Distribution and Habitat Suitability of Artemisia annua in China Under Climate Change Scenarios.

Artemisia annua is a medicinal plant of high therapeutic and economic value, yet its wild resources have declined markedly in recent decades. Climate change is expected to further alter its geographical distribution, posing challenges to both conservation and sustainable utilization. Here, we applied a Maximum Entropy (MaxEnt) model that integrates climatic, topographic, and soil variables to assess the current and future suitable habitats of A. annua in China under three CMIP6 shared socioeconomic pathways (SSP126, SSP245, and SSP585) for the mid-21st century (2050s) and late-21st century (20

Early Life Vascular Risk and Midlife Markers of Brain Aging.

BACKGROUND: Early-life vascular risk factors are increasingly linked to midlife cognitive decline, though critical risk periods remain unclear. These factors also relate to structural brain changes, yet midlife alterations tied to cognitive decline are poorly defined. We leveraged data from the CARDIA study (Coronary Artery Risk Development in Young Adults) to identify critical periods and factors associated with midlife cognition and neuroimaging signatures. METHODS: We utilized longitudinal cognitive and neuroimaging data from the prospective CARDIA study with vascular risk factor data span

Incidence and Disease Course of Polymyalgia Rheumatica During Two Decades: A Nationwide Study Across Health Care Sectors in Denmark.

OBJECTIVE: A new algorithm enables the identification of patients with polymyalgia rheumatica (PMR) across health care sectors in Denmark. We estimated annual incidence rates from 2003 to 2024 and described the disease course, including the occurrence of late-onset giant cell arteritis (GCA) and prednisolone or prednisone (PDN) treatment. METHODS: A Danish nationwide, population-based cohort study was conducted using data linked at the individual level. Cases were identified among people ages 50 years or older between January 1, 2003, and November 1, 2024. RESULTS: A total of 51,401 patients

Defining Difficult-to-Treat Rheumatoid Arthritis in Routine Care: Comparative Performance of Published Criteria and Description of Early and Late Difficult-to-Treat Rheumatoid Arthritis Phenotypes.

OBJECTIVE: Despite advances in treat-to-target strategies, a subset of patients with rheumatoid arthritis (RA) remains refractory to multiple therapies and is classified as having difficult-to-treat RA (D2T-RA). Although EULAR has proposed classification criteria for D2T-RA, these patients remain variably identified in real-world practice because of heterogeneous applications. We aimed to compare the preformance of published applications of the EULAR D2T-RA criteria in a real-world RA cohort and to determine weather the timing of D2T-RA development could distinguish early from late D2T-RA phen

Phosphine-mediated azine C-H couplings with water and ammonia.

The unique reactivity of transition metals has advanced C-H functionalization chemistry, enabling practitioners to directly modify drug and agrochemical compounds during structure-activity relationship (SAR) studies. These metals are particularly adept at transforming the C-H bonds of arenes and aromatic heterocycles, generating new reactions that form C-C and C-heteroatom bonds. However, among the many potential coupling partners, (hetero)arene C-H coupling reactions with water and ammonia are rare1-5. Despite being two of the most abundant chemicals on Earth, their reactivity can be at odds

Dialkyl ether synthesis through heteroatom homolytic substitution.

The modular and selective synthesis of dialkyl ethers, particularly sterically congested variants, remains a longstanding challenge in drug discovery and medicinal chemistry1,2. Hindered alkyl ethers are especially desirable given their prevalence in bioactive natural products and their favourable physicochemical properties3. Classically, dialkyl ether synthesis relies on nucleophilic substitution strategies; however, SN2 reactions are fundamentally limited by steric congestion at the transition state, whereas SN1 pathways proceed through promiscuous carbocation intermediates that are prone to

Eplontersen with and without background transthyretin stabilizers in transthyretin amyloid cardiomyopathy: secondary analysis of a phase 3, randomized controlled trial.

Transthyretin (TTR) amyloid cardiomyopathy (ATTR-CM) therapies include both TTR gene silencers and TTR stabilizers. The role of TTR gene silencers added to background TTR stabilizer therapy is unknown, and the effects of silencer treatment in the absence of stabilizer use are unclear. In the CARDIO-TTRansform study, 1,432 patients (n = 135 (9.4%) women and n = 1,297 (90.6%) men) with ATTR-CM were randomized (1:1) and treated with the antisense oligonucleotide eplontersen (45 mg every 4 weeks) that targets TTR or with placebo for up to 140 weeks. At baseline, 57% of the patients were taking TTR

Fluoroalkyl carbenes: versatile platforms for fluoroalkylated functional molecules.

