Role of LRRK2 in physiological activities, diseases, and therapy.

Leucine-rich repeat kinase 2 (LRRK2) is a critical target for the treatment of Parkinson's disease (PD) and potentially other diseases. LRRK2 is involved in intracellular signaling, immune response, and inflammation, with key roles in both the central nervous system and peripheral tissues. LRRK2 mutations are linked to cellular dysfunction including mitochondrial and neuronal damage and can disrupt signaling pathway balance, thereby contributing to PD and other disorders. Inhibiting LRRK2 kinase activity shows potential for treating PD by correcting cellular imbalances and reducing neuronal da

Peripheral and CSF protein quantification in Parkinson's disease and multiple system atrophy-the nucleic acid-linked immuno-sandwich assay.

There are currently no validated peripheral biomarkers for the diagnosis, differentiation or progression of the neurodegenerative synucleinopathies, Parkinson's disease and multiple system atrophy. Diagnostic biomarkers that reflect the disease mechanisms or progression biomarkers that change with disease severity would be extremely valuable for assessing disease-modifying therapies. Our objective was to explore putative protein biomarkers of Parkinson's disease and multiple system atrophy, in relation to clinical disease severity, using the nucleic acid-linked immuno-sandwich assay central ne

Opening of rod photoreceptor CNG channels by photodynamically generated singlet oxygen.

Molecular oxygen exists in three electronic states: the triplet ground state and two singlet (1O2) excited states. Under physiological conditions, 1O2 can be produced either through photodynamic processes, which require light, photosensitizer, and oxygen, or via metabolic reactions involving enzymes and other reactive oxygen species (ROS). 1O2 readily reacts with biomacromolecules, however, its volatile chemical nature and the lack of precise working models hamper the study of its molecular mechanism and physiological significance. Here we report that human CNG channels from rod photoreceptors

Immunotherapy considerations for distal sensory-predominant and small-fiber peripheral neuropathies.

Increasing knowledge about the common widespread, distal-predominant polyneuropathies is shifting the standard of care beyond merely palliating symptom toward intervention. This is less advanced for the more prevalent but harder-to-ascertain distal sensory polyneuropathies (DSP) than for motor and demyelinating neuropathies. DSP comprises the sensory-predominant large-fiber axonal/demyelinating neuropathies plus the small-fiber sensory/autonomic axonopathies. Small-fiber disturbances can cause premature fatigue, neuropathic pain and itch, postural orthostasis tachycardia, and gastrointestinal

Bupleurum chinense DC. ameliorates depression through hepatic lipopolysaccharide-binding protein-associated inflammation and monoamine biosynthesis.

ETHNOPHARMACOLOGICAL RELEVANCE: Bupleurum chinense DC. (Radix Bupleuri, Chaihu) has traditionally been applied to "soothe the liver and relieve depression." Contemporary pharmacological evidence indicates that Radix Bupleuri (RB) and its active components exhibit anti-inflammatory, antitumor, and antidepressant effects. Nevertheless, the mechanisms underlying its antidepressant activity remain unclear. AIM OF THE STUDY: To evaluate the antidepressant-like effects of RB in mouse models and to determine whether modulation of peripheral inflammatory status is associated with changes in the brain

Reviewing the Peripheral and Central Mechanisms of Visceral Hypersensitivity in Intestinal Disorders.

Visceral hypersensitivity (VH) is a condition where the internal organs have an enhanced sensitization to normal physiological stimuli or mild pathological stimuli, leading to chronic visceral pain or other discomforts, which is a typical characteristic of some intestinal disorders, such as irritable bowel syndrome (IBS) and inflammatory bowel disease (IBD). VH might be caused by gene, psychological disorders, social stress factors, gut microbiota, and some other factors, but the exact mechanisms are not yet clear. This review focuses on recent developments in the effect of intestinal cells on

Sarcoidosis With Muscular and Peripheral Nervous System Involvement: An Atypical Presentation of a Rare Disease.

Neuromuscular sarcoidosis is uncommon and may represent a diagnostic challenge, particularly in the absence of more typical sarcoidosis manifestations such as pulmonary involvement. We report a case of a 71-year-old male presenting with constitutional symptoms, weight loss, progressive gait impairment, hypercalcemia, and acute kidney injury. Initial evaluation excluded malignancy, active infection, and monoclonal gammopathy. Advanced imaging showed diffuse muscular involvement, and muscle biopsy confirmed non-caseating granulomatous myositis, establishing the diagnosis of muscular sarcoidosis.

Liver clock: Malalignment and entrainment therapeutics.

