The Atlantic salmon tongue mucosa functions as an active mucosal immune site capable of responding to a pathogen.

The tongue of teleost fish is a mucosal organ whose role in immunity remains largely understudied. Here, we performed transcriptomic analyses of the Atlantic salmon (Salmo salar) smolt tongue to investigate its baseline gene expression profile and its immune response to a bacterial challenge with Yersinia ruckeri. In the unstimulated state, transcriptomic profiling revealed that the tongue is a highly metabolically active organ with enrichment of genes involved in cytoskeletal organisation, transport, and metabolism. Gene ontology (GO) analysis identified terms associated with immune system pr

Impact of maternal compensation on developmental phenotypes in a zebrafish model of severe congenital muscular dystrophy.

Genetic compensation is a common phenomenon in zebrafish in response to genetic alterations. Differences between genetic and morpholino-mediated zebrafish models of human diseases have led to significant difficulties in phenotypic interpretation and translatability. One form of compensation is the maternal deposit of mRNAs and proteins to the oocyte that supports developmental processes before zygotic genome activation. In this study, we generated a zebrafish model of severe congenital muscular dystrophy (CMD) by targeting protein O-mannose N-Acetylglucosaminyltransferase 2 (pomgnt2), a matern

Tailoring strategies for dyslipidemia management and atherosclerotic cardiovascular disease prevention in muscular dystrophy patients.

OBJECTIVES: Patients with muscular dystrophies are at increased cardiometabolic risk due to reduced mobility, increased adiposity, glucose intolerance, and dyslipidemia. While statins remain the cornerstone of cardiovascular disease prevention in the general population, their use in muscular dystrophies is complicated by the presence of underlying muscle symptoms, which can be difficult to distinguish from statin-associated myopathy. This overlap contributes to heightened concerns about statin tolerance and may impair long-term adherence. The emergence of nonstatin lipid-lowering therapies pre

Exploring Mechanisms of Calcific Tendonitis Using a Novel Turkey Model.

Calcific tendinopathy is a common musculoskeletal disorder marked by calcium deposition within tendons, often accompanied by pain, impaired mobility, and in severe cases, ossification. However, the mechanisms underlying tendon mineralization remain poorly understood. In contrast to pathological human calcification, turkeys naturally develop tendon mineralization during growth without signs of inflammation or ossification. This study had two phases: an observational phase to assess onset and progression of calcification, and an interventional phase to evaluate the role of mechanical loading. In

Preimplantation genetic testing-M for pathogenic variant in CTC1 gene causing cerebroretinal microangiopathy.

PURPOSE: Cerebroretinal microangiopathy with calcifications and cysts (CRMCC) is an autosomal recessive genetic and multisystem disorder characterized by extensive intracranial calcifications, leukoencephalopathy, and retinal vascular abnormalities, which is primarily caused by mutations in the CTC1 gene. Preimplantation genetic testing (PGT) is a procedure that helps patients choose embryos free of chromosome abnormalities and monogenic diseases for uterine transfer, preventing serious genetic disorders from being passed on to future generations. Here we introduce a family with fetal malforma

Difficult to manage axial spondyloarthritis patients have high burden of pain-related comorbidities and adverse long-term outcome.

BACKGROUND: Difficult to manage (D2M) axial spondyloarthritis (axSpA) is an evolving clinical concept. We aimed to assess the prevalence, predictors and long-term outcomes of D2M axSpA. METHODS: Prospective single center cohort study of axSpA patients starting targeted agents (01/01/2007 until 28/02/2024). The ASAS criteria were applied to classify D2M. Baseline parameters were assessed as predictors of D2M development by multivariate logistic regression models. To identify comorbidity clusters and their contribution to D2M, a K-means cluster analysis on binary indicators of most prevalent ch

Glycation Product Synthesized in Anhydrous Conditions Mimics an Epitope in Epithelial and Mesenchymal Tissues.

Background: Advanced glycation end-products (AGEs) are formed and deposited in tissues, contributing to various disorders, including diabetes, other metabolic diseases, and aging. A new epitope, AGE10, was identified in human and animal tissues using a monoclonal antibody raised against synthetic melibiose-derived glycation end-products (MAGE), which were synthesized under anhydrous conditions with bovine serum albumin or myoglobin. The biology of the AGE10 epitope, particularly its role in diseases and in cancer tissues, is not well understood. Methods: The study was aimed at investigating th

The Adipokine Axis in Heart Failure: Linking Obesity, Sarcopenia and Cardiac Dysfunction in HFpEF.

Obesity, sarcopenia, and heart failure with preserved ejection fraction (HFpEF) constitute an interconnected clinical triad driven by multisystem mechanisms centered on the adipokine axis. Adipose tissue, now recognized as a dynamic endocrine organ, undergoes pathological remodeling in obesity, characterized by hypoxia, chronic low-grade inflammation, and dysregulated adipokine secretion. These changes impair endothelial function, promote myocardial fibrosis, and disrupt skeletal muscle metabolism, thereby linking cardiometabolic and musculoskeletal dysfunction. This review integrates current

Longitudinal cohort study of muscle function and metabolic biomarkers in disease-modifying treatment of spinal muscular atrophy.

Disease-modifying treatments (DMT) for spinal muscular atrophy (SMA) developed in the past decade have improved the prognosis of patients with this severe condition. However, treatment effects vary, highlighting the need for sensitive predictive biomarkers to identify individuals more likely to respond to DMT. A previous study detected changes in mitochondrial oxidative capacity in SMA arm muscles, using 31-phosphorus magnetic resonance spectroscopy (31P MRS). Here, we investigated whether 31P MRS can distinguish responders from non-responders to DMT. In this longitudinal observational cohort

Peptide OH-CATH30 Mitigates Cachexia-Induced Muscle Atrophy via Modulation of TLR4-Associated Inflammation.

BACKGROUND: Cachexia, characterized by severe weight loss and muscle atrophy, frequently occurs in chronic conditions such as sepsis, cancer and chemotherapy, with limited effective treatments. Despite similar clinical manifestations, the underlying mechanisms across different disease contexts remain unclear. Identifying common pathways could lead to novel therapies. This study examines the role of Toll-like receptor 4 (TLR4), which is upregulated in various cachexia models, and assesses the therapeutic potential of the TLR4-inhibiting peptide OH-CATH30 in mitigating muscle atrophy. METHODS:

Spatial mapping of CoQ10 repletion by BPM31510 in a genetic mouse model (Coq4F147C) of coenzyme Q deficiency.

Primary coenzyme Q10 (CoQ10) deficiency is a rare mitochondrial disorder caused by mutations in genes involved in CoQ biosynthesis (e.g., COQ4) that result in impaired mitochondrial respiration, oxidative stress, and dysfunction across multiple organ systems because of decreased mitochondrial levels of CoQ10. Although oral CoQ10 supplementation has been examined for standard of care, poor absorption and inadequate tissue and intracellular distribution have resulted in a lack of clinically significant efficacy. BPM31510 is a lipid nanoparticle containing oxidized CoQ10 designed to improve bioav
