Proteomics is a widely applied analysis pipeline in biomedical research and clinical chemistry. In skeletal muscle biology, proteomics has been extensively used to identify and characterize individual proteoforms in normal and diseased contractile tissue. This article outlines mass spectrometric approaches to study the proteome of myofibers and associated cell types. We highlight sample preparation and protein digestion for bottom-up proteomics, as well as mass analyzers and bioinformatic data handling. As an illustrative example, the identification and characterization of the sarcolemmal dyst

Background Trained immunity (TI) refers to a hyperresponsive functional state of innate immune cells, such as inflammatory monocytes, induced in response to a variety of stimuli including signals released from damaged tissue, and characterised by increased cytokine production. Inflammatory monocytes play a critical role in Duchenne muscular dystrophy (DMD) by driving chronic muscle inflammation and disease progression. In this study we examined the role of trained immunity in dystrophic muscle pathology by specifically targeting this process in monocytes. Methods We used the mdx mouse model of

BACKGROUND: Dystrophinopathy comprises a group of X-linked recessive muscular disorders that primarily affect skeletal and/or cardiac muscles. Although more than 90% of dystrophinopathy patients exhibit detectable variants in the DMD gene, a small proportion of cases remain undiagnosed at the molecular level.
METHODS: We employed an integrated multiomics approach, including RNA sequencing (RNA-seq) of muscle tissue, optical genome mapping (OGM), short- and long-read whole-genome sequencing (WGS), and karyotype analysis to elucidate the genetic mechanism in a dystrophinopathy family.
RESULTS:

Duchenne muscular dystrophy (DMD) is a fatal X‑linked genetic disease caused by the absence of functional dystrophin, resulting in muscle membrane fragility, chronic inflammation, and progressive muscle wasting. The lack of effective treatments is further compounded by the slow onset of measurable changes in clinical outcomes, such as muscle strength, which often take a year or longer to become evident. Blood‑based biomarkers that can predict treatment efficacy and monitor disease progression would significantly accelerate therapeutic development. We propose that oxidative modifications to pro

Many fundamental biological processes are governed by mechanical forces, with proteins acting as the key molecular mediators. Elucidating how protein unfolding responds to force is critical for understanding the mechano-pathologies, such as cardiomyopathy and muscular dystrophy. While the unfolding trajectories measured by Single-Molecule Force Spectroscopy (SMFS) map the instantaneous force response against molecular extension, its broader application is limited by time-consuming data collection and high operational costs. Here, we present the first scalable generative diffusion framework for

Duchenne muscular dystrophy (DMD) arises from thousands of distinct mutations at a single X-linked locus that converge on loss of functional dystrophin, and this convergence on one molecular endpoint-rather than any genetic uniformity-has long made the disease appear a candidate for straightforward molecular correction. Three decades of work indicate that the difficulty lies less in restoring dystrophin than in restoring it adequately along four axes: amount, retained domains, breadth of coverage across skeletal muscle, diaphragm, heart and brain-the constraint being extent of coverage rather

Facioscapulohumeral muscular dystrophy (FSHD) is a genetically and epigenetically complex disorder in which the comprehensive characterization of the D4Z4 repeat array remains challenging. We applied Oxford Nanopore long-read sequencing (ONT-LRS) to three patients with FSHD, one unaffected control, and a trio family to simultaneously characterize D4Z4 repeat size, haplotype, DNA methylation, and genomic structure. Results were validated by Southern blotting, single-molecule optical mapping and bisulfite sequencing. ONT-LRS enabled integrated characterization of D4Z4 repeat size, haplotype, sin

Newly discovered genetic subtypes of muscular dystrophy tend to be rarer than older ones, as occurs for other genetically heterogeneous classes of inherited disorders. Epidemiologically, there is a higher likelihood of ascertaining more prevalent genetic subtypes, but it is unclear why pathogenic variants are more abundant in certain genetic subtypes than others. We hypothesized that distributions of pathogenic variant types and molecular consequences of those variants for each genetic subtype vary based on the year of discovery of the muscular dystrophy gene. We compiled a list of genes assoc

Engineered surface topographies have proven to be an efficient method to replicate in vitro the architecture of highly organized tissues by controlling the spatial arrangement of cells. In this work, we studied the ability to guide muscle cell development using femtosecond-laser-induced topography on standard culture plates. This technique allows the creation of highly porous microstructured channels that exhibit strong hydrophobicity while offering mechanical cues for cell guidance. We characterized the resulting engineered surfaces by assessing their capacity to direct cellular organization.

