Enhancing Diagnostic Accuracy in Sinonasal Neuroendocrine Tumors: The Case for DOTATATE PET Over FDG PET.

Neuroendocrine tumors (NET) are a heterogeneous group of neoplasms that arise from neuroendocrine cells and commonly originate in the GI tract, lungs, and pancreas, but can also present in the skull base and paranasal sinuses. While fluorodeoxyglucose positron emission tomography (FDG PET) scans are routinely used in the evaluation of sinonasal malignancies, their limitations in detecting recurrences near the cranium and cervical lymph node metastases in NETs such as olfactory neuroblastoma (ONB) and sinonasal neuroendocrine carcinomas (SNEC) have prompted the search for an alternative imaging

NeuN expression in health and disease: A histological perspective on neuronal heterogeneity.

Neuronal nuclei (NeuN), also known as Rbfox3, is a widely used neuronal marker for identifying postmitotic neurons in both basic neuroscience and diagnostic neuropathology. Since its discovery, NeuN immunoreactivity has enabled accurate neuron counting, injury assessment, and anatomical mapping. However, accumulating evidence demonstrates that NeuN is not universally expressed across all mature neurons. Specific neuron types, such as olfactory mitral cells, cerebellar Purkinje cells, and retinal photoreceptors, consistently lack NeuN immunostaining despite functional maturity. Moreover, NeuN e

Olfactory Groove Meningioma Compressing the Optic Chiasm and Optic Nerve: Improvement of Visual Functions by Surgical Intervention.

OBJECTIVE: Olfactory groove meningiomas (OGMs) are rare but serious tumours that can cause visual loss. Surgical treatment of such meningiomas is critical for the recovery of visual functions. METHODS: In this case report, a 62-year-old patient presented to our clinic with sudden visual loss in the right eye and underwent ophthalmological examination. Visual field (VF), retinal nerve fibre layer (RNFL) thickness test, and contrast-enhanced cranial and orbital magnetic resonance imaging (MRI) were performed. The patient was diagnosed with OGM compressing the optic chiasm and nerve, and was ope

Refinement of Hyams Grading Criteria for Olfactory Neuroblastoma in an Endoscopic Sinus Surgery Predominant Cohort with Extended Follow-up: Worth the Effort?

BACKGROUND: Hyams grading is considered prognostic in olfactory neuroblastoma (ONB), but grading criteria are subjective and predate modern classification and surgical approach. We evaluate the application of granular grading criteria to an endoscopic surgery predominant cohort with extended follow-up. METHODS: 78 ONB patients were identified (1994-2019) with original diagnoses dating to 1979. Original Hyams grade, Modified Hyams grade incorporating more detailed criteria and other histologic features (i.e. divergent differentiation, clear cell/oncocytic change, and spindling) were assessed i

Stem Cell Niches for Olfactory Regeneration and Their Therapeutic Applications.

Olfactory dysfunction affects many individuals and may result from infections, trauma, neurodegenerative diseases, or genetic disorders. Unlike most neurons in the mammalian nervous system, olfactory sensory neurons (OSNs) continuously regenerate throughout life, a process facilitated by specialized stem cell niches in the olfactory epithelium. Horizontal basal cells are typically dormant under normal conditions but become activated following injury, differentiating into globose basal cells (GBCs) and other cell types. GBCs serve as the primary source for OSN regeneration and are regulated by

Differential impacts of social isolation and enriched environment on multi-sensory brain-wide functionality and network segregation.

Environmental factors profoundly affect the brain's sensory processing network, optimizing adaptive strategies for growth and survival. We hypothesized that sensory-specific functional responses and/or functional networks could be altered by manipulating environmental conditions. To elucidate how the environment influences brain development, we exposed male mice to two - socially isolated and enriched environments - during early life (from 4 to 11 weeks) and investigated their effects on mouse brain functionality using sensory stimulus-evoked BOLD fMRI and resting-state fMRI. Here, we show tha

Electrical Stimulation of Trigeminal Nerve at the Anterior Nasal Septum in Healthy Individuals and Patients With Olfactory Dysfunction.

BACKGROUND: The intranasal trigeminal system contributes to the sense of smell. Its integrity in olfactory dysfunction (OD) may be crucial for future treatments. This study assessed trigeminal sensitivity in healthy individuals and patients with OD of different etiologies using electrical stimulation of the nasal mucosa. METHODS: Eighty-two participants were included in this multicentric study: healthy individuals (n = 28) and patients with OD (n = 54), comprising post-viral OD (PVOD, n = 29), post-traumatic OD (PTOD, n = 15), and chronic rhinosinusitis with nasal polyps (SND, n = 10). Olfactory Nerve Transection Transiently Activates Olfactory Ensheathing Cells in Xenopus laevis Larvae.

