caused due to mutations in the copper ATPase gene ATP7B, resulting in accumulation of copper and the consequent disruption of cellular redox balance through reactive oxygen species generation. Current therapies mainly depend on copper chelation to lower metal burden which sometimes also strip copper from cuproproteins and disturb key physiological copper-dependent processes. It also does not directly suppress pathological copper reactivity i.e., free radical generation, a major driver of WD progression. To overcome these limitations, we have rationally designed Gua-Cu-3,

Melanoma in an Unlikely Guise: Diagnostic Challenges of a Solid-cystic Popliteal Mass in Neurofibromatosis Type 1.

Malignant melanoma (MM) uncommonly presents as a solid-cystic soft tissue mass, posing a significant diagnostic challenge. We report a rare case in a 63-year-old woman with neurofibromatosis type 1 (NF-1) who presented with a painful, rapidly progressive swelling on the posterior aspect of her right lower thigh. Imaging revealed a large, well-defined solid-cystic lesion suggestive of a soft tissue sarcoma or malignant peripheral nerve sheath tumor. Initial fine-needle aspiration cytology was nondiagnostic. Definitive diagnosis was achieved following wide surgical excision, with histopathologic

Respiratory function in children and young adults affected by neurofibromatosis type 1 treated with selumetinib.

BACKGROUND: Plexiform neurofibromas (PNs) are benign tumors that can cause pain, disfigurement, and disability in patients with neurofibromatosis type 1. Selumetinib, a MEK1/2 inhibitor, is the first drug who showed efficacy in inducing PNs shrinkage, reducing the associated morbidities in pediatric patients. Overall, selumetinib showed a good safety profile. However, multiple adverse effects have been reported both in pediatric and adult patients, including respiratory ones. Serial spirometry is suggested in patients with thoracic neurofibromas to detect changes that may underly PN's modifica

CSD-GCN: Cross-Modal Self-Distillation Graph Convolutional Network for Alzheimer's Disease Diagnosis.

Alzheimer's disease (AD) is a prevalent neurodegenerative disorder that demands more comprehensive and precise modeling to unravel its complex pathological mechanisms and multidimensional brain network abnormalities. However, current brain network analysis approaches often suffer from limitations such as single-modal data processing and shallow fusion strategies, which hinder their ability to capture the multi-scale connectivity patterns associated with AD effectively. To alleviate these limitations, a novel multi-modal fusion framework is proposed in this paper, namely a cross-modal self-dist

Fluorescence lifetime imaging of G-quadruplex RNA dynamics in Alzheimer's disease using a novel nucleic acid-sensitive probe.

Real-time monitoring of G-quadruplex (G4) RNA dynamics is vital for understanding their biological roles in the progression and treatment of Alzheimer's disease. However, this remains challenging due to two key difficulties. First, current fluorescent probes lack sufficient selectivity for G4 RNA over G4 DNA and other non-G4 secondary structures in live-cell competitive environments. Second, fluorescence intensity-based imaging cannot detect subtle changes in G4 RNA because of variations in fluorophore uptake and photobleaching. Herein, we report a novel thiazole orange derivative (TOGR) for f

Fluorescence imaging-based investigation of tau-mediated promotion of U1 small nuclear ribonucleoprotein 70 kDa co-phase separation.

Neurofibrillary tangles formed by Tau protein are a core pathological hallmark of Alzheimer's disease (AD). Although liquid-liquid phase separation (LLPS) of Tau is thought to induce its aggregation, and the basic-acidic dipeptide (BAD) domain-containing protein U1 small nuclear ribonucleoprotein 70 kDa (U1-70K) abnormally co-localizes with Tau tangles in AD brains, the molecular mechanism underlying their co-aggregation remains unclear. In this study, we found that Tau co-phase separates with two low-complexity domains (LC1/LC2). Both LC1 and LC2 promote the gelation of Tau droplets and the f

EA-DualGAT: An error-aware dual-graph attention framework for Parkinson's disease diagnosis from wearable plantar pressure gait signals.

Timely diagnosis of Parkinson's disease (PD) is essential for timely intervention and effective disease management. Wearable sensing technologies provide objective gait measurements and have become a promising, low-cost solution for automated PD diagnosis. However, existing deep learning and graph-based methods primarily focus on individual gait representations or fixed graph structures, often overlooking relationships among gait samples, difficult samples, and adaptive graph refinement during training. To address these limitations, this study proposes EA-DualGAT, an Error-Aware Dual Graph Att

Nicotine modulates α7nAChR associated ceramide-mitochondrial stress responses in Parkinsonian models.

Parkinson's disease (PD) is characterized by progressive dopaminergic degeneration accompanied by mitochondrial dysfunction and oxidative stress, yet the metabolic mechanisms underlying these pathological changes remain incompletely understood. Here, we investigated whether nicotine preserves dopaminergic integrity by modulating ceramide homeostasis through α7 nicotinic acetylcholine receptor (α7nAChR) signalling. In an MPTP mouse model, nicotine improved motor and exploratory deficits, preserved striatal tyrosine hydroxyl positive fibres, and selectively reversed ceramide accumulation in the

APOE ε4 genotype-dependent reduction of amyloid-related imaging abnormality in real-world lecanemab use: Implications for risk-stratified prescribing.

Real-world amyloid-related imaging abnormality (ARIA) rates with lecanemab are consistently lower than in the Clarity AD trial, yet whether this reduction is genotype-specific is unknown. We compared published APOE ε4-stratified ARIA-E and ARIA-H rates from Clarity AD with a US real-world cohort and Japanese post-marketing surveillance. ARIA rates were largely unchanged in APOE ε4 noncarriers, whereas reductions were concentrated among carriers, especially homozygotes. This pattern was observed for both ARIA subtypes across both settings. These early real-world findings suggest that the appare

Model-based ordering of the ALPS index among Alzheimer's disease biomarkers.

Glymphatic dysfunction has been implicated in Alzheimer's disease (AD), but its position relative to amyloid and tau biomarkers remains unclear. We examined the relative position of the diffusion tensor image analysis along the perivascular space (ALPS) index within the AD biomarker cascade using Alzheimer's Disease Neuroimaging Initiative data. Amyloid PET, tau PET, plasma amyloid-β 42/40 ratio, plasma phosphorylated-tau 217, and ALPS index were analyzed with a data-driven event-based model. The ALPS index was ranked between plasma amyloid-β 42/40 alteration and amyloid PET positivity in the
