Rutin attenuates cuprizone-induced demyelination in C57BL/6 mice through activation of Nrf2 signaling and inhibition of NF-κB.

OBJECTIVES: Multiple sclerosis (MS) is a debilitating demyelinating disease characterized by oxidative stress and neuroinflammation. This study investigated the therapeutic potential of rutin, a natural flavonoid, in a cuprizone (CPZ)-induced mouse model of demyelination. MATERIALS AND METHODS: Male C57BL/6 mice were fed a 0.2% CPZ diet for six weeks to induce central nervous system demyelination and were concurrently treated with rutin (50 or 100 mg/kg, intraperitoneally, for the last 14 days). Behavioral assessments, histochemical examinations, and immunohistochemistry for Nrf2, HO-1, NQO-1

Marchiafava-Bignami Disease with Hyperperfusion on Arterial Spin Labeling and Complete Recovery.

Marchiafava-Bignami disease (MBD) is a rare demyelinating disease, which mostly diagnosed among alcoholics. Previous studies on cerebral perfusion of patients with MBD mostly found hypoperfusion of corpus callosum and extra-callosal lesions. Nevertheless, hyperperfusion of corpus callosum in MBD has rarely been described. The aim of this study was to report a case of MBD with hyperperfusion on arterial spin labeling and completely recovered. We described a case of MBD with hyperperfusion in the corpus callosum on arterial spin labeling perfusion magnetic resonance imaging, presented with altere

White Matter Microstructural Correlates of Auditory Brainstem Responses in Patients With Charcot-Marie-Tooth Disease.

INTRODUCTION: Some patients with Charcot-Marie-Tooth disease (CMT) exhibit prolonged auditory brainstem response (ABR) latencies or abnormal waveforms, suggesting potential damage to the peripheral auditory nerve or central auditory pathways. Diffusion tensor imaging (DTI), a non-invasive neuroimaging technique, can detect the integrity and functional properties of white matter structures with high sensitivity. However, research on the association between DTI characteristics and ABR changes in patients with CMT remains relatively limited, and whether both modalities reflect synergistic damage

Initially Normal MRI, Delayed Splenial Lesion in Marchiafava-Bignami Disease.

Marchiafava-Bignami disease (MBD) may show normal findings on early MRI. In patients with alcohol use disorder or risk of thiamine deficiency, repeat imaging is important because splenial lesions can develop later. Recognizing this pattern is key to avoiding delayed diagnosis.

Acute Marchiafava-Bignami Disease Linked to Tea and Toast Diet: A Case Report.

Marchiafava-Bignami disease (MBD) is a rare demyelinating disorder primarily associated with chronic alcoholism. We present a case of a nonalcoholic patient who developed acute cognitive decline secondary to severe dietary deficiency. Magnetic resonance imaging (MRI) demonstrated an isolated, symmetric lesion in the central corpus callosum, differentiating MBD from acute infarction. Rapid improvement followed Vitamin B supplementation, emphasizing the need for early recognition and treatment. This case expands the clinical spectrum of MBD and underscores the crucial role of MRI in diagnosis an

Marchiafava-Bignami Disease and Pellagra in a Chronic Alcohol User: A Reversible Case of Cognitive Impairment.

Marchiafava-Bignami disease (MBD) is a rare complication of chronic alcoholism characterized by demyelination of the corpus callosum. Pellagra, resulting from niacin (vitamin B3) deficiency, typically presents with dermatitis, diarrhea, and dementia. Both MBD and pellagra are often associated with severe nutritional deficiencies. We describe the case of a 56-year-old man with alcohol use disorder admitted after a syncopal episode, presenting with disorientation, apathy, and photosensitive desquamative lesions on sun-exposed skin. Brain MRI revealed T2 hyperintensity in the splenium and body of

Marchiafava-Bignami disease mimicking acute corpus callosum infarction associated with chronic shochu consumption.

BACKGROUND: Marchiafava-Bignami disease (MBD) is a rare alcohol-related disorder that predominantly affects the corpus callosum. Because MBD frequently demonstrates diffusion restriction on magnetic resonance imaging (MRI), it can closely mimic acute ischemic stroke (AIS), posing a diagnostic challenge in image-driven acute stroke evaluation. Early recognition is crucial, as timely thiamine therapy may result in complete clinical and radiologic recovery. CASE DESCRIPTION: A 38-year-old man with long-standing daily consumption of shochu, a Japanese distilled spirit, developed vertigo and vomit

Protective effects of adenosine A2B receptor antagonism in a cuprizone-induced demyelination model.

Demyelinating diseases are characterized by the progressive loss of myelin in the central nervous system (CNS). Myelin protection can be achieved by fostering the differentiation of oligodendrocyte progenitor cells to mature oligodendrocytes, the only myelinating cells in the brain. Oligodendrocytes maturation is sustained, among others, by the neuromodulator adenosine through the activation of its specific receptors: A1, A2A, A2B, A3, all expressed in the brain on neurons and glial cells. The role of A2B receptors (A2BRs) in a cuprizone-induced demyelination model in male C57BL/6 mice was inv

Case Report: Marchiafava-Bignami disease: anti-parietal cell antibodies as a potential etiological factor in a novel case presentation.

Marchiafava-Bignami disease (MBD) represents a rare neurological disorder predominantly associated with chronic ethanol consumption and thiamine insufficiency. However, its possible correlation with cobalamin deficiency resulting from anti-parietal cell antibodies (APCA) remained inadequately characterized in the literature. Timely diagnosis and therapeutic intervention were critical for optimizing clinical outcomes. This case suggested that APCA screening and vitamin B12 assessment should be considered in the diagnostic workup of MBD, especially in patients without classic thiamine deficiency

Anterior commissure involvement in Marchiafava-Bignami disease: MRI findings.