Olfactory ensheathing cells (OECs) play a crucial role in supporting the continuous turnover of olfactory receptor neurons by promoting axon growth and targeting. While OECs have been extensively studied in mammals for their potential in treating nerve injuries, little is known about these cells in non-mammalian vertebrates. We identified and characterized the morphology of OECs, fibroblasts, and macrophages in the olfactory system of Xenopus laevis larvae. Additionally, we used antibodies against phosphorylated ribosomal protein S6 (p-rpS6) to visualize the activation of non-neuronal cells in

Non-contact radiofrequency stimulation to the olfactory nerve of human subjects.

Damage to the olfactory nerve, caused by aging, trauma, or neurological disorders, can lead to smell loss, negatively impacting quality of life, taste perception, safety, and emotional well-being. Currently, olfactory training, which involves exposure to aromatic odorants, is the most widely used treatment. While this method provides some benefit, it does not offer a fundamental cure. Developing an approach that directly stimulates the olfactory nerves to restore neural activity is critical for effectively addressing smell loss. This study investigates the effects of radiofrequency (RF) stimul

Repetitive Mild Traumatic Brain Injury Disrupts Learning and Memory: A Novel Olfactory Approach to Detection.

Traumatic brain injury (TBI) leads to significant public health concerns due to cognitive decline and increased risks of neurological conditions like Alzheimer's disease and chronic traumatic encephalopathy. Preclinical models are essential for exploring how mild TBI leads to neuronal dysfunction and neurodegeneration. Using a mouse model, we applied repetitive, mild, side-alternating impacts to induce rapid head rotational acceleration-deceleration. A novel odor-based learning and memory task was developed to address TBI-related vision impairments. Our findings revealed that this side-impact

Acupuncture modulates the microbiota-gut-brain axis: a new strategy for Parkinson's disease treatment.

Parkinson's disease is a relatively common neurodegenerative disorder in clinical practice, and its prevalence is increasing worldwide. It not only causes patients to have movement disorders such as tremors and delayed initiation but also makes them suffer from olfactory disorders, gastrointestinal disorders, insomnia and other symptoms, which imposes a heavy burden on both patients and their families. In recent years, some scholars believe that the gut-brain axis may be the key to revealing the pathogenesis of Parkinson's disease. The changes in intestinal flora, or bacterial infections and o

Gut dysbiosis as a potential driver of Parkinson's and Alzheimer's disease pathogenesis.

The prevalence of neurodegenerative diseases such as Parkinson's disease (PD) and Alzheimer's disease (AD) in the U.S. is expected to increase as the population ages. Despite significant advancements in neurodegenerative research, the initiating events remain unclear, and no treatments currently exist to stop or reverse disease pathogenesis. Emerging studies highlight the importance of the gut microbiome and gut-brain-axis in the pathogenesis of many human diseases. This narrative review aims to integrate current research investigating how gut microbial dysbiosis may influence the development

Olfactory identification impairment in cerebral small-vessel disease indicates cognitive network disconnection and predicts accelerated cognitive decline.

BACKGROUND: Olfactory identification impairment is a well-established early screening index for cognitive decline in neurodegenerative disorders. However, whether this simple sensory test can similarly detect or predict cognitive deficits in cerebral small vessel disease (CSVD)-a leading cause of vascular cognitive impairment-remains unclear. METHODS: All participants-including 193 CSVD patients and 164 normal controls (NC)-completed the University of Pennsylvania Smell Identification Test (UPSIT) and a comprehensive neuropsychological battery. Multimodal MRI analyses were conducted to identi

Fisetin Augments OEC Neuroprotection Through Modulating Neurotrophin/Cytokine Signaling and Activating PI3K/Akt/CREB Pathway in an In Vitro Neurodegenerative Injury.

The persistence of deleterious substances at lesion sites severely impedes neuronal survival and axonal regeneration following central nervous system (CNS) injury or in neurodegenerative disorders. Therefore, clearing these harmful components and establishing a regeneration-permissive microenvironment are crucial for functional recovery. In this study, primary olfactory ensheathing cells (OECs) isolated from adult SD rats were pharmacologically treated with fisetin, a pharmacological agent. To model CNS injury conditions, neural debris was generated through mechanical disruption of primary neu

Stem Cells and Cell-Free Therapies for Olfactory Epithelium Regeneration: Insights from Experimental Models.

Olfactory impairment is one of the most common diseases of the sense organs, and it is closely related to quality life. Although the molecular mechanism of olfaction was recently brought to light, the pathophysiology and effective treatments for olfactory dysfunction still remain challenging. Olfactory impairment can be caused by the degeneration of olfactory receptor neurons in the nose and also by the degeneration of the olfactory bulb of the olfactory cortex. Several studies have shown that stem cells promote the regeneration of the olfactory neuroepithelium after permanent damage in anosmi
