Zoonotic potential of uropathogenic Escherichia coli lineages from companion animals.

Escherichia coli is responsible for urinary tract infections (UTI) in humans and pets. This study aims to provide data on the virulome and resistome of E. coli strains isolated during bacteriuria in companion animals and to assess their zoonotic potential. 135 E. coli strains prospectively collected from urine samples of 44 cats and 91 dogs in three French veterinary teaching hospitals were analyzed via antibiotic susceptibility tests and whole genome sequencing. Phylogroup B2 was overrepresented and several sequence types (STs) associated with human extra-intestinal pathogenic E. coli (ExPEC)

Genetic and phenotypic analysis of the virulence plasmid of a non-Shigatoxigenic enteroaggregative Escherichia coli O104:H4 outbreak strain.

Enteroaggregative Escherichia coli O104:H4 is best known for causing a worldwide outbreak in 2011 due to the acquisition of a Shiga-like toxin alongside traditional enteroaggregative virulence traits; however, whilst the 2011 outbreak strain has been well studied, the virulence plasmid of O104:H4 has been subjected to far less experimental analysis. In this paper, we analyse the genetic and phenotypic contribution of the pAA virulence plasmid to a non-Shigatoxigenic O104:H4 strain (1070/13) that was nonetheless implicated in a substantial UK outbreak in 2013. We find that pAA1070 is 99.95% ide

Whole-genome analysis of five Escherichia coli strains isolated from focal duodenal necrosis in laying hens reveals genetic similarities to the E. coli O25:H4 ST131 strain.

Focal duodenal necrosis (FDN) is an intestinal disease causing significant economic losses in the table-egg industry due to reduced egg production in laying hens. Its etiology and pathogenesis remain poorly understood. Between 2021 and 2023, 111 Escherichia coli isolates were collected from FDN lesions and screened for the presence of virulence genes using PCR panels. Five strains-FDN-4, FDN-9, FDN-11, FDN-24, and FDN-50-were selected for whole-genome sequencing due to their high virulence gene content. Core-genome analyses found that the five FDN E. coli belong to different phylogroups and st

Evolution of Escherichia coli strains under competent or compromised adaptive immunity.

Escherichia coli is a commensal of the intestine of most mammals, but also an important human pathogen. Within a healthy human its population structure is highly dynamic, where typically a dominant E. coli strain is accompanied by several low abundance satellite strains. However, the factors underlying E. coli strain dynamics and evolution within hosts are still poorly understood. Here, we colonised germ-free immune-competent (wild-type) or immune-compromised (Rag2KO) mice, with two phylogenetically distinct strains of E. coli, to determine if strain co-existence and within-strain evolution ar

Association of Escherichia coli pathotypes with fecal markers of enteropathy and nutritional status among underweight adults in Bangladesh.

INTRODUCTION: Environmental enteric dysfunction (EED), a subclinical intestinal disorder, is characterized by chronic fecal-oral exposure to entero-pathogens and could be diagnosed by measuring non-invasive biomarkers. Escherichia coli is the one of the key bacterial enteric pathogens that drives EED, but there is a lack of information on the E. coli pathotypes in relation to the biomarkers of EED in malnourished adults. Here, we intended to measure the possible association of these pathotypes with EED biomarkers and nutritional status of adults residing in a slum in Bangladesh. METHOD: Fecal

Enterotoxigenic Escherichia coli as a Modulator of the Entero-Pulmonary Axis in Piglets: Impacts on the Microbiota and Immune Responses.

The high prevalence of enterotoxigenic Escherichia coli (ETEC) in nondiarrheic piglets contributes to its rapid spread; however, few studies have explored the effects of latent gastrointestinal pathogens on animal health. Therefore, using high-throughput sequencing approaches, we explored changes in entero-pulmonary microbiota and immune gene expression in healthy, asymptomatic, and diarrheic piglets. As expected, bacterial communities were less diverse in the respiratory tract than in the gut, with a site-specific composition that was more stable in the gut and highly variable in the lung amo

Residence-colonization trade-off and niche differentiation enable coexistence of Escherichia coli phylogroups in healthy humans.