A 52-year-old man with a 30-year history of chronic alcohol abuse presented withseizures and progressive cognitive decline. Brain MRI demonstrated symmetricdiffusion-weighted hyperintensities involving the genu, body, and splenium of thecorpus callosum, extending to the anterior commissure, with corresponding restricteddiffusion on the apparent diffusion coefficient map. These findings were consistent withMarchiafava-Bignami disease, reflecting a toxic-metabolic demyelinating processassociated with chronic alcohol exposure.

Extensive Osmotic Demyelination Syndrome With a Concomitant Cytotoxic Lesion of the Corpus Callosum in Hyperemesis Gravidarum: A Case Report.

Osmotic demyelination syndrome (ODS) is a rare neurological disorder classically associated with rapid correction of hyponatremia but increasingly recognized in the setting of complex osmotic disturbances. Its radiological spectrum includes central pontine and extrapontine myelinolysis, with variable clinical and imaging presentations. The coexistence of cytotoxic lesions of the corpus callosum (CLOCCs) in this context remains exceptional and poorly understood. We report the case of a 35-year-old pregnant woman presenting with severe hyperemesis gravidarum complicated by profound electrolyte i

Multifaceted response of microglia in the cuprizone model of demyelination.

The cuprizone diet paradigm, which involves selective depletion of oligodendrocytes and subsequent myelin loss, provides an excellent model to study the neuroimmune response to demyelination and remyelination. Microglia, the resident immune cells of the central nervous system, mount a robust response to cuprizone administration. However, the specific triggers and temporal trajectory of microglia activation are not well-defined. Moreover, it is unclear the extent to which microglia respond to cuprizone directly rather than to cuprizone-induced demyelination. In this study, we characterized the

[Marchiafava-Bignami disease: an alcoholic with reduced consciousness and muscle paratonia].

A 69-year-old man presented with decreased consciousness and a puzzling muscle paratonia. MRI showed extensive white matter abnormalities with diffusion restriction in the corpus callosum, consistent with Marchiafava-Bignami disease, a rare complication of chronic alcohol abuse. The phenomenon of paratonia should have raised a clinical suspicion towards endstage neurodegenerative disease.

Acute Binge Alcohol Consumption Presenting With a Cytotoxic Lesion of the Corpus Callosum (CLOCC) in a Young, Healthy Filipino Male: A Case Report With Features Overlapping the Proposed Marchiafava-Bignami Disease Spectrum.

Marchiafava-Bignami disease (MBD) is a rare toxic-metabolic encephalopathy characterized by corpus callosum involvement, classically associated with chronic alcoholism and malnutrition. Cytotoxic lesions of the corpus callosum (CLOCCs) represent a radiologic pattern that may occur in various metabolic, toxic, and infectious conditions and can mimic acute ischemic stroke. We report a 28-year-old previously healthy Filipino male who presented with an acute, remittent, and moderately severe bi-occipital headache following an episode of acute binge alcohol consumption. Spleen-brain axis regulates myelin integrity via TGF-β1 signaling in cuprizone-induced demyelination and remyelination model.

Demyelination and impaired remyelination are hallmark features of many neurodegenerative and psychiatric disorders. Although central mechanisms have been widely investigated, the contribution of peripheral immune organs such as the spleen remains poorly understood. In this study, we examined the role of the spleen and transforming growth factor-β1 (TGF-β1) in cuprizone (CPZ)-induced demyelination and remyelination in mice. Splenectomy performed before CPZ exposure significantly worsened demyelination in the corpus callosum, as indicated by reduced myelinated area and myelin basic protein (MBP)

Diquat exposure disrupts ferroportin-dependent iron homeostasis and drives ferroptosis-associated white matter injury.

Diquat (DQ) is one of the most widely used bipyridyl herbicides and has been identified as a potential neurotoxin; however, its primary central nervous system (CNS) targets and the underlying mechanisms remain unclear. In cases of acute human DQ poisoning, subcortical white matter lesions have been reported to emerge approximately two weeks after exposure, with demyelination representing the predominant pathological finding. We used an acute oral DQ exposure rat model that mimics human DQ poisoning. Magnetic resonance imaging (MRI) revealed prominent white matter abnormalities in the corpus ca

Effect of transplanted oligodendrocyte precursor cells derived from inflammatory and non-inflammatory microenvironment on remyelination in a chronic cuprizone model.

INTRODUCTION: Multiple sclerosis is a chronic demyelinating disease of the central nervous system. Transplantation of oligodendrocyte progenitor cells (OPCs) is a promising approach to enhance remyelination; however, the influence of the OPCs' microenvironmental origin on their therapeutic efficacy remains unclear. This study compared the remyelinating capacity of OPCs isolated from inflammatory (lipopolysaccharide) and non-inflammatory (cuprizone) microenvironments after transplanting into the corpus callosum and examined their effects on extracellular matrix chondroitin sulfate proteoglycans

Early-Age Cuprizone Exposure Induces Region-Specific Demyelination and Neuroinflammation in Mice.

Myelin, essential for rapid nerve conduction and axonal integrity in the central and peripheral nervous systems, is compromised in demyelinating diseases, leading to neurological deficits and progressive neurodegeneration. Although remyelination can occur, regeneration in adults is often limited, resulting in incomplete repair and impaired nerve function. In multiple sclerosis (MS), an immune-mediated demyelinating disease with diverse clinical phenotypes, progression and disability correlate with demyelination and failed remyelination, influenced by genetic and environmental factors.
