Nanomolar Cadmium Disrupts Neurotransmitter Release Timing via a ROS-dependent Mechanism at the Mouse Neuromuscular Junction: Modulation by Nanomolar Zn2.

Among heavy metals, cadmium (Cd2+) is the most widespread pollutant, exhibiting pronounced neurotoxicity and exacerbating neurodegenerative diseases. Even at nanomolar concentrations in plasma, Cd2+ increases the risk of multiple disorders. The mechanisms underlying the detrimental effects of nanomolar Cd2+ on the nervous system are far from fully understood. Using microelectrode recordings and fluorescence approaches, we investigated the effects of low Cd2+ concentrations on acetylcholine release and redox balance at the mouse neuromuscular junction. Similar to voltage-gated Ca2+ channel bloc

Inferior olive lesions in autoimmune glial fibrillary acidic protein astrocytopathy: a report of two cases.

BACKGROUND: Autoimmune glial fibrillary acidic protein (GFAP) astrocytopathy is a newly identified inflammatory nervous system disorder characterized by Immunoglobulin G autoantibodies against GFAP. While several MRI features are considered typical for autoimmune GFAP astrocytopathy, reports of lesions affecting the inferior olivary nucleus (ION) are rare. In this context, we present two cases of autoimmune GFAP astrocytopathy with lesions in the ION. CASE PRESENTATION: Case 1 involved a 64-year-old man with meningoencephalomyelitis, peripheral neuropathy, and autonomic dysfunction. His brain

Inhibition of USP7 Destabilizes the Noncanonical PRC1.1 Complex and Induces Neuroblastoma Differentiation.

UNLABELLED: Pediatric cancers are frequently driven by genomic alterations that result in impaired differentiation during development. To identify complex-level dependencies required for differentiation in neuroblastoma, a pediatric cancer of the developing peripheral nervous system, we curated a list of protein complexes using the CORUM database and mined the Dependency Map using gene set enrichment analysis. This analysis identified the noncanonical polycomb repressive complex 1.1 (PRC1.1) complex, which represses transcriptional activity through ubiquitination of histone 2A, lysine 119 (H2A

The role of prostanoids in the retina and optic nerve in health and disease.

Prostaglandin D2 (PGD2), PGE2, and other prostanoids play important roles in retinal and optic nerve (ON) health and disease, including neovascularization, neuroinflammation, and neurodegeneration. We reviewed the synthesis, expression, regulation, mechanisms, and functions of the principal prostanoids (PGF2α, PGI2, TXA2, PGE2, and PGD2) in the eye. Prostanoids can be difficult to study in-vivo or ex-vivo because they are small, unstable molecules. To address this, we incorporated lipidomics data clarifying the prostanoids present in the mouse retina and ON, and mined single-cell transcriptomi

Human induced pluripotent stem cell-based models for studying neural repair.

Injuries and degenerative diseases of the human nervous system result in irreversible functional loss, reflecting the limited regenerative capacity of the central nervous system and the slow repair rate of the peripheral nervous system. Progress has been hindered by the lack of human-relevant experimental models that accurately capture the cellular diversity, long axonal architecture, and species-specific regulatory mechanisms underlying neural injury and repair. Human induced pluripotent stem cells (iPSCs) have emerged as a transformative platform to bridge this gap, enabling the generation o

Impact of ageing on progressive thinning of the retinal nerve fibre layer in glaucoma.

BACKGROUND/AIMS: To determine the proportion of progressive retinal nerve fibre layer thickness (RNFLT) change that can be attributed to normal ageing. METHODS: This prospective, longitudinal, cohort study included one eye each of 200 patients treated for open-angle glaucoma and 73 healthy control subjects who were followed with optical coherence tomography (OCT) every 6 months. A linear mixed-effects (LME) model was used to estimate the mean RNFLT slope and the difference between the two groups to evaluate factors affecting the slope, while adjusting for other variables. RESULTS: The median

Low-intensity focused ultrasound-activated piezoelectric gel bandage for diabetic wound repair and neuropathic pain relief.

