Solvent and Temperature Dependent Selectivity in the [4 + 2] Annulation-Initiated Cascade Reactions of Nitroindoles with Curcumins: Access to Linear and Angular Polycycles.

A metal-free dearomatization of nitroindole via Michael addition of curcumin in the presence of tetramethylguanidine (TMG) followed by further solvent-dependent cascade intramolecular Michael additions generates tetrahydrocarbazoles and hexahydropyranocarbazolones with high diastereoselectivity, which was rationalized by DFT calculations. Furthermore, an unprecedented and conceptually novel in situ skeletal rearrangement of the tetrahydrocarbazole at elevated temperature leading to angular heterotriquinanes, which exhibit promising photophysical properties, is disclosed. A non-natural analogue

Design of Wheat Protein-Derived Keap1-Targeting Antioxidant Peptides: Integrating Intelligent Screening, ProteinMPNN-Guided Sequence Optimization, and TAT-Fused Delivery.

Wheat gliadin-derived peptides show promising antioxidant potential, but their target-guided discovery remains challenging. Here, we established a computation-driven pipeline for Keap1-targeting antioxidant peptides. From 2798 virtual oligopeptides generated using our Peptide_MDI platform, KCENEECTFP and RELVNSTCFA were selected through multitier screening and molecular dynamics simulations. KCENEECTFP exhibited antioxidant activity and modulated the Keap1-Nrf2 pathway in H2O2-stressed IPEC-J2 cells. Structure-guided ProteinMPNN optimization yielded SCDCPSCLYP, which showed enhanced radical sc

How a Single-Chain Polystyrene Nanoparticle Dictates α-Synuclein Adsorption and Intramolecular β-Sheet Nucleation.

A growing number of studies have detected nanoplastics, particularly polystyrene (PS) nanoparticles, in human blood and various critical tissues. Nevertheless, the molecular mechanisms by which their physical size modulates the aggregation of disease-related proteins remain poorly understood. Here, using well-tempered metadynamics simulations, we demonstrate that single-chain PS nanoparticles with a degree of polymerization N collapse into spherical globules in water, following a scaling law of radius of gyration Rg ∼ N1/3, and that their size critically determines the conformational fate of a

Altered Membrane Protein Abundance in the Cerebral Cortex of the JNPL3 Mouse Model of Tauopathy.

Developing effective therapeutic approaches for tau-associated neurodegenerative diseases, including frontotemporal dementia and Alzheimer's disease, remains extremely challenging due to their complex pathogeneses. Membrane proteins constitute a large proportion of current therapeutic and drug delivery targets because of their key roles in essential cellular processes. Here, we examined alterations in membrane protein abundances in the prefrontal cortex of male and female mouse models of human tauopathy, JNPL3 mice, compared with sex-matched wild-type (WT) mice using quantitative proteomics. The role of polysomnography in Parkinson's disease: objective sleep assessment and clinical indications.

Sleep disorders (SDs) are highly prevalent in Parkinson's disease (PD), affecting up to 80% of patients during the disease and significantly contributing to impaired quality of life, cognitive decline, motor worsening, fall risk, and increased caregiver burden. Polysomnography (PSG) remains the gold standard for objective sleep assessment in PD and plays a central role in differential diagnosis and therapeutic planning. This review examines the spectrum of SDs identifiable by PSG, including rapid eye movement (REM) sleep behavior disorder (RBD), obstructive sleep apnea (OSA), periodic limb mov

The entorhinal cortex atrophy score for diagnostic distinction between phases of the Alzheimer's disease continuum.

BACKGROUND: Alzheimer's disease (AD) is the most common type of dementia, causing functional impairment in millions of individuals and accounting for a large amount of healthcare costs. The entorhinal cortex appears to be one of the first brain structures affected by AD in its early stages. The Entorhinal Cortex Atrophy (ERICA) score is a visual atrophy score used by neuroradiologists in clinical practice for the diagnosis of AD. OBJECTIVE: To analyze the effectiveness of the ERICA score for distinguishing between patients with subjective cognitive decline (SCD), mild cognitive impairment (MC

HSP27, cell stress, and hormesis: From mammals to humanized yeast models.

Hormesis is a biphasic dose-response phenomenon characterized by beneficial or stimulatory effects at low exposure levels to a stressor, followed by inhibitory or toxic effects at higher doses. This effect is related to the level of oxidative stress in the cell. Human HSPB1 (HSP27) is a small heat shock protein involved in the oxidative stress response, whose overexpression correlates with increased longevity in mammalian models. HSP27 protects neurons from damage caused by protein aggregation observed in neurodegenerative diseases; however, this same survival capability also favors tumor cell

Cellular senescence in ageing and neurodegenerative disorders: Molecular mechanisms, biomarkers, and therapeutic strategies.

Cellular senescence has been increasingly associated with neurodegenerative disorders, potentially linking ageing-related cellular impairment with mitochondrial dysfunction, chronic inflammation, and disrupted intercellular signalling. The present study aims to explore the increasing relevance of cellular senescence among key cell types, including neurons, astrocytes, neural progenitor cells, microglia and oligodendrocyte-lineage cells across neurodegenerative ailments such as Alzheimer's disease, Parkinson's disease, amyotrophic lateral sclerosis and multiple sclerosis. This review discusses

Neurodegeneration in Sub-Saharan Africa: Intersections of Biological Ageing, Genomic Diversity and the Lifetime Exposome.

