Intranasal vaccines specifically eliciting mucosal immunity in the upper respiratory tract have shown advantages in protecting against respiratory virus invasion. Yet, no clinically licensed intranasal adjuvant remains a major hurdle for the development of intranasal vaccines with low immunogenic antigens like subunit vaccines. Here, we show that liposomes loading simvastatin (Lipo-SV) serve as potent mucosal adjuvants for the intranasal liposomal subunit vaccine encapsulating the hemagglutinin 1 (HA1) glycoprotein of A/PR/8/34 (PR8) H1N1 influenza (Lipo-HA1), providing robust protection again

Vibrio cholerae is a ubiquitous marine bacterium that solubilizes and consumes chitin in the marine water column. In both the marine environment and the intestinal tract, V. cholerae forms biofilms: how do the diverse surfaces that V. cholerae encounters influence its biofilm formation and, in turn, shape its ecological interactions with other microbes? Here, we use the interaction between the predator Bdellovibrio bacteriovorus and V. cholerae as a model to explore how the environmental chitin substrate alters V. cholerae biofilm formation and predator-prey dynamics. We find that glass-bound

Toxigenic Vibrio cholerae (V. cholerae) was first sensitively and rapidly detected in hospital wastewater in Beijing, China, on 12 June 2025. Viable V. cholerae was successfully isolated from the wastewater sample and identified as the O1 Ogawa serotype. Whole-genome sequencing and comparative genomic analysis revealed that the isolate was closely related to locally circulating strains but clearly distinct from the seventh pandemic lineage, suggesting its link to indigenous transmission chains. In addition, we immediately initiated a comprehensive and multi-source traceback investigation upon

Cholera remains a major public health challenge in Kenya, driven by environmental pollution, poor water, sanitation, and hygiene (WASH) facilities, weak monitoring systems, and climate effects. While there have been improvements in studying the disease, testing, monitoring, and environmental checks, the spread continues, especially in informal settlements, rural areas, and refugee camps. This review looked at 106 peer-reviewed studies published between 1979 and 2024. It aimed to describe the scope, progress, and gaps in cholera research in Kenya. Five databases, including Google Scholar, Web o

Excess mortality is a key consequence of crises due to armed conflicts and natural disasters. Israel's campaign in the Gaza Strip has exposed people to multiple risk factors for increased infectious disease burden. We developed a novel method for forward-projecting mortality due to both endemic and epidemic-prone infections in Gaza between 7 October 2023 and 6 August 2024. After defining alternative crisis scenarios, we used expert elicitation to quantify the probability of specific epidemics and increases in transmissibility and case fatality. We analysed past vaccination coverage and natural

Poultry farming faces fowl cholera caused by Pasteurella multocida, yet genomic data are limited. Isolates from ISA Browns were identified using matrix-assisted laser desorption/ionization time-of-flight (MALDI-TOF) and sequenced (8.6, 1.4, and 1.3 M reads). De novo assemblies revealed genome sizes of ~2.4-2.5 Mb and guanine or cytosine (GC) content of ~40%.

The seventh pandemic of cholera, caused by the seventh pandemic El Tor lineage of Vibrio cholerae, was previously shown to have emanated in three global waves from the Bay of Bengal, bordering Bangladesh and India1. However, the respective roles of the Ganges Delta and Basin regions in seeding these global pandemic waves were not known. Here we show that, although transmission events occur between Bangladesh and India, V. cholerae in the two countries has largely evolved separately over the past 20 years, apparently constrained by national borders rather than by hydrological features, such as

OBJECTIVE: To identify priority areas for multisectoral interventions for cholera control in Cameroon.
METHODS: We collected data on cholera cases from January 2016 to September 2023 in all 10 regions of Cameroon sourced from the DHIS-2 software, national cholera line lists, situation reports and databases of the Centre Pasteur du Cameroun and the National Public Health Laboratory. We entered these data into the Global Task Force on Cholera Control tool to determine a priority index for districts based on four cholera indicators: incidence, mortality, persistence and test positivity. We calcu

UNLABELLED: Environmental pressures and repeated exposure to antimicrobials drive microbial evolution, often through genomic changes or acquisition of mobile genetic elements that enhance fitness and resistance. This has critically influenced the control of cholera, particularly with the emergence of multidrug-resistant Vibrio cholerae strains, such as those recently reported in Yemen. To explore the long-term dynamics of antimicrobial resistance (AMR), we analyzed 149 clinical Vibrio isolates collected in India over 16 years (2008-2023), including five cases of Vibrio paracholerae. Phenotypic

