A single-cell multiomic analysis identifies molecular and gene-regulatory mechanisms dysregulated in developing Down syndrome neocortex.

Down syndrome (DS) is the most common genetic cause of intellectual disability, yet the cellular and molecular mechanisms driving this developmental disorder remain unclear. In this study, we profiled human mid-gestation neocortex with snMultiomics across 26 donors. We observed a reduction in neural progenitors and corticothalamic neurons and an increase of intratelencephalic neurons, accompanied by accelerated neuronal specification. We uncovered widespread changes in gene expression, chromatin accessibility, and cell interaction networks affecting neurogenesis, specification, and maturation;

Electroacupuncture combined with neural stem cell transplantation promotes neurogenesis and functional recovery and is associated with tsRNA expression changes in a rat model of ischemic stroke.

BACKGROUND: Electroacupuncture (EA) combined with neural stem cell transplantation (NSCT) represents a promising therapeutic strategy for neurological recovery after ischemic stroke. This study investigated the enhanced effects of EA at 'Quchi' (LI11) and 'Zusanli' (ST36) acupoints combined with NSCT on motor function and neurogenesis in a rat middle cerebral artery occlusion (MCAO) model, and explored the underlying tsRNA-associated molecular changes. METHODS: Neural stem cells (NSCs) were isolated from neonatal rats and characterized. MCAO rats were randomized into five groups: sham, MCAO,

Robust neurogenesis in chronic stroke monkeys following mesenchymal stem cell transplantation plus intermittent theta-burst stimulation.

A significant population of chronic stroke patients urgently requires more effective therapies because of functional plateaus and limited intervention options. Intermittent theta-burst stimulation (iTBS) has been increasingly adopted in chronic-phase ischemic stroke management. However, the efficacy decreases over time. Mesenchymal stem cells (MSCs) have emerged as a promising therapeutic strategy for chronic stroke management through multiple mechanisms. Whether combined MSC transplantation and iTBS confers superior synergistic efficacy warrants further investigation. In this study, human umb

Mesenchymal stem cells therapy for Ischaemic stroke: Mechanism and progress.

Ischaemic stroke is a leading global cause of death and long-term disability. Current acute interventions, notably thrombolysis and mechanical thrombectomy, are limited by narrow therapeutic windows and primarily focus on vascular recanalization. These approaches often fail to address the sustained neurovascular injury, persistent inflammation, and insufficient endogenous repair that underlie long-term functional impairment. In this context, mesenchymal stem cells (MSCs) have emerged as a highly promising regenerative strategy for ischemic stroke, targeting the multifaceted pathophysiology bey

Genetic correlation analysis of Alzheimer's disease and stroke implicates PHLPP1 as a shared locus in individuals of African ancestry.

INTRODUCTION: Neuropathological studies indicate a strong association between Alzheimer's disease (AD) and stroke, yet the molecular mechanisms underlying this association remain unclear. METHODS: Local genetic correlation analysis was conducted with LAVA (Local Analysis of [co]Variant Annotation) using the results from genome-wide association studies on AD and stroke in individuals of African ancestry. Enhanced Hi-C Capture Analysis (eHiCA) examined chromatin interactions using induced pluripotent stem cell (iPSC) -derived cells from AD brain autopsy samples. RESULTS: LAVA identified a regi

Therapeutic Mechanisms of Stem Cell-Derived Exosomes for Neurological Disorders: An Overview.

The current management of neurological disorders remains largely symptomatic. In recent years, stem cell-derived exosomes have emerged as a promising alternative therapeutic strategy. This narrative review synthesizes evidence from preclinical studies investigating the mechanisms and efficacy of exosome-based therapy for neurological conditions. The included studies encompass animal models and in vitro systems. Accumulating preclinical evidence consistently supports the therapeutic potential of stem cell-derived exosomes across several neurological disorders. In Alzheimer's disease models, ste

Glucose-dependent spatial and temporal modulation of oligodendrocyte progenitor cell proliferation via ACLY-regulated histone acetylation.