Fluoroalkyl carbenes have emerged as versatile and synthetically powerful reactive intermediates for the construction of structurally diverse fluorinated molecules, driven by their distinctive electronic properties and highly tunable reactivity. In particular, the interplay between the electrophilic carbenic center and the adjacent fluoroalkyl group enables unconventional bond-forming processes, including sequential and dual bond formation at the carbenic carbon, thereby providing unique disconnections for complex molecule construction. This review provides a comprehensive and critical analysi

NG2-Glia in Central Nervous System Following Injury: From Pathological Involvement to Repair Potential.

BACKGROUND: Central nervous system (CNS) injury remains a severe neurological condition with limited regenerative capacity. NG2-glia are abundant and functionally versatile glial cells that have attracted increasing attention in CNS repair research. However, their roles are complex and context-dependent, ranging from early pathological involvement to later reparative potential, and a systematic synthesis of this temporal dichotomy is lacking. METHOD: We systematically searched PubMed and Web of Science from inception to 2026 using combinations of keywords including "NG2-glia," "NG2 cells," "o

Quantitative Systems Pharmacology (QSP): Bridging Biology and Mechanism with Clinical Drug Development Decisions.

Quantitative systems pharmacology (QSP) integrates mechanistic representations of biology with quantitative pharmacology to support decision‑making across the drug development continuum. Over the past two decades, QSP has evolved from an exploratory research activity into an established component of model‑informed drug development (MIDD), particularly in settings where system‑level interactions complicate the interpretation of exposure-response relationships or where direct clinical data are limited. This review outlines the historical development and conceptual foundations of QSP and summariz

Post-Shower Cardiovascular and Oxygenation Responses During an Antarctic Inland Traverse: A Single-Participant Repeated-Measures Study.

IntroductionPolar field hygiene may require transitions between severe cold, warm-water exposure, and sheltered recovery. Physiological responses to this sequence are poorly described.MethodsA healthy 35-year-old man completed 13 approximately 5-min showers using approximately 10 L of water at 40 °C late in a 78-day inland Antarctic traverse. Systolic and diastolic blood pressure, heart rate, and peripheral oxygen saturation (SpO2) were measured after 20 min of seated rest before showering, at the first post-shower measurement obtained within 5 min of shower completion, and every 5 min thereaf

The Viennese neurologist Richard Stern and the discrimination and persecution of Jewish physicians in Austria and Slovakia.

This article reconstructs the life and career of the Viennese neurologist and neuropathologist Richard Stern (1878-1942) and situates his trajectory within the longue durée of antisemitic discrimination against Jewish physicians in Austria. Drawing on archival sources and contemporary scientific publications, it examines Stern's academic training in the largely forgotten tradition of Lothar Frankl-Hochwart (1862-1914), his clinical and scientific work, and his professional activities in social insurance medicine. The study emphasizes that the exclusion of Jewish physicians in Austria did not b

Physiological rather than pathological cardiac remodeling in olympic-style elite weightlifters: an imaging and biomarker study.

High-intensity resistance training causes intermittent extreme afterload, but its potential to induce maladaptive myocardial fibrosis remains unclear. This study investigated the molecular and tissue-level cardiac consequences of long-term resistance training. A cross-sectional study of 13 elite male weightlifters (≥ 5 years experience) and 13 age-matched healthy controls. Participants underwent transthoracic echocardiography (longitudinal strain) and cardiac magnetic resonance imaging (native T1, extracellular volume [ECV], and late gadolinium enhancement [LGE]). Glutaric aciduria type 1 presenting with Parkinsonism with presynaptic dopaminergic dysfunction in an adult female: a case report.

Glutaric aciduria type 1 (GA1) is an autosomal recessive neurometabolic disorder caused by pathogenic variants in the GCDH gene, typically presenting in infancy with dystonia following encephalopathic crisis. Late- and adult-onset forms are rare and may manifest with nonspecific neurological features. We describe a 43-year-old woman with levodopa-responsive parkinsonism and presynaptic dopaminergic dysfunction. Genetic analysis revealed compound heterozygous GCDH variants, consistent with GA1. One of the reported variants (c.1178G > A, p. Gly393Glu) is extremely rare and has never been associa

Metabolic and Bariatric Surgery and Change in the AI-ECG-Derived Biological-Age Gap: A Longitudinal Cohort Study.

BACKGROUND: Obesity is associated with accelerated aging, yet whether metabolic and bariatric surgery (MBS) can reverse or attenuate this process, as measured by the artificial intelligence-electrocardiogram (AI-ECG)-derived age gap, remains uncertain. METHODS: We retrospectively studied 2,380 adults who underwent primary MBS from 2008 to 2023 and contributed 6,149 ECG observations. The outcome, δage, was AI-ECG-predicted age minus chronological age. Linear mixed-effects models used continuous time with a clinically prespecified 1-year knot and decomposed weight into between-person and within
