A female patient is described in whom the disease clinical picture, as regards the localization of the multiple lesions in the nervous system (optic nerves, pyramid and cerebellar paths), the undulatory course of the process, and the characteristic manifestations (epileptic seizures, intellect derangement) combined symptoms of both disseminated and diffuse sclerosis. Two hundred and two children with primary nephrotic syndrome were studied. Clinical features at onset and evolution were studied and correlated with microscopic findings and response to treatment. Histologic type and steroid-response were correlated with urinary protein selectivity in a group of them. CT scans of 11 patients with proved white-matter disease were reviewed and divided into three categories. The demyelinating (secondary) and diffuse sclerosis groups frequently exhibited abnormal intravenous enhancement and mass shift. This made differentiation from inflammatory and neoplastic processes difficult. In the dysmyelinating disorders, pathologically noted diffuse loss of white matter and absence of inflammatory response appeared on the scans as areas of diffuse and decreased density in the centrum semiovale. There was no evidence of abnormal contrast enhancement.

The grey lethal mouse is an osteopetrotic mutant in which there is diffuse sclerosis of the entire skeleton and the teeth remain trapped in the jaws. The condition was treated with daily subcutaneous injections of PTE beginning at birth. PTE induced molar tooth eruption in some mutants and improved root formation (without eruption) in others. However, the zone of predentine in the crown of the tooth remained abnormally wide and dentinogenesis remained irregular. Resorption of alveolar bone was adequate to permit complete tooth eruption in some animals and increased root formation in others.
