Laser patterning of ECM-derived biomaterials to direct degradation, site-specific resorption, controlled vascularization and functional repair of large nerve defects.

Controlled degradation of extracellular matrix-derived biomaterials in a site-specific and temporal sequence might facilitate early vascularization and improve tissue regeneration. In this study, we developed a tailored laser patterning treatment that successfully addresses this challenge. We show that application of a focused diode-pumped solid-state laser (532 nm) for 30 s duration leads to local heating and reduction of collagen fibril integrity in localized laser-patterned areas of a collagen biomaterial. When implanted in vivo, these thermally degraded regions then become susceptible to f

Versatile role of fibro-adipogenic progenitors in neuromuscular junction homeostasis and pathology.

The neuromuscular junction (NMJ) is a specialized synapse that enables reciprocal signaling between motor nerves and muscle fibers. NMJ integrity is essential for muscle homeostasis, while retrograde signals from muscle also modulate nerve axon physiology. Nerve injury or neuromuscular diseases often result in functional deficits and histological changes. Fibro-adipogenic progenitors (FAPs) are muscle-resident multipotent mesenchymal cells, and a large body of research illustrated their role in muscle mass maintenance and regeneration. Recently, FAPs have been suggested to be responsive to NMJ

SUPT16H-associated neurodevelopmental disorder and neurocristopathy: genetic and phenotypic spectrum.

SUPT16H encodes a subunit of the FACT (FAcilitates Chromatin Transcription) complex, a histone chaperone essential for maintaining chromatin integrity during transcription, replication, and DNA repair. Pathogenic de novo SUPT16H missense variants have previously been linked to neurodevelopmental disorders in eight individuals. Here, we expand the genotypic and phenotypic spectrum by identifying 24 additional individuals harboring ultrarare heterozygous missense or truncating variants, who share overlapping clinical features including intellectual disability, autism spectrum disorder, hypotonia

Building consensus: construction of a juvenile and adult scRNA-seq meta-atlas for dataset comparisons and harmonizing transcriptomic definitions of enteric neuron subtypes.

BACKGROUND: The enteric nervous system (ENS) is a complex network of interconnected ganglia within the gastrointestinal (GI) tract. Among its diverse functions, the ENS detects bowel luminal contents and coordinates the passing of stool. ENS defects predispose to GI motility disorders. Previously, distinct enteric neuron types were cataloged by dye-filling techniques, immunohistochemistry, retrograde labeling, and electrophysiology. Recent technical advances in single cell RNA-sequencing (scRNA-seq) have enabled transcriptional profiling of hundreds to millions of individual cells from the int

Alterations in Both Caliber and Myelination of Callosal Axons Elicited by Ubiquitous Genetic Ablation of c-Jun Amino-Terminal Kinase 3 (JNK3).

Myelination is a fundamental process supporting appropriate motor, sensory, and cognitive functions. During development, oligodendrocyte progenitor cells (OPCs) proliferate, migrate, and gradually differentiate into mature oligodendrocytes, which produce and assemble myelin sheets that insulate axons in the mammalian central nervous system. Recent evidence suggested a regulatory role of the protein kinase JNK1, one of three mammalian JNK isoforms, on the proliferation and differentiation of OPCs, but whether other JNK isoforms modulate these and other cellular events contributing to myelinatio

Circulating Neutrophil and Monocyte Counts Are Inversely Associated With Heart Rate Variability Indices: The Maastricht Study.

BACKGROUND: Inflammatory biomarkers, including immune cells appear associated with reduced heart rate variability (HRV), an indicator of cardiac autonomic nervous dysfunction. However, the influence of leukocytes on HRV and their relation across sex and glucose metabolism status remains unclear. Hence, we sought to investigate the cross-sectional association between leukocytes and HRV indices and their association across sex and glucose metabolism status in a general population. METHODS: We retrieved cross-sectional data of 1888 participants of the Maastricht Study who completed the baseline

Effectiveness and cost-effectiveness of ozone treatment in patients with paraesthesia (numbness, tingling) secondary to chemotherapy-induced peripheral neuropathy: randomized, triple-blind clinical trial (OzoParQT).