Nociceptive hypersensitivity and prolonged wound healing due to the interaction of peripheral neuropathy and local immune disorders are key issues that need to be addressed in diabetic foot ulcer (DFU) regenerative repair. Here, we present a low-intensity focused ultrasound (LIFU)-activated piezoelectric gel bandage for DFU wound repair and neuropathic pain relief. Acting as an artificial "skin", this self-powered, functionalized bandage not only temporarily shields tissue from external environment, but also serves as a drug reservoir that can in situ release nitric oxide upon LIFU to promote

Hyperhidrosis Improved by Samhwangsasim-Tang: A Case Report.

Hyperhidrosis is a disorder characterized by excessive sweating beyond physiological requirements, which significantly interferes with daily functioning and quality of life. It affects approximately 4%-5% of the population worldwide, yet its underlying pathophysiology remains unclear, with proposed mechanisms involving autonomic nervous system dysregulation, metabolic imbalance, and psychological factors. Here, we describe a case of chronic hyperhidrosis in which spontaneous sweating and facial flushing improved markedly following administration of Samhwangsasim-tang as monotherapy. A 66-year-

Structural and Functional Impact of Spinal Cord Stimulation in Diabetic Peripheral Neuropathy.

Diabetic peripheral neuropathy (DPN) is a common and debilitating complication of diabetes that leads to chronic pain, sensory loss, and functional decline. While spinal cord stimulation (SCS) is FDA-approved for painful diabetic neuropathy, its potential to modify disease pathology rather than merely suppress symptoms remains poorly understood. This narrative review bridges this knowledge gap by elucidating the role of SCS in mitigating structural and functional changes in the spinal cord and the peripheral nervous system in DPN. Severe Juvenile Dermatomyositis With Peripheral Nervous System Involvement: A Case Report.

Juvenile dermatomyositis is the most common inflammatory myopathy of childhood. While many cases achieve favorable outcomes, severe forms may develop persistent disease activity, complications, and long-term disability. A five-year-old girl was admitted after 20 days of Gottron's papules and heliotrope rash, with intermittent fever during the first 10 days. She then developed mild proximal-predominant muscle weakness in all four limbs, rapidly progressing to severe generalized weakness with signs of axonal neuropathy over the subsequent 15 days. Despite aggressive immunosuppression with IV met

Influenza A-Associated Acute Necrotizing Encephalitis With Complete Recovery Following Early Immunomodulatory Therapy.

We describe a case of a previously healthy 19-month-old female who experienced rapid neurological deterioration following infection with influenza A (H1N1) and was found to have neuroimaging features characteristic of acute necrotizing encephalitis (ANE), including bilateral thalamic involvement and additional multifocal, symmetric gray- and white-matter lesions. At initial presentation, multiple proinflammatory cytokines were markedly elevated in the peripheral blood. Interestingly, while multiple cytokines were also elevated in cerebrospinal fluid (CSF), interleukin-6 (IL-6) and IL-8 levels

Peptidoglycan recognition proteins in the brain: Role in neuroinflammation and behavioral consequences.

Peptidoglycan recognition proteins (PGRPs) constitute an evolutionarily conserved family of pattern recognition molecules that detect bacterial peptidoglycan. While their antimicrobial functions have been well characterized in peripheral immunity, recent discoveries have unveiled critical roles for PGRPs in central nervous system inflammation and behavior. Among the four mammalian PGRP family members, PGLYRP1 exhibits unique expression in brain microglia and demonstrates potent pro-inflammatory properties in neurological contexts. Recently, PGLYRP1 has been shown to function as a key amplifier

Efficacy and Safety of Ultrasound-Guided Right-Sided Stellate Ganglion Block for Reducing the Severity and Duration of Acute Paroxysmal Sympathetic Hyperactivity in Patients with Acquired Brain Injury: A Randomized Controlled Clinical Trial.