Despite abundant literature on pathogenicity and virulence of the opportunistic pathogen Escherichia coli, much less is known about its ecological and evolutionary dynamics as a commensal. Based on two detailed longitudinal datasets on the gut microbiota of healthy adults followed for months to years in France and the USA, we identified a robust trade-off between the ability to establish in a new host (colonization) and to remain in the host (residence). Major E. coli lineages (phylogroups or subgroups) exhibited similar fitness but diverse strategies, from strong colonisers residing few days

Aptamer-functionalized graphene quantum dots combined with artificial intelligence detect bacteria for urinary tract infections.

OBJECTIVES: Urinary tract infection is one of the most prevalent forms of bacterial infection, and prompt and efficient identification of pathogenic bacteria plays a pivotal role in the management of urinary tract infections. In this study, we propose a novel approach utilizing aptamer-functionalized graphene quantum dots integrated with an artificial intelligence detection system (AG-AI detection system) for rapid and highly sensitive detection of Escherichia coli (E. coli). METHODS: Firstly, graphene quantum dots were modified with the aptamer that can specifically recognize and bind to E.

Emphysematous Cystitis: A Case Report and Literature Review.

Urinary tract infections (UTIs) are one of the most common infectious diseases worldwide, predominantly affecting women due to anatomical factors. Among UTIs, emphysematous cystitis (EC) is a rare but clinically significant entity characterized by gas presence in the bladder wall or lumen, often caused by gram-negative bacteria like Escherichia coli and Klebsiella pneumoniae. Diagnosis is challenging due to nonspecific symptoms such as dysuria, abdominal pain, and occasionally pneumaturia, with computed tomography being the diagnostic method of choice. We report the case of a 65-year-old male

A Comprehensive Review of the Role of Virulence Factors in Enteropathogenic Escherichia coli-Induced Intestinal Injury.

Escherichia coli (E. coli) is a rod-shaped gram-negative bacterium that includes the diarrheagenic strains, an identical group of intestinal pathogens.E. coli diarrhea is transmitted through the feco-oral route, through contaminated food and water. Enteropathogenic E. coli (EPEC) is one of the leading causes of diarrhea in the pediatric age group in developing and developed countries. Depending on the absence or presence of E. coli adherence factor plasmids, they are classified as typical or atypical isolates. The distinguishing feature of EPEC's pathology is the attaching and effacing lesions

Large-scale genomic analysis reveals the distribution and diversity of type VI secretion systems in Escherichia coli.

The type VI secretion system (T6SS) is a versatile nanomachine that injects effectors into target cells, playing a role in bacterial competition and virulence. While widespread in Gram-negative bacteria, T6SS prevalence varies across species and strains, and its distribution in Escherichia coli remains underexplored despite it being an important intestinal (IPEC) and extraintestinal (ExPEC) pathogen. Here, we examined the prevalence of T6SS subclasses (T6SSi) across 131,610 E. coli genomes, which we annotated for clinical relevance. T6SS genes were identified while focusing on the three subcla

Oral and parenteral treatment with a third-generation cephalosporin promotes the proliferation of diverse ESBL-producing Escherichia coli in the chicken intestinal tract.

The global rise of antimicrobial resistance is a major public health threat, with Escherichia coli facilitating the spread of extended-spectrum beta-lactamase (ESBL) genes like blaCTX-M, which confer resistance to third-generation cephalosporins (3GCs). This study examines the impact of 3GC treatment on resistant E. coli clones and horizontal gene transfer (HGT) of ESBL genes in broiler chickens in Quito, Ecuador. Fifteen-day-old Ross broilers were divided into three groups: oral ceftriaxone (100 mg/kg), parenteral ceftriaxone (100 mg/kg intramuscular), and control (no treatment). The study in

Emergence of porcine-derived hybrid Escherichia coli with dual-disease manifestations in intestinal and extraintestinal niches.