The causative agent of cholera, Vibrio cholerae, is a bacterium native to the aquatic environment and commensal to zooplankton, namely copepods. V. cholerae thrives in warm, moderately saline water and its incidence is strongly influenced by environmental factors, which have proven critical for predictive awareness of cholera by identifying outbreak locations and timing. Susceptible-Infected-Recovered (SIR) models provide useful information for understanding transmission dynamics and epidemic curves of disease outbreaks. Previous such models lacked predictive ability due to limited data in reg

Healthcare worker (HCW) training on epidemic preparedness is vital for effective outbreak detection, reporting and response, especially in resource-limited settings. However, traditional training methods often fail to address the specific needs of frontline HCWs, causing significant delays in disease surveillance and response. This paper explores the Clinical Integrated Disease Surveillance and Response (cIDSR) training model, designed to supplement existing IDSR efforts and address key barriers in epidemic detection and notification. Piloted in Nigeria and Uganda, cIDSR provides HCWs with free

Spinal cord injury induces profound molecular changes in surrounding tissue. Deciphering these changes with cell type-specific resolution shall facilitate discovery of new molecular targets that promote recovery. Here, we performed a proteomic analysis of laser-dissected motoneurons (MNs), ependymal cells (ECs), and dorsal root ganglia (DRG) obtained from adult rats 2 or 6 weeks after thoracic spinal cord transection at T11-T12 level, and control animals (n = 5-6 rats per group). We traced with fluorescent cholera toxin and laser-microdissected MNs innervating soleus muscle (SOL, n = 172 ± 39

Factors associated with severe dehydration from cholera among children in the community is still not fully understood. We analyzed the characteristics of the pediatric cases who were hospitalized in the community cholera treatment centers in the capital Lusaka between October 2023 and May 2024. Presence of underlying conditions (e.g. human immunodeficiency virus (HIV) infection, severe acute malnutrition), specific catchment areas (e.g. low-income residential areas), and an early phase of the outbreak was associated with increased numbers of severe cases in the community by multivariate analys

BACKGROUND: Housing conditions play a fundamental role in infectious diseases transmission, particularly in endemic regions. Structural modifications to homes-such as screened windows and doors, sealed eaves, elevated flooring, and insecticide-treated barriers-are increasingly recognized as key strategies for reducing mosquito entry and, consequently, malaria risk. Similarly, improved latrine, proper waste disposal, and the possibility to hand wash can help to reduce the spread of waterborne diseases such as cholera. However, their effectiveness and feasibility vary across geographical and soc

Cholera is a diarrheal disease caused by Vibrio cholerae, which secretes cholera enterotoxin (CTX) in the small intestinal epithelium. The pentameric B subunit (CTB) of CTX binds to glycans on the cellular surface through a primary binding site for GM1 glycosphingolipid or galactose. However, GM1 is undetectable in the human SI, and fucose is a key alternative ligand. Previously, we developed linear norbornenyl glycopolymers that block CTB binding by forming CTB-glycopolymer aggregates. Here, we evaluate CTB-glycopolymer binding kinetics under flow conditions using surface plasmon resonance. A

Vaccinations against bacterial pathogens play a crucial role in infection prevention, especially given the growing prevalence of antibiotic resistance. Historically, vaccines against cholera, diphtheria, and pertussis (whooping cough) marked the first significant advances in the fight against bacterial diseases. Later, vaccines against meningococcus and tetanus, among others, were added. These active immunizations expose the immune system to components of the pathogen, stimulating the production of antibodies, T helper cells, B cells, and memory cells, thereby providing long-term protection. BACKGROUND: Cholera continues to be a significant public health threat, particularly in underserved regions with overcrowding and poor sanitation. The 2022-2023 cholera outbreak in Syria, particularly in Aleppo, overwhelmed medical staff, who faced extreme stress, extended work hours, and department closures. This cross-sectional study examines the association between this outbreak and the psychological distress, fatigue, and post-traumatic stress disorder (PTSD) experienced by healthcare workers.
METHODS: This cross-sectional study, conducted at Aleppo University Hospital between September 2

Light is one of the most pervasive physical cues in aquatic environments, yet its impact on nonphototrophic pathogens remains largely unexplored. Here, we show that a strain of cholera bacterium Vibrio cholerae directly couples illumination to motility through cyclic AMP (cAMP) signaling. Exposure to visible light rapidly elevates intracellular cAMP and increases swimming speed, whereas deletion of the single adenylyl cyclase gene (cyaA) abolishes both responses; complementation or addition of exogenous cAMP restores the phenotype. Heterologous expression of V. cholerae CyaA in an Escherichia