Neurodegenerative diseases are emerging as an important consequence of population ageing in sub-Saharan Africa (SSA), but their scientific relevance extends beyond a projected rise in case numbers. SSA combines exceptional human genomic diversity with marked heterogeneity in vascular and metabolic risk, infection, nutrition, occupational exposures, air pollution, and access to health care. This combination creates an underused setting in which to study why biological ageing produces neurodegeneration in some individuals but relative resilience in others. This review critically synthesises evid

Drosophila Models of Human Neurodegenerative Diseases.

In his book Insects and the Life of Man: Collected Essays on Pure Science and Applied Biology, Sir V. B. Wigglesworth examined how a fundamental understanding of insect physiology and biology can be applied to some of society's greatest challenges, including human health. Here we reflect on how, in the 50 years since his book's publication, Drosophila melanogaster have played a critical role in advancing our understanding of human neurodegenerative diseases. Focusing on Parkinson's disease, Huntington's disease, and the frontotemporal dementia / amyotrophic lateral sclerosis spectrum, we exami

Tau isoform mapping across tissues and biofluids reveals a signature distinguishing Alzheimer's disease from amyotrophic lateral sclerosis.

Blood phosphorylated tau 217 (p-tau217) accurately detects Alzheimer's disease (AD) pathology but is also elevated in amyotrophic lateral sclerosis (ALS), likely reflecting peripheral (big) tau released from muscle. Assays selectively measuring brain-derived (BD-) tau and BD-p-tau217 are therefore needed. We analysed serum BD-tau and p-tau217 in ALS (n = 139), AD (n = 101) and controls (n = 75) using direct immunoassays and after BD-Tau immunoprecipitation (BD-Tau-IP), and tau species in brain (n = 16), spinal cord (n = 11) and muscle (n = 10) by targeted mass spectrometry. Emerging links between gut dysbiosis, aryl hydrocarbon receptor activation, and Parkinson's disease.

Parkinson's disease (PD), the second most common progressive neurodegenerative disorder, is classically defined as a motor disorder characterized by dopaminergic neuron loss in the substantia nigra and motor dysfunction. Increasing evidence, however, identifies PD as a multisystem disorder with early non-motor features, including sleep, psychiatric, and gastrointestinal disturbances, implicating the gut-brain axis. Tryptophan metabolism through serotonin, kynurenine, and indole pathways generates bioactive metabolites, many of which regulate the aryl hydrocarbon receptor (AhR). AhR is a ligand

Molecular functions, cellular responses and pathological implications of the RNA modification N6-methyladenosine.

The RNA modification N6-methyladenosine (m6A) precisely regulates eukaryotic gene expression. The deposition of m6A is governed by chromatin environment, transcription and RNA processing. m6A modifications influence chromatin function and RNA splicing, stability, localization and translation, and enable cellular adaptation to environmental changes. Dysregulation of m6A has been implicated in pathological processes, including neurodegenerative diseases and cancer. In this Review, we dissect the m6A machinery by describing how writers and erasers coordinate m6A deposition and reader proteins tra

Acoustofluidic enrichment of small extracellular vesicles: physical principles, microfluidic implementations, and prospects for Parkinson's disease liquid biopsy.

Reliable blood-based small extracellular vesicle (sEV) analysis for Parkinson's disease (PD) is constrained by co-isolation of lipoproteins and proteins from plasma. This review examines acoustofluidics as a sample-preprocessing and integration-enabling strategy for PD liquid biopsy. We first summarize the analytical value and limitations of sEV-associated α-synuclein, microRNA panels, and multi-omics cargo, and then compare conventional and microfluidic enrichment methods with particular attention to lipoprotein co-isolation, recovery, fraction purity, throughput, and compatibility with downs

MEK inhibitors in NF1-associated high-grade glioma: partial responses and outcomes from a single-institution cohort and literature review.

PURPOSE: NF1-associated high-grade gliomas (HGGs) are rare, biologically heterogeneous tumors with poor outcomes. Although MEK inhibitors (MEKis) are established options for selected NF1-associated low-grade gliomas, their role in NF1-associated HGG remains undefined. We describe clinical and radiographic outcomes with MEKis, including mirdametinib. METHODS: We retrospectively reviewed adult patients with NF1-associated HGG treated at The University of Texas MD Anderson Cancer Center who initiated selumetinib, mirdametinib, or dabrafenib plus trametinib from December 1, 2019, to January 31, 2

Monitoring amyloid formation from natively folded proteins to amyloid fibrils by NMR spectroscopy.

Amyloids are insoluble protein aggregates that arise through protein misfolding and are associated with numerous neurodegenerative disorders, systemic amyloidosis, and other human diseases. Nuclear magnetic resonance (NMR) spectroscopy, encompassing both solution-state and solid-state approaches, offers unique capabilities to investigate each stage of the complex amyloid formation pathway, from soluble monomeric precursors through transient intermediate states and oligomeric assemblies to highly ordered mature fibrils. This residue-specific and atomic-level characterization of the aggregation
