Combined phage therapy and faecal microbiota transplantation to treat recurrent urinary tract infection: a case series.

Recurrent urinary tract infections are recalcitrant and difficult-to-treat bacterial infections that primarily affect women. Here we combine phage therapy with faecal microbiota transplantation to decolonize the urinary and intestinal reservoirs of patients with recurrent urinary tract infections. Three women received oral and intravesical phage therapy for 8 days, and two underwent subsequent faecal microbiota transplantation. Treatments were well tolerated, and although Escherichia coli was detected in follow-up samples, patients experienced none or fewer and less severe episodes of urinary

The impact of human milk oligosaccharides and galacto-oligosaccharides on the growth of early life pathogenic gut bacteria.

Antibiotics are widely used to prevent and treat early life gut infections, but their aberrant use has raised global concerns about antibiotic resistance. As breastfed infants have fewer intestinal infections than formula-fed infants, this stimulated scientific interest in the anti-pathogenic properties of breastmilk, specifically human milk oligosaccharides (HMOs). HMOs are non-digestible sugars that support infant gut microbiome development, immune maturation, and protection against pathogens. However, to determine how specific HMO structures differentially affect microbial growth and pathog

Regional Prevalence of Carbapenemase-Producing Enterobacterales and Extended-Spectrum Beta-Lactamase-Producing Enterobacterales in Okayama and Neighboring Prefectures, Japan.

Carbapenemase-producing Enterobacterales (CPE) and extended-spectrum β-lactamase (ESBL)-producing Enterobacterales (ESBL-E) are representative antimicrobial-resistant pathogens. We investigated the prevalence of CPE and ESBL-E in Okayama, Japan to clarify their current epidemiology and potential clinical burden. Active surveillance for CPE and ESBL-E was performed first, using stool samples submitted to the Okayama Medical Laboratory (Japan) in the years 2024-2025. Screening was performed using a selective agar (CHROMagar mSuper CARBA/ESBL). Phenotypic identification was conducted with the use

Microbiome features associated with persistent intestinal carriages of Escherichia coli ST131 in a Southeast Asian cohort study.

Escherichia coli sequence-type 131 (ST131) is the dominant global extraintestinal pathogen capable of asymptomatic intestinal carriage and sustained household transmission, challenging infection control. Despite its clinical significance, the ecological determinants of gut persistence remain poorly understood. We performed shotgun metagenomics on fecal samples to investigate gut microbiome features associated with ST131-positive samples, distinct host carrier statuses (persistent, intermittent and non-carriers) and household risks in a study of a Southeast Asian cohort. Here, we show that ST13

Clonal diversity and genetic characteristics of extraintestinal pathogenic Escherichia coli in northern Japan: identification of the potentially prevalent clones phylogroup B2-ST357 and ST1193.

OBJECTIVES: Extraintestinal pathogenic Escherichia coli (ExPEC) is a leading cause of bloodstream and urinary tract infections, often showing multiple drug resistance associated with production of extended-spectrum beta-lactamases (ESBLs). The present study aimed at revealing the clonal structure and diversity of ExPEC in northern Japan and its relation to antimicrobial resistance (AMR) profiles and determinants, and genetic features. METHODS: Phylogenetic group and sequence type (ST) based on MLST scheme were determined for clinical isolates of ExPEC. PCR and sequencing were used to identify

Therapeutic potential of a novel virulent bacteriophage XQ-1 against avian pathogenic Escherichia coli infection in broiler chickens.

Avian Pathogenic Escherichia coli (APEC) represents a significant subgroup within extraintestinal pathogenic Escherichia coli strains and constitutes a substantial threat to the global poultry industry. Although the negative impacts of APEC have been mitigated considerably through antibiotic use, this practice has concurrently facilitated the widespread emergence and dissemination of antibiotic-resistant APEC strains worldwide. Consequently, bacteriophage has emerged as a promising alternative to antibiotics. In the current study, a novel virulent bacteriophage, designated XQ-1, was isolated f

Prophage-encoding engineered bacteria enable prophylactic lytic phage therapy for enteric infection in mice.

Phage therapy is a promising antibacterial approach but it has limited efficacy against enteric bacterial infections, such as Salmonella enterica Typhimurium (STm). Here we hypothesized that establishing high concentrations of a pathogen-targeting phage in the gut before infection would prophylactically protect against future infection. To this end, we engineered non-pathogenic Escherichia coli to encode a prophage that produces obligately lytic phage progeny targeting STm while overcoming its host defence systems, including restriction-modification systems. We show that prophylactic gut colon

FUT1 genetic polymorphism influences susceptibility to F18 enterotoxigenic Escherichia coli infection in pigs.

F18 enterotoxigenic Escherichia coli (ETEC) causes post-weaning diarrhea in nursery pigs, with susceptibility determined by a polymorphism in the α(1,2)-fucosyltransferase (FUT1) gene at nucleotide M307, where FUT1-M307A/A pigs are deemed F18 resistant and FUT1-M307G/A and FUT1-M307G/G pigs are deemed F18 susceptible to bacterial attachment. This post hoc analysis evaluated the effect of FUT1 genotype on disease susceptibility following F18 ETEC challenge over 3 experiments. It was hypothesized that FUT1 susceptible pigs (G/G and G/A) would incur greater F18 attachment than FUT1 resistant pigs

Inhibitory Effect of Terminalia chebula Retz. on MDR Shigella flexneri Invasion of HeLa Cells.