BACKGROUND: Chemotherapy-induced peripheral neuropathy (CIPN) is a frequent, disabling side effect of taxanes and platinum drugs, often compromising quality of life and treatment continuity. Existing therapies are few and largely ineffective. Given this unmet need, our prior experience suggests ozone therapy may offer clinical benefit as an adjuvant treatment. METHODS: OzoParQT is a Phase II–III randomized, triple-blind trial including 42 adults (≥ 18 years) with any cancer and Grade ≥ 2 CIPN (numbness and/or tingling) lasting ≥ 3 months. Eligible patients must be off neurotoxic chemotherapy f

Pyroptosis in Peripheral Neuropathy: From Molecular Mechanisms to Therapeutic Targeting.

BACKGROUND: Peripheral neuropathy (PN) is a common consequence of peripheral nervous system (PNS) disorders, yet its effective treatment remains a significant clinical challenge. Pyroptosis, an inflammatory form of programmed cell death (PCD) triggered by gasdermin A-E (GSDMA-E), contributes to the pathogenesis of PN and represents a promising therapeutic target. While reviews of pyroptosis in other diseases are extensive, comprehensive reviews focusing on PN are lacking. METHOD: We systematically searched PubMed, Scopus, Web of Science, and Google Scholar (1986-2025). Only original studies i

Characteristics of motor polyneuropathy in Fabry disease: a case-control study.

In Fabry disease (FD) neuropathic pain is often the first symptom due to impairment of the small fibres of peripheral nerves. The problem of large fibre involvement in FD has not been described well. This prompted us to investigate electrophysiological changes in motor fibres with different conduction velocities in FD patients. The study group consisted of 20 patients with FD (8 women and 12 men); mean age was 40.65 ± 11.94 years, and a matched control group. Medical history, neurological examination, together with standard neurography and conduction velocity distribution (CVD) tests of the uln

A CNS-Directed, AAV9 Gene Therapy Restores Expression and Biochemical Function of Guanidinoacetate Methyltransferase in Models of GAMT Deficiency.

Guanidinoacetate methyltransferase (GAMT) is an essential enzyme in the biosynthesis of creatine, an important molecule in energy recycling. GAMT loss of function leads to GAMT deficiency (GAMT-D), an autosomal recessive disorder resulting in low creatine levels and the accumulation of a toxic intermediate, guanidinoacetate (GAA). GAMT-D patients present with intellectual disability and epilepsy, emphasizing the detrimental consequences of disturbed creatine metabolisms in the central nervous system (CNS). Current treatments are not curative and may not restore creatine metabolism in the brain

TEP-1, a glial thioester protein is required for cilia organization and intraflagellar transport in ensheathed sensory neurons in Caenorhabditis elegans.

Age-related macular degeneration (AMD), the leading cause of blindness in the elderly, is characterized by progressive degeneration of retinal photoreceptors. Traditional disease models suggest that defective repression of thioester protein C3 activity by complement factor H (CFH) is a major contributor to pathogenesis in AMD and a related disease, early-onset drusen maculopathy (EODM). Our previous study identified novel functions for human CFH and Caenorhabditis elegans CFH-1 in the maintenance of inversin compartment integrity in photoreceptors and mechanosensory neurons, indicating that CF

Double-blind, sham-controlled, pilot study of trigeminal nerve stimulation for autism spectrum disorder.

Trigeminal nerve stimulation (TNS) is a minimal-risk, noninvasive neuromodulation method with growing evidence of efficacy across psychiatric conditions. However, its safety and potential effects in autism spectrum disorder (ASD) remain underexplored. This exploratory pilot study aimed primarily to evaluate the safety and tolerability, and secondarily to explore changes in ASD-related symptoms - including impairments in social communication and reciprocity, attention, executive functioning, emotional regulation, sleep, and sensory processing - in children with ASD, and to examine associated ch

Artery Tertiary Lymphoid Organs Establish Biological Platforms to Connect Atherosclerotic Plaques to the Brain.

