Alcohol-induced structural and cellular brain alterations: molecular and histopathological mechanisms.

Chronic alcohol consumption is a leading cause of acquired neurodegeneration with well-documented structural and ultrastructural brain alterations. This review analyzes the cellular and molecular mechanisms underlying alcohol neurotoxicity, integrating findings from animal models, human post-mortem studies, and neuroimaging investigations. Ethanol crosses the blood-brain barrier and generates toxic metabolites including acetaldehyde and reactive oxygen species, triggering oxidative stress, lipid peroxidation, and mitochondrial dysfunction. Chronic exposure induces glutamatergic and gamma-amino

Functional and Structural Characterization of a Large Animal Model of RDH5-Associated Retinopathy.

PURPOSE: Inherited retinal diseases are a group of hereditary diseases that cause variable levels of blindness and affect a multitude of adults and children. One such disease is fundus albipunctatus (FA). FA is caused by autosomal recessive retinol dehydrogenase 5 (RDH5) mutations and results in rod dysfunction leading to night blindness and, in a subset of patients, macular degeneration (MD). We previously reported a spontaneous feline model of FA due to an RDH5 missense mutation. The affected cats showed rod dysfunction and a proportion developed degeneration of the area centralis (AC), equi

Opposing Roles of CYP2E1 and ALDH2 in Binge Alcohol-Mediated Cerebellar Damage.

To explore the opposing roles between the toxic acetaldehyde-producing enzyme Cytochrome P450-2E1 (CYP2E1) and acetaldehyde-clearing protein mitochondrial Aldehyde Dehydrogenase-2 (ALDH2) in alcohol-mediated cerebellar damage, two separate experiments were conducted. Young Svj/129 wild-type (WT) vs. Cyp2e1-Knockout (KO) mice were administered consecutive doses of 5 g/kg/dose ethanol (EtOH) at 12 h intervals, and C57BL/6J WT vs. Aldh2-KO mice were administered 4 g/kg/dose EtOH at 12 h intervals. Control mice were given dextrose, and cerebella were collected one hour after the last ethanol dose.

Noradrenergic infraslow rhythm during sleep is the critical link between heart-rate dynamics and memory consolidation.

Recent work shows that the brain's arousal system remains active during sleep, with rhythmic locus coeruleus (LC) activity shaping sleep architecture and supporting memory consolidation. The LC releases norepinephrine (NE) in infraslow (~0.02 Hz) bouts that gate NREM sleep spindles. Here, we demonstrate that heart rate (HR) fluctuations during NREM are tightly phase-locked to these NE rhythms, identifying the LC as a key driver of very-low-frequency HR variability (VLF-HRV), an understudied autonomic signal. Using optogenetics, transient LC inhibition blunts HR slowing, whereas LC activation p

Loss of endogenous tau suppresses APOE4-induced patterned behavioral decline and axon dysmorphia in a model of Alzheimer's disease.

Alzheimer's disease (AD) causes a characteristic spatiotemporal pattern of neurodegeneration. The factors that account for this pattern of degeneration, including associated neuronal dysfunction and dysmorphia, are unclear. Previously, we generated a model of AD using the nematode Caenorhabditis elegans with the AD risk variant of apolipoprotein E (APOE4). We showed that the soma of HSN class neurons degenerate in early-adult animals. Here, we performed behavioral analyses to deduce the effect of APOE4 on the function of distinct neuronal circuits. We found evidence that APOE4 induces dysfunct

6-gingerol alleviates Alzheimer's disease in rats through the Pumilio RNA binding family member 1/microRNA-340-5p/pleomorphic adenoma gene like-2/estimated glomerular filtration rate/hypoxia-inducible factor-1alpha/2alpha pathway.

This study explores the modulatory role of Pumilio RNA-binding family member 1 (circ_PUM1) in the neuroprotective mechanisms of 6-gingerol against Alzheimer's disease (AD)-related neuronal injury, providing new insights into circRNA-mediated regulation of neurodegeneration. Variations in circRNA expression indirectly influenced by 6-gingerol (6-GIN) were comprehensively assessed within AD experimental frameworks. The AD phenotype was not absent when hippocampal amyloid-β (Aβ) was introduced into rats, nor when SK-N-SH and SK-SY5Y cells were subjected to Aβ challenge, jointly constructing compl

Fingolimod Effects on Motor Function and BDNF-TrkB Signaling in a Huntington's Mouse Model Are Disease-Stage-Dependent.

Huntington's Disease (HD) is characterized by prominent degeneration of the principal neurons of the striatum and by progressive motor and cognitive deterioration. Striatal neurons degenerate in HD due to multiple cell-autonomous and non-autonomous factors. Impaired neurotrophin signaling by brain-derived neurotrophic factor (BDNF) and its cognate receptor Tropomyosin receptor kinase B (TrkB) is an important mechanism underlying neuronal loss in HD. Fingolimod, a clinically approved oral drug for Multiple Sclerosis, was originally developed based on its anti-inflammatory properties. Recent wor

Association between leukocyte telomere length and neurodegenerative diseases: a prospective cohort in the UK Biobank.