INTRODUCTION: Shigella flexneri causes bacillary dysentery by invading the colon epithe-lium, leading to inflammation and tissue damage. The emergence of multidrug-resistant (MDR) Shi-gella strains and the lack of effective vaccines have highlighted the need for alternative treatments. Natural plant compounds have been explored as potential antimicrobial agents. This study evaluates the antibacterial activity of ethanolic (70%-96%) and methanolic (70%-99.9%) extracts of Termi-nalia chebula Retz. against MDR S. flexneri and their effect on bacterial invasion of HeLa cells in vitro. We assessed

Lecture Capture Transcripts: A Valuable Learning Tool for Students of all Abilities.

Escherichia coli is a Gram-negative bacterium which colonizes the intestinal tract of man and other animals. In addition to being a part of the normal bacterial flora of the human intestine, there are a number of enteropathogenic strains of E. coli which cause infections ranging in consequence from diarrhoea to colitis. Antigen 43 (Ag43) is the major phase-variable protein in the outer membrane of E. coli. One benefit for bacteria resulting from phase variation of surface antigens is usually ascribed to evasion of host defences. However, results of recent studies indicate that infection of E.

Trichomonad Disease in Wild Turkeys (Meleagris gallopavo): Pathology and Molecular Characterization of Histomonas, Tetratrichomonas, Tritrichomonas, and Simplicimonas spp.

The Wild Turkey (Meleagris gallopavo) is a geographically widespread, popular game bird and conservation icon in North America. Following successful reestablishment in parts of the US and Canada, regional declines have fueled population health concerns; therefore, understanding mortality causes and spatiotemporal patterns of disease is important to uncover potential ongoing and future health risks. Histomonosis, caused by the trichomonad Histomonas meleagridis, is a well-established and potentially fatal disease in Wild Turkeys; however, its prevalence and potential population health impacts r

Major F plasmid clusters are linked with ColV and pUTI89-like marker genes in bloodstream isolates of Escherichia coli.

BACKGROUND: F plasmids are abundant in E. coli, carrying a variety of genetic cargo involved in fitness, pathogenicity, and antimicrobial resistance. ColV and pUTI89-like plasmids have drawn attention for their potential roles in various forms of extra-intestinal pathogenicity. However, the rates of their carriage and the overall diversity of F plasmids in E. coli bloodstream infections (BSI E. coli) remain unknown. METHODS: We performed a t-SNE-based cluster analysis of predicted F plasmids from a collection of 4711 BSI E. coli draft genomes to describe their diversity and abundance. We also

Enterocloster clostridioformis protects against Salmonella pathogenesis and modulates epithelial and mucosal immune function.

BACKGROUND: Promoting resistance to enteric pathogen infection is a core function of the gut microbiota; however, many of the specific host-commensal interactions that mediate this protection remain uncharacterised. To address this knowledge gap, we monocolonised germ-free mice with mouse-derived commensal microbes to screen for microbiota-induced resistance to Salmonella Typhimurium infection. RESULTS: We identified Enterocloster clostridioformis as a protective species against S. Typhimurium infection. E. clostridioformis selectively upregulates resistin-like molecule β and cell cycle pathw

Identification and characterization of amphipathic antimicrobial peptides with broad spectrum activity against multi-drug resistant bacteria.

Antimicrobial peptides (AMPs) are potential alternatives to antibiotics given the reduced likelihood of resistance and their high selectivity towards bacteria. AMPs with activity against antibiotic-resistant bacteria have been reported. The aim of this study is to characterize the activity of novel BP100 analogues against multidrug-resistant bacteria. Eleven bacterial strains representing five pathogenic species were used to evaluate the antimicrobial activity of 26 peptides. An initial screen was performed at 50 µg/ml, and those peptides that inhibited ≈90 % of growth of all strains were sele

Whole-Genome Sequence Reveals Genetic Determinants of the Colonization of Shiga Toxin-Producing Escherichia coli O22:H8 Isolates in Cattle.

Shiga toxin-producing Escherichia coli (STEC) is a zoonotic foodborne pathogen responsible for different cases and outbreaks of severe enteric diseases in humans worldwide. Our group previously showed that Argentine O22:H8 isolates (155 and 182) interfere with the colonization of O157:H7 in an experimental infection bovine model. In this study, we applied a comparative genomic and genome-wide association study (GWAS) analysis to explore the differences between the genetic bases of O22:H8 and O157:H7 strains isolated from cattle. The whole-genome sequence of O22:H8 155 and 182 isolates was comp

In Silico Detection of Virulence Genes in Whole-Genome Sequences of Extra-Intestinal Pathogenic Escherichia coli (ExPEC) Documented in Countries of the Andean Community.

E. coli pathotypes, which cause extra-intestinal infections, pose significant public health challenges, emphasizing the need for virulence gene surveillance to understand their dynamics. Key virulence genes have been identified in E. coli from Andean community countries, predominantly linked to human and animal sources. However, detailed data on virulence profiles from environmental and food sources remain limited. This study utilized an in silico approach to analyze 2402 whole-genome sequences from EnteroBase, known for associations with antimicrobial resistance genes. Of the isolates, 30% we