How it is determined whether postnatal oligodendrocyte progenitor cells (OPCs) will survive, proliferate or differentiate remains unclear. Here we suggest that temporal and brain regional fluctuations of glucose, concomitant with changes in vascularization, modulate OPC population dynamics. We found that regions with high glucose levels exhibited greater OPC proliferation and histone acetylation than regions with low glucose and that this was mediated by the enzyme ATP-citrate lyase (ACLY), which converts glucose-derived citrate to acetyl-CoA. Mice with Acly deletion in OPCs showed a transient

Mesenchymal Stem cell therapy with GHRH receptor analog resolves post-stroke vasogenic edema via modulating AQP4 and mitochondria-ER crosstalk.

BACKGROUND: Post-stroke edema is one of the fatal consequences of ischemic stroke where increased intracranial pressure can exacerbate neurological dysfunction leading to mortality. To date, conventional post-stroke therapy has failed to manage cerebral edema and render neuroprotection. Earlier, our lab reported the benefits of intra-arterially administered mesenchymal stem cells (IA MSCs) in alleviating post-stroke vasogenic edema. Recently, post-stroke neuroendocrine regulation involving growth hormone releasing hormone receptor (GHRH-R) analog is gaining attention as one of the potential ta

Collecting Eggs, Not Killing Chickens: Why Stem Cell Secretome and Exosomes Are Redefining Regenerative Medicine for Healthspan Extension.

Regenerative medicine is becoming more widely integrated with longevity-oriented and preventive care as populations age and chronic degenerative diseases burden healthcare systems. Mesenchymal stem cell (MSC) therapies have progressed from experimental interventions to approved products, yet scalability, safety, cost, and regulatory complexity constrain widespread implementation in medical wellness contexts. The predominant therapeutic effects of MSCs are mediated via paracrine mechanisms, leading to cell-free approaches based on the MSC secretome-a complex mixture of bioactive factors includi

Added Value of Thin-Section Coronal DWI for Lesion Visualization in Acute Brainstem Infarction: A Retrospective Analysis.

Background and Objectives: Brainstem infarctions remain challenging to identify due to their small size, complex anatomy, and known limitations of conventional axial diffusion-weighted imaging (DWI), particularly in the posterior fossa. Thin-section coronal DWI may improve lesion conspicuity by providing higher spatial resolution and an orthogonal imaging perspective. To evaluate whether 3 mm thin-section coronal DWI improves lesion visualization and delineation compared with standard 4 mm axial DWI in patients with MRI-confirmed acute brainstem infarction. Materials and Methods: In this retro

Post-ischemic modification of neurogenesis and oligodendrogenesis in rodent models.

Neurogenesis and oligodendrogenesis occur throughout life under both physiological and pathophysiological conditions. Brain insults such as ischemia, trauma, epilepsy, or Alzheimer disease result in the promotion of neurogenesis and oligodendrogenesis; however, the mechanisms and the roles of this promotion are not well elucidated. Neurogenesis occurs in two distinct regions in the brain, namely, the subventricular zone (SVZ) of the lateral ventricle and the subgranular zone (SGZ) of the dentate gyrus. Neural stem cells (NSCs) have the potential to self-renew, proliferate, and differentiate in

RVG-Modified BMSCs-Derived Small Extracellular Vesicles Loaded With miR-21 Alleviate Neuronal Injury Resulted From Excessive Autophagy via Targeting PTEN/Akt/mTOR Pathway After Cerebral Ischaemia.

Ischaemic stroke (IS) leads to tragic disability and high adult mortality, while there are limited therapeutic measures for it. Small extracellular vesicles (sEVs) derived from bone marrow mesenchymal stem cells (BMSCs) have been suggested to have satisfactory therapeutic effects on IS by the delivery of their packed microRNA (miRNA). However, systematically administered naïve sEVs are difficult to cross the blood-brain barrier (BBB) and enter the brain parenchyma; also, the low abundance of sEVs cargo miRNAs restricts the possibility of maximising their regulatory functions. Hence, the brain

MAGMAS Inhibition Enhances Temozolomide Efficacy in Chemotherapy-Resistant Glioblastoma Models.

UNLABELLED: Glioblastoma (GBM, isocitrate dehydrogenase wild-type grade 4 astrocytoma) is the most aggressive and common brain tumor, characterized by increased proliferation, invasiveness, mitochondrial-dependent changes, and necrosis. GBM recurrence is universal despite the standard-of-care treatment with maximal surgical resection, radiation, and temozolomide (TMZ). Most patients relapse 6 to 9 months following initial diagnosis, and the median survival after recurrence is less than a year. Therefore, effective therapeutic strategies are needed to overcome glioma resistance mechanisms and i

Human Amnion-Derived Mesenchymal Stromal Cell Exosomes Promote Neuroprotection and Neurovascular Remodeling after Cerebral Ischemia.