Mitochondrial dysfunction is an important component in the pathogenesis of Duchenne muscular dystrophy (DMD). Although the mitochondrial voltage-dependent anion channel (VDAC) inhibitor olesoxime (TRO19622) has demonstrated efficacy in a severe DMD model, its effects in the classic mdx mouse model with a mild phenotype remain unclear. In this study, we administered olesoxime to 4-week-old mdx mice (20 mg/kg every other day for 4 weeks) and evaluated key parameters of mitochondrial function and muscle pathology. We found that olesoxime exerted no beneficial effects on impaired oxidative phospho

BACKGROUND: Obesity is heterogeneous; conventional anthropometric measures combine fat distribution, ectopic fat deposition, and muscle volume, limiting refined causal inference for chronic airway diseases. We used four magnetic resonance imaging (MRI)-defined obesity axes to assess their causal effects on chronic obstructive pulmonary disease (COPD), asthma, and allergic rhinitis (AR), and to evaluate mediation by circulating inflammatory proteins.
METHODS: Genetic instruments were selected for four MRI-defined obesity axes. Two-sample Mendelian randomization (MR) used inverse-variance weigh

PURPOSE OF REVIEW: Pediatric osteoporosis is a heterogeneous group of primary and secondary disorders that creates challenges for orthopaedic surgeons. This review summarizes evidence on conditions commonly encountered in pediatric orthopedic practice, medical management strategies, and perioperative care for children with bone fragility.
RECENT FINDINGS: Diagnostic approaches have been evolving toward integrating fracture characteristics, skeletal risk factors, and bone mineral density (BMD). A single low-trauma vertebral compression fracture is now sufficient for diagnosis of osteoporosis i

Deep chest wall abscesses may be difficult to distinguish from primary pyomyositis and necrotizing soft-tissue infection because of overlapping clinical and radiographic findings. Although soft-tissue gas raises concern for necrotizing infection, it is not independently diagnostic and may reflect anaerobic bacterial activity, communication with the skin, or prior instrumentation. Infections of the pectoral region involving uncommon polymicrobial organisms are rarely reported. A 61-year-old man with prediabetes, hypertension, chronic kidney disease, and a prior cerebrovascular accident presente

UNLABELLED: Early postprandial glucagon concentrations are higher in type 1 diabetes (T1D) than in individuals with no diabetes (ND). To determine the cause, we infused stable [13C9, 15N1]glucagon before, during, and after a mixed meal in 16 ND and 16 T1D individuals to measure glucagon turnover. In a subcohort of 9 ND and 12 T1D individuals, we estimated [13C9, 15N1]glucagon kinetics during steady state. A linear, single-compartment model described [13C9, 15N1]glucagon kinetics and allowed precise estimation of the volume of distribution (VD) and clearance rate (CL). Model parameters were sim

Duchenne/Becker muscular dystrophy (DMD/BMD) is caused by variants in the X-linked DMD gene-most commonly deletions, duplications, or point mutations, and only rarely by complex chromosomal rearrangements (CCRs). In this study, structural variant sequencing (SVseq) was applied to a 14-year-old male patient with DMD. Although the patient exhibited typical symptoms, conventional tests-including karyotype analysis, gene panel, whole-exome sequencing (WES), and multiplex ligation-dependent probe amplification (MLPA)-were all negative. SVseq identified a novel complex rearrangement on the X chromos

Structural variants (SVs) comprise copy-number variants (e.g., deletions and duplications) and copy-neutral SV (e.g., perfect inversions). Short-read high-throughput DNA-sequencing is the established technology for small-variant calling in clinical genomics, whereas Oxford nanopore or PacBio long-read NGS technologies improve SV identification but remain challenging to implement in clinical settings. We developed and validated a novel experimental protocol and bioinformatics pipeline for SV genotyping from short-read whole-genome sequencing (WGS) of restriction-circles of DNA. The experimental

Background Fat embolism syndrome (FES) is a rare but potentially life-threatening complication classically associated with long-bone fractures and orthopedic procedures. Patients with Duchenne muscular dystrophy (DMD) are at increased risk because of severe osteoporosis, chronic corticosteroid therapy, loss of ambulation, and fatty bone marrow replacement. However, FES remains underrecognized in this population, particularly when triggered by minor trauma without radiographic evidence of fracture. Case Presentation We report the case of a 12-year-old boy with DMD receiving long-term corticoste