Escherichia coli, a significant zoonotic pathogen, continuously evolves to produce hybrid strains with diverse pathogenic characteristics, threatening public health. These hybrid strains are capable of causing multiple diseases across various tissues within a single host or among genetically diverse hosts. They have been reported to exhibit heightened virulence, enhanced infectiousness, and increased resistance to environmental clearance. Here, we identified a hybrid aEPEC/ExPEC isolate, PE164, from the bloody stool of a piglet with severe diarrhoea, which exhibited both intestinal and extrain

The role of gastrointestinal PCR in inflammatory bowel disease flares: A double-edged sword or a diagnostic breakthrough?

Inflammatory bowel disease (IBD), encompassing Crohn's disease (CD) and ulcerative colitis (UC), is characterized by chronic gastrointestinal inflammation, leading to unpredictable flares and substantial morbidity. Gastrointestinal polymerase chain reaction (GI-PCR) testing has emerged as a widely used diagnostic tool to identify enteric pathogens during IBD exacerbations. It is a novel, quick and sensitive diagnostic test to detect Clostridioides difficile infection (CDI), as well as other enteric pathogens. However, the clinical significance of non-C. difficile pathogens detected by GI-PCR r

Characterization of the ligand binding pocket of the virulence regulator Rns, a member of the AraC/XylS family of transcription factors.

UNLABELLED: Diarrheal disease caused by Gram-negative enteric pathogens, such as enterotoxigenic Escherichia coli (ETEC), Vibrio cholerae, Shigella spp., and Salmonella spp., is a leading cause of morbidity and mortality of children, especially in low resource nations. While progress has been made in reducing this burden, there remains a need to develop effective therapies. Recently, we determined the structure of Rns, a member of the AraC/XylS family that regulates the expression of pili and other virulence factors in ETEC. The structure revealed decanoic acid bound between the N- and C-termi

Therapeutic potential of thymol, carvacrol, and biogenic silver nanoparticle against avian pathogenic Escherichia coli (APEC): alternatives to the use of antimicrobials.

Antimicrobial resistance represents a significant threat to global public health, and poultry production is a critical sector within this context. Since the use of antimicrobials in poultry farming can promote the selection of resistant bacterial strains, the search for effective alternatives is essential. Thymol and carvacrol, two aromatic monoterpenes, as well as biogenic silver nanoparticles (bio-AgNP) have antimicrobial potential. Therefore, the aim of this study was to evaluate the action of thymol, carvacrol, and bio-AgNP against multidrug-resistant (MDR) Escherichia coli strains with vi

ETEC biofilms are regulated by magnesium and lactate bioavailability.

The prevailing dogma is that enterotoxigenic Escherichia coli (ETEC) use plasmid-borne colonization factors (CFs) to adhere to the intestinal epithelium, where the organisms proliferate and produce their diarrhea-causing virulence factors, the heat-stable (ST) and/or heat-labile (LT) enterotoxins. However, vaccines that target major CF antigens fail to induce complete protective immunity, indicating that ETEC may also use other antigens to colonize the small intestines. We previously demonstrated that ST intoxication limits magnesium bioavailability in the intestinal lumen, but the role of mag

Colorectal cancer-associated Streptococcus gallolyticus: a hidden diversity expose.

Streptococcus gallolyticus subsp. gallolyticus (SGG) is a bacterial pathogen implicated in bacteremia and endocarditis and is often associated with colon tumors in elderly individuals. The development of colorectal cancer (CRC) has been linked to intestinal dysbiosis, characterized by increased proportions of SGG and other intestinal microbes. In this study, we present the complete nucleotide sequence of five novel clinical isolates of SGG associated with colorectal cancer, revealing unexpected genetic diversity. Sequencing an additional 30 SGG clinical isolates provided a more comprehensive d
