AA and Parkinson's Disease.

Parkinson's disease (PD) is due to degeneration of dopaminergic neurons in substantia nigra pars compacta. PD neuropathology is characterized by the presence of Lewy bodies that contain aggregated α-synuclein, which has been implicated in a range of interactions with phospholipid membranes and free fatty acids. I propose that α-synuclein interacts with arachidonic acid (AA) to regulate the function of dopaminergic neurons. α-Synuclein sequesters AA, whereas AA induces the formation of oligomers that are alpha-helical; resistant to fibril formation; more prone to disaggregation, enzymatic diges

AA in Brain-Memory, Dementia, Depression, Anxiety, Alzheimer's Disease, Affective and Psychiatric Disorders, and Coronary Heart Disease (CHD).

The human brain is rich in AA and DHA, which are crucial for normal brain growth and development and memory. They form a major structural component of neurons of the cerebral cortex and photoreceptor cells in the retina. AA and DHA support synaptic plasticity and contribute to efficient neurotransmission. Adequate amounts of AA and DHA support neuronal growth and the formation of new synapses and thus are crucial for memory and cognitive function.Depression, anxiety, schizophrenia, and other neurological diseases occur more frequently in those with coronary heart disease (CHD) and vice versa.

Systematic Dissection of Key Driver Perturbation Signatures in Single Cells via ECCITE-seq.

CRISPR screens, such as expanded CRISPR-compatible cellular indexing of transcriptomes and epitopes by sequencing (ECCITE-seq), enable the simultaneous measurement of transcriptomes, gRNA identity, and cell-surface protein expression at single-cell resolution to systematically interrogate gene function. This platform provides a powerful and scalable experimental approach for validating disease-associated regulators identified by large-scale association studies and other computational methods, including network-based analyses of multi-omics data. Here, as an example application, we describe an

Exercise attenuates obesity-related cognitive and sleep-circadian dysfunctions by attenuating neuroinflammation via JAK/STAT in sex and age specific manner.

BACKGROUND: Obesity including genetic obesity accelerates age-related cognitive decline and disrupts circadian regulation, increasing the risk of neurodegenerative disorders. These effects are mediated in part by metabolic dysregulation, neuroinflammation, and altered sleep-circadian cycles, yet the molecular and lifestyle factors that modify obesity-associated brain dysfunction remain poorly defined. OBJECTIVES: We investigated whether exercise mitigates metabolic, cognitive, and circadian dysfunction associated with genetic obesity, and whether exercise responses differ by sex and age. MET

SPECT hybrid approach combining conventional and deep features for early detection of Parkinson's disease.

The early diagnosis of Parkinson's disease (PD) using SPECT imaging continues to be challenging due to the subtle dopaminergic deficits present in the early stages of the disease. This study proposes a novel hybrid approach combining conventional and deep learning features to improve PD classification, and applies it to reclassify scans without evidence of dopaminergic deficit (SWEDD) cases. We used SPECT images from early PD patients and healthy controls (HC) and extracted SBR metrics, morphometric, and deep learning features. Our multi-stage feature selection pipeline employed near-zero vari

Predicting disease spread from host movement data: Chronic wasting disease in North America as a case study.

Rare long-distance movements can increase the spatial spread of invasive species and shifts in species ranges. For wildlife disease spread, however, seasonal migrations may only matter if they lead to transmission to individuals using different seasonal ranges, and the importance of juvenile dispersal depends on whether those juveniles are infected before they disperse. High-resolution animal movement data are increasing, but it is unclear how to leverage these data to improve predictions of future disease spread particularly in regions without pre-existing movement data. We predicted the spee

Irisin-integrin αV/β5 coupling of α-synuclein phagocytosis and clearance.

Parkinson's disease-associated cognitive impairment (PD-CI) is closely linked to α-synuclein (α-syn) accumulation and synaptic dysfunction, yet effective disease-modifying strategies remain limited. Irisin is an exercise-inducible myokine with neuroprotective potential, but its receptor mechanisms and its role in α-syn clearance in PD-CI are poorly defined. Here, we observed that aerobic exercise markedly increased circulating irisin levels, reduced serum α-syn levels, and improved cognitive performance in a cohort of 21 PD patients. In addition, irisin signals through integrin αV/β5 to enhanc

Combined Olfactory Testing and Substantia Nigra Hyperechogenicity for Diagnostic Differentiation of Parkinson's Disease.

INTRODUCTION: Olfactory impairment and substantia nigra (SN) hyperechogenicity are well-recognized features of Parkinson's disease (PD). Their complementary diagnostic value and demographic influences warrant further clarification. This study aimed to assess the relationship and combined diagnostic value of SN hyperechogenicity and olfactory dysfunction in patients with clinically diagnosed PD. METHODS: We conducted a cross-sectional study including 89 PD patients and 113 healthy controls. Olfactory function was evaluated using the 16-item Sniffin' Sticks test, and SN echogenicity was measure

Identification of senescence-related genes in Parkinson's disease reveals candidate therapeutic targets and pathological processes.

