Effects of Finasteride exposure on lifespan and locomotor phenotypes.

Finasteride is one of the core medications for treating androgen-associated diseases in men. It exhibits multiple therapeutic functions, including inhibiting 5α-reductase, reducing dihydrotestosterone levels, and improving conditions, including prostate enlargement and hair loss. Using Drosophila as the primary model and a mouse behavioral assay as a complementary approach, we combined developmental and behavioral assessments, morphological analyses of motor and sensory neurons, transcriptomics, and genetic methods to investigate developmental, neuronal, and behavioral changes associated with

Peripheral and cerebrospinal fluid inflammatory markers in sporadic amyotrophic lateral sclerosis an exploratory cross-sectional association study.

Amyotrophic lateral sclerosis (ALS) is a progressive neurodegenerative disorder with an ambiguous etiology, whose onset may be linked to innate immune system activation outside the central nervous system (CNS). We investigated the association between peripheral blood and CNS inflammatory parameters in 45 sporadic ALS patients compared to 30 controls, analyzing peripheral blood tests (C-reactive protein [CRP], blood count, fibrinogen) and cerebrospinal fluid (CSF) immunological measures (Tumor Necrosis Factor alpha [TNF-α], TNF receptor type 1 [TNFR-1], CRP) alongside clinical characteristics.

Development of Italian centile curves for the North Star Ambulatory Assessment and timed items.

Centile curves may improve the interpretation of motor outcomes in specific populations. This study aimed to develop Italian age-specific centile curves for NSAA, rise-from-floor velocity (RFFV), and 10-meter walk/run velocity (10MWRV) in boys with Duchenne muscular dystrophy (DMD) and to assess their agreement with existing UK reference curves. Data from 627 patients (5080 assessments) with an age between 3-18 years were extracted from the Italian DMD Registry. GAMLSS models were used to estimate centile curves. Agreement with UK data (restricted to ages 5-16 years and who are actively on ste

Suspected Bulbar-Predominant Myasthenia Gravis Exacerbation in an Older Adult With Respiratory Deterioration and Silent Aspiration: A Case Report.

Myasthenia gravis (MG) is an autoimmune disorder of the neuromuscular junction characterized by fluctuating skeletal muscle weakness. Bulbar-predominant exacerbations may be difficult to recognize in older adults with multiple comorbidities, particularly when respiratory deterioration is initially attributed to cardiopulmonary disease. We report the case of an 83-year-old woman with generalized acetylcholine receptor antibody-positive MG, dementia with behavioral disturbance, moderate chronic obstructive pulmonary disease (COPD), chronic kidney disease, and previous deep vein thrombosis who wa

Demyelination of the central nervous system and peripheral neuropathy associated with sintilimab treatment for lung adenocarcinoma: a case report.

BACKGROUND: Immune checkpoint inhibitor (ICI) therapy unleashes the body's natural immune system to fight cancer. Neurological immune-related adverse events (NirAEs) are rare but can be severe. CASE PRESENTATION: In this report, we present a 47-year-old man of Han ethnicity who was diagnosed with lung adenocarcinoma with intracranial metastasis. Initially, the patient underwent intracranial radiotherapy followed by first-line and maintenance chemotherapy with pemetrexed, cisplatin and bevacizumab, and a partial response was achieved without significant toxicity. After progression, the patient

A Severe Steroid-Induced Myopathy in a Child With a Pharmacogenetic Predisposition: A Case Report and Systematic Review of the Literature.

BACKGROUND: Glucocorticoids (GCs) are essential in paediatric inflammatory and immune-mediated diseases but are associated with dose- and duration-dependent toxicity. Pharmacogenetic variability in metabolic and transporter pathways may contribute to inter-individual differences in GC adverse effects. CASE PRESENTATION: A 7-year-old girl with recurrent IgA vasculitis required prolonged corticosteroid therapy. During treatment, she developed marked Cushingoid features, progressive proximal muscle weakness, and severe fatigability consistent with steroid-induced myopathy. Pharmacogenetic analys

Protein disulphide isomerase protects against ALS/FTD phenotypes associated with cyclin F in zebrafish and cellular models.