The mammalian circadian system comprises of a central clock in the suprachiasmatic nucleus (SCN) and peripheral clocks in body tissues and organs i.e. liver clock. Both the central and liver clocks are based on clock genes and their protein products, which regulate circadian rhythms in metabolism, physiology, and behavior by modulating nervous and harmone signaling, including the sympathetic nervous system, fasting and eating behavior. The SCN is primarily entrained by light-dark cycles. Liver clock genes can be influenced by other factors, such as feeding cycles and specific nutrients. Light

Multidimensional profiling of heterogeneity in supratentorial ependymomas.

Supratentorial ependymomas are aggressive childhood brain cancers that retain features of neurodevelopmental cell types1 and segregate into molecularly and clinically distinct subgroups2,3, suggesting different developmental roots. The developmental signatures, as well as microenvironmental factors, underlying aberrant cellular transformation and behaviour across each supratentorial ependymoma subgroup are unclear. Here we integrated single-cell and spatial transcriptomics, as well as in vitro and in vivo live-cell imaging, to define supratentorial ependymoma cell states, spatial organization

Shared autonomic phenotype of long COVID and myalgic encephalomyelitis/chronic fatigue syndrome.

INTRODUCTION: Long COVID and myalgic encephalomyelitis/chronic fatigue syndrome (ME/CFS) are relatively common and disabling multisystem disorders that share overlapping features, including post-infectious onset and similar clinical manifestations such as brain fog, fatigue, muscle pain, and dysautonomia with orthostatic intolerance. These similarities suggest that Long COVID and ME/CFS may share common pathophysiological mechanisms, though the underlying mechanisms remain poorly understood, partly due to the difficulty in quantifying many of the symptoms. MATERIALS AND METHODS: This retrospe

Translocation of bacteria from the gut to the brain in mice.

Recent advances suggest a correlation between gut dysbiosis and neurological diseases, however, relatively little is known about how gut bacteria impact the brain. Here, we reveal that bacteria can translocate directly from the gut to the brain in small numbers when mice are fed an atherogenic, high-fat diet (Paigen diet) that causes alterations in gut microbiome composition and gut barrier permeability. The bacteria were not found in other systemic sites or the blood, but were detected in the vagus nerve. Right cervical vagotomy reduced bacterial burden in the brain, implicating the vagus ner

Structural and functional investigation of RD3-GCAP1 interaction in retinal photoreceptors under normal and disease conditions.

Guanylate Cyclase Activating Protein 1 (GCAP1) and Retinal Degeneration Protein 3 (RD3) are key regulators of retinal guanylate cyclase 1 (GC1), whose dysregulation leads to inherited retinal dystrophies (IRDs). While GCAP1 mutations cause constitutive GC1 activation and photoreceptor degeneration, RD3 acts as a potent cyclase inhibitor essential for proper GC1 trafficking. Here, we investigated the molecular interaction between GCAP1 and RD3 as well as its perturbation by IRD-associated GCAP1 mutations (D100G, N104H, E111V, E155G) using NMR spectroscopy, surface plasmon resonance, AlphaFold3

miRNA 183 Knockout Alters Cone Subtype Distribution, Transcriptional Activity and ERG Signals in the Tetrachromatic Zebrafish Visual System.

miRNA 183 is part of the miRNA-183/96/182 cluster, which is known to play a decisive role in fine-tuning the activity of gene expression in sensory systems, particularly in the retina. Although miR-183 is essential for retinal gene expression in mammals, the contributions of miR-183 to mRNA expression and photoreceptor development and function in other classes of animals have not been fully elucidated. Danio rerio have a diverse photoreceptor system, with cone photoreceptors sensitive to red, green, blue and ultraviolet (UV) light. We generated knockout zebrafish by deleting the whole seed seq

Vasomotor Function and Its Modulation by Perivascular Adipose Tissue Vary Depending on Metabolic Syndrome Stage.

BACKGROUND: Perivascular adipose tissue (PVAT), found outside blood vessels, enhances vasorelaxation when endothelium-mediated vasorelaxation fails, as observed in the mesenteric arteries of rats with metabolic syndrome (MetS) and hypertension; however, whether high blood pressure contributes to PVAT dysfunction remains unclear. We, therefore, aimed to investigate vascular function and PVAT modulation in the mesenteric arteries of rats with MetS but no hypertension. METHODS: Mesenteric arteries were isolated from both lean and obese male Zucker fatty (ZF) and ZF diabetes mellitus (ZFDM) rats

Multisystem Lomentospora prolificans progression on fosmanogepix despite in vitro activity: a call for expanding clinical research on novel antifungals.

BACKGROUND: Lomentospora prolificans is a rare and often fatal cause of invasive mold disease (IMD), particularly in immunocompromised individuals. Treatment remains difficult due to intrinsic resistance to most antifungals and the challenges of achieving therapeutic drug levels in the central nervous system. Fosmanogepix and olorofim are novel antifungal agents with promising activity against difficult-to-treat molds, including L. prolificans, and show potential for excellent central nervous system penetration. CASE SUMMARY: We report a case of disseminated L. prolificans infection with pulm