BACKGROUND: Ischemic stroke remains a leading cause of death and long-term disability worldwide, and effective neuroprotective therapies applicable to a broad patient population are still limited. Although mesenchymal stromal cell-derived exosomes (MSC-EXO) have emerged as promising cell-free therapeutic agents, evidence regarding exosomes derived from human amnion-derived mesenchymal stromal cells (AMSC-EXO) in cerebral ischemia remains scarce. METHODS: Human AMSC-EXO were isolated from conditioned media of cultured human amnion-derived mesenchymal stromal cells and characterized by nanopart

Edaravone delivery of exosome-liposome hybrid nanoparticles for the treatment of ischemic stroke.

Ischemic stroke (IS) triggers neuroinflammatory cascades mediated by microglial polarization, where pro-inflammatory M1 phenotypes exacerbate neuronal damage, and anti-inflammatory M2 phenotypes promote repair. Edaravone (Eda), a free radical scavenger, shows potential for modulating microglial polarization but faces challenges in penetrating the blood-brain barrier (BBB) and achieving targeted delivery. This study develops a hybrid nanoparticle (SHp-NSCsome) that integrates hypoxia-preconditioned neural stem cell-derived exosomes with Eda-loaded liposomes functionalized with the ischemic-homi

A new perspective on mesenchymal stem cells and their derivatives in alleviating cerebral ischemia-reperfusion injury dynamically regulating mitochondrial function to remodel the immune network.

Cerebral Ischemia-Reperfusion Injury (CIRI) is a common pathological process in ischemic stroke. Its core detriment lies in the cascade of subsequent injuries triggered by blood flow restoration after cerebrovascular recanalization, primarily including oxidative stress burst, calcium overload, immune-inflammatory imbalance, and mitochondrial dysfunction, ultimately leading to massive neuronal apoptosis and necrosis. Mitochondria, as central hubs of cellular energy metabolism and apoptosis regulation, exhibit functional disturbances that represent the initiating and core link in CIRI. CIRI trig

Serum Neurofilament Light Chain Predicts Stroke Outcome and is a Potential Marker for Treatment Effects of Neural Stem Cell-derived Extracellular Vesicles in a Rat Stroke Model.

Acute ischemic stroke (AIS) remains a leading cause of disability worldwide, and effective treatments are urgently needed beyond reperfusion therapy. Translating preclinical success to clinical impact has been hindered by variability in animal models and the lack of translational biomarkers that predict outcomes across species. To overcome these barriers, we developed a robust rat AIS model optimized for consistency and severity, enabling rigorous therapeutic testing. Additionally, we tested a panel of common clinical serum biomarkers to improve translation from rodents to humans. We demonstra

Proteomic profiling reveals region-specific neuroimmune and apoptotic signatures induced by Bothrops jararaca venom toxins in the murine brain.

Human envenomation by Bothrops species can lead to systemic effects such as cardiovascular disturbances, coagulopathy, renal failure, and central nervous system (CNS) complications, including hemorrhagic and ischemic strokes. Although Bothrops jararaca is a medically significant species implicated in cases of human stroke, the CNS-specific effects of its venom remain poorly understood. In this study, we employed label-free mass spectrometry-based proteomics to characterize venom-induced alterations in the cerebral cortex and brainstem of mice following intramuscular injection of B. jararaca ve

Serial brain FDG-PET and IMZ-SPECT following intracerebral MSC transplantation in patients with subacute ischemic stroke.

Ischemic stroke is a leading cause of mortality and long-term neurological disability worldwide, and cell-based therapies represent a promising approach. Although clinical studies have reported favorable outcomes following cell transplantation, the effects on host neuronal integrity remain incompletely understood. This study investigated temporal and spatial changes in fluorodeoxyglucose positron emission tomography (FDG-PET) and 123I-iomazenil single-photon emission computed tomography (IMZ-SPECT) after intracerebral cell transplantation in patients with subacute ischemic stroke and examined
