CRISPR screen reveals FTO-mediated m6A modification of ATF3 as protective in ischaemic stroke.

RNA modifications, especially N6-methyladenosine (m6A), have emerged as pivotal regulators of gene expression in neurological disorders, yet their roles in ischaemic stroke remain unclear. Here, we performed a CRISPR-Cas9 loss-of-function screen targeting 118 RNA modification-related genes in neuronal cells subjected to oxygen-glucose deprivation/reoxygenation (OGD/R) and validated the findings in a mouse model of middle cerebral artery occlusion/reperfusion (MCAO/R). We identified Fto as a top negatively selected gene, with its expression progressively reduced and global m6A levels increased

Neutrophil-Related Gene Signatures for Ischemic Stroke Diagnosis.

INTRODUCTION: Ischemic stroke (IS) is a major cause of death and disability worldwide. The transcriptional mechanism of neutrophil extracellular trap-related genes (NRGs) and their diagnostic potential remain unknown. This study aims to explore the mechanism of NRGs in IS through machine learning and single-cell RNA sequencing (scRNA-seq). METHODS: We conducted differential analysis and functional enrichment analysis on the GEO dataset. Machine learning algorithms were used to identify NRGs related to IS. ScRNA-seq analysis was employed to verify the expression of NRGs in different cell types

Stent angioplasty in patients with vertebral artery ostial stenosis: clinical and angiographic outcomes in 525 patients.

BACKGROUND: The optimal treatment approach for patients with severe atherosclerotic vertebral artery ostial stenosis (VAOS) is not yet supported by evidence from large-scale studies. Even with optimal medical treatment and effective risk factor management, substantial numbers of patients experience symptoms or recurrent strokes. Endovascular treatment is a potential solution, particularly when medicinal therapy alone is insufficient for symptom and stroke risk reduction. This study was aimed at assessing the safety and effectiveness of stent angioplasty in patients with VAOS. METHODS: This si

Persistent Steal Physiology during a Ramp Cerebrovascular Reactivity Assessment Correlates with Severe Cortical Thinning in Steno-Occlusive Disease.

BACKGROUND AND PURPOSE: Steal physiology, a marker of severe cerebrovascular impairment, is associated with cortical thinning, a morphologic phenomenon seen in patients with chronic supply artery steno-occlusive disease. This link has been previously established only with a sudden "step" hypercapnic stimulus. The significance of steal patterns derived from a gradual "ramp" stimulus, and their association with structural brain injury, is unclear. We investigated how steal patterns derived from both step and ramp stimuli compare and relate to the severity of cortical thinning in patients with st

The CCL11-CCR3 Axis Regulates the Aggravation of Neuroinflammation in Astrocytic Necroptosis After Cerebral Ischemia.

In ischemic stroke (IS), the programmed death of neurons is a key factor contributing to poor prognosis and neurological dysfunction. Recently, it has been discovered that necroptosis, as a form of programmed cell death, promotes neuroinflammatory damage after IS; however, its mechanism is still unclear. Recent studies have shown that CCL11, as an eosinophil chemokine, may induce neuronal cytotoxicity by triggering the production of reactive oxygen species (ROS) in microglia. Meanwhile, CCR3, serving as the major functional membrane receptor target of CCL11, plays a crucial role in transducing

Longitudinal sex differences in cerebrovascular ageing in older adults: results from the brain in motion study.

Older females are at an increased risk of dementia, cardiovascular disease and stroke. However sex differences in cardiovascular and cerebrovascular ageing remain poorly understood. We examined longitudinal changes in vascular function in 82 adults (mean age: 65.4 ± 5.1 years; range: 55-75; 39 females) at two timepoints (T1 and T2) 6.1 ± 1.4 years apart. We measured middle cerebral artery velocity (MCAv) and pulsatility (PI) using transcranial Doppler ultrasound, mean arterial blood pressure (MAP) with finger photoplethysmography and cerebrovascular conductance and resistance (CVCi, CVRi) at r

Calycosin alleviates blood-brain barrier disruption after cerebral ischemia-reperfusion injury by inhibiting NLRP3-mediated pyroptosis.

Ischemic stroke, a severe neurological disorder with a multifactorial pathogenesis, presents significant therapeutic challenges. Calycosin, a natural flavonoid, has diverse biological activities, including antioxidant, anti-inflammatory, and antitumor effects. In this study we investigate the protective effects of calycosin against blood-brain barrier (BBB) damage following cerebral ischemia-reperfusion injury (CIRI) and explore the underlying mechanisms. We employ middle cerebral artery occlusion (MCAO) in rats and oxygen-glucose deprivation (OGD) in bEnd.3 brain microvascular endothelial cel

Diurnal differences in cerebral microvascular blood flow and oxygen delivery across brain regions in awake mice.

SIGNIFICANCE: Elucidating the diurnal differences in cerebral hemodynamics is essential for both advancing our understanding of brain function and improving therapeutic strategies for neurological disorders. However, it remains unclear how the inactive and active diurnal phases influence the microvascular-scale distributions of cerebral blood flow and oxygenation and whether these distributions exhibit brain-region-specific variations. AIM: We aim to characterize the time-of-day variations in cerebral microvascular blood flow and oxygenation across brain regions in awake mice. APPROACH: We u

"Is larger always better?" An in vitro comparison of cyclical and static aspiration with different catheters in a neurovascular flow model.

BACKGROUND: Recent advances in mechanical thrombectomy for large vessel occlusion suggest that cyclical aspiration (CyA) may enhance clot ingestion and reduce embolic complications compared with static aspiration (StA). We aimed to identify the experimental conditions under which CyA outperforms StA. METHODS: A 3D-printed neurovascular model in a flow loop (137/89 mmHg, 72 bpm) simulated middle cerebral artery M1 segment (MCA-M1) occlusions with soft and stiff clot analogs. CyA (4.5 Hz and Δp=85-105 kPa) was performed with a dual-solenoid device coupled to a vacuum pump. Eighty experiments we

Is Insulin Resistance Associated With Clinical Outcomes after Endovascular Treatment for Acute Stroke?

BACKGROUND AND PURPOSE: Insulin Resistance (IR) impact on Acute Ischemic Stroke (AIS) outcomes has been challenging to assess due to the lack of a readily available IR biomarker in acute settings. The Triglyceride-Glucose (TyG) index is a novel surrogate marker for IR that may help bridge this gap. We aimed to evaluate the relationship between IR and clinical outcomes after AIS in patients with large vessel occlusion (LVO). METHODS: This single-center retrospective cohort study included patients presenting between 2010 and 2022 with internal carotid artery (ICA) or middle cerebral artery (M1)