BACKGROUND: This study aims to investigate the association between leukocyte telomere length (LTL) and the risk of incident NDDs using a large-scale cohort from the UK Biobank. METHODS: Data from 459,902 subjects were analyzed using Cox proportional hazards models and machine learning (ML) algorithms to assess LTL's association with NDD risk. RESULTS: Shorter LTL was associated with an increased risk of NDDs, including Alzheimer's disease (HR: 0.52, 95% CI 0.40-0.67, P < 0.001), dementia in AD (HR: 0.53, 95% CI 0.39-0.73, P < 0.001), unspecified dementia (HR: 0.74, 95% CI 0.58-0.95, P < 0.05

Neuroimmunology of Oral Cancer: Tumor Progression, Systemic Inflammation, and Neural Circuit Dysfunction from the Cerebellum to the Frontal Cortex.

Oral cancer, predominantly oral squamous cell carcinoma, ranks among the ten most common malignancies worldwide and carries a disproportionate burden across South and South-East Asia, where tobacco, betel quid, areca nut, and alcohol use remain widespread. Although conventionally framed as a localised disease, accumulating evidence indicates that oral cancer may behave as a systemic condition with under-recognised neuroimmunological consequences. This narrative review synthesises current literature on the pathways through which tumour-driven inflammation extends beyond the primary site to infl

Regenerating Locus Coeruleus-Norepinephrine (LC-NE) Function: A Novel Approach for Neurodegenerative Diseases.

Pathological changes in the locus coeruleus-norepinephrine (LC-NE) neurons, the major source of norepinephrine (NE, also known as noradrenaline) in the brain, are evident during the early stages of neurodegenerative diseases (ND). Research on both human and animal models have highlighted the therapeutic potential of targeting the LC-NE system to mitigate the progression of ND and alleviate associated psychiatric symptoms. However, the early and widespread degeneration of the LC-NE system presents a significant challenge for direct intervention in ND. Recent advances in regenerative cell therap

Novel Tissue-Specific Multifunctionalized Nanotechnological Platform Encapsulating Riluzole Against Motor Neuron Diseases.

BACKGROUND: Motor neuron diseases are neurological disorders characterized by progressive degeneration of upper and/or lower motor neurons. Amyotrophic Lateral Sclerosis (ALS) is the most common form of motor neuron diseases, where patients suffer progressive paralysis, muscle atrophy and finally death. Despite ALS severity, no treatment is safe and fully effective. In this area, Riluzole was the first drug approved and it constitutes the gold-standard for this pathology. However, to obtain suitable therapeutic efficacy, Riluzole requires high doses that are associated with severe adverse effe

PNPLA6 regulates retinal homeostasis by choline through phospholipid turnover.

Although mutations in human patatin-like phospholipase PNPLA6 are associated with hereditary retinal degenerative diseases, its mechanistic action in the retina is poorly understood. Here, we uncover the molecular mechanism by which PNPLA6 dysfunction disturbs retinal homeostasis and visual function. PNPLA6, by acting as a phospholipase B, regulates choline mobilization from phosphatidylcholine and subsequent choline turnover for phosphatidylcholine regeneration in retinal pigment epithelial cells. PNPLA6-driven choline is supplied from retinal pigment epithelial cells to adjacent photorecepto

Loss of excitatory inputs and decreased tonic and evoked activity of locus coeruleus neurons in aged P301S mice.

Tau pathology in the locus coeruleus (LC) is associated with several neurodegenerative conditions including Alzheimer's disease and frontotemporal dementia. Phosphorylated tau accumulates in the LC and results in inflammation, synaptic loss, and eventually cell death as the disease progresses. Loss of LC neurons and noradrenergic innervation is thought to contribute to the symptoms of cognitive decline later in disease. While loss and degeneration of LC neurons has been well studied, less is known about changes in LC physiology at advanced stages of tau pathology that precedes neurodegeneratio

Neuroglia pathology in genetic and epigenetic disorders of the central nervous system.

Glial cells are increasingly recognized for their important interactions with both developing and mature neurons, in particular for maintenance of dendritic ramifications and spines, synapses, and neurotransmitter uptake. MicroRNA abnormalities are demonstrated in individual astrocytes with alterations in neurological diseases. Alexander disease is a prototype astrocytic disease because of genetically altered glial fibrillary acidic protein (GFAP) filaments. Other genetic diseases are now recognized as involving glial cells in their pathogenesis: Rett, Fragile-X, Aicardi-Goutières, and Down sy

Noradrenergic signaling controls Alzheimer's disease pathology via activation of microglial β2 adrenergic receptors.

Norepinephrine (NE) is a potent anti-inflammatory agent in the brain. In Alzheimer's disease (AD), the loss of NE signaling heightens neuroinflammation and exacerbates amyloid pathology. NE inhibits surveillance activity of microglia, the brain's resident immune cells, via their β2 adrenergic receptors (β2ARs). Here, we investigate the role of microglial β2AR signaling in AD pathology in the 5xFAD mouse model of AD. We found that loss of cortical NE projections preceded the degeneration of NE-producing neurons and that microglia in 5xFAD mice, especially those microglia that were associated wi