BACKGROUND: Mutations in CCNF, encoding cyclin F, cause a small proportion of amyotrophic lateral sclerosis (ALS) and frontotemporal dementia (FTD) cases. Mutant cyclin F mislocalises from the nucleus to the cytoplasm, impairs the ubiquitin proteasome system, disrupts endoplasmic reticulum (ER) to Golgi transport, and triggers ER stress and apoptosis. Protein disulphide isomerase (PDI), a redox-active ER chaperone of the thioredoxin superfamily, protects against ALS phenotypes induced by mutant SOD1, TDP-43, and FUS, but its activity against mutant cyclin F has not been examined. OBJECTIVES:

OGT regulates RNA-binding protein nucleocytoplasmic partitioning, but this fails in amyotrophic lateral sclerosis.

O-linked N-acetylglucosamine transferase (OGT) catalyzes the O-linked β-N-acetylglucosamine (O-GlcNAc) modification to proteins and has been implicated in a range of diseases, including amyotrophic lateral sclerosis (ALS), a rapidly progressive and currently incurable neurodegenerative disorder. However, the precise mechanisms by which OGT influences ALS pathogenesis remain largely unresolved. In this study, we used human-induced pluripotent stem cell (hiPSC)-derived spinal motor neurons (hiPSC-MNs), the principal cell type that undergoes degeneration in ALS, to systematically reveal that OGT-

Association between malocclusion and temporomandibular joint disorders: evidence from a Mendelian randomization study.

BACKGROUND: Although malocclusion and temporomandibular joint disorders (TMD) frequently co-occur in clinical practice, controversy persists regarding whether this overlap reflects a pathophysiological causal relationship or incidental concurrence. This study aimed to investigate the potential association between malocclusion and TMD using a bidirectional two-sample Mendelian randomization (MR) approach. METHODS: Bidirectional two-sample MR was performed to evaluate the potential causal relationship between malocclusion and TMD. Summary statistics for malocclusion were obtained from the FinnG

Electroacupuncture Attenuates Peripheral Neuropathy Development in Hyperglycemic Rats and Is Associated With NLRP3 Inflammasome Modulation.

BACKGROUND: This study employed a high-energy diet and streptozotocin (STZ) injection to establish a rat model of diabetic peripheral neuropathy (DPN), focusing on inflammatory responses. The aim was to investigate the mechanism by which electroacupuncture (EA) improves the neuroinflammatory microenvironment in diabetic rats and attenuates DPN development. METHOD: Twenty-four male SD rats were randomly assigned using a random number table into control, model, and EA groups, with eight rats in each group. A DPN model was established using a high-energy diet combined with STZ injection. The EA

Integrated Single-Myofibre Multi-Omics Correlates Inflammatory State to Myosin Energetics in ICU-Acquired Weakness.

BACKGROUND: Skeletal muscle atrophy is a pervasive complication of critical illness that worsens survival and recovery, yet remains poorly explained by current clinical or molecular markers. METHODS: To directly connect disease-associated molecular states to the contractile machinery, this study combined sequential functional, transcriptomic and proteomic profiling of the same single human skeletal myofibres from critically ill patients in the intensive care unit with acquired weakness (ICU-AW) and controls. This integrated approach provides a framework for validating molecular signals direct

Efgartigimod-responsive autoimmune peripheral neuropathy with concurrent anti-GM2 IgG and anti-contactin-1 IgG antibodies: a rare case report.

BACKGROUND: Immune-mediated peripheral neuropathies arise from dysregulated immune responses targeting peripheral nerves. Contactin-1 (CNTN1) antibodies, typically of the IgG4 subclass, are associated with autoimmune nodo/paranodopathies and often respond poorly to intravenous immunoglobulin (IVIG). Anti-GM2 IgG antibodies are rare and are usually linked to complement-mediated axonal injury. The coexistence of these antibodies has not, to our knowledge, been previously reported. CASE PRESENTATION: A 73-year-old man developed rapidly progressive quadriparesis, bulbar dysfunction, and respirato

Musculoskeletal symptoms and pain-response behaviors among university students in Palestine: a cross-sectional study.

BACKGROUND: Musculoskeletal (MSK) symptoms are a major contributor to disability worldwide, and university students may be increasingly affected because of prolonged sedentary behavior, intensive digital-device use, and sustained academic workloads. In Palestine, these risks may be compounded by psychosocial stressors and constrained healthcare access. This study assessed the prevalence of MSK symptoms and hand discomfort and examined associated behavioral, ergonomic, and device-use factors among Palestinian university students. METHODS: A cross-sectional study was conducted among 1,052 unive
