Odor Experience Stabilizes Glomerular Output Representations in Two Mouse Models of Autism.

Novel stimuli can be stressful for individuals with autism spectrum disorders (ASD), though repeated exposure can reduce this effect. In Cntnap2-/- and Shank3B+/- mouse models of ASD, novel background odors impaired behavioral target odor recognition but that deficit improved with training. To investigate the neural basis of this improvement, we used wide-field calcium imaging to measure olfactory bulb responses in Cntnap2-/- and Shank3B+/- mice and WT mice of either sex. Training with background odors enhanced both behavioral performance and neural discriminability of odor mixtures in both Cn

Olfactory Mucosa Mesenchymal Stem Cells (OM-MSCs)-Derived Exosomes Mitigate Neuroinflammation and Nerve Injury After Intracranial Hemorrhage (ICH) Through miR-30-5p and Its Associated Mechanisms.

Exosomes derived from olfactory mucosa mesenchymal stem cells (OM-MSCs-EXOs) offer a novel therapeutic strategy for various diseases due to their role in regulating intercellular signaling. However, the potential of OM-MSCs-EXOs to improve intracranial hemorrhage (ICH) and its underlying mechanisms is unclear. In this study, we found that OM-MSCs-EXOs can perfect neurofunctional recovery after ICH. The level of miR-30b-5p declined in thrombin-triggered OM-MSCs. Suppression of miR-30b-5p in OM-MSCs-EXOs inhibited mitophagy and promoted reactive oxygen species (ROS) and inflammasome-related prot

A familial case of Kallmann syndrome: novel variants in ANOS1 and GNRHR genes.

Kallmann syndrome (KS) is a rare genetic disorder characterized by hypogonadotropic hypogonadism and anosmia or hyposmia, stemming from the defective migration of GnRH and olfactory neurons during embryogenesis. This study investigated a multigenerational family with KS, identifying novel mutations in the ANOS1 (c.78_108del, X-linked) and GNRHR (c.974del, autosomal recessive) genes through genetic testing. Affected males carrying the ANOS1 mutations displayed a range of phenotypes, all of which were associated with hypogonadism and varying degrees of anosmia. Furthermore, isolated mutations in

Nasal-to-brain siRNA delivery based on trace amine associated receptor for improving cognitive function.

Gene-based therapies for central nervous system (CNS) disorders face substantial challenges in overcoming the blood-brain barrier (BBB) to effectively target brain tissues. The nasal-to-brain delivery route has gained increasing attention as it bypasses the BBB, facilitating faster drug delivery to the lesion site while minimizing systemic side effects. Here, we developed a nasal-to-brain delivery system to administer small interfering RNA (siRNA) for the treatment of radiation-induced brain injury (RBI). RNA sequencing revealed that the p53 signaling pathway was predominantly enriched in the

Stage-Dependent Disruptions in Neurogenesis and Neurotrophins' Production Following Prenatal and Postnatal Valproic Acid Exposure: Implications for Autism Spectrum Disorders.

Autism spectrum disorders (ASD) are neurodevelopmental conditions involving impaired neuronal processes such as connectivity, synaptogenesis, and migration. Prenatal exposure to valproic acid (VPA), an anticonvulsant and mood stabilizer, is linked to increased ASD risk, with timing as a key factor. However, the molecular mechanisms of VPA-induced neurodevelopmental disruptions remain unclear. Building on our previous study, which characterized VPA-induced prenatal and postnatal ASD models with impaired social behavior, repetitive patterns, and altered brain connectivity, this study examines mo

PGC-1α mediates hypoxia-preconditioned olfactory mucosa mesenchymal stem cells improved neuroinflammatory response via inhibiting microglial ferroptosis in ischemic stroke.

BACKGROUND: Cerebral ischemia/reperfusion injury can induce nerve cell apoptosis and ferroptosis and activation of the neuroinflammatory cascade plays a critical role in ischemic stroke progression. Mesenchymal stem cells (MSCs) can aid in repairing nerve injuries and regulating inflammatory responses, but their roles in ischemic stroke are unknown. METHODS: Here, we investigate the impact of microglial susceptibility to ferroptosis on neuronal apoptosis using an oxygen-glucose deprivation/reperfusion (OGD/R) model. Furthermore, we examined the effects of hypoxia-preconditioned olfactory mucos

Key Signals Produced by Gut Microbiota Associated with Metabolic Syndrome, Cancer, Cardiovascular Diseases, and Brain Functions.

Gut microbiota have a significant impact neurotransmitters, short-chain fatty acids (SCFAs), immune signaling molecules, and gut hormones. These signaling molecules interact with receptors on the gut wall, immune cells, or the enteric nervous system (ENS), and reach the central nervous system (CNS) via the Vagus nerve (VN). SCFAs interact with G protein-coupled receptors (GPCRs), Toll-like receptors (TLRs), and proliferator-activated receptors (PPARs), influencing inflammatory reactions, gut motility, nutrient absorption, hormone secretion, neurochemical signaling, and brain functions. First-in-human intranasal [13N]oxytocin PET: evaluation of feasibility, biodistribution, and radiation dosimetry.

BACKGROUND: Oxytocin is a neuropeptide with therapeutic potential for several neuropsychiatric and pain-related disorders. Intranasal delivery is proposed to enable access to the central nervous system via the trigeminal nerve and olfactory nerves, thereby bypassing the blood-brain barrier. However, direct evidence of biodistribution following intranasal administration in humans is limited. This study evaluated the feasibility of imaging oxytocin uptake using a novel PET tracer, [13N]oxytocin, in healthy volunteers. RESULTS: Six participants received intranasal [13N]oxytocin and underwent who

Age-related alterations in the expression of mesencephalic astrocyte-derived neurotrophic factor in the brain and their impact on neurobehavioral functions.

Mesencephalic astrocyte-derived neurotrophic factor (MANF) is a neurotrophic protein localized in the endoplasmic reticulum (ER) and pivotally involved in maintaining ER homeostasis. The ER is central to protein synthesis, folding, degradation and secretion (proteostasis), and experiences considerable stress in neurodegenerative diseases (NDDs), which activates the unfolded protein response (UPR). Aging, the primary risk factor for NDDS, is associated with impaired ER function. MANF is shown to be protective in various experimental models of NDDs. We hypothesized that the expression of MANF in

Advances in Intranasal Delivery of Exosomes for Central Nervous System Disorders.

Central nervous system disorders are major global health challenges that contribute to significant morbidity and mortality. Traditional therapeutic strategies often face substantial limitations, primarily due to the blood-brain barrier, which restricts the delivery of pharmacological agents to the brain and consequently affects treatment effectiveness. In recent years, in order to enhance the efficacy of the central nervous system treatments, exosome-based approaches have gained interest. Exosomes, small extracellular vesicles (30-150 nm) secreted by cells, present a feasible therapeutic strat

Cortical excitatory and inhibitory neuron deficits may underlie the cognitive and social impairments in a mouse model of schizophrenia with exonic Reln deletion.

Reelin is an essential extracellular matrix glycoprotein that regulates cortical layers formation and has been implicated in several neuropsychiatric conditions, including schizophrenia (SCZ). To explore its role in brain function and its potential involvement in SCZ, we developed a Reln heterozygous deletion (Relndel/+) mouse model that replicates a genetic deletion identified in a Japanese patient with SCZ. In previous studies, we demonstrated that Relndel/+ mice exhibit cognitive impairments in a visual discrimination test. In the present study, we found that Relndel/+ mice displayed impair