PURPOSE OF REVIEW: The ability of the nervous system to sense cues from medium and large arteries is critical for maintaining vascular integrity and function. However, there is currently no evidence that atherosclerotic plaques of arteries other than arterioles are directly hard-wired by axons to transmit signals to the brain or that efferent axons from the brain gain access to plaques. Yet, studies in experimental mice have shown that the outer connective tissue coat of arteries, i.e. the adventitia, adopts sentinel functions to indirectly and faithfully connect plaques to the brain forming a

Protocadherin 20 Is a POU Class 2 Homeobox 3 Target Gene Required for Proper Tuft Cell Microvillus Organization.

BACKGROUND & AIMS: Tuft cells play protective roles in infection, inflammation, and tumorigenesis through the secretion of cytokines and eicosanoids. Tuft cells are known for their tall, blunt microvilli, thought to be analogous to mechanosensory hair cell stereocilia; however, a functional role for the microvillar apparatus has not been identified. POU Class 2 Homeobox 3 (POU2F3) is the master regulator transcription factor for tuft cells, yet how POU2F3 drives formation of this unique structure is unknown. Here, we aimed to identify POU2F3 target genes and commonalities between tuft and hair

Immune cells in cerebral small vessel disease: Emerging roles and potential targeting therapies.

Cerebral small vessel disease is an age-related condition that severely affects the quality of life of older adults; however, there are currently no definitive treatments or preventive measures. Cells from both the peripheral and central immune systems significantly impact the development of cerebral small vessel disease. By analyzing the effects of different immune cells on brain dysfunction associated with this disease, we aim to explore the various mechanisms through which autoimmune and acquired immune cells in peripheral circulation and the central nervous system contribute to disease dev

Single cell analysis of developing Merkel cells reveals the emergence of non-coding RNA biotypes as a hallmark of terminal differentiation.

RNA processing generates diverse protein-coding and non-coding transcripts, yet RNA biotype diversity during cellular differentiation is not well characterized. Merkel cells (MCs) are cutaneous mechanosensors. We analyzed full-length transcripts of FACS-sorted single mouse MCs at all stages of development and discovered that their terminal differentiation was accompanied by an emergence of non-coding transcripts associated with genes related to MC function. Non-coding RNAs upregulated during terminal differentiation included retained intron transcripts capable of forming nuclear condensates th

Microglial, astrocytic, oligodendrocyte, B-/T-cell and neutrophil dysregulation in neuroinflammation of Alzheimer's disease and related dementias.

Wide number of basic, preclinical and clinical research have led to increase our knowledge on understanding the potential roles of neuroinflammation in neurodegenerative diseases such as Alzheimer's disease (AD). In fact, neuroinflammation is considered as one of the key pathological factors of neuronal dysfunction leading to dementia in AD and other forms of dementias or related dementias (RD). Central nervous system (CNS) network communicates and control the immune system signaling pathways within the brain as well as peripheral part of the body. Hence, disturbance in the physiological immun

Generation and characterization of iPSC-derived microglia for in vitro modeling of stimuli-specific neuroimmune responses.

INTRODUCTION: Microglia are macrophage-like brain resident immune cells known to express numerous Alzheimer's disease risk genes. Here we generated a human induced pluripotent stem cell (iPSC) derived microglia cell culture model for use in neuroimmune modeling and therapeutic testing. METHODS: We generated iPSC lines using episomal reprogramming for subsequent stepwise differentiation of iPSC-derived microglia (iMG) without commercial kits. We characterized the responses of this model to immunogenic stimuli and recombinant TREM2 antibodies. RESULTS: The iMG expressed several key microglia s