BACKGROUND: Parkinson's disease (PD), a progressive neurodegenerative disease, is closely linked to the aging process; however, its precise molecular mechanisms remain unclear. This work aimed at clarifying the function of senescence-related genes (SRGs) in PD and investigating their molecular underpinnings, as well as their potential diagnostic and therapeutic significance. METHODS: By integrating bioinformatics with experimental validation, the CellAge database provided 278 SRGs for this study. A total of 2925 differentially expressed genes (DEGs) and 31 differentially expressed SRGs (DESRG

Gene Correction Enhances Dopaminergic Cell Therapy in a Nonhuman Primate Model of Parkinson's Disease.

Human induced pluripotent stem cell (iPSC)-derived dopaminergic progenitors offer a promising strategy for cell replacement in Parkinson's disease (PD), yet their long-term efficacy and safety in primates remain underexplored. Here, we generated isogenic iPSC lines from a PD patient carrying compound LRRK2 mutations (R50H and M2397T) and used CRISPR/Cas9 and prime editing to correct these variants. Both mutant and gene-corrected lines were differentiated into midbrain dopaminergic progenitors (DA-NPCs), characterized in vitro, and transplanted bilaterally into the striatum of MPTP-lesioned cyn

A coarse-to-fine machine-learning framework for identifying functional connectivity markers of cognitive impairment in Parkinson's disease.

BACKGROUND: Resting-state functional MRI (rs-fMRI) has been applied to investigate cognitive impairment (CI) in Parkinson's disease (PD). Nevertheless, reported functional connectivity (FC) alterations remain heterogeneous, partly due to reliance on linear analytical approaches and limited validation across datasets. OBJECTIVE: To develop a machine-learning framework for identifying generalizable FC markers of CI in PD. METHODS: Rs-fMRI data were obtained from an online cohort (for model training) and an independent local cohort (for external validation) of individuals with PD. Subjects were

The role of microglia glucose metabolism reprogramming in Alzheimer's disease treatment.

Alzheimer's disease (AD) is a neurodegenerative disorder characterized by progressive cognitive impairment, and there remains a lack of effective treatments capable of reversing or significantly slowing disease progression. Accumulating evidence indicates that the immune function of microglia, the resident immune cells of the central nervous system, is a critical factor in regulating AD pathogenesis. Emerging research in immunometabolism further reveals that glucose metabolic reprogramming serves as a central driver of microglial phenotypic and functional differentiation. This review systemati

Fear-related psychological experiences in people with Parkinson's disease: a systematic review of qualitative studies.

BACKGROUND: Fear is a multifaceted and pervasive phenomenon among people with Parkinson's disease. Although some qualitative studies have explored fear-related psychological experiences in this population, there remains a lack of comprehensive integration of these insights. This study aimed to synthesize existing qualitative research on fear-related emotional experiences in people with Parkinson's disease, and to provide a theoretical basis for developing effective psychological interventions in clinical practice. METHODS: A comprehensive computer-based search was conducted across 11 database

Muscular torque variability in Parkinson's Disease with and without dopaminergic medication.

We examined whether dopaminergic medication improves sustained muscular torque/force steadiness in Parkinson's disease (PD). Ten individuals with PD performed 40 second isometric ankle dorsiflexion contractions at 10%, 20%, and 30% maximum voluntary torque (MVT) in OFF- and ON-medication states. Torque variability was quantified using the coefficient of variation (CoV). In the OFF-medication state, torque CoV increased progressively over time, indicating reduced force steadiness during sustained contraction. Medication significantly reduced overall CoV. Higher contraction intensities were asso

Key circuits through which sleep affects memory and their role in Alzheimer's disease.

Sleep is a critical physiological process for maintaining the cognitive function of brain, particularly about memory consolidation. This review systematically elaborates on the multiscale regulatory mechanisms of the sleep-wake cycle, with a focus on the neural circuits involved in sleep-dependent memory consolidation, such as the hippocampus-prefrontal-thalamus network, characteristic neural oscillations, and microglia-mediated synaptic changes. Building upon these foundations, the article delves into Alzheimer's disease (AD), providing an in-depth analysis of the complex bidirectional relati

Exploring Complex Genetic Mechanisms in Brain Imaging Genetics via a New Multi-Task Learning Method.

Brain imaging genetics generally combines genotype data with brain structure and functional measures to investigate the genetic basis of neurological disorders. Multimodal brain imaging data carry different but complementary information, which can clearly measure the same brain. Multi-task sparse canonical correlation analysis (MTSCCA) is often used to analyze the bi-multivariate correlation between genotype data and multimodal imaging features; however, it lacks the capacity to identify class-specific biomarkers and to deeply capture complex genetic mechanisms. In this paper, a linear discrim

Presenilin-1 controls glycolysis and identity of pancreatic beta cells.

Presenilin-1 (PS1) is an endoplasmic reticulum protein, most known for its role in pathogenesis of familial Alzheimer's Disease (AD). PS1 has been attributed roles in intracellular calcium homeostasis in the brain, as well as in the pancreatic beta cells, where it has been shown to be fundamental for the initial phase of glucose-induced insulin secretion. To gain mechanistic insight into beta cell autonomous function of PS1, we have examined various beta cell models and found that PS1 controls glycolytic flux of pancreatic beta cells by regulating sub-mitochondrial Ca2+ homeostasis.