Paroxysmal sympathetic hyperactivity (PSH) is a severe autonomic disorder following acquired brain injury for which there are no effective targeted therapies. The aim of this study was to evaluate whether ultrasound-guided right-sided stellate ganglion block (RSGB) can safely and effectively attenuate acute episodes of PSH. A prospective, single-blind randomized controlled trial was conducted. Eighty patients with PSH were randomized to receive either conventional care or RSGB combined with conventional care. Primary outcomes included PSH episode intensity and duration as assessed by the Clini

Gastroesophageal reflux-induced cough: the positive feedback loop of microaspiration and neurogenic inflammation.

Gastroesophageal reflux-related cough (GERC) is a common etiology of chronic cough, characterized by a complex pathogenesis. The limited efficacy of conventional acid suppression therapy suggests the presence of critical mechanisms that extend beyond mere gastric acid irritation. This review systematically explores the interaction between the two core pathways-"microaspiration" and "neurogenic inflammation"-in the context of GERC, aiming to elucidate their synergistic pathogenic mechanisms. On one hand, the gastric contents involved in microaspiration (such as gastric acid and pepsin) directly

A Combinatorial Strategy for Peripheral Nerve Repair Utilizing 3D-Printed Collagen Nerve Guidance Conduits and Photobiomodulation Therapy: A Pilot Study in a Rat Sciatic Nerve Transection Model.

BACKGROUND: Nerve guidance conduits (NGCs) are a clinically approved option for peripheral nerve repair but remain ineffective compared with autograft "gold standard" treatment options. A combinatorial approach to conduit-based peripheral nerve repair could improve repair outcomes. Photobiomodulation therapy (PBMT) has shown potential to enhance axonal regeneration, but its efficacy in combination with NGCs requires further evaluation. OBJECTIVE: This pilot study investigated the therapeutic potential of a novel 3D-printed, collagen-based NGC combined with PBMT for peripheral nerve regenerati

Replating Induces mTOR-Dependent Rescue of Protein Synthesis in Charcot-Marie-Tooth Diseased Neurons.

Charcot-Marie-Tooth disease (CMT) is an inherited peripheral neuropathy characterized by sensory dysfunction and muscle weakness, manifesting in the most distal limbs first and progressing more proximal. Over a hundred genes are currently linked to CMT with enrichment for activities in myelination, axon transport, and protein synthesis. Mutations in tRNA synthetases cause dominantly inherited forms of CMT, and animal models with CMT-linked mutations in these enzymes display defects in neuronal protein synthesis. Rescuing protein synthesis in CMT-mutant neurons could offer exciting therapeutic

Gelsolin-3: A Novel Myelin Autoantigen in Demyelinating Guillain-Barré Syndrome.

OBJECTIVE: Although immunoglobulin G (IgG) autoantibodies against gangliosides are well-established in axonal Guillain-Barré syndrome (GBS), specific autoantibodies in demyelinating GBS-including acute inflammatory demyelinating polyneuropathy (AIDP) and its variant, bifacial weakness with paresthesias (BFP)-remain unidentified. We aimed to identify novel autoantibodies and clarify the immunological link between AIDP and BFP. METHODS: Immunoprecipitation and mass spectrometry identified gelsolin from BFP serum IgG targeting non-compact, periaxonal myelin surrounding the nodes of Ranvier. West

OPA1 Deficiency Impairs NGF Signaling and Drives Sympathetic Neurodegeneration.

Peripheral sympathetic neurodegeneration drives cardiac dysfunction in dominant optic atrophy, revealing a critical neuro-cardiac link. Optic atrophy factor-1 haploinsufficiency disrupts mitochondrial dynamics and neurotrophic signaling, causing targeted sympathetic denervation and arrhythmias. Restoring nerve growth factor transport and mitochondrial health in sympathetic neurons represents a promising therapeutic avenue for cardiac autonomic disorders. Future research must unravel mechanisms of neurocardiac crosstalk to develop precise interventions against neurogenic cardiac disease progres
